Example 2:
referring to the drawings, this example prepares an alcoholic animal model of rat gastric ulcer and performs utility evaluation using the mixed powder of radix tetrastigme and charcoal-fried obtained in example 1; the specific experimental procedures are referenced as follows:
1. preparation of rat gastric ulcer model. Taking 60 rats with half each sex, randomly dividing into 3 groups, wherein each group comprises 20 rats, namely a control group, an experimental group and a blank group, diet prohibition is performed for 24 hours, the experimental group and the control group are filled with stomach alcohol according to 1.5ml/100g of body weight to manufacture an acute gastric ulcer model, the blank group is filled with physiological saline, and the experiment is performed after 12 hours;
2. grouping into a control group, an experimental group and a blank group; wherein the control group and blank group are fed with normal feed and drinking water; an experimental group uses the radix tetrastigme fried carbon powder obtained in the example 1 to prepare an additive feed for feeding, normal drinking water is carried out, the general anatomical condition of the stomach is observed on 0, 1, 2, 3, 4 and 5 days, an HE staining section is prepared, and the gastric ulcer repairing condition is observed.
3, analysis of experimental results:
(1) and (5) carrying out feed consumption statistics. Recording the weight of the rat, weighing the same feed amount according to the weight proportion, and counting the feed consumption condition, wherein the result shows that the feed consumption of the blank group is obviously higher than that of the experimental group and the control group; the feed consumption of the experimental group is obviously higher than that of the control group (all rats are randomly raised in cages, 2 rats are raised per cage); about 5 days, the feed consumption of the experimental group is not different from that of the blank group, and is obviously different from that of the control group; the stomach of the molded rat is generally shown in figure 1; in fig. 1, numbers 1-3 are experimental groups, 7-9 are control groups, 4-6 are blank groups, 3 patients in each group are observed for molding, and it can be seen that the control group and the experimental group have more food residues, the stomach fundus is inflated and swollen, and a blood sample is presented;
(2) dissection and pathology. Anatomical display shows that after the model is built for 12 hours, the ulcer surfaces of rats in the control group and the experimental group are in a severe ulcer state, and mucous membranes fall off, and particularly, refer to fig. 2; pathological HE section shows that mucosa of the ulcer surface is shed, inflammatory cells are infiltrated, a slight edema zone appears on the stomach basement and the muscle layer as shown in figures 5 and 6, and the blank group presents normal stomach section pathology as shown in figure 4; after 3 days, the experimental group showed the basic characteristics of ulcer in healing phase, the cases showed a decrease in myocyte edema, the cases showed ulcer in repairing phase locally, and the feces were blackish, as shown in fig. 8; after 4 days, the mucous membrane is basically repaired and the normal gastric mucous membrane state is presented.
The results of the independent evaluation test of the radix tetrastigme fried carbon mixed powder show that the radix tetrastigme fried carbon mixed powder has a good repairing effect on the gastric ulcer of a rat molded by alcohol.
