The invention also provides a preparation method of the traditional Chinese medicine composition, and the preparation method comprises the steps of mixing, extracting, concentrating, drying, crushing and sieving the gynura divaricata, the ginseng, the purslane, the sophora flower bud/sophora flower, the mulberry leaf, the gardenia, the kudzu vine root, the grape seed and the corn stigma according to the formula amount to obtain the traditional Chinese medicine composition.
Preferably, the extraction is water extraction and alcohol extraction.
The invention also provides application of the traditional Chinese medicine composition in preparation of hypoglycemic drugs and/or health foods.
The invention also provides a medicament for reducing blood sugar, which comprises the traditional Chinese medicine composition and medically acceptable auxiliary materials.
Preferably, the medicament includes, but is not limited to, granules, tablets, pills and capsules.
Compared with the prior art:
(1) the traditional Chinese medicine composition for reducing blood sugar can enhance and restore the function of pancreatic insulin secretion or/and eliminate insulin resistance through the synergistic compounding of the gynura divaricata, the ginseng and the purslane, thereby achieving the effect of effectively reducing blood sugar.
(2) The traditional Chinese medicine composition for reducing blood sugar takes medicine-food homologous traditional Chinese medicines as main raw materials, has high safety and no toxic or side effect, can be taken for a long time, effectively avoids the risk of liver injury possibly caused by the common use of a plurality of traditional Chinese medicine components, and improves the safety of medicines and the compliance of patients.
(3) The preparation method disclosed by the invention is simple in process, low in cost and suitable for large-scale production.

Wherein the insulin kit is purchased from Yiprises technologies, Inc. of Beijing; the blood glucose kit is purchased from Shenzhen Merrill biomedical electronics GmbH; the flos Sophorae Immaturus/flos Sophorae Immaturus is flower and bud of flos Sophorae.
Basic embodiment
A traditional Chinese medicine composition for reducing blood sugar comprises the following components in parts by weight: 2-50 parts of gynura divaricata, 1-30 parts of ginseng and 2-50 parts of purslane.
Preferably, the traditional Chinese medicine composition comprises the following components in parts by weight: 5-48 parts of gynura divaricata, 1.5-28 parts of ginseng and 5-45 parts of purslane.
In some preferred embodiments, the traditional Chinese medicine composition further comprises the following components in parts by weight: 1-30 parts of sophora flower bud/sophora flower, 1-30 parts of mulberry leaf and 1-20 parts of cape jasmine.
Preferably, the weight ratio of the mulberry leaves to the gardenia is 1: 1.
In some preferred embodiments, the traditional Chinese medicine composition further comprises the following components in parts by weight: 1-20 parts of kudzu root, 1-20 parts of grape seed, 1-20 parts of medlar and 1-30 parts of corn stigma.
Preferably, the weight ratio of the kudzu root to the grape seed to the corn silk is 1:1: 2.
In some preferred embodiments, the Chinese medicinal composition comprises the following components in parts by weight: 2-50 parts of gynura divaricata, 1-30 parts of ginseng, 2-50 parts of purslane, 1-30 parts of sophora flower bud/sophora flower, 1-30 parts of mulberry leaf, 1-20 parts of gardenia, 1-20 parts of kudzu root, 1-20 parts of grape seed, 1-20 parts of wolfberry fruit and 1-30 parts of corn stigma.
In some preferred embodiments, the Chinese medicinal composition comprises the following components in parts by weight: 20 parts of gynura divaricata, 2 parts of ginseng, 20 parts of purslane, 4 parts of sophora flower bud/sophora flower bud, 2 parts of mulberry leaf, 2 parts of cape jasmine, 2 parts of kudzu root, 2 parts of grape seed, 2 parts of wolfberry fruit and 4 parts of corn stigma.

First, acute toxicity test
1. Laboratory animal
ICR mice (purchased from Shanghai Spiker laboratory animals Co., Ltd.) weighed 18-22g, male.
2. Grouping and administration method
The ICR mice are randomly divided into 23 groups according to body weight, each group comprises 8 mice, and the traditional Chinese medicine compositions prepared in the examples 1-14 and the comparative examples 1-8 are prepared into suspensions with the concentration of 0.2g/mL respectively; the administration volume is 0.4mL/10g, and the administration dose of the mouse is determined to be 8.0 g/kg; the control group was administered with an equal volume of deionized water, all administered by gavage once a day.
3. Results of the experiment
After the gastric lavage, the mouse is quieter and can move normally. Other conditions such as diet, activity, hair color, feces, etc. are found to be abnormal. After 14 days, the weight of each animal was increased (see table 4), and no death was observed.

As can be seen from the above table, the maximum tolerated dose MTD of the traditional Chinese medicine composition provided by the invention for mice after gastric lavage is greater than 8.0g/kg, and no obvious abnormality of the mice is seen. Therefore, the traditional Chinese medicine composition has no toxic or side effect and high safety, can be taken for a long time, effectively avoids the risk of liver injury caused by the common use of various traditional Chinese medicine components, and improves the safety of the medicine and the compliance of patients.
Second, hypoglycemic test
1. Establishment of diabetes animal model
ICR mice are intraperitoneally injected with 200mg/kg · bw 4% alloxan solution, before injection, fasting is carried out for 12h without water prohibition, after 72h, blood collection is carried out on orbital venous plexus to measure fasting blood glucose, and the mice with blood glucose value larger than 11.1mmol/L are experimental diabetes animal models.
2. Grouping and administration of drugs
The successfully modeled diabetes animal models are randomly divided into 23 groups of 8 animals, wherein 22 groups are experimental groups, the traditional Chinese medicine composition for reducing blood sugar prepared in examples 1-14 and comparative examples 1-8 is respectively gavaged every day, the gavage dose is 200mg/kg, 1 group of model control groups are gavaged every day with physiological saline with the same volume. Meanwhile, 1 group of normal control groups (8 normal mice) is provided, and the normal saline with the same volume as the stomach is infused every day; the stomach 28d is perfused continuously.
Fasting Blood Glucose (FBG) values were measured once per week for each group of mice; fasting for 12h before the last administration, blood sampling of orbital vein after 2h administration, serum centrifugation, and serum insulin level determination by kit method.

Note: the # P is less than 0.01, and has very significant difference compared with a normal control group; p < 0.5, with significant difference compared to model control; p < 0.01, with a very significant difference compared to the model control group.
As can be seen from Table 5, the fasting blood glucose of the mice of the normal control group and the model control group of the invention is very significantly different (P is less than 0.01), which indicates that the model of the diabetes animal model of the invention is successfully modeled.
The fasting blood glucose monitored by 21d and 28d of the mice filled with the traditional Chinese medicine composition prepared in the groups 1 to 6 of the invention is significantly different (P is less than 0.5) compared with the control group of the model, which shows that the traditional Chinese medicine composition prepared by the invention has the effect of reducing blood glucose.
Meanwhile, it was found that the fasting blood glucose monitored at 14d and 21d of the mice filled with the Chinese medicinal composition prepared in the gavage example 6 is significantly different (P < 0.5) compared with the control group; the fasting blood glucose monitored at 28d was significantly different (P < 0.01) from the model control group. The prepared traditional Chinese medicine composition has a good blood sugar reduction effect when the weight ratio of the gynura divaricata, the ginseng and the purslane is 10:1: 10.
As can be seen from the above Table 5, the mice fed with the Chinese medicinal composition prepared in the gavage examples 7-9 showed significant differences in fasting blood glucose (P < 0.5) in 14d and 21d, compared with the model control group; the fasting blood glucose monitored at 28d is very significantly different (P is less than 0.01) compared with the model control group; in mice fed with the traditional Chinese medicine composition prepared in the gavage example 10, fasting blood glucose monitored at 21d and 28d is very significantly different (P is less than 0.01) compared with a model control group; the weight ratio of the mulberry leaves to the gardenia is 1:1, so that the prepared traditional Chinese medicine composition has a good blood sugar reduction effect.
As can be seen from the above Table 5, the mice fed with the Chinese medicinal composition prepared in the gavage examples 11-13 showed significant differences (P < 0.5) in fasting blood glucose monitored at 7d and 14d, compared with the control group; the fasting blood sugar monitored by 21d and 28d is very different from that of the model control group (P is less than 0.01); the 7d fasting blood glucose of the mice with the Chinese medicinal composition prepared in the gavage example 14 is significantly different (P < 0.5) compared with the control group; the fasting blood glucose monitored at 14d, 21d and 28d is very significantly different (P < 0.01) compared with the model control group; the prepared traditional Chinese medicine composition has a good blood sugar reducing effect when the weight of the kudzu root, the grape seed and the corn stigma is 1:1: 2.

As can be seen from table 6, the serum insulin level of the mouse of the model control group is significantly lower than that of the normal control group and shows a significant difference (P < 0.01), and the insulin level of the mouse of the gavage traditional Chinese medicine composition prepared in the embodiments 1 to 14 of the present invention is significantly higher (P < 0.5) than that of the model control group, which indicates that the traditional Chinese medicine composition prepared in the present invention may have the abilities of repairing islet β cells and increasing insulin secretion of the islet β cells, thereby increasing the serum insulin level of the diabetic mouse and reducing the blood sugar of the diabetic mouse.
In conclusion, the traditional Chinese medicine composition for reducing blood sugar can recover the function of pancreatic insulin secretion or/and eliminate insulin resistance through the synergistic compounding of the gynura divaricata, the ginseng and the purslane, so that the effect of effectively reducing blood sugar is achieved.
The traditional Chinese medicine composition for reducing blood sugar takes medicine-food homologous traditional Chinese medicines as main raw materials, has high safety and no toxic or side effect, can be taken for a long time, effectively avoids the risk of liver injury possibly caused by the common use of a plurality of traditional Chinese medicine components, and improves the safety of medicines and the compliance of patients.
In addition, the preparation method provided by the invention is simple in process, low in cost and suitable for large-scale production.
It should be emphasized that the embodiments described herein are illustrative rather than restrictive, and thus the present invention is not limited to the embodiments described in the detailed description, but also includes other embodiments that can be derived from the technical solutions of the present invention by those skilled in the art.

Claims (10)

1. The traditional Chinese medicine composition for reducing blood sugar is characterized by comprising the following components in parts by weight: 2-50 parts of gynura divaricata, 1-30 parts of ginseng and 2-50 parts of purslane.
2. The traditional Chinese medicine composition according to claim 1, which is characterized by comprising the following components in parts by weight: 5-48 parts of gynura divaricata, 1.5-28 parts of ginseng and 5-45 parts of purslane.
3. The traditional Chinese medicine composition according to claim 1, further comprising, in parts by weight: 1-30 parts of sophora flower bud/sophora flower, 1-30 parts of mulberry leaf and 1-20 parts of cape jasmine.
4. The Chinese medicinal composition according to claim 3, wherein the weight ratio of the mulberry leaves to the gardenia is 1: 1.
5. The traditional Chinese medicine composition according to claim 3, further comprising, in parts by weight: 1-20 parts of kudzu root, 1-20 parts of grape seed, 1-20 parts of medlar and 1-30 parts of corn stigma.
6. The traditional Chinese medicine composition of claim 5, wherein the weight ratio of the kudzu root to the grape seed to the corn silk is 1:1: 2.

The traditional Chinese medicine composition according to claim 5, which is characterized by comprising the following components in parts by weight: 2-50 parts of gynura divaricata, 1-30 parts of ginseng, 2-50 parts of purslane, 1-30 parts of sophora flower bud/sophora flower, 1-30 parts of mulberry leaf, 1-20 parts of gardenia, 1-20 parts of kudzu root, 1-20 parts of grape seed, 1-20 parts of wolfberry fruit and 1-30 parts of corn stigma.

The traditional Chinese medicine composition according to claim 7, which comprises the following components in parts by weight: 20 parts of gynura divaricata, 2 parts of ginseng, 20 parts of purslane, 4 parts of sophora flower bud/sophora flower bud, 2 parts of mulberry leaf, 2 parts of cape jasmine, 2 parts of kudzu root, 2 parts of grape seed, 2 parts of wolfberry fruit and 4 parts of corn stigma.

The method for preparing a Chinese medicinal composition according to any one of claims 1 to 8, wherein the method comprises mixing, extracting, concentrating, drying, crushing and sieving the gynura divaricata, ginseng, purslane, sophora flower bud/flower bud, mulberry leaf, gardenia, kudzu vine root, grape seed and corn silk in formula amounts to obtain the Chinese medicinal composition.
10. Use of the Chinese medicinal composition according to any one of claims 1 to 8 in the preparation of hypoglycemic drugs and/or health foods.
