Abstract
本发明属于医学生物材料领域,具体公开了一种颗粒酶测定的近红外分子探针,此外还公开了所述的分子探针的制备方法以及将其用于颗粒酶近红外测定的方法。本发明所述的测定探针,当在颗粒酶B存在时,其能特异性的催化水解探针中的片段,再经分子化的环化反应,释放出荧光团。本发明制备出来的分子探针对颗粒酶B具有高选择性、灵敏度高;且制备方法简单合成条件不苛刻,操作方便。可实现对细胞毒性T细胞内颗粒酶B的原位检测,具有免洗、无荧光背景干扰等优势。
Description
技术领域
本发明属于医学生物材料领域,特别涉及一种快速检测细胞毒性T细胞分泌的颗粒酶B 的近红外荧光探针的制备,以及通过监测颗粒酶B预测移植排斥反应。
背景技术
细胞毒性T淋巴细胞(CTL)可以通过多种机制识别和杀伤病毒感染细胞和肿瘤细胞。其中最主要途径是向靶细胞分泌颗粒成分,释放致命的溶细胞分子,这些颗粒含有颗粒酶原及其它蛋白酶原,包括穿孔蛋白。由于CTL细胞与靶细胞结合(经靶细胞表面的CTL受体和MHC分子的抗原结合),颗粒的内容物释放,颗粒酶进入了靶细胞,穿孔蛋白进入了靶细胞通过在细胞膜的聚合形成了靶细胞膜的小孔,使细胞膜穿孔,最后穿孔蛋白使颗粒酶的膜穿孔引起颗粒酶的释放。现已发现CTL可以分别产生至少11种颗粒酶:颗粒酶A-颗粒酶M.在人类中发现了五种,即颗粒酶A(GzA),颗粒酶B(GzB),颗粒酶H(GzH),颗粒酶K(GzK)和颗粒酶M(GzM)。其中,GzB是诱导细胞凋亡最活跃的。在细胞浆内,颗粒酶B能通过几种不同的途径激起细胞的死亡,首先激起caspases的链锁反应,引起靶细胞DNA降解活动,然后裂解。
在组织和器官衰竭的晚期,移植已成为最有效的治疗方法。然而,移植排斥仍然是移植后最关键的并发症,也是影响移植物功能和存活的主要因素。当前用于检测移植排斥的金标准是组织活检,但其提供的预测信息有限,并且组织活检是一种侵入性工具,可能引起严重的并发症。与移植排斥相关的生物标志物的无创检测可以帮助临床医生在临床上明显的器官功能障碍之前采取个性化治疗并改善预后。许多研究表明,外周血,移植组织和受体尿中的 GzB mRNA水平较高。在急性排斥反应评估中,使用GzB做标志物可能是更好的选择,以指导异体移植活检的执行和更早的治疗干预。综上所述,免疫排斥部位GzB浓度的增加表明了移植排斥反应的发生,GzB可以用作急性排斥反应的早期预测指标。
目前,已经开发了一些用于动态监测和成像细胞内GzB的荧光传感器。Packard和Choi 等设计的探针,可基于荧光蛋白变异体之间的共振能量转移(FRET)报告蛋白酶活性。还有研究者开发了GzB的PET示踪剂(68Ga-NOTA-GZP)来监测肿瘤的免疫。然而这些显像剂的发射波长阻碍了它们在体内生物成像中的应用。
发明内容
为解决现有GzB测定存在的不足,本发明第一目的在于,提供了一种利用近红外测定 GzB的全新测定思路,并提供了一种可实现GzB近红外测定的全新结构的分子探针。
本发明第二目的在于,提供了一种所述的近红外分子探针的制备方法。
本发明第三目的在于,通过了一种所述的近红外分子探针在用于制备测定GzB的测试试剂中的应用。
本发明第四目的在于,提供了一种可测定GzB的测试试剂。
一种近红外分子探针,为具有式1结构式的化合物:
本发明开拓性地提出了一种基于近红外手段对颗粒酶B进行测定的全新思路;并创新地提供了一种可实现颗粒酶B近红外测定的全新结构的小分子探针化合物。本发明研究发现,该全新的分子探针能够特异地被GzB识别,可实现颗粒酶B的近红外实时检测和成像,不仅如此,还具有干扰小,灵敏度高、稳定性高等优势。
本发明还提供了一种所述的近红外分子探针的制备方法,包括以下步骤:
步骤(1):将具有式2结构式的化合物、丙烯酰卤、缚酸剂进行反应,得到式3中间产物;
步骤(2):将式3中间产物与式4结构化合物反进行反应,即得;
步骤(1)中,所述的丙烯酸卤为丙烯酸氯。
作为优选,优选地,所述的丙烯酸卤的摩尔量不低于理论反应量,优选为理论反应量的 1.0~2.2倍。
作为优选,步骤(1)中,所述的缚酸剂为TEA。
优选地,所述的缚酸剂的摩尔量不低于理论反应量,优选为理论反应量的1.0~2.2倍。
作为优选,步骤(1)中,在保护性气氛下进行反应;所述的保护性气氛例如为氮气或者氩气。
优选地,步骤(1)的反应温度优选为室温,例如为15~40℃。
优选地,步骤(1)的反应溶剂为CH2Cl2。
本发明步骤(1)中,可预先将式2原料和缚酸剂在反应溶剂中搅拌溶解,再添加丙烯酸卤原料,进行酯化反应,随后再进行纯化分离,即得到所述的中间体。
本发明中,可获得的中间体用溶剂溶解后,再加入式4的原料溶液,进行反应。
作为优选,步骤(2)中,溶解中间体的溶剂可以为二甲亚砜(DMSO)或N,N-二甲基甲酰胺(DMF)中的至少一种。
作为优选,所述的式4的原料溶液例如为式4的水溶液,进一步优选为式4的Tris-HCl 缓冲溶液(Ph=7.4)。
作为优选,所述的式3与式4化合物的摩尔比为2~4:1。
作为优选,步骤(2)在保护气氛下进行反应,例如为氮气或者氩气。反应的温度为20~45℃。
本发明优选的制备方法,具体步骤为:
步骤(1):将式2结构式的化合物和三乙胺溶解在有机溶剂中,在惰性气体的保护下加入丙烯酰氯溶液,在室温下得到探针分子中间体(式3)。优选地,所述将式2结构式的化合物和三乙胺溶解在CH2Cl2中,然后加入丙烯酰氯在惰性气体中于室温搅拌,获得分子探针中间体;进一步优选,将式2结构式的化合物加入三乙胺溶解在CH2Cl2中,在惰性气体保护下,加入丙烯酰氯,20~30℃下搅拌,减压缩得到粗固体,用硅胶柱层析,然后用第一有机溶剂为淋洗液进行纯化,获得分子探针中间体。所述第一有机溶剂为二氯甲烷(CH2Cl2)或甲醇(Methanol)。
步骤(2):将所述分子中间体溶于有机溶剂,将式4化合物溶于水中,随后将两者溶液在惰性气体保护下,混合并搅拌反应,获得分子探针。优选地,将所述分子探针中间体溶于第一有机溶剂中,在惰性气体保护下加入到溶解于水中的式4溶液,在35~40℃下剧烈搅拌过夜,获得分子探针;进一步优选,将式4化合物溶解于Tris-HCl缓冲溶液(Ph=7.4)中,然后将溶有中间体的的DMSO溶液逐滴加入上述溶液中。滴加完毕后,反应混合物置于35~40℃下,剧烈搅拌过夜。反应结束后,用三氯甲烷萃取反应后的混合物,以除去没有反应的中间体,再将水层在高真空条件下干燥,得到终产物分子探针。
本发明还提供了一种所述的近红外分子探针在用于制备检测颗粒酶B检测试剂中的应用。
本发明提供了一种全新的利用近红外手段测定颗粒酶B的全新思路,并提供了一种能够实现该全新近红外测定思路的全新结构的分子探针,并进一步研究发现,利用所述的近红外探针,能够实现颗粒酶B的特异识别以及高效测定。本发明研究发现,基于所述的探针的近红外测定,其检测过程简单,无背景荧光干扰,反应快速灵敏,可实现早期预测移植排斥反应的发生。
作为优选,所述的应用,将其用于制备检测细胞毒性T细胞内颗粒酶B检测试剂。
进一步优选,所述的应用,将其用于制备检测移植后的免疫排斥的测试试剂的应用。
本发明还提供了一种检测颗粒酶B的检测试剂,包含所述的近红外分子探针。
优选地,所述的检测试剂为检测细胞毒性T细胞内颗粒酶B检测试剂。
进一步优选,所述的检测试剂为检测移植后的免疫排斥的测试试剂。
本发明克服现有方法存在的上述不足之处,增强探针在活体成像中的应用,优化并设计出一种新型的近红外小分子荧光探针,并将其应用于免疫重建的皮肤移植小鼠模型中免疫排斥反应的早期原位可视化检测。本发明利用off-on型荧光探针,通过酶的特异切割序列原则能够被颗粒酶B识别并切割,从而使原本淬灭的荧光发光,从而达到可视化检测颗粒酶B的目的,确定细胞毒性T淋巴细胞的活化,从而提示移植排斥反应的发生。
本发明中,将式2化合物利用丙烯酰氯封闭淬灭后再与式4化合物进行点击化学合成,获得所述全新结构的探针分子,研究发现,该全新分子不易被其他丝氨酸的酶类识别切割。其识别位点为半胱氨酸与天冬氨酸之间的肽键。当探针与细胞内的颗粒酶B反应,随后发生分子内环化,再释放出近红外荧光团的同时,产生七元环的副产物。从而实现颗粒酶B的无干扰、可视化测定。
本发明所述的测定方法,包括如下步骤:
1)皮肤移植模型:6~8周龄雄性NSG小鼠背部皮下移植人类皮肤组织10mm×10mm,由于NSG小鼠为高度免疫缺陷鼠,缺乏T细胞、B细胞以及NK细胞,在接受异种移植后,不会产生免疫排斥反应。在皮肤建模完成后14天,取小鼠背部移植皮肤及正常皮肤组织标本固定于4%多聚甲醛中,常规脱水,石蜡包埋,5μm切片,苏木素-伊红(HE)染色,光学显微镜下观察细胞形态变化,鉴定是否成功建模。之后,将所有移植小鼠分为实验组和对照组。实验组移植小鼠进行免疫重建,通过尾静脉注射人外周血淋巴细胞1×107个/150μl;对照组小鼠经尾静脉注射等体积的PBS缓冲液。继续观察小鼠背部移植皮肤的色泽湿润度,然后于小鼠移植局部注射近红外荧光探针50μl/只。
2)在注射探针之后的不同时间点,移植小鼠置于LuminaII小动物活体荧光成像仪中,分别拍摄移植小鼠的近红外图像。
3)近红外荧光探针注射入小鼠体内后,由于近红外荧光的背景干扰小,穿透度深,我们能够非常轻松且明显的观察到移植皮肤局部与探针的响应情况,从而判断移植排斥反应的发生。
4)根据荧光结果确定细胞毒性T淋巴细胞的活化,确定排斥反应状态达到指导临床选择免疫治疗方案的目的。
与现有技术相比,本发明具有以下有益效果:
1、本发明首次提供利用近红外手段测定颗粒酶B的测定思路;并提供了一种能够成功实现颗粒酶B的近红外测定的、且具有优异测定效果的近红外小分子探针。
2、本发明所述全新结构的近红外探针,为免洗、无背景荧光干扰off-on型探针,可实现对靶标颗粒酶B的原位监测。此外,探针具有很好的水溶性,而在细胞内当目标底物颗粒酶 B存在时,其能特异性地催化水解、再经过分子内的环化反应,使荧光团失去淬灭剂,恢复该荧光团的荧光和难溶于水的性质,进而实现对活细胞内颗粒酶B活性的高选择、高灵敏的定位检测及高分辨荧光成像分析。显著提高探针在生物复杂环境中的稳定性,大大提高了复杂生物样本或活体内的检测能力;而近红外荧光染料可避免生物发光的干扰,明显提高信背比,增加探针灵敏性。
简而言之,本发明所制备的近红外探针能够实现生物复杂环境中(小鼠皮肤移植模型) 对颗粒酶B的高灵敏度、高特异性的快速原位检测,从而提示排斥反应的发生。

具体实施方式
以下结合实施例对本发明进行进一步的说明,而非限制本发明。除非本申请上下文中另有其他说明,否则本申请中所用技术术语及缩写均具有本领域技术人员所知的常规含义;除非另有说明,否则下述实施例中所用原料化合物均为商购获得。
本发明以移植排斥反应中细胞毒性T淋巴细胞作用于靶细胞的颗粒酶B为研究对象,设计近红外的荧光探针用以快速监测排斥反应中所产生的颗粒酶B,举例验证其在细胞实验以及动物模型中的可行性。按照制备本发明所提到的近红外荧光探针的制备以及基于此探针的皮肤移植排斥反应的检测方法,其具体实施方式如下:
实施例1
近红外荧光探针的设计与合成:该分子探针的合成线路如反应式1所示,

具体制备步骤如下:
(1)化合物2合成:
将化合物1(式2化合物,1.0eqv)和TEA(2eqv)溶解在CH2Cl2中的溶液中,加入丙烯酰氯(2eqv),在25℃下搅拌10分钟后,将反应混合物在减压下浓缩,得到粗固体,将其通过硅胶柱纯化,层析得到最终产物。
(2)化合物2合成:
将Ac-VGPDC-NH 2(式4化合物,1eqv)溶解在4.5mL的10mM Tris-HCl缓冲溶液(pH7.4)中;将0.5mL的溶有式3化合物(反应式1中的化合物2;3eqv)的色谱级DMSO滴加到上述溶液中。添加完成后,将反应混合物置于37℃并搅拌过夜(10~12h)。反应完成后,将反应混合物用氯仿萃取以除去未反应的化合物2,并将水层在高真空下干燥以获得探针。制得的探针的MS图见图5。
实施例2
测定分子探针对颗粒酶B(Granzyme B,GzB)的响应:将重组人颗粒酶B的试剂稀释在分析缓冲液中(在50mM Tris,pH 7.4,100mM NaCl,10%甘油中),设置相同浓度的GzB 探针组(即实施例1中得到的分子探针配置成10μM),在每组中添加具有不同浓度梯度的重组人颗粒酶B(浓度增加),同时,设置不含GzB的空白组(添加等量的磷酸盐缓冲液PBS)。反应后,对混合物进行荧光测量。激发波长为670nm(λem=698nm),激发和发射缝均为5nm,采集发射波长在685至800nm范围内。如图1所示,在吸收波长为698nm处,荧光强度随着颗粒酶B浓度的升高而逐渐增强。
实施例3
测定分子探针对颗粒酶B的选择性:将实施例1中得到的分子探针配置成3mL浓度为 10μM的分子探针溶于PBS缓冲液,然后分别于颗粒酶B,以及同为丝氨酸蛋白酶家族的糜蛋白酶、胰蛋白酶以及同为颗粒酶成分的颗粒酶A进行分析。如图2所示,可以看到在吸收波长为698nm处,只有颗粒酶B产生强烈的荧光信号,其他酶类并没有明显改变。表明该分子探针对颗粒酶B的检测具有高选择性,特别是在波长为680~725nm、优选685~705nm波长下,具有更优的选择性。
实施例4
对实施例1制得的探针的检测能力进行分析:
测定分子探针在活细胞中对颗粒酶B的检测能力:将细胞分为三组,第一组为对照组,正常培养的CD8(+)T淋巴细胞直接与探针共培养;第二组为实验组,CD8(+)T细胞预先接受佛波酯PMA和离子霉素ION刺激(终浓度PMA为10ng/mL,ION为1μg/mL),再与探针分子共培养;第三组为抑制组,CD8(+)T细胞预先接受PMA和ION刺激以及颗粒酶B抑制剂(Z-ADD-CH2Cl购于Abcam公司)共同孵育后,再加入实施例1中得到的探针分子。先将培养好的CD8(+)T淋巴细胞用RPMI-1640培养基接种在底部有10毫米孔的35 毫米共聚焦培养皿中,于37℃恒温细胞培养箱中培养24小时,然后每1mL细胞RPMI-1640 培养基中用10μM实施例1中得到的探针分子(或与等体积的PBS)一起处理4小时。待细胞静止稳定后,使用共聚焦显微镜进行荧光成像分析,采集荧光图像。如图3,可以看到第一列(A列)的对照组细胞是未经处理的,与探针共孵育后没有产生荧光信号。这表明在GzB 不存在时,探针也不会和细胞内的其他蛋白酶类产生响应。而当CD8(+)T淋巴细胞经PMA 和ION刺激后(B列),在探针存在时,细胞内观察到明显红色荧光信号。这是由于细胞被 PMA和ION激活引起了GzB的释放,进而催化水解探针分子释放荧光团(B2)。第三列(C 列)则为加入PMA和ION以及GzB抑制剂一起孵育的细胞,可以看到细胞内的红色荧光信号明显减弱(C2)。这是由于Z-AAD-CH2Cl抑制了GzB的活性,不能对探针起到有效的水解作用。这些实验结果表明,该设计的探针可以特异性的检测细胞内的GzB活性。
实施例5
小分子近红外荧光探针(实施例1制得)用于快速检测移植排斥反应的步骤:选择高度免疫缺陷的NSG小鼠作为研究对象,通过腹膜内注射4%水合氯醛麻醉小鼠,然后在无菌操作台中将小鼠用胶带适当地固定在棉垫上,用复合碘擦拭并消毒,并切掉约10×10mm的背部皮肤。将人的皮肤切成约10×10mm的大小,并浸泡在少量无菌生理盐水中以使其保持湿润。间歇缝合后,取适量无菌纱布压紧包裹。每周对皮肤移植物进行3次视觉和触觉检查,以发现排斥或移植物损伤的迹象。用人外周血单核细胞对重度免疫缺陷的NSG小鼠进行免疫重建,这是非常经典且成熟的人源化小鼠模型。将人外周血单核细胞重悬于PBS中,并通过尾静脉注射到皮肤移植的NSG小鼠中,每只小鼠接受1×107PBMC,注射体积为150μl。对照小鼠接受等体积的PBS。然后观察免疫重建后的皮肤移植物,在PBMCs注射后五天,给小鼠皮下注射了GzB敏感探针(实施例1制得的式1探针)。然后分别于探针给药后2小时和 48小时,麻醉小鼠并进行活体荧光成像。使用前将探针溶于无菌PBS中(终浓度10μmol/L),每只小鼠接受50μL溶液。
根据图4结果,荧光探针通过尾静脉注射入小鼠体内后,我们可以非常直观的检测到荧光信号,根据荧光信号的有无则可推知细胞毒性T细胞是否激活,据此可以提示免疫排斥反应的发生,并可指导临床选择免疫治疗方案。

5.如权利要求2所述的近红外分子探针的制备方法,其特征在于,步骤(1)中,所述的缚酸剂为TEA。
6.如权利要求5所述的近红外分子探针的制备方法,其特征在于,步骤(1)中,所述的缚酸剂的摩尔量不低于理论反应量。
7.如权利要求6所述的近红外分子探针的制备方法,其特征在于,步骤(1)中,所述的缚酸剂的摩尔量为理论反应量的1.0~2.2倍。
8.如权利要求2所述的近红外分子探针的制备方法,其特征在于,步骤(1)中,在保护性气氛下进行反应。
9.如权利要求2所述的近红外分子探针的制备方法,其特征在于,步骤(1)的反应温度优为15~40℃。
10.如权利要求2所述的近红外分子探针的制备方法,其特征在于,步骤(1)的反应溶剂为CH2Cl2。
11.如权利要求2所述的近红外分子探针的制备方法,其特征在于,步骤(2)在保护气氛下进行反应,反应的温度为20~45℃。
12.一种权利要求1所述的近红外分子探针在用于制备检测颗粒酶B检测试剂中的应用。
13.如权利要求12所述的应用,其特征在于,将其用于制备检测细胞毒性T细胞内颗粒酶B检测试剂。
14.如权利要求12所述的应用,其特征在于,将其用于制备检测移植后的免疫排斥的测试试剂的应用。
15.一种检测颗粒酶B的检测试剂,其特征在于,包含权利要求1所述的近红外分子探针。
16.如权利要求15所述的检测颗粒酶B的检测试剂,其特征在于,所述的检测试剂为检测细胞毒性T细胞内颗粒酶B检测试剂。
17.如权利要求16所述的检测颗粒酶B的检测试剂,其特征在于,所述的检测试剂为检测移植后的免疫排斥的测试试剂。
