技术领域
本发明属于天然药物技术领域,尤其涉及黄夹次乙苷作为抗肿瘤药物的应用。
背景技术
肿瘤是机体在各种致癌因素作用下,局部组织的某一个细胞在基因水平上失去对其生长的正常调控,导致其克隆性异常增生而形成的新生物,分为良性肿瘤和恶性肿瘤两大类,其中恶性肿瘤对人类生命健康存在更大威胁。目前,恶性肿瘤的治疗方法很多,早期肿瘤以手术治疗为主,中晚期肿瘤以放、化疗为主。恶性肿瘤在早期容易转移,给外科手术切除带来了极大的困难;放、化疗在消灭肿瘤细胞的同时,患者受其毒副作用影响而表现出较差的预后。并且目前现有技术中的一线化疗药物均存在容易耐药、易复发、副作用大等问题。
在各种恶性肿瘤中,骨肉瘤(OS)是长骨中最常见的原发性恶性骨肿瘤,其中儿童和青少年特别危险,儿童发病率每年为0.2-3例/10万,青少年为0.8-11/10万。转移是OS相关死亡的主要原因。尽管有效的化疗药物以及新辅助化疗使患者的5年生存率增加到66-82%,只有30%的转移性OS患者达到5年无事件生存期。那些可检测到转移的患者5年生存率低至19%。此外,即使在没有原发转移的患者中,40%将继续发展为继发转移;在一项研究中,非转移性高级别骨肉瘤随后转移的患者的生存率分别为5%和23%,4年分别为肺和骨转移。同时,传统化疗药物同时伴有较大的副作用,骨肉瘤患者对传统化疗药物耐受较差是骨肉瘤患者生存率停滞不前的另一个重要原因。
发明内容
针对现有技术中的一线化疗药物均存在容易耐药、易复发、副作用大等问题,本发明提供了黄夹次乙苷作为抗肿瘤药物的应用。
以及,本发明还提供了一种抑制体外骨肉瘤增殖细胞增殖的方法。
以及,本发明还提供了一种广谱抗肿瘤药物组合物。
为达到上述发明目的,本发明实施例提供了黄夹次乙苷作为抗肿瘤药物的应用。
相对于现有技术而言,黄夹次乙苷是一种强心苷类的天然产物,来源于中药夹竹桃科黄花夹竹桃(Thevetia peruviana(Pers.)K.Schum.)提取物。本发明在研究发现了黄夹次乙苷的广谱抗癌作用,其抗肿瘤机制包括抑制肿瘤细胞增殖、促进凋亡、周期阻滞、抑制迁移、抑制血管新生和肿瘤免疫等,对多种肿瘤细胞具有抑制增殖作用和直接杀死作用。但黄夹次乙苷对正常的内皮细胞与心肌细胞并无明显影响,如图24、图15所示,黄夹次乙苷对小鼠体重,脏器指数,心电图并无明显影响,证明了黄夹次乙苷对肿瘤的选择性以及安全性。。
优选地,所述肿瘤包括骨肉瘤、乳腺癌、宫颈癌、卵巢癌、神经母细胞瘤和肝癌。黄夹次乙苷对上述肿瘤的肿瘤细胞具有抑制增殖作用或直接杀死作用。
优选地,所述的抗肿瘤药物是抗骨肉瘤药物。
优选地,所述的抗肿瘤药物是抗乳腺癌药物。本研究经过体内实验发现,黄夹次乙苷能抑制小鼠乳腺癌生长。目前,通过在临床中使用靶向免疫检查点调控蛋白(如PD-1)已见证了激活免疫系统能够治疗癌症。黄夹次乙苷能通过多种途径显著激活肿瘤免疫并下调PD-1的表达,从而抑制4T1乳腺癌的生长。并且发明人经过研究发现,黄夹次乙苷虽然在体外并没有抑制4T1乳腺癌细胞增殖的作用,但在体内却能明显抑制4T1乳腺癌的生长。
优选地,所述的抗肿瘤药物是激活肿瘤免疫的药物。黄夹次乙苷能显著激活肿瘤免疫并下调PD-1的表达,从而达到抗肿瘤作用。
优选地,所述的抗肿瘤药物是抑制肿瘤血管新生的药物。新血管形成(血管生成)是肿瘤生长的基础。由于缺氧,肿瘤组织产生并释放血管生成生长因子,例如血管内皮生长因子(VEGF),酸性和碱性成纤维细胞生长因子(aFGF和bFGF)以及血小板衍生的内皮细胞生长因子(PD-ECGF),来形成新血管。黄夹次乙苷能显著抑制内皮细胞血管形成,具有抑制肿瘤血管新生的活性。
以及,本发明实施例还提供了一种抑制体外骨肉瘤增殖细胞增殖的方法,将黄夹次乙苷加入到肿瘤细胞的培养液中,黄夹次乙苷在所述培养液中的终浓度为10~100nM。在10~100nM浓度范围内,黄夹次乙苷能抑制内皮细胞血管形成,抑制人骨肉瘤细胞迁移。
以及,本发明实施例还提供了一种广谱抗肿瘤药物组合物,其特征在于,所述的抗肿瘤药物组合物中含有黄夹次乙苷。
优选地,将所述的广谱抗肿瘤药物组合物制成片剂、胶囊剂、颗粒剂、口服液、缓释制剂、控释制剂、纳米制剂或者注射剂,以方便临床使用。

实施例1
本发明实施例提供了黄夹次乙苷作为广谱抗肿瘤药物对多种肿瘤细胞体外增殖活性的影响,具体如下:
1、实验方法
1.1实验细胞株
EA.hy926(人脐静脉细胞融合细胞株),来自于天津中医药大学现代中药国家重点实验室;
MYO(乳鼠原代心肌细胞系),来自于天津中医药大学现代中药国家重点实验室;
U2OS(人骨肉瘤细胞株),来自于天津中医药大学现代中药国家重点实验室;
A2780(人宫颈癌细胞株),来自于天津中医药大学现代中药国家重点实验室;
Hela(人卵巢癌细胞株),来自于天津中医药大学现代中药国家重点实验室;
MCF7(人三阳性乳腺癌细胞株),来自于天津中医药大学现代中药国家重点实验室;
SK-N-SH(神经母细胞瘤细胞),来自于天津中医药大学现代中药国家重点实验室;
MDA-MB-231(人三阴性乳腺癌细胞株),来自于天津中医药大学现代中药国家重点实验室;
4T1-luc(带有荧光素酶的小鼠乳腺癌细胞株),来自于天津中医药大学现代中药国家重点实验室。
1.2细胞培养方法
采用含有10%FBS的DMEM高糖培养基,在37℃、5%CO2培养箱中对上述细胞进行常规培养,选择第3-9代细胞,融合至80%~90%时,用0.25%胰酶消化1min左右,用于后续实验。
1.3样品制备
将黄夹次乙苷用DMSO配置成10-1mol/L、10-2mol/L、10-3mol/L的溶液,并分成10μL/管,置于-20℃保存。取10-2M母液,用含有1%CS-FBS的无酚红培基进稀释成10-3M、10-4M、10- 5M、10-6M、10-7M、10-8M、10-9M的样品溶液。1.4高内涵细胞成像系统检测黄夹次乙苷对不同细胞活性的影响
用1%的FBS将上述1.2中的细胞接种于96孔细胞培养板中,每孔8000个细胞,贴壁培养24h后,加入1.3中不同浓度黄夹次乙苷样品(10mmol/L、1mmol/L、100nmol/L、10nmol/L、1nmol/L)孵育24h后将药吸出,用Hoechst 33342(1μg/L)对细胞核进行染色,37℃孵育30min,1×PBS清洗3次。在高内涵分析仪器下每孔选取中间5个视野,10倍物镜进行拍照,计算各孔内细胞的个数。采用SPSS16.0统计软件对各组数据进行单因素方差分析(n=3),最小显著差异水平设定为*P<0.05,**P<0.01,并用Graph Pad Prism 6作统计图,如图1所示,分别为MCF-7、Hela、U2OS、A2780、SN-N-SH、hy926、MYO、4T1和MDA-MB-231对药物黄夹次乙苷的IC50(被抑制一半时抑制剂的浓度)曲线。各细胞的IC50值如表1所示。

由图1及表1可见,黄夹次乙苷对A2780、Hela、MCF7、SK-N-SH、U2OS这五种细胞有显著性抑制增殖的作用,同时对正常细胞(EA.hy926、MYO)无显著作用,对4T1和MDA-MB-231无体外抑制作用。IC50值如表1所示,提示黄夹次乙苷抑制细胞增殖具有选择性,可能对人源肿瘤细胞的杀伤作用显著性强于正常细胞,黄夹次乙苷可能是一个相对安全的抗肿瘤药物。
1.5 CCK-8试剂盒检测黄夹次乙苷对不同细胞活性的影响
用1%的FBS将上述1.2中的细胞接种于96孔细胞培养板中,每孔8000个细胞,贴壁培养24h后,加入1.3中不同浓度黄夹次乙苷样品(10mmol/L、1mmol/L、100nmol/L、10nmol/L、1nmol/L)孵育24h。参照CCK-8试剂盒说明书,每孔加10%CCK-8溶液,37℃培养箱孵育2h,应用3多功能读板机检测450nm波长各孔吸光度值。结果如图2和表2所示,图2为MCF7、MDA-MB-231、Hela、A2780、SK-N-SH、U2OS、hy926、MYO和4T1对药物黄夹次乙苷的EC50曲线。CCK-8试剂盒检测得到的结果与高内涵细胞成像分析结果一致,黄夹次乙苷对A2780、Hela、MCF7、SK-K-SH、U2OS有显著的增殖抑制作用,对4T1无体外抑制作用。

用1%的FBS将上述1.2中的细胞接种于96孔细胞培养板中,每孔8000个细胞,贴壁培养24h后,对照组给DMEM,给药组给含黄夹次乙苷样品浓度分别为100nmol/L、10nmol/L、1nmol/L的DMEM。每组3个复孔,细胞于37℃、5%CO2、饱和湿度条件培养,作用达24小时后,进行免疫荧光染色:用荧光染料Alexa488(绿色)标记EDU,标识出S期细胞;用磷酸化组蛋白PHH3及其荧光染料Alexa647标记(红色),标识出M期细胞;用荧光染料Hochest33342(蓝色)标识出所有细胞核,决定G1期与G2期细胞阈值(G1期细胞染色体为二倍体期,而G2期为有丝分裂期,细胞内包含两套完整的二倍体染色体,所以其G2期细胞荧光表达量是G1期荧光表达量的二倍,以此设定阈值范围,从而区分G1与G2期)。在高内涵分析仪器实验设置中每孔选取中间13个视野,均用10倍物镜进行拍照,选取三个通道(Alexa647、Alexa488和Hoechst33324),曝光时间分别为3000ms、1000ms和200ms,计算被不同荧光染料标记的细胞核的个数,运用高内涵分析软件计算被不同抗体或染料标记的细胞核数及被EdU标记的细胞核荧光强度,用均数±标准差表示,采用SPSS16.0统计软件对各组数据进行单因素方差分析,最小显著差异水平设定为*P<0.05,**P<0.01,***P<0.001。并用Graph Pad Prism 6作统计图,结果如图3~4所示。
如图3和4所示U2OS细胞在不同细胞周期的细胞数占整个细胞周期细胞数的比值:在细胞周期G0/G1期时,与对照组比,含有100nM和10nM黄夹次乙苷的给药组细胞所占的比例显著下降(P<0.01),含有10nM和1nM黄夹次乙苷的给药组细胞所占的比例无统计学差异;在细胞周期S期时,与对照组比较,含有100nM黄夹次乙苷的给药组细胞所占的比例显著上升(P<0.01),10nM和1nM的黄夹次乙苷细胞所占的比例无统计学差异;在细胞周期G2期和M期时,与对照组比较,各给药组细胞所占的比例无统计学差异。即100nM的黄夹次乙苷能显著诱导U2OS细胞发生细胞周期S期阻滞(P<0.01)。该结果与100nM浓度黄夹次乙苷显著抑制细胞增殖的结果相呼应。
2.流式细胞术方法检测黄夹次乙苷对骨肉瘤细胞的细胞周期的影响
选用第3~9代U2OS细胞,用含有10%FBS的DMEM高糖培养基,在37℃、5%CO2培养箱中进行常规培养,待融合至80%左右时,换1%的FBS同步24h,对照组给DMEM,给药组给含黄夹次乙苷样品浓度分别为100nmol/L、10nmol/L、1nmol/L的DMEM,孵育24h;收集细胞,数目约为1×106~5×106个/mL,1000r/min离心3min,弃去培养液;用PBS洗涤1次,离心去PBS,加入冰预冷的70%v/v乙醇,4℃固定12小时以上;离心弃去固定液,加3mL PBS重悬5min,500~1000r/min离心5min,弃去PBS;用1mL PI染液染色,4℃避光30min。
流式细胞仪检测:PI用氩离子激发荧光,激光光波波长为488nm,发射光波波长大于630nm,产生红色荧光分析PI荧光强度的直方图;
在分析PI荧光的直方图时,先用门技术排除成双或聚集的细胞以及发微弱荧光的细胞碎片,在PI荧光的直方图上,凋亡细胞在G1/G0期前出现一亚二倍体峰。如以G1/G0期所在位置的荧光强度为1.0,则一个典型的凋亡细胞样本其亚二倍体峰的荧光强度为0.45,可用鸡和鲑鱼的红细胞的PI荧光强度做参照标准,两者分别为0.35和0.7,可以确保在两者之间的不是细胞碎片而是完整的细胞。结果如图5、图6所示。
如图5、图6所示:U2OS细胞在不同细胞周期的细胞数占整个细胞周期细胞数的比值:在细胞周期G0/G1期时,与对照组比,含有10nM和100nM黄夹次乙苷的给药组细胞所占的比例显著下降(P<0.01),含有1nM黄夹次乙苷的给药组细胞所占的比例无统计学差异;在细胞周期S期时,与对照组比较,含有100nM黄夹次乙苷的给药组细胞所占的比例显著上升(P<0.01),10nM和1nM的黄夹次乙苷细胞所占的比例无统计学差异;在细胞周期G2期和M期时,与对照组比较,各给药组细胞所占的比例无统计学差异。即100nM的黄夹次乙苷能显著诱导U2OS细胞发生细胞周期S期阻滞(P<0.01)。该结果与100nM浓度黄夹次乙苷显著抑制细胞增殖的结果相呼应。
实施例3
本发明实施例提供了黄夹次乙苷作为抗骨肉瘤药物对骨肉瘤细胞凋亡的影响。
1、流式细胞术检测黄夹次乙苷对U2OS细胞凋亡的影响
选用第3~9代U2OS细胞,用含有10%FBS的DMEM高糖培养基,在37℃、5%CO2培养箱中进行常规培养,待融合至80%左右时,换1%的FBS同步24h,对照组给DMEM,给药组分别给含黄夹次乙苷样品浓度分别为100nmol/L、10nmol/L、1nmol/L的DMEM,孵育24h;吸掉培养基上清,加入适量PBS润洗,轻轻摇晃细胞培养器皿,让PBS覆盖整个器皿表面,之后将PBS尽可能吸干净。
加入适量胰酶,37℃孵育,每1分钟左右时取出一次,轻轻摇晃细胞培养器皿或轻拍细胞培养器皿侧壁,观察细胞是否顺利漂起。待细胞能顺利漂起时,加入所加胰酶3倍体积的培养基(含10%FBS)终止消化反应。1000rpm离心5分钟,将上清尽可能的去除干净。
用binding buffer重悬细胞沉淀,置于50-55℃热水中,作为待测骨肉瘤细胞。
空白对照(Control):取未处理、不染色的细胞来初步调电压。
Annexin V单染管:取上述待测骨肉瘤细胞,用Annexin V单染。
核酸单染管:取上述待测骨肉瘤细胞,用PI染料单染。
阴性对照(Negative Control):按上述方法制备不含黄夹次乙苷的细胞悬液作为阴性对照样品,Annexin V和PI双染,用来画门。
阳性对照(Positive Control):取上述待测骨肉瘤细胞,Annexin V和核酸染料双染。
细胞重悬在100μL binding buffer中(细胞数一般为105个),按照说明书加入适量试剂。
室温避光孵育15分钟。
在染色样品中加入400μL binding buffer后放置在冰上,并在1天之内使用Attune NxT流式细胞仪检测。
流式细胞术检测U2OS细胞凋亡的结果如图7、图8所示,10nM和100nM的黄夹次乙苷可能诱导U2OS细胞凋亡。
2、通过Western blot分析研究黄夹次乙苷对人骨肉瘤细胞U2OS相关凋亡蛋白表达的影响
结果如图9所示,以肌动蛋白(β-Actin)为内参,与空白对照组比较,10nM与100nM的黄夹次乙苷促凋亡蛋白Bax蛋白表达有显著性增加(P<0.05,P<0.05),1nM的黄夹次乙苷无统计学差异;抑凋亡蛋白Bcl-2蛋白表达有显著性减少(P<0.05,P<0.01),1nM的黄夹次乙苷无统计学差异;Caspase-3活化的与未活化的比值有显著性增加(P<0.01,P<0.01),1nM的黄夹次乙苷无统计学差异。
Western blot结果证明,与空白对照组比,10nM与100nM的黄夹次乙苷能促进促凋亡蛋白Bax的表达,抑制抑凋亡蛋白Bcl-2的表达,激活凋亡执行蛋白Caspease-3。
实施例4
本发明实施例提供了黄夹次乙苷作为抗骨肉瘤药物对骨肉瘤细胞迁移能力的影响。
1、黄夹次乙苷对抑制骨肉瘤细胞(U2OS)迁移能力研究
U2OS细胞进行体外划痕实验。如图10所示,100nM与10nM的黄夹次乙苷显著抑制U2OS细胞迁移。
U2OS细胞不同时间点迁移率折线图11所示,在12h时间点,与对照组比,100nM与10nM的黄夹次乙苷显著抑制U2OS细胞迁移(P<0.05);1nM的黄夹次乙苷抑制U2OS细胞迁移无统计学差异;在24h时间点,与对照组比较,100nM与10nM的黄夹次乙苷显著抑制U2OS细胞迁移(P<0.01),1nM的黄夹次乙苷抑制U2OS细胞迁移无统计学差异。
实施例5
本发明实施例提供了黄夹次乙苷作为抗乳腺癌药物抑制小鼠乳腺癌生长的作用。
采用ICR小鼠,18只,4-5周龄,雌性,购于中国食品药品检定研究院实验动物资源研究所,SPF级。室温饲养,12h交替照明,光照时间8:30a.m-8:30p.m。动物的使用经天津国际生物医药联合研究院动物伦理委员会批准,符合伦理审查要求。
实验前将黄夹次乙苷固体粉末用DMSO配成500mg/mL的母液,分装到4mLEP管中,5.5μL/管,-20℃储存。
将处于对数期生长的小鼠乳腺癌细胞用PBS稀释成5×107/mL的细胞悬液,按5×106个/只的接种量,接种于裸鼠的腋下皮下,待其生长到300mm3(接种肿瘤细胞后第六天),按肿瘤体积的大小将小鼠随机分为三组,即黄夹次乙苷高剂量组(1mg/kg)、黄夹次乙苷低剂量组(0.5mg/kg)和空白对照组(含0.2%DMSO的生理盐水),每组各6只,每天腹腔注射给药1次,并观察小鼠的状态。给药时长15天,在整个治疗期间每2天测量一次小鼠体重及肿瘤体积,并使用以下公式计算肿瘤体积:肿瘤的体积=长*宽的平方/2。
由于黄夹次乙苷为强心苷类化合物,因此在接种后第19天,三溴乙醇麻醉,心电仪检测小鼠的心功能,检测黄夹次乙苷的安全性。第20天摘眼球取血,小鼠脱颈椎处死,取肝、心、脾、肺、肾、胸腺称湿重。取肿瘤,称湿重、拍照,然后放入液氮中速冻2h后转入-80℃冰箱保存,用作后续实验考察。
由图12~14可见,与空白对照组相比,黄夹次乙苷高剂量组能显著抑制肿瘤生长(P<0.01),但三组4T1肿瘤模型小鼠的体重随时间变化无统计学差异。三组小鼠的心、肝、脾、肺、肾、胸腺及脑的湿重与其自身体重的比值如图15所示,较对照组比,高剂量及低剂量给药组小鼠的心、肝、脾、肺、肾、胸腺及脑的湿重与其自身体重的比值都无统计学差异,说明黄夹次乙苷对肿瘤的选择性和安全性。
小动物心电仪检测的三组小鼠的心电图如图16所示,心功指标如表3所示,与对照组比,各项心功指标无统计学差异。
实施例6
本发明实施例提供了黄夹次乙苷作为抗乳腺癌药物对肿瘤血管新生能力的影响。
1、黄夹次乙苷对抑制血管内皮细胞小管形成能力研究
选用EA.hy926(人脐静脉细胞融合细胞,购买自中国科学院上海生命科学研究院细胞资源中心),用含有10%FBS的DMEM高糖培养基孵育,置于37℃,5%CO2培养箱进行常规培养。选择第3~6代细胞,融合至80%~90%时,用0.25%胰酶消化1min左右,用于后续实验。
将消化后的EA.hy926细胞常规接种于75cm2培养瓶中,贴壁培养12h后,换成无血清培养基饥饿24h。BD基质胶提前4℃过夜解冻,200μL灭菌枪头、96孔板预冷。于冰盒上进行操作,将基质胶迅速加入各孔底部中间位置(45μL/孔),剧烈震荡使基质胶铺满整个孔底,平放于4℃冰箱静置15min,室温离心2500rpm,10min后,于37℃、5%CO2培养箱中平衡30min。胰酶消化收集细胞,细胞计数。以每孔1.5×104个细胞的密度接种于96孔板中,空白对照组为含0.1%FBS的培养基,干预组的细胞培养基中加入100nmol/L的黄夹次乙苷,阳性对照组加50ng/mL血管内皮生长因子(VEGF),分别于37℃、5%CO2培养箱中培养16h。在高内涵分析仪器下每孔选取中间9个视野,用10倍物镜进行拍照,如图17所示。
计算各孔内血管网眼结构的个数,用SPSS16.0统计软件对各组数据进行单因素方差分析(n=3),最小显著差异水平设定为*P<0.05或**P<0.01。如图18所示,与空白对照组相比,100nM的黄夹次乙苷显著抑制内皮细胞血管形成(P<0.01),50ng/mL的VEGF能显著促进内皮细胞血管形成(P<0.01)。
由图17和图18的结果可见,100nM的黄夹次乙苷能显著抑制内皮细胞血管形成,提示黄夹次乙苷可能有抑制肿瘤血管新生的活性。
2、黄夹次乙苷对血管内皮细胞迁移能力的影响
EA.hy926细胞进行体外划痕实验。如图19和20所示,100nM的黄夹次乙苷显著抑制内皮细胞迁移。
以不加药的1%FBS的对照组作为阴性对照,内皮细胞不同时间点迁移率折线图如图20所示,在12h时间点,与对照组比,100nM的黄夹次乙苷显著抑制内皮细胞迁移(P<0.05);10nM和1nM的黄夹次乙苷抑制内皮细胞迁移无统计学差异,在24h时间点,与对照组比较,100nM的黄夹次乙苷显著抑制内皮细胞迁移(P<0.01),10nM和1nM的黄夹次乙苷抑制内皮细胞迁移无统计学差异。
图19和图20的实验结果与血管新生实验结果相一致,提示黄夹次乙苷可能具有抑制肿瘤血管新生的潜能。
3、黄夹次乙苷对抑制小鼠体内肿瘤血管新生能力研究
肿瘤接种模型小鼠在分组给药第16天(详细造模及给药过程见实例5),三溴乙醇麻醉,桡动脉给造影剂,micro-CT检测肿瘤周围血管的分布状况。
图21为micro-CT扫描的4T1肿瘤模型小鼠的肿瘤图片,圆圈区域是小鼠皮下的肿瘤,图22为ImageJ提取的血管分布图,圆圈区域是小鼠皮下肿瘤上的血管分布情况。图23是分支血管的数目的柱状图。可以直观的看到肿瘤周围的血管分布情况,可见黄夹次乙苷高剂量组肿瘤周围的血管分布明显比溶剂对照组明显稀疏,分支血管的数目明显少于溶剂对照组。
实验例7
本实验例提供了黄夹次乙苷作为抗乳腺癌药物的抗肿瘤免疫作用机制研究,用常规RNA提取方法对实施例5中冰箱保存的肿瘤进行RNA提取,将提取所得RNA进行肿瘤基因的考察,检测黄夹次乙苷抑制小鼠乳腺癌作用机制。
1、实验方法
1.1样品采集和准备
在1%琼脂糖凝胶上监测RNA降解和污染:使用NanoPhotometer分光光度计(IMPLEN,CA,USA)检查RNA纯度,使用2.0Flurometer中的RNAAssay Kit测量RNA浓度(Life Technologies,CA,USA),使用Bioanalyzer 2100系统的RNA Nano6000Assay Kit(Agilent Technologies,CA,USA)评估RNA完整性。
1.2转录组测序的文库制备
每个样品用1μgRNA作为RNA样品制备的输入材料。使用UltraTMRNA文库制备试剂盒(NEB,USA)产生测序文库,并将索引代码添加到每个样品的属性序列中。最后,纯化PCR产物(AMPure XP系统),并在Agilent Bioanalyzer 2100系统上评估文库质量。
1.3聚类和测序
使用TruSeq PE Cluster Kit v3-cBot-HS(Illumia)在cBot ClusterGeneration System上进行索引编码样本的聚类。在簇生成后,文库制备物在IlluminaHiseq平台上测序,并产生125bp/150bp的配对末端读数。
1.4数据分析
a质量控制
fastq格式的原始数据(原始读取)首先通过内部perl脚本处理。在此步骤中,通过删除包含适配器的读取,包含ploy-N的读取和从原始数据中读取的低质量读取来获得干净数据(干净读取)。同时,Q20,Q30和GC含量计算了干净的数据。所有下游分析均基于高质量的清洁数据。
b读取映射到参考基因组
使用Hisat2 v2.0.4构建参考基因组的索引,并将配对末端清洁读数与参考基因组比对。选择Hisat2作为映射工具。
c基因表达水平的量化
用HTSeq v0.9.1计录映射到每个基因的读数,然后基于基因的长度计算每个基因的FPKM,并将读数计数映射到该基因。
d差异表达分析
使用DESeq R包(1.18.0)进行两个条件/组的差异表达分析(每个条件两个生物学重复)。使用Benjamini和Hochberg的方法调整所得的P值以控制错误发现率。由DESeq发现的具有整的P值<0.05的基因被指定为差异表达的。
e(对于没有生物学重复的DEGSeq)在差异基因表达分析之前,对于每个测序文库,通过edgeR程序包通过一个缩放归一化因子调整读取计数。使用DEGSeq R包(1.20.0)进行两种条件的差异表达分析。使用Benjamini&Hochberg方法调整P值。将校正的P值0.005和log2(倍数变化)设定为显着差异表达的阈值。
f通过IPA(Ingenuity Pathway Analysis)软件实施差异表达基因的分析。
2、实验结果
将肿瘤组织进行转录组分析,如图24所示,其中,X轴表示不同样品中基因表达的变化,Y轴表示基因表达差异的统计学显着性,右上方区域中为上调的基因,左上方区域中为下调的基因。由图24中可以看出,空白对照组和给药组差异表达基因共102个,其中上调55个,下调47个。然后对这102个基因用IPA分析软件进行分析,从图25中可以看到,对照组和给药组差异基因的差异程度,两组基因颜色差异越大表示基因差异越大。
在图26的十条通路中,第一条为树突状细胞(DCs)成熟通路,第二条为肾上腺髓质信号通路,第三条为自然杀伤细胞信号通路,第四条为结直肠癌转移信号通路,第五条为Ephrin A信号通路,第六条为eNOS信号通路,第七条为通过JAK1/JAK3作用的γ-链细胞因子信号通路,第八条为巨噬细胞,成纤维细胞和内皮细胞在类风湿性关节炎中的作用通路,第九条为活化T细胞核因子在免疫反应中的作用途径,第十条为瘦素在肥胖中的作用通路。从图26中可以看出有4条通路与免疫紧密相关。在图27中的十条通路中,第一条为辅助T细胞分化通路,第二条为激酶介导的免疫应答通路,第三条为糖皮质激素受体信号通路,第四条为巨噬细胞和T辅助细胞通过IL-17A和IL-17F产生相关细胞因子信号通路,第五条为模式识别受体在细菌和病毒识别中的作用通路,第六条为高迁移率族蛋白B1(HMGB1)信号通路,第七条为哮喘炎症通路,第八条为肠上皮细胞通过IL-17A和IL-17F产生相关细胞因子通路,第九条为白介素15(IL-15)信号通路,第十条为激活T辅助细胞1和T辅助细胞2信号通路。从图27中可以看出,前十条通路中有80%跟免疫通路紧密相关。空白对照组和给黄夹次乙苷组差异表达基因的上游调控基因进行分析发现P值最高的通路为辅助T细胞分化通路,辅助T细胞分化在免疫反应中为中间过程,进一步证明黄夹次乙苷通过激活免疫途径抑制4T1乳腺癌。此外,对Gene Card里能够识别的差异基因进行分析,如图28所示,肿瘤模型空白对照组和肿瘤模型给药组差异基因在肿瘤和免疫相关靶点中所占的比例为73%。
本研究对空白对照组和给黄夹次乙苷组差异表达基因进行分析发现最核心值最高的通路为树突状细胞(DCs)成熟通路,树突状细胞是机体功能最强的专职抗原递呈细胞,它能高效地摄取、加工处理和递呈抗原,未成熟DCs具有较强的迁移能力,成熟DCs能有效激活初始型T细胞,处于启动、调控、并维持免疫应答的中心环节。DCs与肿瘤的发生、发展有着密切关系,大部分实体瘤内浸润的DCs数量多则患者预后好。此分析结果同样说明黄夹次乙苷通过激活免疫途径抑制4T1乳腺癌。利用RT-PCR方法对在DCs成熟通路中与肿瘤免疫相关的差异基因进行分析验证,结果发现黄夹次乙苷可以明显上调FCGR3A和HLA-DRB1,下调FGFR4,如图29所示,该结果与转录组分析结果一致。此外,我们对肿瘤免疫的标志性基因PD-1的转录水平进行了分析,发现黄夹次乙苷明显下调了PD-1的表达,更进一步证明了黄夹次乙苷通过肿瘤免疫途径而抑制小鼠体内4T1乳腺癌细胞的增殖。
