Abstract
The invention discloses a hyaluronic acid essential oil composition for nourishing and repairing oral cavity and application thereof; the composition comprises hyaluronic acid, essential oil, licorice root, loquat leaf and centella asiatica extracts, wherein the hyaluronic acid is hyaluronic acid with the mass ratio of 1: 4-5: 3-4 and above 120kDa, hyaluronic acid with the mass ratio of 80-120kDa and hyaluronic acid with the mass ratio of 1-80 kDa. The composition is used for preparing oral cavity repairing product. The invention selects hyaluronic acid with proper molecular weight and plant extracts of hyaluronic acid, liquorice root, asiatic pennywort herb and the like with scientific proportion to be extracted; more importantly, the HA is used as a carrier for drug delivery, multiple target points of glycyrrhizic acid, loquat leaf glycoside, asiaticoside and the like are promoted to act on cells and cells, and the effects of oxidation resistance, free radical resistance, cell nourishing, repairing and the like are achieved; thereby creating a good and lasting microenvironment for the oral cavity.
Description
Technical Field
The invention relates to a hyaluronic acid essential oil composition for nourishing and repairing oral cavity and application thereof.
Background
With the improvement of living standard, people pay more and more attention to oral hygiene, oral health care and oral health, and have relied on toothpaste as a cleaning product in daily life. Toothpaste can remove stains and bacterial plaques on the surfaces of teeth, has no stimulation and toxicity on oral mucosa, and is very important for oral cavity cleaning and oral cavity health care, but only the toothpaste is used. The root of common oral problems such as gingivitis, oropharynx, oral mucositis and the like is the restoration of oral micro-damage after a long time. To address these oral problems, an oral care product must be selected that is effective in repairing the micro-lesions of the oral cavity. Most of the toothpaste in the current market is mainly clean, and other health-care toothpastes with different efficacies are: the traditional Chinese medicine composition has the advantages of preventing caries by adding fluorine, resisting allergy by adding zinc, stopping bleeding by adding a white medicine, disinfecting and resisting bacteria, treating headache, mainly treating symptoms by applying medicines according to symptoms, being difficult to persist and fundamentally determining.
Most toothpaste formulations today are generally only concerned with cleaning alone, or with problems with the teeth alone; but ignores: toothache, bleeding, halitosis, periodontitis and other symptoms are not only problems of teeth, but also problems of oral barrier and mucosal cell damage and defects. If compare the tooth and the tree, the oral cavity is soil and is taken down earth greatly promptly. Trees need to be kept branches and cleaned, but more importantly, the teeth and the trees have the same effect of absorbing various nutrition and growth and development factors from soil. The microenvironment of the oral cavity is poor, so that the probability of a series of problems of the teeth is certainly greatly improved. Therefore, the invention provides a moisturizing and cleaning binary system formula of oral products such as toothpaste, mouthwash and the like, which comprises the following components in parts by weight: a scheme for strengthening body resistance to eliminate pathogenic factors, protecting good micro-ecology of the oral cavity and repairing micro-damage of the oral cavity; moisturizing is performed from each cell in the oral cavity; the flow of water among cells is accelerated, nutrition is conveyed, information is transmitted, metabolism is promoted, the self-repairing capability of the cells is promoted, the creation and repair of the mouth microenvironment are enhanced by the healthy and active cells, and the problems of the mouth and teeth can be avoided fundamentally by the healthy and active cells.
Hyaluronic Acid (HA) is an acidic mucopolysaccharide, an unbranched macromolecular glycosaminoglycan composed of repeating units of N-acetylglucosamine and D-glucuronosyl, present in the stroma of animal tissues and in the capsule of certain bacteria. Hyaluronic acid is widely used in the fields of medicines, cosmetics, foods and the like. Hyaluronic acid has a number of physiological functions, such as anti-inflammatory, anti-infective, anti-edema, tissue remodeling and wound healing, in addition to its functions of retaining water, lubricating, regulating osmotic pressure, etc. With the intensive research and the accumulation of clinical experience, HA HAs many new application fields, wherein the application of HA in the oral cavity field HAs been gradually noticed.
In the prior art, oral care products containing much hyaluronic acid are available, for example, CN 106420493 a discloses a hyaluronic acid toothpaste with anti-inflammatory and antibacterial effects and a preparation method thereof, which utilizes the anti-inflammatory and antibacterial effects of hyaluronic acid, and ascorbic acid and composition fluid extract which have synergistic, anti-inflammatory and antibacterial effects with hyaluronic acid, so as to have better anti-inflammatory and antibacterial effects. CN 104666112 a discloses a bioactive hyaluronic acid toothpaste; the toothpaste accessory thickener sorbitol and the bioactive hyaluronic acid synergistically enhance the anti-inflammatory effect of the bioactive hyaluronic acid. CN107536725A discloses a multi-effect oral composition containing a hyaluronic acid mixture and an application thereof, the invention compounds hyaluronic acid with low molecular weight and hyaluronic acid with high molecular weight to form a mixture, and the mixture has a function compounding effect, the hyaluronic acid with low molecular weight has a good mucous membrane repairing effect, the hyaluronic acid with high molecular weight has a good mucous membrane anti-inflammatory effect, and the hyaluronic acid mixture and zinc citrate are compounded to play a synergistic bacteriostasis effect, inhibit oral pathogenic bacteria and achieve the effect of maintaining oral health. The patent technologies all utilize the anti-inflammatory and bacteriostatic effects of hyaluronic acid. For another example, CN112618389A discloses a baking soda whitening toothpaste containing hyaluronic acid and a preparation method thereof, wherein the compound hyaluronic acid or salt thereof is formed by compounding high and medium molecular weight and oligomeric hyaluronic acid or salt thereof, and the compound hyaluronic acid or salt thereof can alleviate the adverse effects of enamel damage, gingival inflammation and the like caused by long-term use of the baking soda toothpaste, and reduce the formation of sensitive teeth.
The survival period of the human oral cells is only 14 days, and the cells are updated at a speed of about 7 percent per day, so that the cells can provide good habitat for the new cells and repair the new cells in time, and the cells are key to oral health. However, neither the above-mentioned patent techniques nor the prior art relate to nor enable the creation of a good microenvironment of the mouth.
Disclosure of Invention
Aiming at the defects in the prior art, the invention provides a hyaluronic acid essential oil composition for nourishing and repairing the oral cavity and application thereof. The invention selects hyaluronic acid with proper molecular weight and plant extracts of hyaluronic acid, liquorice root, loquat leaf, asiatic pennywort herb and the like with scientific proportion to be extracted; the HA not only utilizes the moisturizing, anti-inflammatory and bacteriostatic effects, but also plays a more important role in drug delivery by taking the HA as a carrier, so that the drugs permeate into the dermis, multiple target points of glycyrrhizic acid, loquat leaf glycoside, asiaticoside and the like are promoted to act between cells, the effects of antioxidation, free radical resistance, cell repair and the like are fully played, the hyaluronic acid and plant extract advantages are mutually supplemented, the synergistic effect is achieved, and a good and lasting oral microenvironment is further created.
The purpose of the invention is realized by the following technical scheme:
the invention provides a hyaluronic acid essential oil oral care composition, which contains hyaluronic acid, essential oil, a licorice root extract, a loquat leaf extract and a centella extract.
The hyaluronic acid, the licorice root extract, the loquat leaf extract and the centella asiatica extract in the composition are all food grade, and the mass ratio of the hyaluronic acid, the licorice root extract, the loquat leaf extract and the centella asiatica extract is (0.01-1): (0.01-0.5): (0.01-0.3). Preferably 0.2:0.25:0.1: 0.05.
According to one embodiment of the invention, the hyaluronic acid is selected from high molecular weight hyaluronic acid, medium molecular weight hyaluronic acid and low molecular weight hyaluronic acid with a mass ratio of 1: 4-5: 3-4.
The molecular weight of the high molecular weight hyaluronic acid is more than 120 kDa.
The molecular weight of the hyaluronic acid with the medium molecular weight is 80-120 kDa.
The molecular weight of the low molecular weight hyaluronic acid is 1-80 kDa.
As one embodiment of the present invention, the glycyrrhizic acid content in the licorice root extract is 55-75%; the content of asiaticoside in the centella extract is 50-70%; the content of loquat glycoside in the loquat leaf extract is 20-50%. In some embodiments, the centella asiatica extract contains 60% of centella asiatica sweet.
As an embodiment of the present invention, the licorice root extract further contains licoflavone and glycyrrhiza polysaccharide. The Glycyrrhrizae radix extract contains 2-5% of licoflavone and 8-12% of Glycyrrhiza polysaccharide. In some embodiments, the licorice root extract has a glycyrrhizic acid content of 65%, a licorice flavone content of 3%, and a licorice polysaccharide content of 10%.
As an embodiment of the present invention, the mass ratio of the essential oil to the hyaluronic acid is: (0.03-0.6):(0.01-1).
As an embodiment of the present invention, the essential oil is a combination of multiple of licorice essential oil, centella essential oil, chamomile essential oil, ylang-ylang essential oil. The licorice essential oil can also be derived from licorice root extract; the centella essential oil can be derived from centella asiatica extract.
The invention also relates to application of the hyaluronic acid essential oil nourishing and repairing composition for the oral cavity in preparation of an oral product.
As an embodiment of the present invention, the oral product has hyaluronic acid content of 0.01-1 wt%, licorice root extract content of 0.01-1 wt%, loquat leaf extract content of 0.01-0.5 wt%, and centella asiatica extract content of 0.01-0.3 wt%.
As an embodiment of the invention, the content of essential oils in the oral product is 0.03-0.6 wt%. Preferably 0.3 to 0.6 wt%.
As an embodiment of the present invention, the oral product comprises a restorative toothpaste, mouthwash, mouth restorative gel, mouth spray, mouth freshener, or pop sugar.
The invention also relates to hyaluronic acid essential oil nourishing and repairing toothpaste which comprises the following components in percentage by weight:
and the balance purified water. In some embodiments, the purified water is present in the toothpaste in an amount of 20 to 50 wt%, preferably 24% to 45%.
As an embodiment of the present invention, the essential oil is a combination of multiple of licorice essential oil, centella essential oil, chamomile essential oil, ylang-ylang essential oil. Preferably 0.3 wt% essential oil. The preferred essential oil composition is in a mass ratio of 1:1:0.5: 0.5. The oral cavity is a habitat with higher flowing frequency, and the essential oil in the proportion is favorable for fast adsorption and permeation into gum and mucosal tissues, so that the efficacy of the essential oil is exerted persistently and effectively.
Three basic elements of oral hygiene: cleaning, moisturizing, nourishing and repairing. The human oral cavity has its own system of cleaning and moisture preservation, relating to the theory of anatomy, oral biology and human immunology. The oral cavity is an important inlet and outlet channel of a human body, and the oral cavity micro-ecology is formed by the human body and microorganisms, so that each cell of the oral cavity can be normally metabolized and moved, and the health of the oral cavity is maintained. Oral health maintenance requires less employment of agents such as antibacterial agents, and relies on the powerful repair capabilities of cells and probiotics themselves. While clean, moist, fresh breath are oral health signs.
Compared with the prior art, the invention has the following beneficial effects:
1) different from glycerin, hyaluronic acid is a substance of a human body, and is reasonably applied to supplement the blank that the secretion of the human body is reduced along with the age, oral epithelial cells and skin need hyaluronic acid to absorb water and preserve moisture, the metabolism of the cells is kept, the moistening of oral mucosa is kept, and the repair function of an oral barrier is protected; after being applied for a few days, the oral cavity sub-health symptoms such as dry mouth, greasy mouth, breath and the like are obviously relieved and disappeared, the oral mucosa of the gum is ruddy, semitransparent and elastic, and the breath is fresh and long-lasting.
2) The invention has no antibacterial agent component, and aims to protect probiotics and maintain oral micro-ecology; by means of the function of HA as a carrier for transferring drugs, factors such as liquorice, centella and the like enter a cell basal layer through epithelial cells, and the functions are better played. In addition, the liquorice has the function of promoting the secretion of saliva by promoting the production of body fluid and removing blood stasis; saliva is rich in HA, peptide, enzyme and the like, and is the best cleaning humectant for human bodies; before the application of the product: the mouth cleaning agent can be used for brushing dry teeth, toothpaste paste dry bodies and dry oral cavities without mouth rinsing, after teeth are brushed for several seconds, people feel that a large amount of saliva is secreted, the whole oral cavity is filled with foams, namely, the liquorice promotes the secretion of saliva and promotes metabolism, the hyaluronic acid locks moisture, and the plant extract is accompanied with micromolecular HA to enter various mucous membranes and cells of the oral cavity to repair the micro-damage of the oral cavity; the cells are healthy, the oral cavity is also healthy, the gum, the periodontal and the teeth are healthier, and a green oral ecology is created.
3) The invention provides a new choice for multi-target repairing oral cell and oral health by using the humectant and bioactive components aiming at oral micro-injury, such as immunity, microbial balance and the like; the oral cavity barrier dysfunction and the oral cavity sub-health state can be solved from different angles, and the oral cavity barrier dysfunction prevention method belongs to the field of good oral cavity prevention learning prospect.

The present invention will be described in detail with reference to examples. The following examples will assist those skilled in the art in further understanding the invention, but are not intended to limit the invention in any way. It should be noted that it would be apparent to those skilled in the art that several modifications and improvements can be made without departing from the inventive concept. All falling within the scope of the present invention.
In the hyaluronic acid essential oil repairing oral composition, the mass ratio of hyaluronic acid to licorice root extract to loquat leaf extract to centella asiatica extract is (0.01-1) to (0.01-1): (0.01-0.5): (0.01-0.3). The composition is specifically applied to oral products, and comprises 0.01-1 wt% of hyaluronic acid, 0.01-1 wt% of licorice root extract, 0.01-0.5 wt% of loquat leaf extract, 0.01-0.3 wt% of centella asiatica extract and 0.1-0.6 wt% of essential oil. The invention realizes the construction of good mouth microenvironment by hyaluronic acid with proper molecular weight and hyaluronic acid, glycyrrhizic acid and asiaticoside with scientific proportion. Wherein the hyaluronic acid is selected from 120kDa hyaluronic acid, 80-100kDa hyaluronic acid and 1-60kDa hyaluronic acid with the mass ratio of 1: 4-5: 3-4. The polymer HA covers the cells, locks water and preserves moisture, forms a warm-wet effect for the cells under mucosa, and the medium and small molecular HA can permeate into different layers and different depths to play a role in transferring carrier drugs and promote glycyrrhizic acid, loquat leaf glycoside, asiaticoside and the like to fully play a role in the cells.

Examples 1 to 5 relate to hyaluronic acid essential oil repair toothpaste, the composition of which is shown in table 1; wherein the Glycyrrhrizae radix extract contains glycyrrhizic acid, licoflavone and Glycyrrhiza polysaccharide, and wherein glycyrrhizic acid content is 65%, licoflavone content is 3%, and Glycyrrhiza polysaccharide content is 12%; the content of asiaticoside in the centella extract is 60%; the content of loquat glycoside in the loquat leaf extract is 40%. The essential oil comprises licorice essential oil, centella essential oil, chamomile essential oil and cananga odorata essential oil in a mass ratio of 1:1:0.5: 0.5.
Comparative example 1
Comparative example 1 relates to a repair toothpaste having the composition shown in table 1; it differs from example 5 only in that: the oral care composition is free of hyaluronic acid essential oil, and is free of hyaluronic acid, essential oil, Glycyrrhrizae radix extract, folium Eriobotryae extract and herba Centellae extract.
Comparative example 2
Comparative example 2 relates to a hyaluronic acid essential oil repair toothpaste, the composition of which is shown in table 1; it differs from example 5 only in that: it does not contain Glycyrrhrizae radix extract.
Comparative example 3
Comparative example 3 relates to a hyaluronic acid essential oil repair toothpaste, the composition of which is shown in table 1; it differs from example 5 only in that: it does not contain centella asiatica extract.
Comparative example 4
Comparative example 4 relates to a hyaluronic acid essential oil repair toothpaste, the composition of which is shown in table 1; it differs from example 5 only in that: it does not contain loquat leaf extract.
Comparative example 5
Comparative example 5 relates to a hyaluronic acid essential oil repair toothpaste, the composition of which is shown in table 1; it differs from example 5 in that: all hyaluronic acid was selected as 100kDa HA.

Verification example
1. Water retention detection
The products of each example and comparative example are sent to the cosmetic technology center of the Chinese inspection and quarantine scientific research institute for water retention rate detection. The raw weights of the toothpastes of the examples and comparative examples were measured and the corresponding weights were recorded after 2, 4, 6, 8, 10, 12, 24, 36, 48, 72, 96 hours, converted to water retention rates of the examples and comparative examples, as shown in table 2 below:

As can be seen from the analysis of the relative water retention results in Table 2, the water retention time of the product in example 5 is obviously increased by 103% from 29.8% to 60.7% compared with that in comparative example 1 after 8 hours; increased by 251% in 10 hours; the increase was 23.8 times for 12 hours. In addition, comparative example 5 HAs HA, but the moisture-keeping effect is not good when the mixture ratio is not proper.
2. Antibacterial testing (suspension quantitative method)
The products of each embodiment and the comparative example are sent to the cosmetic technical center of China inspection and quarantine scientific research institute for antibacterial detection, and the detection and evaluation are based on WS/T650-plus 2019 antibacterial and bacteriostatic effect evaluation method; the test was repeated three times under the test conditions; the 40% concentration sample toothpaste (suspension) acted on porphyromonas gingivalis for 3min (the 40% concentration suspension is prepared from toothpaste paste and water 1: 2, the proportion of the paste is 1.15, the concentration is 39.8%), the average value of each bacteriostasis rate is shown in the following table 2: the test is repeated three times under the condition of 20 +/-1 ℃; the 40% sample toothpaste (suspension) acts on helicobacter pylori for 3min, and the average value of each bacteriostasis rate is shown in the following table 3 (more than 90%, the toothpaste is judged to have strong bacteriostasis):

3. Anti-free radical test: the effect of obviously removing DPPH free radicals is achieved when the test concentration of each example and each comparative example is more than 12.5 percent according to the condition that the paste is 100 percent; the results are given in table 4 below:

In table 4, the control refers to a blank, i.e., a commercial toothpaste of ten dollars or less.
4. Anti-inflammatory and anti-allergic efficacy was evaluated in vitro (IL-6), and dexamethasone was used as a negative control.
The example 5 is sent to the cosmetic technology center of the Chinese inspection and quarantine science research institute for in vitro anti-inflammatory and allergy-relieving efficacy evaluation experiment, and the analysis of the cell viability detection result (action 24h) is shown in figure 1; wherein, the HA concentration of the paste is 100 percent, and the concentration is converted into the test concentration; example 5 toothpaste containing 0.8% hyaluronic acid, i.e. 1g of toothpaste containing 8mg HA, was diluted with water to HA concentrations as shown in FIG. 1(a), respectively.
FIG. 2 is the raw data of IL-6 content measurement, the standard curve is shown in FIG. 3, and the analysis of the test results is shown in FIG. 4:
considering that the control group and the test group are separately arranged for biological cell detection, the control component is negative control, namely no stimulation and no medicine addition are carried out, and the model control makes the negative control cells have inflammation stimulation. Positive control negative control cells were stimulated for inflammation and dexamethasone was added. The comparison shows that the sample group is superior to the positive group, and the effect is better than that of dexamethasone.
FIG. 4 is a graph of the effect of toothpaste of example 5 on IL-6 levels, # # vs. negative control, p < 0.01; comparing with model control, p is less than 0.01. Compared with a negative control group, the secretion level of the inflammatory factor IL-6 of the model control group is obviously increased (p is less than 0.01), which indicates that the modeling is successful, and the model can be used for evaluating the desensitization effect of the test object. Compared with a model control group, the IL-6 release amount is not reduced after the HOEC cells act on the samples with the concentration of 0.16mg/ml and 0.31 mg/ml; the samples at a concentration of 0.08mg/ml had a statistically significant (p < 0.01) decrease in IL-6 release after HOEC cells were treated. Example 5 at a concentration of 0.08mg/ml, the release amount of IL-6 secreted by HOEC cells induced by IFN-y and LPS together can be remarkably reduced, and the slow repair effect on mucosal cells is shown.

Claims (10)
1. A hyaluronic acid essential oil composition for nourishing and repairing oral cavity, which is characterized by comprising hyaluronic acid, essential oil, licorice root extract, loquat leaf extract and centella extract.
2. The hyaluronic acid essential oil nourishment and restoration oral composition according to claim 1, wherein the mass ratio of hyaluronic acid, licorice root extract, loquat leaf extract and centella asiatica extract in the composition is (0.01-1) to (0.01-1): (0.01-0.5): (0.01-0.3).
3. The hyaluronic acid essential oil nourishing and repairing composition for the oral cavity as claimed in claim 1, wherein the hyaluronic acid is hyaluronic acid with a mass ratio of 1: 4-5: 3-4 and above 120kDa, hyaluronic acid with a mass ratio of 80-120kDa and hyaluronic acid with a mass ratio of 1-80 kDa.
4. The hyaluronic acid essential oil nourishment and restoration oral composition according to claim 1, wherein the glycyrrhizic acid content in the licorice root extract is 55-75%; the content of asiaticoside in the centella extract is 50-70%; the content of loquat glycoside in the loquat leaf extract is 20-50%.
5. The hyaluronic acid essential oil nourishment and restoration oral composition according to claim 4, wherein the licorice root extract further comprises licoflavone and glycyrrhiza polysaccharide; the Glycyrrhrizae radix extract contains 2-5% of licoflavone and 8-12% of Glycyrrhiza polysaccharide.
6. The hyaluronic acid essential oil nourishing and restoring oral composition as defined in claim 1, wherein the mass ratio of hyaluronic acid to hyaluronic acid is (0.03-0.6): (0.01-1); the essential oil is a combination of multiple of licorice essential oil, centella essential oil, chamomile essential oil and cananga odorata essential oil.
7. Use of a hyaluronic acid essential oil trophy-repairing oral composition according to any of claims 1-6, for the preparation of an oral product.
8. Use according to claim 7, characterized in that the content of hyaluronic acid in the oral product is between 0.01 and 1 wt%, the content of licorice root extract is between 0.01 and 1 wt%, the content of loquat leaf extract is between 0.01 and 0.5 wt% and the content of centella asiatica extract is between 0.01 and 0.3 wt%.
9. The use of claim 7, wherein the oral product comprises a restorative toothpaste, a mouthwash, a mouth repair gel, a mouth spray, a mouth freshener, or an exploding bead.
