Abstract
本发明公开了一种Bortezomib的药物新用途。所述新用途是其在下述方面的应用:1)在制备细胞辐射保护剂中的应用;2)在制备预防和/或抗辐射损伤的药物中的应用;3)在制备促进辐射后细胞增殖的产品中的应用。本发明的药效学实验表明,Bortezomib提高细胞照射后的存活率及增殖能力;提高了辐射后人或哺乳动物的存活率;提高了辐射后人或哺乳动物体内白细胞数量;提高了辐射后人或哺乳动物的脾脏指数和胸腺指数。为制备预防和/或抗辐射损伤的药物提供了新的思路。
Description
技术领域
本发明属于医药技术领域,具体涉及一种蛋白酶体(proteasome)选择性抑制剂硼替佐米(Bortezomib)在抗辐射损伤中的用途。
背景技术
电离辐射已经出现在人类生产生活的各行各业,在工业、农业、国防、科技以及医药卫生等领域都得到了广泛应用,人类在享受其带来巨大利益的同时,自身暴露于辐射环境的危险性也日益增加。电离辐射是一种不需要任何介质参与即可传递的能量,能对生物体造成相当大的损害,可分为直接损伤和间接损伤。直接损伤是指放射线直接作用于DNA、蛋白质等生物大分子改变其分子结构及生物学特性;间接损伤是指放射线作用于机体内水分子,使其发生电离,产生大量自由基,从而引发多种相关损伤性疾病。目前研究开发的抗辐射损伤药物主要包括氨巯基类、细胞因子类、激素类、氮氧自由基和天然药物等几类,但这些药物的综合疗效还不理想,一些药物的治疗效果未得到认可,一些药物使用后产生了严重的副作用,限制了抗辐射损伤药物在临床上的应用。因此,开发高效、低毒或无毒的抗辐射损伤药物具有极大的应用前景和社会效益。
泛素-蛋白酶体系统是细胞内蛋白质降解的主要途径,参与细胞内80%以上蛋白质的降解,其主要负责降解错误折叠和突变的蛋白质以及许多参与调控发育、分化、细胞增殖、信号转导和凋亡的蛋白质。蛋白质先被泛素标记,然后被蛋白酶体识别和降解,通过这样一个多种蛋白质参与的多步骤反应过程,细胞对不需要的蛋白进行降解,对整个蛋白平衡极为重要。
Bortezomib是第一个应用于临床治疗的蛋白酶体抑制剂,它是一种人工合成的硼酸二肽化合物,属于可逆性蛋白酶体抑制剂,可以选择性地与蛋白酶体活性位点的苏氨酸结合,抑制蛋白酶体20S亚单位的糜蛋白/胰蛋白活性。Bortezomib作为世界上首个以蛋白酶为靶标的抗肿瘤药物,通过多种不同机制发挥抗肿瘤作用,主要用于多发性骨髓瘤和套细胞淋巴瘤的治疗。此外,Bortezomib作为单一药物或者联合治疗的一部分在其它血液系统恶行肿瘤和实体瘤中进行研究,但未能得到理想的疗效,应用受限。
目前,关于蛋白酶体抑制剂Bortezomib在抗辐射损伤中的应用尚无报道。
发明内容
本发明的目的是提供了一种蛋白酶体抑制剂Bortezomib(硼替佐米)的应用。
本发明所提供的蛋白酶体抑制剂Bortezomib的用途是其在下述方面的应用:
1)在制备细胞辐射保护剂中的应用;
2)在制备预防和/或抗辐射损伤的药物中的应用;
3)在制备促进辐射后细胞增殖的产品中的应用。
本发明中,所述细胞可为真核生物的正常细胞。
所述真核生物可为人或哺乳动物;所述正常细胞为来源于正常组织的细胞;所述细胞包括肝细胞、肺细胞、脐静脉内皮细胞、小肠上皮细胞、胃上皮细胞。
所述肝细胞可为L-O2;所述肺细胞可为MRC-5;所述脐静脉内皮细胞可为HUVEC;所述小肠上皮细胞可为HIEC;所述胃上皮细胞可为GES-1。
所述细胞辐射保护剂具体体现在提高了细胞辐射后的存活率。
所述预防和/或抗辐射损伤具体体现在下述至少一方面:
1)提高了辐射后人或哺乳动物的存活率;
2)提高了辐射后人或哺乳动物体内白细胞数量;
3)提高了辐射后人或哺乳动物的脾脏指数;
4)提高了辐射后人或哺乳动物的胸腺指数;
5)保护辐射后人或哺乳动物的脏器(如脾脏、胸腺等)。
所述辐射为电离辐射,包括α射线辐射、β射线辐射、γ射线辐射、X射线辐射、中子射线辐射,具体如60Coγ辐射。所述60Coγ辐射的辐射剂量可为6Gy-12Gy。
所述辐射损伤可为辐射引起的脏器损伤,如脾脏损伤、胸腺损伤、性腺损伤、胰腺损伤和肺损伤等。
本发明还保护一种体外促进辐射后的细胞增殖的方法。
该方法包括用Bortezomib处理辐射后的细胞。
所述Bortezomib在处理体系中的浓度可为2.5nM-25nM。
所述细胞包括肝细胞、肺细胞、脐静脉内皮细胞、小肠上皮细胞、胃上皮细胞。
所述肝细胞可为L-O2;所述肺细胞可为MRC-5;所述脐静脉内皮细胞可为HUVEC;所述小肠上皮细胞可为HIEC;所述胃上皮细胞可为GES-1。
以Bortezomib为活性成分制备的真核生物正常细胞辐射保护剂或预防和/或抗辐射损伤的药物也属于本发明的保护范围。
所述真核生物正常细胞辐射保护剂或预防和/或抗辐射损伤的药物可通过注射、喷射、滴鼻、滴眼、渗透、吸收、物理或化学介导的方法导入机体如肌肉、皮内、皮下、静脉、粘膜组织;或是被其他物质混合或包裹后导入机体。
需要的时候,在上述药物中还可以加入一种或多种药学上可接受的载体。所述载体包括药学领域常规的稀释剂、赋形剂、填充剂、粘合剂、湿润剂、崩解剂、吸收促进剂、表面活性剂、吸附载体、润滑剂等。
以Bortezomib为活性成分制备的真核生物正常细胞辐射保护剂或预防和/或抗辐射损伤药物可以制成注射液、片剂、粉剂、颗粒剂、胶囊、口服液、膏剂、霜剂等多种形式。上述各种剂型的药物均可以按照药学领域的常规方法制备。

下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
下述实施例中所涉及的定量实验,均设置至少3个重复,结果取均值。
下述实施例中所使用的永生化人脐静脉内皮细胞HUVEC来自中国医学科学院基础医学研究所细胞资源中心。
实施例1、辐射后体外细胞活力情况检测
材料与方法
1.HUVEC细胞培养
HUVEC细胞在含10%胎牛血清的DMEM培养基中,于37℃,5%CO2培养箱中培养。细胞处于对数生长期时胰酶消化,用细胞计数仪计算细胞密度,按每孔5000个细胞接种于96孔板中,继续在培养箱中孵育24小时。待细胞贴壁并稳定生长后进行给药实验,共同孵育24小时,待细胞密度长至70~80%时进行60Coγ射线照射处理。
2.Bortezomib对60Coγ射线导致的HUVEC细胞活力丧失影响的检测
HUVEC细胞在含10%胎牛血清的DMEM培养基中,于37℃,5%CO2培养箱中培养。细胞处于对数生长期时胰酶消化,用细胞计数仪计算细胞密度,按每孔5000个细胞接种于96孔板中,继续在培养箱中孵育24小时。待细胞贴壁并稳定生长后,将Bortezomib以终浓度5nM给药,共同孵育24小时后60Coγ射线8Gy照射,剂量率1Gy/min,同时设置未处理对照组、单纯给药组、单纯照射组和无细胞空白组,继续于37℃,5%CO2培养箱中培养。72小时后进行检测,每孔加入CCK-8溶液,37℃避光孵育1小时,用酶标仪在450nm处测定吸光值D。
3.Bortezomib作用结果的处理与统计
利用不同组别吸光度值显示细胞的活力,反映细胞照射后活力状态的改变,以及Bortezomib对细胞照射后活力丧失的影响。细胞存活率为实验组的吸光度值与无药物未照射组(即未处理对照组)的吸光度值的比值,用百分比表示:
存活率=(D450实验组-D450空白组)/(D450对照组-D450空白组)*100%
结果用GraphPad Prism 7.04软件进行统计分析,与单纯照射组相比,*P<0.05。
结果
实验独立重复三次,CCK-8检测结果见图1。Bortezomib可以抑制60Coγ射线导致的HUVEC细胞活力丧失,提高细胞照射后的存活率。
实施例2、辐射后体外细胞增殖情况检测
材料与方法
1.HUVEC细胞培养
同实施例1。
2.Bortezomib对HUVEC细胞经60Coγ射线照射后增殖情况的检测
利用HUVEC细胞在照射和给药条件下进行克隆形成实验,细胞在含10%胎牛血清的DMEM培养基中,于37℃,5%CO2培养箱中培养。细胞处于对数生长期时胰酶消化,用细胞计数仪计算细胞密度,按每孔1500个细胞接种于6孔板中,继续在培养箱中孵育24小时。待细胞贴壁并稳定生长后,将Bortezomib以终浓度5nM给药,共同孵育24小时后60Coγ射线8Gy照射,剂量率1Gy/min,同时设置未处理对照组、单纯给药组和单纯照射组,继续于37℃,5%CO2培养箱中培养。每隔3天进行换液,弃去旧的培养基,缓慢加入新鲜的培养基,并观察细胞状态,持续培养14天。克隆完成后,PBS洗涤1次,每孔加入1mL 4%多聚甲醛室温固定30分钟,PBS洗涤1次。每孔加入结晶紫染液1mL,染色15分钟,PBS洗涤3次。晾干,拍照,利用ImageJ 1.48v软件统计克隆形成数量N。
3.Bortezomib作用结果的处理与统计
利用不同组别的克隆形成数量显示细胞的增殖情况,反映细胞照射后增殖能力的改变,以及Bortezomib对细胞照射后增殖能力的影响。克隆存活率为实验组的克隆形成数量与无药物未照射组(即未处理对照组)的克隆形成数量的比值,用百分比表示:
存活率=N实验组/N对照组*100%
结果用GraphPad Prism 7.04软件进行统计分析,与单纯照射组相比,*P<0.05。
结果
实验独立重复三次,细胞克隆形成的结果见图2。Bortezomib可以抑制60Coγ射线导致的HUVEC细胞增殖能力减弱,提高细胞照射后的增殖能力。
实施例3、小鼠辐射后30天存活率实验
材料与方法
1.C57BL/6J小鼠饲养
选用40只18~22g雄性C57BL/6J小鼠,购自北京维通利华实验动物技术有限公司。小鼠饲养环境温度为22℃±2℃,相对湿度为50%~60%。
2.Bortezomib对60Coγ射线8Gy一次全身照射C57BL/6J小鼠30天存活率的影响
药物配制:精密称取Bortezomib粉末0.05mg,依次缓慢加入50μL DMSO、400μLPEG、50μL Tween 80和500μL水,涡旋混匀至溶液澄清,配制成0.05mg/mL的溶液。
动物分组及处理:40只C57BL/6J小鼠随机分为4组,每组10只,设置未处理对照组、单纯给药组、单纯照射组和给药照射组。未给药组小鼠腹腔注射等体积的上述溶剂(5%DMSO+40%PEG+5%Tween 80+50%水),Bortezomib给药组小鼠分别于照射前24小时和1小时按0.5mg/kg的剂量各进行腹腔注射1次。
3.Bortezomib抗辐射作用结果的处理与统计
每天观察各组小鼠的活动状态,记录30天内小鼠的存活情况,结果用GraphPadPrism 7.04软件进行统计分析,反映Bortezomib预防给药对60Coγ射线8Gy一次全身照射C57BL/6J小鼠30天存活率的影响。与单纯照射组相比,*P<0.05。
结果
Bortezomib以50mg/kg剂量预防用药处理的经60Coγ射线8Gy照射的C57BL/6J小鼠30天存活率的结果见图3。未处理对照组和单纯给药组的小鼠均未死亡,存活率为100%,故图中的曲线发生重合。Bortezomib预防给药能够提高60Coγ射线8Gy照射的C57BL/6J小鼠的存活率。
实施例4、小鼠辐射后指标分析
材料与方法
1.C57BL/6J小鼠饲养
同实施例3。
2.Bortezomib对60Coγ射线8Gy一次全身照射C57BL/6J小鼠各指标分析
动物分组及处理:20只C57BL/6J小鼠随机分为4组,每组5只,设置未处理对照组、单纯给药组、单纯照射组和给药照射组。未给药组小鼠腹腔注射等体积的上述溶剂(5%DMSO+40%PEG+5%Tween 80+50%水),Bortezomib给药组小鼠分别于照射前24小时和1小时按0.5mg/kg的剂量各进行腹腔注射1次。
白细胞数量分析:照射后第7天小鼠眼球取血200μL,加入到100μL的乙二胺四乙酸二钠盐(EDTA-2Na)的抗凝溶液中,用全自动血细胞分析仪检测白细胞数量。
脾脏和胸腺指数分析:照射后第7天小鼠颈椎脱臼处死,解剖取下小鼠的脾脏和胸腺,用PBS冲洗干净血迹,分析天平称量重量,记录结果。
3.Bortezomib抗辐射作用结果的处理与统计
记录不同组别小鼠的白细胞数量,分析白细胞数量变化的异常,反映Bortezomib在小鼠抗辐射中的作用。
记录不同组别小鼠的脾脏和胸腺质量,以及解剖前小鼠的重量,分析各组别小鼠的脾脏指数和胸腺指数,反映Bortezomib在小鼠抗辐射中的作用。
脏器指数=脏器质量(mg)/体重(g)。
结果用GraphPad Prism 7.04软件进行统计分析,与单纯照射组相比,*P<0.05。
结果
Bortezomib预防给药处理的照射后小鼠白细胞数量的结果见图4。与单纯照射组相比,Bortezomib可以提高照射后小鼠的白细胞数量,在一定程度上具有抗辐射损伤作用。
Bortezomib预防给药处理的照射后小鼠的脾脏指数和胸腺指数的结果见图5和图6。与单纯照射组相比,Bortezomib可以提高照射后小鼠的脾脏指数和胸腺指数,在一定程度上对照射小鼠的脏器具有保护作用。
Claims (10)
1.Bortezomib在制备预防和/或抗辐射损伤的药物中的应用。
2.根据权利要求1所述的应用,其特征在于:所述预防和/或抗辐射损伤体现在下述至少一方面:
1)提高辐射后人或哺乳动物的存活率;
2)提高辐射后人或哺乳动物体内白细胞数量;
3)提高辐射后人或哺乳动物的脾脏指数;
4)提高辐射后人或哺乳动物的胸腺指数;
5)保护辐射后人或哺乳动物的脾脏;
6)保护辐射后人或哺乳动物的胸腺。
3.Bortezomib在制备细胞辐射保护剂中的应用。
4.根据权利要求3所述的应用,其特征在于:所述细胞辐射保护剂提高了细胞辐射后的存活率。
5.Bortezomib在制备促进辐射后细胞增殖的产品中的应用。
6.根据权利要求3-5中任一项所述的应用,其特征在于:所述细胞为真核生物的正常细胞;所述真核生物为人或哺乳动物;
所述正常细胞为来源于正常组织的细胞;所述细胞包括肝细胞、肺细胞、脐静脉内皮细胞、小肠上皮细胞、胃上皮细胞。
7.根据权利要求6所述的应用,其特征在于:所述肝细胞为L-O2细胞;所述肺细胞为MRC-5细胞;所述脐静脉内皮细胞为HUVEC细胞;所述小肠上皮细胞为HIEC细胞;所述胃上皮细胞为GES-1细胞。
8.根据权利要求1-7中任一项所述的应用,其特征在于:
所述辐射为电离辐射,包括α射线辐射、β射线辐射、γ射线辐射、X射线辐射、中子射线辐射,
具体的,所述辐射为60Coγ辐射;所述60Coγ辐射的辐射剂量为6Gy-12Gy;
所述辐射损伤为辐射引起的脏器损伤。
9.一种体外促进辐射后的细胞增殖的方法,包括用Bortezomib处理辐射后的细胞。
10.根据权利要求9所述的方法,其特征在于:所述Bortezomib在处理体系中的浓度为2.5nM-25nM。
