Abstract
本发明涉及基于P53基因获得小型猪肿瘤疾病模型的方法,属于医学动物疾病模型领域。本方法主要包括P53基因BE3 sgRNA的设计与组装,BE3和sgRNA质粒的转染,筛选并获得P53基因关键碱基突变的阳性细胞系,利用体细胞核移植技术构建重构胚,将重构胚移植到代孕母猪体内继续发育后获得P53基因突变仔猪,通过检测克隆仔猪组织中P53基因RNA及蛋白质表达水平来验证P53基因的功能,采用组织病理学及免疫组化法鉴定仔猪组织中的肿瘤;该肿瘤疾病模型可以精准模拟人类P53基因致病突变,对开展人类肿瘤的发生、形成、临床研究以及药物的研发具有重要的意义,为人类肿瘤疾病的研究和治疗提供了可靠的模型。

技术领域
本发明属于医学动物疾病模型领域,具体的说,涉及基于P53基因获得小型猪肿瘤疾病模型的方法。
背景技术
P53基因是迄今为止最为有效的肿瘤抑制基因,大约一半的肿瘤有P53突变。此外,在一些最难治疗的癌症中,至少80%的样本中P53发生突变。为了更好地了解P53基因突变导致肿瘤发生的机制,开发治疗和预防方法,动物模型是可行的肿瘤研究平台。
猪在解剖学和生理学上与人类相似。与实验啮齿类动物相比,构建带有关键基因的基因编辑猪是研究人类疾病的优秀生物医学模型,对提高对人类遗传疾病病因和潜在治疗的理解发挥了重要作用。与人类一样,猪的自发性癌症也很罕见,人类和猪之间的相似性在癌症生物学的分子水平上甚至是保守的。另外猪的主要生理结构、生化指标、代谢特点都与人极为相似,小型猪体形小,更利于实验过程中的操作和管理,相对于猴等动物建立的疾病模型而言,繁殖更快、成本也较低,在生命科学的研究中具有独特的优越性和应用价值。因此,针对癌症特异性基因的基因编辑猪可以用作癌症模型。
虽然之前已经产生了几种类型的P53基因敲除猪,但这些基因敲除猪都依赖于DSB(DNA双链断裂)的产生,这种突变是随机的,不能精确模拟人类P53基因致病突变。因此,为了研究疾病的致病机制并进行基因治疗,构建精准的疾病模型是非常必要的,而且目前还未见报道。
P53基因关键碱基突变小型猪能够精确模拟人类P53基因致病突变,可为研究人类肿瘤的形成和转移提供新的有力工具,对于开发肿瘤药物和制定治疗方案具有重要的指导意义。
BE3基因编辑系统是在CRISPR/Cas9n基础上融合表达胞苷脱氨酶和尿嘧啶糖苷酶抑制蛋白,在细胞中通过sgRNA实现目标序列靶向C-T转化,由于该系统不会产生DSBs(double strand breaks)或依赖模板DNA,具备精准和无损编辑的特点。目前该编辑系统已成功应用于多种动植物中的单核苷酸编辑。单碱基突变是绝大多数人类遗传疾病产生的原因,因此BE3基因编辑系统在构建精准人类疾病模型和基因治疗上展现了强大的技术优势。
但将BE3基因编辑技术用于P53基因关键碱基突变,存在的位置效应会影响基因编辑效率,并且某些碱基的突变可能会出现胚胎致死的问题,严重影响了该技术在制备猪肿瘤疾病模型中的高效应用。
发明内容
针对上述问题,本发明提供了一种基于P53基因获得小型猪肿瘤疾病模型的方法,该方法操作简单、科学合理、可靠性高、且易于实现,为肿瘤的发病机理、基因治疗方法的研究提供了重要的参考。
本发明提供了一种基于P53基因获得小型猪肿瘤疾病模型的方法,采用BE3碱基编辑系统对P53基因进行编辑,获得P53基因编辑猪胎儿成纤维细胞;通过体细胞核移植及胚胎移植,获得P53基因编辑的猪肿瘤疾病模型。
进一步优选,P53基因的sgRNA为Exon4;识别序列分别为
CCTTCTCAGAAGACCTACCCTGG。
进一步优选,Exon4的PCR扩增引物P53-F1/R1的序列为
GGGAAGCACAGACCTATACTGACTC/ATGGAGAGCGAACAGAAGGTCAGAG。
本发明的有益效果:
利用BE3技术突变P53基因关键碱基获得小型猪肿瘤疾病模型,与利用其他诱导方法建立的动物疾病模型相比,本模型的建立效率高、实用性强、可控性高、复制性强、精准性高,可以从根本上阐明和研究肿瘤的致病机理和遗传机制,为肿瘤的“治本”药物研究和基因治疗指明了研究方向;再者,小型猪与小鼠、大鼠、兔等动物相比,小型猪的体重、生理结构上与人极为相似,建立相应的疾病模型指标与人的疾病临床指标更为相近,这为在不同水平上来探讨人类疾病的致病机制以及遗传因素提供了依据,并且小型猪的器官和肿瘤大小与人类也更为相近,更利于在肿瘤诊断、放射治疗等方面的研究。
本发明通过比较不同位点sgRNA的碱基突变效率,获得两个编辑效率较高的sgRNA碱基突变位点,通过转染筛选获得这两个sgRNA碱基突变位点的体细胞,利用体细胞核移植和胚胎移植技术,验证得到其中一个sgRNA编辑的位点会导致胚胎发育停滞。本发明对sgRNA碱基突变位点的限定,在这个参数选择下,利用BE3技术突变P53基因关键碱基获得小型猪肿瘤疾病模型的方法,不仅能够使得敲除周期较短,小型猪发生肿瘤的概率更大。
综上所述,小型猪作为人类的食用动物用于基因编辑实验动物的疾病研究,也符合伦理要求,而且成本低,价格便宜,不易给实验者造成伤害,利用BE3技术突变P53基因关键碱基获得小型猪肿瘤疾病模型对于人类肿瘤的研究和治疗具有重要的参考价值,在未来的医学领域中易于推广使用。

具体实施方式
下面结合实施例对本发明中利用BE3技术突变P53基因关键碱基获得小型猪肿瘤疾病模型的方法做更详细的阐述,以便今后技术人员的理解。
实施例
以滇南小耳猪作为实验动物,采用本发明方法分别构建小型猪肿瘤疾病模型(实施例1、实施例2),实施例1和实施例2不同点见表1;以野生型滇南小耳猪作为对比例。
基于P53基因获得小型猪肿瘤疾病模型的方法(如图1-2所示),具体操作过程按以下步骤进行:
第一步P53基因BE3碱基编辑系统设计、构建及活性检测
1)根据人类P53基因致病突变,在猪P53基因序列第四外显子和/或第五外显子区域设计BE3编辑靶点;
2)组装sgRNA表达质粒pP53-sgRNA;
3)将获得的质粒pCMV-BE3和pP53-sgRNA去内毒素质粒大提,-20℃保存备用;
4)将无内毒素质粒pCMV-BE3/pP53-sgRNA通过脂质体2000转染至滇南小耳猪胎儿成纤维细胞内,转染48h后观察并记录荧光,随后使用流式细胞仪分选出1000个阳性细胞,收集到1.5mL离心管内。离心去上清后用10μL的微量细胞裂解缓冲液重悬并处理,取1μL处理液作为模板,使用P53-F/R为引物进行PCR扩增。取200ng纯化后的PCR产物T7E1酶切,37℃水浴30min,琼脂糖凝胶电泳检测酶切结果。另取5μL经过72℃30min加A处理后连接到pMD18-T载体中,转化涂板后各挑取30-50个细胞克隆,送样测序,通过测序结果分析P53-sgRNA在细胞内的活性和BE3系统的碱基编辑效率。

第二步细胞转染、筛选及P53基因关键碱基突变阳性细胞系的获得
1)转染前3天复苏滇南小耳猪原代胎儿成纤维细胞,利用胰酶消化贴壁细胞,离心收集细胞,收集细胞数为7×105,利用电击缓冲液进行悬浮,同时加入无内毒素质粒pCMV-BE3/pP53-sgRNA,调节使最终体积为700μL,再将混合液放入到0.4cm的电击杯中,选择方波,在电压250V,25ms条件下进行1次脉冲电击,将电击后的细胞悬浮液转移到100mm培养皿中,加入DMEM,继续培养;
2)转染48h后,利用胰酶消化转染后贴壁的细胞,采用极度稀释培养法培养,最终稀释为100μL悬液中细胞的数目为95-105个,稀释培养10-13天细胞即可长成所需的单细胞克隆;
3)提取的各细胞克隆DNA为模板,使用特异性引物进行扩增,将PCR产物通过纯化、加A、TA克隆、测序的方法,以确定各细胞克隆P53基因碱基突变情况,鉴定为双敲的细胞克隆继续培养及冻存,留作体细胞核移植备用;
第三步体细胞核移植及胚胎移植
1)卵母细胞的成熟培养
a、从屠宰场采集猪的卵巢并在4h内运回实验室;
b、挑选直径为3-6mm的卵泡从中抽取卵丘-卵母细胞复合体,放入添加0.1mg.ml-1丙酮酸、0.1mg.ml-1L-半胱氨酸盐酸、10%猪卵泡液、10ng.mL-1EGF的TCM-199成熟培养液滴中,在5%CO2,38.5℃的培养箱中培养38-42h;
2)构建P53基因关键碱基突变的重构胚
a、取P53基因关键碱基突变的成纤维细胞,用含有0.5%FBS的DMEM培养48h,使体细胞处于G0或G1期;
b、卵丘-卵母细胞复合体经体外成熟培养后,用透明质酸酶去除卵母细胞外围的卵丘细胞,将排出第一极体的卵母细胞放入预处理液中处理0.5-1h,在操作液中用显微注射针将极体及其周围胞质一起吸去,然后将P53基因关键碱基突变的成纤维体细胞导入到去核的卵母细胞的卵间隙中;
3)电融合和电激活重构胚
再将每批20枚重构胚胎,移入融合液中清洗三遍,在25V/mm脉冲电压,脉冲时程为20μs的电融合参数下进行1次脉冲重构胚的电融合处理,完成后移入NCSU23培养基中培养2h,再把重构胚在激活液中清洗三遍,然后在150V/mm脉冲电压,脉冲时程为100μs的参数下进行1次电激活,重构胚移入含有2.2μg.mL-1CB的培养液中放入5%CO2、5%O2、38.5℃的三气培养箱中平衡2h;
4)体外培养
将已平衡好的重构胚胎移入到PZM3培养液中,置于三气培养箱中培养48h和7d后分别观察记录重构胚胎的卵裂率和囊胚率,并用Hoechst33342对囊胚染色,测定囊胚的细胞数;
5)选取自然发情的代孕母猪进行胚胎移植,将重构胚移植到其体内继续发育,待代孕母猪妊娠约114天后分娩获得P53基因关键碱基突变的克隆仔猪;
第四步克隆猪P53基因关键碱基突变的验证
仔猪出生后用手术剪刀剪取0.5×0.5cm的耳组织,按照Tissue DNA Kit(OMEGA,D3396-02)的步骤提取各仔猪DNA,使用引物P53-F/R扩增靶点序列,通过T7E1酶切、测序以判断各仔猪P53基因关键碱基是否突变;
第五步P53基因关键碱基突变克隆仔猪组织、器官中肿瘤的鉴定
采用组织免疫学以及免疫组化染色的方法,鉴定小型猪中各组织器官中肿瘤的发生以及形成情况。
实验分析
1.P53基因BE3碱基编辑系统活性检测
表2 P53-sgRNA碱基编辑效率
如图3和表2所示,通过PCR产物T7E1酶切及测序对四个sgRNA进行活性检测,结果显示BE3系统在4个靶点都有编辑活性,突变效率分别为55.8%、60.7%、12.3%和10.0%。选取突变效率较高的P53-sgRNA-Exon4和P53-sgRNA-Exon5进行后续实验。
2.对所得的细胞克隆点进行P53基因关键碱基是否突变的检测
嘌呤霉素筛选后的细胞,稀释培养2周左右,P53-sgRNA-Exon4、P53-sgRNA-Exon5各获得细胞克隆32个、28个,通过PCR产物的TA克隆和Sanger测序,分别鉴定出22个、21个细胞克隆在靶位置发生C>T的转换,其中单等位基因突变的细胞克隆分别有9个、7个,双等位基因突变的细胞克隆分别有13个、14个。(图4)
3.对所得的克隆仔猪进行P53基因关键碱基是否突变的检测
通过基因组DNA提取、PCR扩增、Sanger测序对17头新生仔猪进行鉴定,结果显示17头新生仔猪在靶位置全部发生关键碱基突变,并且编辑位点都是sgRNA-Exon4,编辑效率达到100%。(图5)sgRNA-Exon5编辑的克隆胚胎同样移植6头代孕母猪,全部妊娠失败,由此推断sgRNA-Exon5编辑位点可能导致胚胎死亡。
4.P53基因关键碱基突变型滇南小耳猪表型鉴定
通过荧光定量检测P53基因的表达,结果显示相比野生型滇南小耳猪,P53基因双敲克隆猪在不同组织中P53的mRNA表达均有明显下降。
通过Western Blot和免疫荧光检测P53在蛋白水平的表达,结果显示在野生型滇南小耳猪的肝脏和肺中P53有大量表达,而在P53基因编辑克隆猪的肝脏和肺中没有P53表达。
结果说明P53基因关键碱基的突变导致P53基因功能缺失。(图6)
5.P53基因关键碱基突变型滇南小耳猪组织病理检测
通过组织病理学检测,与野生型猪相比,P53基因关键碱基突变型滇南小耳猪表现出严重的肺部异常。(图7)

综上所述,小型猪作为人类的食用动物用于基因编辑实验动物的疾病研究,符合伦理要求,而且成本低,价格便宜,不易给实验者造成伤害,利用BE3技术突变P53基因关键碱基获得的小型猪肿瘤疾病模型对于人类肿瘤的研究和治疗具有重要的参考价值,在未来的医学领域中易于推广使用。

1.基于P53基因获得小型猪肿瘤疾病模型的方法,其特征在于:采用BE3碱基编辑系统对P53基因进行编辑,获得P53基因编辑猪胎儿成纤维细胞;通过体细胞核移植及胚胎移植,获得P53基因编辑的猪肿瘤疾病模型;
P53基因的sgRNA为Exon4,其识别序列为CCTTCTCAGAAGACCTACCCTGG;所述Exon4的PCR扩增引物P53-F1/R1的序列为GGGAAGCACAGACCTATACTGACTC/ATGGAGAGCGAACAGAAGGTCAGAG。
