Abstract
本发明公开一种具有助眠功能的食品配方及制备方法和应用,所述食品配方原料重量份比包括药食同源提取物1~4份;新食品原料1~3.5份;食品基料1~10份;本发明还公开一种具有助眠功能的食品配方制备方法和应用。本发明提供一种预防失眠所导致的精神紧张、抑郁等症状,同时促进失眠导致的疲劳的快速恢复,迅速和持久的改善睡眠症状的一种具有助眠功能的食品配方及其制备方法和应用。

技术领域
本发明涉及食品、食品生产及应用技术领域,特别涉及一种具有助眠功能的食品配方及制备方法和应用。
背景技术
失眠是指人们对睡眠时间和(或)睡眠质量不满足并影响日间社会功能的一种主观体验。中国睡眠研究会2016年公布的睡眠调查结果显示,中国成年人失眠发生率高达38.2%,超过3亿中国人有睡眠障碍。另外,中国睡眠研究会还发布了《2017年中国青年睡眠现状报告》,对近6万名青年人的调查显示,76%的人表示入睡困难,青年女性更为突出。并且,随着社会竞争的加剧和电子类产品的普及以及社交方式的改变,青年群体睡眠不足和睡眠障碍的人群比例仍在逐渐增加,而青年人是社会的中坚力量,同时也是家庭的支柱,一旦罹患失眠,将会增加社会和家庭负担。另外,大量的人体试验和动物实验证明,失眠等原因所引起的睡眠不足可导致白天无精打采和精力不集中,并且长期失眠会导致学习记忆功能降低[张丹伟,汪周兵,李月峰,等.失眠障碍患者记忆功能与海马结构及功能影像的相关性.精神医学杂志,2019,32(1):33-37.]、抑郁[于蕊,王越.原发性失眠患者失眠与焦虑抑郁及认知功能的相关性研究.饮食保健,2021,(21):2-3.],严重者会导致生物节律的改变及激素分泌的异常,引发精神心理性疾病,并增加高血压、糖尿病、肥胖等慢性疾病的发病风险,严重损害患者的健康。因此,失眠及其由其引起的一系列后续反应已经不单纯是一个生理问题,还是一个普遍存在的社会问题[陈智文.中青年失眠患者中医证型分析.中国卫生标准管理,2021,12(11):109-112.]。
目前对失眠的常规治疗方法包括西医镇静催眠疗法、中医的针灸和中药疗法以及心理疗法等。西药疗法主要是使用镇静催眠药物,但是这类药物如果长期应用会产生依赖性且远期疗效欠佳。而中医药在辨证论治的基础上,通过不同药物和针灸、理疗等物理疗法的组合,则可在调理脏腑功能的基础上,逐渐改变失眠产生的条件,从而长久改善失眠症状[兰济乐,阮叶萍,蒋东晓.基于数据挖掘和网络药理学的失眠中药配方规律及作用机制研究.浙江中医药大学学报,2020,44(12):1230-1238,1241.]。根据中医的基本理论,近年来人们相继申请了一批具有助眠作用的中医药方面的专利,例如具有助眠作用的中药组合物(专利申请号202110032699.X,201811192779.6)、药食同源物质组合物(专利申请号201811277847.9)、固体饮料(专利申请号202110542480.4)、龟苓膏(专利申请号202011455552.3)和红糖(专利申请号202011195918.8)等。但是,上述研究成果和专利大部分属于中药范畴,即使标注为食品形式的固体饮料、龟苓膏和红糖,其原料内也含有超出药食同源和新食品原料的中药成分,因此在推广过程中必将受到剂型和法律法规的限制。
近年来随着社会潮流的发展和网红经济的引导,青年人追求时髦、返璞归真和药食同源的理念已经深入人心,以时尚新颖、功能显著、色香味俱佳的食品形式取代西药和中成药已经形成一种新的趋势和潮流,而膏丸散酊等传统的中药剂型逐渐被冷落。因此,为了迎合青年人的喜好和大面积推广的需要,本发明在现有中医药研究的基础上,全部采用被社会普遍接受的药食同源类物质的提取物和新食品原料类物质为主要功效成分,加上调味剂、赋形剂和其他食品基料,制备为青年人喜闻乐见、几乎无任何成瘾性和毒副作用且助眠效果明显的软糖、饼干、膏方、固体饮料和压片糖果,并取得较大的社会和经济效益。
中医认为,失眠的发生与五脏功能失调有关,大多由于思虑过度,过度疲劳,而至肝阳上亢、阴虚火旺、心肾不交、脾胃不和所致,因此在调理失眠时重点是养心安神、平息肝火、调和脾胃,以达到助眠益智的效果。据此原理,本发明发挥助眠作用的主要功效成分为新食品原料中的杜仲雄花以及γ-氨基丁酸(GABA),药食同源类物品包括酸枣仁、茯苓、百合、莲子、黄精、益智仁、栀子、甘草。
本发明以酸枣仁、茯苓为主要成分,酸枣仁含有酸枣仁皂苷、黄酮、生物碱,具有养肝、宁心、安神的作用,现代研究证明低浓度酸枣仁提取物可以缓解小鼠的焦虑状态,高浓度时能有效催眠小鼠,与地西泮相比,酸枣仁的作用效果稍慢,但作用时间更长久,服用后对患者学习记忆的损害也比较小[刘莹,管彤,梁昊都,等.酸枣仁改善睡眠药理作用及其机制研究.中医药信息,2021,38(3):82-86.]。茯苓主要含茯苓多糖,具有渗湿利水,健脾和胃的作用,与酸枣仁合用能够促进睡眠并保护睡眠不足导致的脏器功能障碍[孙梦娜,叶辉宇,李强明,等.茯苓不同溶剂提取物改善小鼠睡眠作用的比较.现代食品科技,2021,37(6):43-49.]。酸枣仁与茯苓配合,养心安神,健脾利湿,迅速改善患者的心肾不交现象从而改善失眠症状[张华,郭霞珍.从中医"心肾不交"理论论安神剂的选药.世界中医药,2011,6(5):381-384.]。
在此基础上,通过添加新食品原料杜仲雄花强化镇静安神作用。杜仲雄花含有环烯醚萜类、苯丙素类、黄酮类等活性物质,具有抗氧化、抗疲劳和强身健体的作用,近年来的研究更进一步证明杜仲雄花水提物和乙酸乙酯提取物均具有明确的镇静催眠作用[刘少阳,李欣,朱文学,等.杜仲雄花水提物镇静催眠作用的研究.农产品加工·学刊,2010(04):31-33+36;李欣,王卉,朱文学,等.杜仲雄花乙酸乙酯提取物镇静催眠作用研究.食品科学,2009(21):360-363.]。百合性甘微寒,具有养心安神、调理脾胃的功效,主要含磷脂、生物碱、皂苷及多糖,可调节神经系统活动节律并使人产生愉悦感,有助于缓解失眠导致的不良情绪[胡超群,徐德平.百合助眠活性组分的筛选和分析.食品与生物技术学报,2017,36(10):1096-1100.]。莲子健脾和胃,与百合合用镇静安神。益智仁暖肾固精、温脾止泻,现代医学研究证明益智仁水提物能抑制小鼠的自发活动,与镇静药戊巴比妥钠有协同作用,有一定的镇静安眠及对学习记忆改善的作用[林耕,郭屹,斯建勇,等.益智仁水提物的镇静催眠作用及对大鼠学习记忆影响的实验研究.中国中医药咨讯,2011,003(010):34-35.]。栀子主要成分为栀子苷,具有促进睡眠,改善失眠导致的学习记忆障碍的作用[曹静,汪萌芽.栀子苷对睡眠剥夺大鼠学习记忆的影响.安徽医学,2020,41(5):497-500.]。黄精补中益气,养阴润肺生津,强壮筋骨的作用,改善具有认知障碍生物的学习记忆能力,缓解失眠、乏力、健忘等症状[韦妮娜,徐曼曼,高少才,等.黄精治疗认知障碍的研究进展.中国医药导报,2019(30):42-45.],与莲子、百合合用具有补益脾胃、强筋健体、促进失眠导致疲劳的快速恢复作用,而甘草具有调和药性,解百草之毒的作用。
现代医学研究证明以酸枣仁和茯苓为主的复方能够通过提高脑内的γ-氨基丁酸(GABA)、5-HT等抑制性神经递质的含量而发挥催眠作用[李欣坪,牛延菲,王蒙蒙,等.以酸枣仁、茯苓为主的复方固体饮料改善睡眠的作用研究.食品安全质量检测学报,2021,12(11):4453-4458]。对于失眠较严重或时间较长的患者,上述药食同源配方的作用强度可能仍然不够,这时可以添加新食品原料GABA。原卫生部批准的新食品原料GABA以L-谷氨酸钠为原料,经短乳杆菌发酵、纯化、过滤浓缩、结晶、分离,喷雾干燥等工艺而制成,纯度可以达到20%以上且价格适中,添加GABA后可通过外源性补充GABA而增加中枢神经系统的抑制作用,从而起到强化镇静安神和促眠作用[王建峰,任举.γ-氨基丁酸的生理作用与制备方法综述.山东化工,2013,42(2):51-52,55.]。
总之,上述新食品原料和药食同源物质的合用,即可益气安神、补益脾胃,放松心情促进睡眠,又可预防失眠所导致的精神紧张、抑郁等症状,促进失眠导致的疲劳的快速恢复,迅速和持久的改善睡眠症状。
发明内容
本发明的主要目的是提供一种预防失眠所导致的精神紧张、抑郁等症状,同时促进失眠导致的疲劳的快速恢复,迅速和持久的改善睡眠症状的一种具有助眠功能的食品配方及其制备方法和应用。
为实现上述目的,本发明提出一种具有助眠功能的食品配方,所述食品配方原料重量份比包括药食同源提取物1~4份;新食品原料1~3.5份;食品基料1~10份。
所述的药食同源提取物,其原料包括至少两种药食同源中药材,其重量份比为:酸枣仁2~5份、茯苓1~3份、百合2~3份、莲子2~3份、黄精2~4份、益智仁2~3份、栀子2~3份、甘草1~2份;
所述的新食品原料的重量份比为:杜仲雄花1~2份、γ-氨基丁酸0.1~1份;
所述的食品基料包括重量份比为:标准粉1~3份、果葡糖浆2~3份、蔗糖1~2份、明胶3~4份。
所述药食同源中药材和新食品原料其至少有一种组方重量比为:杜仲雄花2份、酸枣仁3份、茯苓2份、百合2份、莲子3份、黄精4份、益智仁2份、栀子3份、甘草2份。
所述具有助眠功能的食品包括软糖或果冻或饼干或固体饮料或压片糖果。

该方法包括以下步骤:
1)制备药食同源和新食品原料提取物;
2)该药食同源和新食品原料提取物的制作方法包括:购买中药饮片,按重量份比例称取杜仲雄花1~2份、酸枣仁2~5份、茯苓1~3份、百合2~3份、莲子2~3份、黄精2~4份、益智仁2~3份、栀子2~3份、甘草1~2份;通过水提的工艺进行提取,制作成食品配方原料。
优选地,所述通过水提的工艺进行提取方法为:先将所述药食同源和新食品原料混合后粉碎至20目,加10倍体积的纯净水浸泡0.5小时;然后加水量分别为原材料体积的10倍或8倍或6倍,煮沸提取3次,提取时间分别为2.0小时、1.5小时和1.5小时,合并三次煎煮液,经减压浓缩至相对比重为1.05~1.10g/ml,经喷雾干燥制成粉剂。
所述具有助眠功能的食品具体呈现形式包括软糖或果冻或饼干或固体饮料或压片糖果。
优选地,所述软糖制作方法包括:将药食同源和新食品原料提取物粉剂的重量比为1~2份,包括由果葡糖浆、蔗糖、明胶制备的食品基料的重量比为6~10份,通过混匀、调色、调味、脱气、浇模、烘干而成的软糖。
优选地,所述压片糖果制作方法包括:将药食同源和新食品原料提取物粉剂的重量比为3~4份,新食品原料γ-氨基丁酸的重量比为0.25~1份,充分混匀后添加适量微晶纤维素、二氧化硅和硬脂酸镁充分混匀后,压制成压片糖果。
本发明另一发明目的是:提供一种具有助眠功能的食品配方在失眠患者干预和对小动物催眠中的应用。
本发明提供技术方案的有益效果是:本发明以青年人喜闻乐见的食品形式推向社会,改善广大失眠症患者的失眠症状并预防失眠所引发的不良反应和并发症,改善失眠症患者的心境并促进失眠导致智力减退和疲劳的恢复,提高该人群的生活质量和工作激情与效率,化解家庭和社会矛盾,以取得较好的社会效益和经济效益。
本发明通过自制药食同源和新食品原料中药提取物一方面可以更好保证功效,另一方面有利于控制成本而在更大范围内进行推广,扩大收益范围。

本实施例,优选地,所述的药食同源提取物,其原料包括至少两种药食同源中药材,其重量份比为:酸枣仁2~5份、茯苓1~3份、百合2~3份、莲子2~3份、黄精2~4份、益智仁2~3份、栀子2~3份、甘草1~2份;
所述的新食品原料的重量份比为:杜仲雄花1~2份、γ-氨基丁酸(简称GABA)0.1~1份;
所述的食品基料包括重量份比为:标准粉1~3份、果葡糖浆2~3份、蔗糖(包括红糖、白糖、砂糖)1~2份、明胶3~4份。

本发明还提供一种具有助眠功能的食品配方制备方法。
该一种具有助眠功能的食品配方制备方法,该方法包括以下步骤:
1)制备药食同源和新食品原料提取物;
2)该药食同源和新食品原料提取物的制作方法包括:购买中药饮片,按重量份比例称取杜仲雄花1~2份、酸枣仁2~5份、茯苓1~3份、百合2~3份、莲子2~3份、黄精2~4份、益智仁2~3份、栀子2~3份、甘草1~2份;通过水提的工艺进行提取制作成食品配方原料。
本实施例,优选地,所述通过水提的工艺进行提取方法为:先将所述食品配方原料混合后粉碎至20目,加10倍体积的纯净水浸泡0.5小时;然后加水量分别为原材料体积的10倍或8倍或6倍,煮沸提取3次,提取时间分别为2.0小时或1.5小时或1.5小时,合并三次煎煮液,经减压浓缩至相对比重为1.05~1.10g/ml,经喷雾干燥制成粉剂。

(1)购买原材料:从中药饮片厂购买符合中国药典标准的中药饮片酸枣仁、茯苓、百合、莲子、黄精、益智仁、栀子、甘草和符合原卫生部2014年第6号通告的杜仲雄花。
(2)称重和粉碎:准确称取,杜仲雄花1000g,酸枣仁1500g、茯苓1000g、百合1000g、莲子1500g、黄精1000g、益智仁1000g、栀子1500g、甘草1000g,粉碎至20目~30目。
(3)提取方法:先将所述食品配方原料混合后粉碎至20目,加10倍体积的纯净水浸泡0.5小时;然后加水量分别为原材料体积的10倍、8倍、6倍,煮沸提取3次,提取时间分别为2.0小时或1.5小时或1.5小时,合并三次煎煮液,经减压浓缩至相对密度为1.05~1.10的浓缩液,加适量糊精喷雾干制成助眠原料的粉剂备用。
进一步优化的,为了更进一步提高提取效率,采用超声辅助提取方法,其具体步骤为:将粉碎后中药材放置提取罐后,用10倍的料液比加入纯净水,浸泡2小时,然后加水量分别为原材料体积的10倍、8倍、6倍,在频率为20KHz、65℃的超声条件下提取三次,每次45分钟。将三次提取液混匀,经减压浓缩至相对密度为1.05~1.10的浓缩液,加适量糊精喷雾干制成助眠原料的粉剂备用。经上述方法制备的粉剂的得率为12%左右。
(4)助眠原料的制备方法:最终的助眠原料包括两个组成配方,助眠原料1为通过上述超声辅助提取方法制备的粉剂;助眠原料2为上述超声辅助提取方法制备的粉剂与20%的GABA按4:1的重量比充分混匀后制备的复合粉剂。
所述的新食品原料GABA为市购符合原卫生部2009年第12号公告要求的20%的GABA。
实施例3
提供一种具有助眠功能的食品配方在失眠患者治疗食品中或对小动物催眠的应用。
本实施例,还提出一种不同助眠原料提取物对小鼠镇静催眠作用
(1)参考文献(林耕,郭屹,斯建勇,等.益智仁水提物的镇静催眠作用及对大鼠学习记忆影响的实验研究.中国中医药咨讯,2011,003(010):34-35.)报道的方法,选用健康成年昆明种小鼠(购买于上海某实验动物中心)120只,体重18-22g,其中60只用于对戊巴比妥钠睡眠时间影响的观察,另外60只用于对戊巴比妥钠阈下睡眠剂量影响的观察。
(2)不同助眠原料提取物对戊巴比妥钠睡眠时间影响的实验观察
将60只昆明小鼠随机分为空白对照组、助眠原料1组、助眠原料2组和阳性对照组,每组15只,空白对照组给予等体积的生理盐水作为对照,助眠原料1组、助眠原料2组分别给予1ml/100g体重的15%的实施例2制备的助眠原料1和助眠原料2的水溶液,阳性对照组给予1ml/100g体重的15%的枣仁安神颗粒(泰华天然生物制药有限公司生产),连续灌胃15日。末次灌胃后30min,每组小鼠腹腔注射戊巴比妥钠40mg/kg。记录小鼠的睡眠持续时间(以翻正反射消失为入睡时间,从翻正反射消失至恢复时间为睡眠持续时间)。时间结果见表1。

(3)不同助眠原料提取物对戊巴比妥钠阈下睡眠剂量影响的实验步骤
将60只昆明小鼠,分组和方法同样分为空白对照组、助眠原料1组、助眠原料2组和阳性对照组,每组15只,分组和干预方法同上,连续灌胃15日。末次灌胃后30min,每组小鼠腹腔注射戊巴比妥钠30mg/kg,观察给戊巴比妥钠后30min内各组发生睡眠的动物数(以翻正反射消失1min以上为指标)。

从上述结果可以看出,助眠原料1和2均可以提高大鼠域上戊巴比妥钠的睡眠时间和域下戊巴比妥钠的入睡率,在动物实验证明助眠原料1和2均有助眠作用,助眠原料2的效果更为明显,与阳性对照药物枣仁安神颗粒相近。
实施例4
本实施例,还提出一种助眠软糖的制备方法
所述具有助眠功能的食品包括软糖或果冻或饼干或固体饮料或压片糖果。
所述软糖制作方法包括:将药食同源和新食品原料提取物粉剂的重量比为1~2份,包括由果葡糖浆、蔗糖、明胶制备的食品基料的重量比为6~10份,通过混合、调色、调味、脱气、浇模、烘干而成的软糖。
为了改善软糖的口味和外观,首先要改善助眠提取物的口味和吸水性,在实施例2制作的助眠原料1的基础上,添加不同重量的甜菊糖苷增加甜味,柠檬粉增加酸味,β-环糊精促进成型和抑制苦味,甘露醇增加醇厚感并抑制原料的吸水性,组成调制的助眠原料1、2、3。

通过20~29岁的男员工3人,女员工7人共10人的试用,将调配好的饮料配制为15%的水溶液,然后分别评价调配好的助眠原料水溶液的口感,经测试10个人中有7人认为方案2调制的助眠原料2的口味(包括酸甜度、涩味、苦味、中药异味的综合评价)最为适宜,1人接受方案1调制助眠原料1的口味,2人接受方案2调制助眠原料3的口味,因此选择方案2调制的助眠原料2做为后面制作软糖的功能成分配料。
(2)助眠软糖的制作方法
首先制备软糖的基料,软糖的基料包括主要包括葡萄糖和明胶,将两者按一定的比例进行称重配比并粗混一体,按调制的助眠原料2与基料的重量比为1:8的比例,准确称取上述调制的助眠原料2。然后将葡萄糖和明胶混合物加入搅拌罐加适量的水进行搅拌以制备粘稠的糖体,在搅拌过程中逐渐添加调制的助眠原料2并混匀,经调色、调味、脱气后的混合料注入特定模具内,彻底粘固后从模具中移出并进一步变冷固化。最后,固化的软糖进入蒸汽室,蒸汽中的湿气帮助裹上一层糖衣,滚动后表面只剩下糖渣,最后进行包装。
实施例5助眠压片糖果的制备方法
所述压片糖果制作方法包括:将实施例2制备的助眠原料1(药食同源和新食品原料提取物粉剂)的重量比为3~4份与新食品原料γ-氨基丁酸的重量比为0.25~1份,充分混匀后制备为助眠原料2,再添加适量微晶纤维素、二氧化硅和硬脂酸镁充分混匀后,压制成压片糖果。
进一步优化的,取实施例2制备的助眠原料2的粉剂共500g,添加微晶纤维素20g、二氧化硅5g,硬脂酸镁7.5g,充分混匀后造粒,压制成0.8g/片的压片糖果,每瓶60粒装瓶备用。
实施例6助眠软糖对失眠患者的睡眠质量影响的临床观察
选取2020年3月-2020年6月期间在上海某二甲医院失眠门诊被确诊为失眠症的患者68例,病程为3个月~3年不等,平均1.1年;其中男性27例,女性41例,随机分为观察组和对照组,每组各34例。两组间性别、年龄和失眠时间均无差异。观察组每晚睡前吃4粒实施例4制作的具有助眠功能的软糖,对照组吃4粒不含助眠原料、但外观和口味一样的普通软糖,连续服用14天和28天时分别评估睡眠改善效果。根据睡眠改善情况,干预效果分为显著、有效和无效。其中,显著的标准是:睡眠质量明显好转,睡眠时间恢复正常或保持在6h以上,第二天醒后精力充沛无疲倦感;改善的标准是:睡眠质量有所提高,睡眠时间明显高于干预前或睡眠时间维持在4h以上,第二天精神状态明显好转;无效的标准是:与干预前相比睡眠质量、时间和第二天精神状态几乎无变化。干预14天和28天后的具体结果见表4。通过表4可以看出,干预14天和28天后给予助眠软糖组对睡眠改善的效果明显高于对照组(经卡方检验,与对照组相比较,干预14天时p<0.05,干预28天时p<0.01)。

实施例7助眠压片糖果改善失眠症状的典型案例
张X倩,女,28岁,某大型公司会计近半年来感到莫名的烦恼,晚上入睡困难,12点后才能入睡,早晨4点醒后就再也不能入睡,第二天昏昏沉沉,工作精力不集中,最近一个月症状更为明显,到多家医院应用安眠药治疗,效果不理想。2021年2月15日通过人介绍了解了本公司生产一种药食同源助眠产品,虽前来咨询。根据其情况,让其停用所有镇静催眠的西药,服用实施例5制备的助眠压片糖果每日4片,睡前1小时一次服用,服用2~3天后感觉睡眠状况不断好转,半个月后睡眠明显改善,每天实际睡眠时间达到6小时以上,且白天工作更有精神,进一步减为每日睡前1小时服用2片助眠压片糖果,同时鼓励其放宽心胸,保持乐观的生活态度,并于白天增加30分钟的慢跑。总共服用1个月后,最终减为每日服用1粒助眠压片糖果,随访3个月没有发生反弹,效果特别满意。
综上所述,本发明成分利用药食同源物质和新食品原料的各自优势和特性,以中医理论为指导,以现代科学研究为依据,通过严格选材、反复进行动物实验和人体试食以调整各组分之间的比例,再结合现代提取和分离工艺,制备的一种具有助眠功能的药食同源和新食品原料组合物。以该组合物为功能成分添加适当的食品基料制备的软糖口味酸甜可口,深得中青年群体的喜好,也可制备为固体饮料、压片糖果、果冻和饼干等形式,方便在大众中推广。经过动物实验和人体试验证明,该助眠配方及以此配方为基础制备的各种食品,包括软糖和压片糖果,具有明显改善失眠症状及由失眠导致的身心症状的作用。
本发明的益处是在健康管理的基础上,通过多靶点的精准干预,改善睡眠质量,降低由于睡眠不足导致的精神和躯体症状,提高失眠患者的生活质量,造福于社会和大众。
