Test example 1 test of the weight-reducing effect of the composition of the present invention
1. Test materials
The feed is selected from commercial feed, and the high-fat feed comprises 70% of basal feed, 4% of cholesterol, 0.5% of sodium cholate, 18% of lard and 7.5% of egg yolk.

2. Test method
2.1 test grouping
The 10 groups of rats are respectively an experiment 1-8 group, a model group and a blank group, wherein the experiment 1-8 group and the model group are fed with high-fat feed, and the blank group is fed with basic feed.
2.2 methods of administration
Experiments 1-8 groups 200mg/kg of the compositions prepared in example 1, example 5, example 9, example 10, comparative examples 1-4, respectively; and (3) adding 100mL of pure water into each composition, uniformly mixing to prepare a suspension, and performing intragastric administration on the suspension of the animals in each experimental group in an oral mode, wherein the pure water with the same volume is administered to the model group and the blank group once a day for 8 weeks continuously.
3. Test data detection and processing
Body weights were recorded weekly for each group of rats, and the rats were sacrificed at the end of 8 weeks of administration, dissected, isolated for perirenal fat, perigenital fat, and omental fat, and weighed to calculate the lipid/body ratio. Fat/body ratio (perirenal fat weight + perigenital fat weight + omental fat weight)/body weight × 100%.
Statistical processing is carried out on experimental data by using anova, and single-factor anova is carried out by using SPSS 25.0 software.
4. Test results
The results of body weight and lipid/body ratio of each group of rats 6 weeks after administration are shown in table 1.
Note: # P < 0.01 compared to blank; comparison with model groups, P < 0.05 and P < 0.01, respectively, are given below.
As can be seen from Table 1, after 8 weeks of experiments, the body weight and the lipid/body ratio of the rats in the model group are greater than those in the blank group, and have very significant difference (P < 0.01), indicating that the obesity model is established. After 8 weeks of experiments, the body weight and the fat/body ratio of the rats in the experimental group 1-2 are obviously lower than those in the model group, the difference is very significant (P is less than 0.01), and the body weight and the fat/body ratio of the rats in the experimental group 2 are lower than those in the experimental group 1, which shows that the purple kudzuvine root added in the composition can further improve the weight-losing effect, because the purple kudzuvine root added is beneficial to the body to discharge metabolites out of the body.
The body weight and the body fat/body ratio of the rats in the experimental group 3, the experimental group 5, the experimental group 6 and the experimental group 7 are obviously lower than those in the model group, the difference is very significant (P is less than 0.01), and the body weight and the body fat/body ratio of the rats in the experimental group 4 and the experimental group 8 are lower than those in the model group, and the difference is significant (P is less than 0.05).
The weight and the fat/body ratio of the rats in the experimental group 1 are obviously higher than those in the experimental groups 5-8, which shows that the agaricus bisporus, the aronia serrulata, the immature bitter orange and the polygonatum odoratum in the composition have better synergistic effect, and the prepared composition lacks one of the agaricus bisporus, the aronia serrulata, the immature bitter orange and the polygonatum odoratum, so that the optimal effect cannot be exerted when the weight and the fat/body ratio of the rats are reduced.
The body weight and lipid/body ratio of the rats of experimental group 3 were higher than those of experimental group 1, indicating that the cassia seed prepared by the method of example 1 used in the composition has an effect of reducing the body weight and lipid/body ratio of the rats better than that of the preparation method of example 9.
The body weight and lipid/body ratio of the rats of experimental group 4 were high compared to that of experimental group 1, indicating that the effect of polygonatum odoratum prepared by the method of example 1 in reducing the body weight and lipid/body ratio of the rats in the composition is due to the preparation method of example 9.
It was tested that the compositions prepared in examples 2-4 had similar effects to example 1 and that the compositions prepared in examples 6-8 had similar effects to example 4.
Test example 2 acute toxicity test of the composition of the present invention
Test methods reference is made to the paper published by the small eastern Sheeter et al: acute toxicity and subchronic toxicity of Astroside powder, acute toxicity test of Astroside powder on mice, recorded in 48 vol 8, page 3097 and 3108 of veterinary drug of animal husbandry, China 2021.
1. Test grouping
30 healthy Kunming mice, each half of male and female, are randomly divided into 3 groups, each group comprises 10 mice, and each half of male and female is respectively an experimental group 9, an experimental group 10 and a control group.
2. Method of administration
The suspension prepared by administering the composition of example 1 daily in the experimental group 9 was subjected to single gavage at the maximum concentration and the maximum volume, the suspension prepared by administering the composition of example 5 daily in the experimental group 10 was subjected to single gavage at the maximum concentration and the maximum volume, the amount of the composition of example 1 and example 5 administered daily was 10g/kg, and the control group was subjected to single gavage with the same volume of physiological saline. Mice were fasted for 12 hours prior to dosing, followed by 14 consecutive days. The behavior, mental state, respiration, feed intake, water intake and death of the mice were observed and recorded for 14 consecutive days after administration, the body weights of the mice were recorded on 7 th and 14 th days, and the mice were sacrificed and dissected at the end of 14 days after administration to examine whether or not there was abnormality in the organs of the mice.
3. Test results
When observed within 14 days, all the mice fed with the composition have normal physiological activities, no poisoning and death, and no pathological change visible to naked eyes after dissection, which shows that the composition has no acute toxicity to the mice after the mice are gavaged at 10 g/kg.
The body weight data of each group of mice was statistically processed using anova, and one-way anova was performed using SPSS 25.0 software, and the results are shown in table 2.

As can be seen from table 2, the body weights of the mice of the experimental group 9 and the working group 10 were lower than those of the control group without significant difference, indicating that the compositions prepared in examples 1 and 5 had no toxic effect on the mice and high safety.
Tests show that the compositions prepared in examples 2 to 4 have no toxic or side effects similar to those of example 1, and the compositions prepared in examples 6 to 8 have the effects similar to those of example 4 and have no toxic or side effects.
The composition prepared by the invention can be added with conventional pharmaceutical excipients and can be prepared into tablets, pills, granules, oral liquid, suspension or hard capsules according to the conventional process, and the effect of the composition is the same as that of the prepared powder.
As can be seen from the test examples 1 and 2, the composition prepared by the invention can healthily and reasonably reduce the weight, reduce the absorption of the body to oil and fat, reduce the percentage of the fat in the weight, has no side effect, and is beneficial to improving the immunity and the resistance by adding the aronia melanocarpa, the polygonatum odoratum and the citrus powder.
While there have been shown and described what are at present considered the fundamental principles and essential features of the invention and its advantages, it will be apparent to those skilled in the art that the invention is not limited to the details of the foregoing exemplary embodiments, but is capable of other specific forms without departing from the spirit or essential characteristics thereof. The present embodiments are therefore to be considered in all respects as illustrative and not restrictive, the scope of the invention being indicated by the appended claims rather than by the foregoing description, and all changes which come within the meaning and range of equivalency of the claims are therefore intended to be embraced therein.
Furthermore, it should be understood that although the present description refers to embodiments, not every embodiment may contain only a single embodiment, and such description is for clarity only, and those skilled in the art should integrate the description, and the embodiments may be combined as appropriate to form other embodiments understood by those skilled in the art.
Claims (10)
1. The composition for blocking grease absorption is characterized by comprising the following raw materials in parts by weight: 3-8 parts of needle mushroom, 5-10 parts of agaricus bisporus, 3-5 parts of wild cherry, 1-3 parts of immature bitter orange, 0.5-2 parts of cassia seed, 2-5 parts of radix polygonati officinalis, 2-5 parts of citrus reticulata powder and 8-15 parts of fructo-oligosaccharide.
2. The composition for assisting in blocking oil absorption according to claim 1, further comprising the following raw materials in parts by weight: 3-6 parts of radix puerariae.
3. The composition for helping to block oil absorption according to claim 1, wherein the preparation method of the flammulina velutipes or the agaricus bisporus comprises the following steps:
1) enzymolysis: freeze-drying needle mushrooms or agaricus bisporus, crushing to obtain mushroom powder, adding cellulase, protease and water into the mushroom powder to obtain a mixed solution, adjusting the pH of the mixed solution to 4-5, performing enzymolysis at 45-55 ℃ for 3-5 hours to obtain an enzymolysis solution, wherein the addition amount of the cellulase is 0.1-0.3% of the weight of the mushroom powder, the addition amount of the protease is 0.1-0.3% of the weight of the mushroom powder, and the addition amount of the water is 30-100 times of the weight of the mushroom powder;
2) and (3) post-treatment: and (3) decocting the enzymolysis liquid at 100-110 ℃ for 0.5-1 h, cooling, centrifuging the enzymolysis liquid, filtering the obtained supernatant to remove residues, precipitating with ethanol, and freeze-drying to obtain the product, wherein the dosage of the ethanol is 2-5 times of the volume of the supernatant.
4. The composition for helping to block oil absorption according to claim 1, wherein the aronia melanocarpa is prepared by the following method: cleaning the aronia melanocarpa, crushing and pulping, carrying out filter pressing on pulp to obtain filter residue, adding ethanol and hydrochloric acid into the filter residue, leaching for 1-3 h at 50-60 ℃, carrying out ultrasonic-assisted leaching for 10-15 min, wherein the microwave power is 300-400W, filtering, concentrating the filtrate, and drying, wherein the volume percentage of the ethanol is 90-95%, the molar concentration of the hydrochloric acid is 1-2 mol/L, and the dosage of the filter residue, the ethanol and the hydrochloric acid is 1-3 g, 200-300 mL and 30-60 mL.
5. The composition for assisting in blocking oil absorption according to claim 1, wherein the immature bitter orange is prepared by a method comprising the following steps of: the method comprises the steps of ultrasonically cleaning immature bitter oranges, fully drying the immature bitter oranges, adding clear water, decocting for 2-4 hours, filtering, taking filtrate, and performing spray drying to obtain the immature bitter oranges, wherein the mass ratio of the immature bitter oranges to the clear water is 1: 6-10.
6. The composition for helping to block oil absorption according to claim 1, wherein the cassia seed is prepared by the following steps:
1) pretreatment: crushing cassia seeds, freeze-drying the crushed cassia seeds at the temperature of-5 to-10 ℃, putting the frozen cassia seed powder into a puffing tank, and puffing at variable temperature and pressure difference to obtain a puffed material of the cassia seeds, wherein the pressure in the puffing tank is increased from normal pressure to 0.3 to 0.6MPa, the puffing temperature is increased to 160 to 250 ℃, the pressure in the puffing tank is instantly reduced to vacuum, and the puffing time is 50 to 100 s;
2) extraction: adding cellulase and water into the semen cassiae puffed material, extracting for 3-6 hours at 50-65 ℃, wherein the addition amount of the cellulase is 0.5-1% of the weight of the semen cassiae puffed material, the addition amount of the water is 30-100 times of the weight of the semen cassiae puffed material, extracting by microwave assistance after enzymolysis is finished, the microwave power is 300-400W, the microwave treatment time is 5-15 min, filtering, and drying filtrate to obtain the semen cassiae puffed material.
7. The composition for assisting in blocking lipid absorption according to claim 1, wherein the method for preparing Yuzhu comprises: sequentially carrying out ultrasonic cleaning, drying and crushing treatment on polygonatum odoratum, soaking for 12-20 h, carrying out reflux extraction by using 60-85% ethanol in percentage by volume to obtain an extraction solution, adsorbing the extraction solution by using macroporous adsorption resin, eluting by using 10% ethanol until an eluent is colorless, and carrying out reduced pressure concentration and drying to obtain the polygonatum odoratum extract.
8. The preparation method of the composition for blocking oil absorption according to any one of claims 1 to 7, wherein the raw materials are respectively crushed and sieved, 100-200 mesh powder is collected, the collected raw materials are fully mixed according to the corresponding weight part ratio, 80-90% by volume of ethanol is added, the mixture is mixed for 10-20 min, the mixture is dried for 2-4 h at the temperature of 60-80 ℃, and the mixture is ground to obtain the composition.
9. A preparation of a composition for helping to block oil absorption, which is characterized in that the preparation is prepared by matching the composition for helping to block oil absorption in any one of claims 1 to 7 with other pharmaceutic adjuvants.
10. The preparation of the composition for helping to block oil absorption according to claim 9, wherein the preparation is in the form of one of tablets, pills, powder, granules, oral liquid, suspension and hard capsules.
