Abstract

本发明公开了一种以L‑酪氨酸为底物生物合成白藜芦醇的工程菌、构建及应用,属于生物技术领域。本发明为白藜芦醇的安全生物转化提供了简单且有效的实施方法,通过对TAL、4CL和STS模块采取共表达,在细胞内实现了L‑酪氨酸至白藜芦醇的生物合成,并通过表达乙酰辅酶A羧化酶基因和乙酰辅酶A合成酶基因,显著地提高了产物白藜芦醇的含量,该工程菌生物转化白藜芦醇的产量高达5.5‑6g/L。

技术领域
本发明属于生物技术领域,尤其涉及一种以L-酪氨酸为底物生物合成白藜芦醇的工程菌的构建及应用。
背景技术
白藜芦醇,又名芪三酚,是一种非黄酮类多酚有机化合物,是许多植物受到刺激时产生的一种抗毒素,可在葡萄叶及葡萄皮中合成,是葡萄酒和葡萄汁中的生物活性成分。口服容易吸收,代谢后通过尿液及粪便排出。白藜芦醇在植物中主要有4种存在形式:反式白藜芦醇、顺式白藜芦醇、反式白藜芦醇苷和顺式白藜芦醇苷,其中以反式形式为主,且反式白藜芦醇的生物活性高于顺式白藜芦醇。目前已知至少有34科、69属,100种不同的植物可产生白藜芦醇,主要涉及植物的根、茎、叶、花、果实和种子等部位,不同部位其白藜芦醇含量差异较大。天然白藜芦醇的主要来源植物是虎杖和葡萄。
白藜芦醇一种植物次级代谢产物,白藜芦醇已被证明在抗氧化、抗炎、抗癌、雌激素、神经保护、心脏保护、抗动脉粥样硬化、抗衰老、抗糖尿病、抗骨质疏松和减肥中的调节作用,备受人们的青睐,在医药、保健品、化妆品等方面有着广阔的应用前景。尤其是近年来在临床上对细胞癌、乳腺癌、结肠癌、胃癌、白血病等多种肿瘤细胞均有显著抑制作用,所以成为人们研究的热点。目前单纯依靠天然植物提取白藜芦醇的方法已经无法满足人们的需要,化学合成法又因为污染严重和安全性受质疑而推广困难,随着微生物代谢工程与合成生物学技术的进步,在大肠杆菌、酿酒酵母等宿主中设计与构建高效的异源合成途径,结合工程菌株的大规模发酵是实现白藜芦醇等植物源珍稀天然产物工业化生产,可为大规模工业化生产白藜芦醇提供重要支撑。
发明内容
本发明的目的在于提供一种以L-酪氨酸为底物生物合成白藜芦醇的工程菌的构建及应用。该工程菌可以L-酪氨酸为底物生物合成白藜芦醇,产量高,利于白藜芦醇的大规模生产。
以L-酪氨酸为底物生物合成白藜芦醇的工程菌的构建方法,具体包括以下步骤:
(1)利用酶切方法对质粒载体进行线性化处理,回收产物为线性化载体片段;
(2)利用特异性引物分别对L-酪氨酸氨解酶基因、对香豆酰基辅酶A连接酶基因和白藜芦醇合成酶基因进行PCR扩增,回收得到目的片段;
(3)利用DNA连接酶将所述含有L-酪氨酸氨解酶、对香豆酰基辅酶A连接酶和白藜芦醇合成酶编码基因的目的片段与所述线性化载体进行连接、转化,得到重组表达质粒载体-TAL、质粒载体-4CL、质粒载体-STS;
(4)利用前述重组表达质粒载体-TAL、质粒载体-4CL、质粒载体-STS构建重组质粒-TAL-4CL-STS
(5)将重组质粒-TAL-4CL-STS经酶切,线性化后利用转入解脂耶氏酵母Po1f中,培养挑取阳性转化子,获得能够产生白藜芦醇的初始工程菌;
按上述方案,步骤(4)所述的构建重组质粒TAL-4CL-STS的具体步骤为:
分别使用特异性引物扩增hp4d-4CL-XPR2、hp4d-STS-XPR2表达框,并使用OverlapPCR连接,扩增得到hp4d-4CL-XPR2-hp4d-STS-XPR2双重表达框,通过无缝克隆插入线性化质粒pINA1269-TAL,最终得到重组质粒TAL-4CL-STS;
或分别使用特异性引物扩增hp4d-4CL-XPR2、hp4d-TAL-XPR2表达框,并使用OverlapPCR连接,扩增得到hp4d-4CL-XPR2-hp4d-TAL-XPR2双重表达框,通过无缝克隆插入线性化质粒pINA1269-STS,最终得到重组质粒pINA1269-TAL-4CL-STS;
或分别使用特异性引物扩增hp4d-TAL-XPR2、hp4d-STS-XPR2表达框,并使用OverlapPCR连接,扩增得到hp4d-TAL-XPR2-hp4d-STS-XPR2双重表达框,通过无缝克隆插入线性化质粒pINA1269-4CL,最终得到重组质粒pINA1269-TAL-4CL-STS;
按上述方案,上述构建方法还包括:
(6)以人工合成的ACC基因为模板,使用特异性引物进行扩增,酶切连接后构建得到重组质粒-ACC;
或以人工合成的ACC、ACS基因为模板,使用Overlap PCR扩增得到融合基因:ACC-ACS酶切连接后构建得到重组质粒-ACC-ACS;
(7)将重组质粒质粒-ACC或-ACC-ACS用限制性内切酶酶切,线性化后转化入前述能够产生白藜芦醇的初始工程菌,培养挑取阳性转化子,获得能够生成白藜芦醇的工程菌株。
一种以L-酪氨酸为底物生物合成白藜芦醇的工程菌,过表达L-酪氨酸氨解酶、对香豆酰基辅酶A连接酶和白藜芦醇合成酶,同时过表达乙酰辅酶A羧化酶基因,或过表达乙酰辅酶A羧化酶基因和乙酰辅酶A合成酶基因。
按上述方案,所述过表达L-酪氨酸氨解酶、对香豆酰基辅酶A连接酶和白藜芦醇合成酶的方法是将所述L-酪氨酸氨解酶基因、对香豆酰基辅酶A连接酶基因和白藜芦醇合成酶基因全部置于表达质粒的启动子之后进行表达,所述L-酪氨酸氨解酶基因、白藜芦醇合成酶基因序列和对香豆酰基辅酶A连接酶基因通过整合型质粒在解脂耶氏酵母中进行共表达,所述L-酪氨酸氨解酶基因、香豆酰基辅酶A连接酶和藜芦醇合成酶在工程菌株中通过hp4d启动子启动转录。通过整合L-酪氨酸氨解酶、对香豆酰基辅酶A连接酶和白藜芦醇合成酶,利用hp4d启动子使L-酪氨酸氨解酶、对香豆酰基辅酶A连接酶和白藜芦醇合成酶在解脂耶氏酵母中共表达,从而提高了白藜芦醇产物产量,为大规模工业化生产白藜芦醇提供了支撑。

按上述方案,所述的应用方法包括以下步骤:
(3)将工程菌在YPD培养基中培养10-15h,得到种子液,所述种子液的OD600=5-10;
(4)将所述的种子液接入到YPD培养基中,进行发酵培养;
(3)发酵一定时间后添加诱导剂进行诱导表达,诱导至一定时间后,添加底物L-酪氨酸进行生物转化,合成白藜芦醇。
按上述方案,所述YPD培养基(质量百分比)为20g/L葡萄糖为碳源,含酵母提取物10g/L,蛋白胨20g/L;
按上述方案,所述种子液的培养条件为28-40℃,160-230rpm,培养时间为1-3h,优选为2h.;
按上述方案,所述种子液接入YPD培养基的比例为1%-2%,优选为2%。
按上述方案,底物L-酪氨酸的浓度为5-30g/L,优选为20g/L。
本发明采用具有较高的乙酰辅酶A内源水平的解脂耶氏酵母,使用酶偶联法同时催化三个反应体系,L-酪氨酸在L-酪氨酸氨解酶催化下生成对香豆酸、对香豆酰基辅酶A连接酶催化对香豆酸生成对香豆酰基辅酶A,然后白藜芦醇合成酶酶体系下生白藜芦醇;同时利用乙酰辅酶A羧化酶基因和/或乙酰辅酶A合成酶基因,乙酰辅酶A合成酶促进乙酰辅酶A的生成,乙酰辅酶A羧化酶催化乙酰辅酶A生成丙二酰辅酶A,白藜芦醇合成酶以对香豆酰辅酶A为底物结合三分子丙二酰辅酶A合成白藜芦醇,从而大幅度提高产物白藜芦醇的产量。
与现有技术相比,本发明具有如下有益效果:
本发明为白藜芦醇的安全生物转化提供了简单且有效的实施方法,通过对TAL、4CL和STS模块采取共表达,在细胞内实现了L-酪氨酸至白藜芦醇的生物合成,并通过表达乙酰辅酶A羧化酶基因和乙酰辅酶A合成酶基因,显著地提高了产物白藜芦醇的含量,该工程菌生物转化白藜芦醇的产量高达5.5-6g/L。

具体实施方式
实施例1pINA1269-TAL-4CL-STS质粒的构建方法
1.pINA1269-TAL、pINA1269-4CL和pINA1269-STS质粒的构建
(1)以人工合成的RgTAL基因(为模板,以引物RgTAL-F和RgTAL-R进行PCR扩增,回收得到目的片段RgTAL。
(2)以人工合成的Gm4CL基因为模板,以引物Gm4CL-F和Gm4CL-R进行PCR扩增,回收得到目的片段Gm4CL。
(3)以人工合成的AhSTS基因为模板,以引物AhSTS-F和AhSTS-R进行PCR扩增,回收得到目的片段AhSTS。

2.hp4d-4CL-XPR2-hp4d-STS-XPR2双重表达框片段的获得
(1)分别以3种pINA1269-4CL质粒DNA为模板,使用引物Expression Cassette-F1、R1进行PCR扩增,回收获得包含不同4CL基因的3种hp4d-4CL-XPR2 DNA片段。
(2)分别以3种pINA1269-STS质粒DNA为模板,使用引物Expression Cassette-F2、R2进行PCR扩增,回收获得包含不同STS基因的3种hp4d-STS-XPR2 DNA片段。
(3)分别以1种上述hp4d-4CL-XPR2 DNA片段、1种hp4d-STS-XPR2 DNA片段同时作为模板,使用引物Expression Cassette-F1、R2进行PCR扩增,回收获得包含不同4CL、STS基因的3种DNA片段,3个代表DNA片段列举如下:hp4d-Gm4CL-XPR2-hp4d-AhSTS-XPR2、hp4d-At4CL-XPR2-hp4d-VvSTS-XPR2、hp4d-Nt4CL-XPR2-hp4d-PcSTS-XPR2。
3.pINA1269-TAL-4CL-STS质粒的构建
(1)使用限制性内切酶SpeI对上述构建的3种pINA1269-TAL质粒进行酶切,回收酶切产物pINA1269-RgTAL、pINA1269-CmTAL、pINA1269-SaTAL。
(2)分别将酶切产物与双重表达框片段进行无缝连接,获得3种重组质粒:pINA1269-RgTAL-Gm4CL-AhSTS、pINA1269-CmTAL-At4CL-VvSTS、pINA1269-SaTAL-Nt4CL-PcSTS。
上述质粒构建方法中所用引物如下表:

实施例2解脂耶氏酵母工程菌的构建及摇瓶发酵
(1)将实施例1所得的质粒pINA1269-RgTAL-Gm4CL-AhSTS、pINA1269-CmTAL-At4CL-VvSTS、pINA1269-SaTAL-Nt4CL-PcSTS经限制性内切酶NotⅠ酶切,线性化后利用LiAC转化法转入解脂耶氏酵母Po1f中,并涂布到SC-Leu酵母缺陷型培养基上,于30℃培养至长出转化子,挑取阳性转化子,获得能够产生白藜芦醇的初始工程菌株1、工程菌株2和工程菌株3。
(2)将工程菌株1、工程菌株2和工程菌株3进行划线到YPD培养基上培养至长出单菌落,挑取单菌落,接种至种子培养基中,培养15h,得到种子液,所述种子培养基为YPD培养基:20g/L葡萄糖为碳源,含酵母提取物10g/L,蛋白胨20g/L,所述种子液的培养条件为30℃,225rpm,所述种子液的OD600=10;
(3)将2%所述的种子液接入到含50mLYPD培养基的摇瓶中,进行摇瓶发酵培养;
(4)培养2h时间后,添加20g/L底物L-酪氨酸进行生物转化,转化36h结束后利用高效液相色谱(HPLC)检测白藜芦醇的产量。
高效液相色谱检测白藜芦醇:
(1)向1mL上述转化液中加入1mL甲醇进行溶解,得到产物溶解物;
(2)所述产物溶解物经5000rpm离心10min,取上清液;
(3)所述上清液经0.22μm滤膜过滤于棕色液相瓶中,得到待测样品;
(4)高效液相色谱的色谱条件为:色谱柱为C18(250mm*4.6mm,5μm)或等效色谱柱,流动相为72wt%的水和28wt%乙腈,流速为1mL/min,进样量为10μL,柱温箱温度为30℃;
(5)根据白藜芦醇标准品的峰面积,最终计算白藜芦醇的含量,结果见表1。
实施例3表达乙酰辅酶A羧化酶(ACC)增加白藜芦醇产量
1.pINA1311-ACC质粒的构建
(1)以人工合成的CgACC(序列如SEQ ID No.10所述,长度1776bp)、AsACC(序列如SEQ ID No.11所述,长度6699bp)、CaACC基因(序列如SEQ ID No.12所述,长度6693bp)为模板,分别利用CgACC-F和CgACC-R,AsACC-F和AsACC-R,CaACC-F和CaACC-R进行PCR扩增,所述扩增产物经回收后得到CgACC、AsACC、CaACC的目的片段,片段大小分别为1776bp、6699bp、6693bp,所述引物CgACC-F序列5'-CCCCACGTGATGTCTGTTGAAACAAGAAAAATAACCAAAGTGC-3',如,SEQ ID No.47所述,所述引物CgACC-R序列5'-CGGGGTACCTTATTTGATTTCCAATAAAACCACTCCTTTGTTAACAG-3',如SEQ ID No.48所述;所述引物AsACC-F序列5'-CCCCACGTGATGAGCGCGTATGATCAT-3',如SEQ ID No.49所述;所述引物AsACC-R序列5'-CGGGGTACCGCTGCTATGCGCATAGCT-3',如SEQ ID No.50所述;所述引物CaACC-F序列5'-CCCCACGTGATGAGCGAAGAACATGCG-3',如SEQ ID No.51所述;所述引物CaACC-R序列5'-CGGGGTACCGCTGCCGGTATCCAGG-3',如SEQ ID No.52所述。
(2)使用限制性内切酶PmlI、KpnI同时对CgACC、AsACC、CaACC回收产物与pINA1311质粒DNA进行双酶切,连接后得到重组质粒pINA1311-CgACC、pINA1311-AsACC、pINA1311-CaACC。
2.解脂耶氏酵母工程菌的构建及摇瓶发酵

(1)以人工合成的CgACC基因为模板,利用引物CgACC-VaACS-F1和CgACC-VaACS-R1进行PCR扩增,所述扩增产物经回收后得到CgACC的目的片段所述引物CgACC-VaACS-F1序列5'-CCCCACGTGATGTCTGTTGAAACAAGAAAAATAACCAAAGTGC-3',如SEQ ID No.53所述,所述引物CgACC-VaACS-R1序列5'-GAACCACCTCCACCTTTGATTTCCAATAAAACCACTCCTTTGTTAACAG-3',如SEQ ID No.54所述。

Claims (10)

1.以L-酪氨酸为底物生物合成白藜芦醇的工程菌的构建方法,具体包括以下步骤:
(1)利用酶切方法对质粒载体进行线性化处理,回收产物为线性化载体片段;
(2)利用特异性引物分别对L-酪氨酸氨解酶基因、对香豆酰基辅酶A连接酶基因和白藜芦醇合成酶基因进行PCR扩增,回收得到目的片段;
(3)利用DNA连接酶将所述含有L-酪氨酸氨解酶、对香豆酰基辅酶A连接酶和白藜芦醇合成酶编码基因的目的片段与所述线性化载体进行连接、转化,得到重组表达质粒载体-TAL、质粒载体-4CL、质粒载体-STS;
(4)利用前述重组表达质粒载体-TAL、质粒载体-4CL、质粒载体-STS构建重组质粒-TAL-4CL-STS;
(5)将重组质粒-TAL-4CL-STS经酶切,线性化后利用转入解脂耶氏酵母Po1f中,培养挑取阳性转化子,获得能够产生白藜芦醇的初始工程菌。
2.根据权利要求1所述的构建方法,其特征在于:步骤(4)所述的构建重组质粒TAL-4CL-STS的具体步骤为:
分别使用特异性引物扩增hp4d-4CL-XPR2、hp4d-STS-XPR2表达框,并使用Overlap PCR连接,扩增得到hp4d-4CL-XPR2-hp4d-STS-XPR2双重表达框,通过无缝克隆插入线性化质粒pINA1269-TAL,最终得到重组质粒TAL-4CL-STS;
或分别使用特异性引物扩增hp4d-4CL-XPR2、hp4d-TAL-XPR2表达框,并使用OverlapPCR连接,扩增得到hp4d-4CL-XPR2-hp4d-TAL-XPR2双重表达框,通过无缝克隆插入线性化质粒pINA1269-STS,最终得到重组质粒pINA1269-TAL-4CL-STS;
或分别使用特异性引物扩增hp4d-TAL-XPR2、hp4d-STS-XPR2表达框,并使用OverlapPCR连接,扩增得到hp4d-TAL-XPR2-hp4d-STS-XPR2双重表达框,通过无缝克隆插入线性化质粒pINA1269-4CL,最终得到重组质粒pINA1269-TAL-4CL-STS。
3.根据权利要求1所述的构建方法,其特征在于:还包括:
(6)以人工合成的ACC基因为模板,使用特异性引物进行扩增,酶切连接后构建得到重组质粒-ACC;
或以人工合成的ACC、ACS基因为模板,使用Overlap PCR扩增得到融合基因:ACC-ACS酶切连接后构建得到重组质粒-ACC-ACS;
(7)将重组质粒质粒-ACC或-ACC-ACS用限制性内切酶酶切,线性化后转化入前述能够产生白藜芦醇的初始工程菌,培养挑取阳性转化子,获得能够生成白藜芦醇的工程菌株。
4.一种以L-酪氨酸为底物生物合成白藜芦醇的工程菌,过表达L-酪氨酸氨解酶、对香豆酰基辅酶A连接酶和白藜芦醇合成酶,同时过表达乙酰辅酶A羧化酶基因,或过表达乙酰辅酶A羧化酶基因和乙酰辅酶A合成酶基因。
5.根据权利要求4所述的工程菌,其特征在于:所述过表达L-酪氨酸氨解酶、对香豆酰基辅酶A连接酶和白藜芦醇合成酶的方法是将所述L-酪氨酸氨解酶基因、对香豆酰基辅酶A连接酶基因和白藜芦醇合成酶基因全部置于表达质粒的启动子之后进行表达,所述L-酪氨酸氨解酶基因、白藜芦醇合成酶基因序列和对香豆酰基辅酶A连接酶基因通过整合型质粒在解脂耶氏酵母中进行共表达,所述L-酪氨酸氨解酶基因、香豆酰基辅酶A连接酶和藜芦醇合成酶在工程菌株中通过hp4d启动子启动转录。
6.根据权利要求4所述的工程菌,其特征在于:所述L-酪氨酸氨解酶基因、对香豆酰基辅酶A连接酶基因和白藜芦醇合成酶来源于异源酶,所述L-酪氨酸氨解酶基因来源于粘红酵母(Rhodotorula glutinis)(RgTAL),核苷酸序列如SEQ ID No.1所述;铜锈菌(Cupriavidus metallidurans)(CmTAL),核苷酸序列如SEQ ID No.4所述;白色链霉菌(Streptomyces albulus)(SaTAL),核苷酸序列如SEQ ID No.7所述;
所述4-香豆酰辅酶A连接酶基因来源于大豆(Glycine max)(Gm4CL),核苷酸序列如SEQID No.2所述;拟南芥(Arabidopsis thaliana)(At4CL),核苷酸序列如SEQ ID No.5所述;烟草(Nicotiana tabacum)(Nt4CL),核苷酸序列如SEQ ID No.8所述;
所述白藜芦醇合酶基因来源于花生(Arachis hypogaea)(AhSTS),核苷酸序列如SEQID No.3所述;葡萄(Vitis vinifera)(VvSTS),核苷酸序列如SEQ ID No.6所述;虎杖(Polygonum cuspidatum)(PcSTS),核苷酸序列如SEQ ID No.9所述。
7.根据权利要求4所述的工程菌,其特征在于:所述异源乙酰辅酶A羧化酶基因来源于谷氨酸棒状杆菌(Corynebacterium glutamicum)(CgACC),核苷酸序列如SEQ ID No.10所述;蜜环菌(Armillaria solidipes)(AsACC),核苷酸序列如SEQ ID No.11所述;鸡油菌(Cantharellus anzutake)(CaACC),核苷酸序列如SEQ ID No.12所述;优选为谷氨酸棒状杆菌(Corynebacterium glutamicum)(CgACC),核苷酸序列如SEQ ID No.10所述。
所述异源乙酰辅酶A合成酶基因来源于小豆(Vigna angularis)(VaACS),核苷酸序列如SEQ ID No.13所述;辣椒(Capsicum annuum)(CaACS),核苷酸序列如SEQ ID No.14所述;蓝球藻(Bathycoccus prasinos)(BpACS),核苷酸序列如SEQ ID No.15所述,优选为小豆(Vigna angularis)(VaACS),核苷酸序列如SEQ ID No.13所述。
8.权利要求1所述的一种以L-酪氨酸为底物生物合成白藜芦醇的工程菌株在白藜芦醇生物合成中的应用。
9.根据权利要求8所述的应用,包括以下步骤:
(1)将工程菌在YPD培养基中培养10-15h,得到种子液,所述种子液的OD600=5-10;
(2)将所述的种子液接入到YPD培养基中,进行发酵培养;
(3)发酵一定时间后添加诱导剂进行诱导表达,诱导至一定时间后,添加底物L-酪氨酸进行生物转化,合成白藜芦醇。
10.根据权利要求8所述的应用,所述YPD培养基(质量百分比)为20g/L葡萄糖为碳源,含酵母提取物10g/L,蛋白胨20g/L;所述种子液的培养条件为28-40℃,160-230rpm;所述种子液接入YPD培养基的比例为1%-2%;底物L-酪氨酸的浓度为5-30g/L。
