Description

Toothpaste containing centella extract and preparation method thereof
Technical Field
The invention belongs to the technical field of oral cleaning products, and particularly relates to a toothpaste containing a centella extract and a preparation method thereof.
Background
The oral cavity is one of the ways of pathogenic microorganisms invading human body, and the temperature, humidity and food residues in the oral cavity are suitable for the growth and reproduction of microorganisms. Effective oral care maintains the oral cavity clean and prevents the occurrence of oral infections. Oral health has become one of the important indicators for the world health organization to evaluate the level of human health, and maintaining oral health is a physiological requirement of humans, and oral care is an integral part of body care. Common oral problems include: swelling and aching of gums, tooth decay, canker sores, bleeding gums, gingivitis, periodontitis, gingival atrophy, and loss of cementum, among others. The health of teeth is very important to the overall health of a human. Poor oral health has been reported to be closely related to systemic diseases such as heart disease, diabetes and some respiratory diseases.
The toothpaste matrix mainly comprises one or more of glycerol, sorbitol, propylene glycol or polyethylene glycol used as a wetting agent, sodium hydroxymethyl cellulose, hydroxyethyl cellulose, carrageenan or xanthan gum used as an adhesive singly or in combination, sodium lauryl sulfate used as a surfactant, calcium carbonate, aluminum hydroxide, silicon dioxide or calcium bicarbonate used as a friction agent singly or in combination, essence and deionized water. Sodium lauryl sulfate is an excellent anionic surfactant, is used as a most common foaming agent in toothpaste, is one of important raw materials for toothpaste production, and endows the toothpaste with good foaming capacity and also plays roles in emulsification, cleaning and decontamination.
However, sodium lauryl sulfate has some irritation to the oral cavity, and in order to reduce irritation, the skilled person will generally reduce the amount of sodium lauryl sulfate or choose not to use sodium lauryl sulfate, or to use a milder foaming agent (such as sodium lauroyl sarcosinate) to reduce irritation to the oral cavity. In recent years, with the increasing living standard of the public, the requirement of people for health is improved, and toothpaste which is safe, effective and has small side effect is favored by more and more consumers.
The plant extract is a substance which exists in a plant body and is extracted by a physical or chemical method to have biological activity, and effective components of the substance, such as alkaloid, volatile oil, flavone, polysaccharide, organic acid, tannin, grease and the like, have antibacterial and anti-inflammatory effects with different degrees. Chinese patent CN 107184473B discloses a toothpaste containing spina gleditsiae extract, comprising the following components: 0.5-8 parts of spina gleditsiae extract; 10-30 parts of a wetting agent; 15-45 parts of a friction agent; 0.5-1.5 parts of foaming agent; 0.4-1.6 parts of a binder; 0.15-0.3 part of a stabilizer; 0.2-0.8 part of sweetening agent. The invention adds the natural plant extract components such as the spina gleditsiae extract and the like, has obvious effects of resisting bacteria, diminishing inflammation and the like, but the added foaming agent contains sodium lauryl sulfate, has certain irritation to the oral cavity, has poor stability, and is easy to generate the phenomena of layering and the like.
Therefore, there is a need to develop a toothpaste containing natural plant extracts, which has the basic functions of toothpaste, and also has the functions of bacteriostasis, anti-inflammation, anti-allergy, hemostasis and the like. Meanwhile, the oral liquid has the characteristics of safety, mildness, no stimulation to the oral cavity, high stability and the like.

Examples 1-3, comparative examples 1-4, and blank control, 10 mice per group were depilated with sodium sulfide as an abdominal depilatory for preinitiation mice, and the toothpaste containing centella asiatica extract prepared in examples 1-3 and comparative examples 1-4 was applied to an area of 2cm × 1cm (about 10% of the total body surface area) and used in an amount of 0.5g/cm2The toothpaste to be tested is respectively and uniformly coated on the shaving area 1 time a day by locally coating a glass rod; smearing toothpaste matrix on blank control group; after the last administration for 1h, coating 0.02 ml/mouse of 100% dimethylbenzene on the front and back two faces of the left ear of a mouse, separating 4 hours, dislocating and killing the cervical vertebra of the mouse, cutting two ears along the base line of auricle, weighing by an electronic balance, and calculating swelling degree, wherein the formula is as follows: swelling degree is the weight of left ear piece to right ear piece.
3. Test results
The test results are shown in table 1.
TABLE 1 Effect on swelling of mouse ears causing inflammation
Group of Number of animals Swelling degree (mg)
EXAMPLE 1 group 10 14.7±5.02*
EXAMPLE 2 group 10 14.1±4.79**
EXAMPLE 3 group 10 15.2±4.36
Comparative example 1 group 10 22.9±2.98*
Comparative example 2 group 10 23.4±6.71
Comparative example 3 group 10 21.2±5.26**
Comparative example 4 group 10 16.3±2.64*
Blank control group 10 26.3±4.75
As can be seen from Table 1: the toothpastes containing centella asiatica extracts prepared in example 1, example 2 and example 3 significantly reduced swelling degree of mouse ear pinna due to xylene and had anti-inflammatory effect, compared to the blank control group, wherein the group of example 2 was the best example of the present invention. While the toothpaste containing centella asiatica prepared in comparative examples 1-3 (when the anti-allergy soothing agent added in the present invention was changed) was significantly less effective in reducing the swelling degree of mouse ear swelling caused by xylene than the results of the experiments in examples 1-3. It can be seen that the anti-allergy soothing agent provided by the invention has the advantages of no component, obvious synergistic effect with the centella asiatica extract, good anti-inflammatory effect and capability of relieving swelling and pain of gum.
Test example two, stability test
1. Test materials: examples 1-3, comparative examples 1-4 toothpaste containing centella asiatica extract was prepared.
2. The test method comprises the following steps: toothpaste packs containing centella asiatica extracts prepared in examples 1 to 3 and comparative examples 1 to 4
Placing into a toothpaste tube for storage, storing at 5 deg.C and 40 deg.C for three months, and observing each tooth
Storage of the paste.
3. Test results
The test results are shown in table 2.
Table 2 stability test results for each toothpaste
Figure BDA0002556127570000101
As can be seen from Table 2: after the toothpaste containing the centella asiatica extract prepared in the groups 1-3 is stored at 5 ℃, room temperature and 40 ℃ for three months respectively, the toothpaste body is uniform and fine, and the solid-liquid separation phenomenon does not occur, which shows that the toothpaste containing the centella asiatica extract prepared by the invention has good stability. In the comparative example 4, the toothpaste cannot be stably stored after the components in the humectant are changed, and the phenomena of layering, separation and water diversion of the paste body and the like can occur after the toothpaste is placed for a certain time.
The foregoing embodiments are merely illustrative of the principles and utilities of the present invention and are not intended to limit the invention.
