Abstract

本发明公开了一种快速检测阴道病原微生物的多重PCR试剂盒制作方法,先分别确定念珠菌属、加德纳菌和毛滴虫的特异引物序列;制备适合念珠菌、加德纳菌和毛滴虫这三种病原的核酸提取试剂盒,适用于阴道分泌物标本;找到适合念珠菌、加德纳菌和毛滴虫这三种病原的荧光PCR反应条件,制作荧光定量PCR检测试剂;最后组装成完整试剂盒;本发明通过制备念珠菌属、加德纳菌和毛滴虫三种病原的多重PCR试剂盒,可以一个PCR反应体系内快速检测出阴道内常见感染的三种病原微生物是哪一种或混合感染,以便于指导临床尽早选择针对性治疗药物。

一种快速检测阴道病原微生物的多重PCR试剂盒制作方法
技术领域
本发明涉及一种快速检测阴道病原微生物的多重PCR试剂盒制作方法,属于生物医药技术领域。
背景技术
阴道炎会导致外阴阴道症状,如瘙痒、灼痛、刺激和异常流液等;正常情况下有需氧菌及厌氧菌寄居在阴道内,形成正常的阴道菌群,当身体机能下降,或收到外界影响,会导致阴道与菌群之间的生态平衡打破,形成致病菌支原体,常见的有念珠菌、加德纳菌和毛滴虫;目前,加德纳杆菌(GD)、白色念珠菌(CA)、阴道毛滴虫(Tv)性阴道炎,缺少快速检测出阴道内感染的三种病原微生物是哪一种或混合感染。
发明内容
针对上述存在的技术问题,本发明的目的是:提出了一种快速检测阴道病原微生物的多重PCR试剂盒制作方法。
本发明的技术解决方案是这样实现的:一种快速检测阴道病原微生物的多重PCR试剂盒制作方法,包含以下步骤:
①分别制备念珠菌、加德纳菌和毛滴虫的特异引物序列;
②制备适合念珠菌、加德纳菌和毛滴虫这三种病原的核酸提取试剂盒,适用于阴道分泌物标本;
③找到适合念珠菌、加德纳菌和毛滴虫这三种病原的荧光PCR反应条件,制作荧光定量PCR检测试剂;
④组装成完整试剂盒。
所述的步骤①中,念珠菌、加德纳菌和毛滴虫的特异引物序列包括:
SEQ ID NO:1~2所示核苷酸序列的引物对;SEQ ID NO:4~5所示核苷酸序列的引物对;SEQ ID NO:7~8所示核苷酸序列的引物对。
所述的步骤②中,适合念珠菌、加德纳菌和毛滴虫这三种病原的核酸提取试剂盒包括:
SEQ ID NO:3所示核苷酸序列的探针;SEQ ID NO:6所示核苷酸序列的探针;SEQID NO:9所示核苷酸序列的探针。
SEQ ID NO:3所示核苷酸序列的探针的5’端标记有荧光报告基团CY5,3’端标记有荧光淬灭基团BHQ3;
SEQ ID NO:6所示核苷酸序列的探针的5’端标记有荧光报告基团FAM,3’端标记有荧光淬灭基团BHQ1;
SEQ ID NO:9所示核苷酸序列的探针的5’端标记有荧光报告基团ROX,3’端标记有荧光淬灭基团MGB。
优选的,所述的步骤③中,荧光定量PCR检测试剂包括qPCR Master Mix酶混合液、qPCR Master Mix反应缓冲液。
由于上述技术方案的运用,本发明与现有技术相比具有下列优点:
本发明通过制备念珠菌、加德纳菌和毛滴虫三种病原的多重PCR试剂盒,可以快速检测出阴道内感染的三种病原微生物是哪一种或混合感染,以便于指导临床选择针对性治疗药物。
具体实施方式
下面结合实施例来说明本发明。
本发明所述的一种快速检测阴道病原微生物的多重PCR试剂盒制作方法,包含以下步骤:
①分别制备念珠菌、加德纳菌和毛滴虫的特异引物序列,包括SEQ ID NO:1~2所示核苷酸序列的引物对;SEQ ID NO:4~5所示核苷酸序列的引物对;SEQ ID NO:7~8所示核苷酸序列的引物对;
②制备适合念珠菌、加德纳菌和毛滴虫这三种病原的核酸提取试剂盒,适用于阴道分泌物标本;试剂盒包括:SEQ ID NO:3所示核苷酸序列的探针;SEQ ID NO:6所示核苷酸序列的探针;SEQ ID NO:9所示核苷酸序列的探针。
③找到适合念珠菌、加德纳菌和毛滴虫这三种病原的荧光PCR反应条件,制作荧光定量PCR检测试剂;
SEQ ID NO:3所示核苷酸序列的探针的5’端标记有荧光报告基团CY5,3’端标记有荧光淬灭基团BHQ3;
SEQ ID NO:6所示核苷酸序列的探针的5’端标记有荧光报告基团FAM,3’端标记有荧光淬灭基团BHQ1;
SEQ ID NO:9所示核苷酸序列的探针的5’端标记有荧光报告基团ROX,3’端标记有荧光淬灭基团MGB;
荧光定量PCR检测试剂包括qPCR Master Mix酶混合液、qPCR Master Mix反应缓冲液。
④最后组装成完整试剂盒。

①分别制备念珠菌、加德纳菌和毛滴虫的特异引物序列;
②制备适合念珠菌、加德纳菌和毛滴虫这三种病原的核酸提取试剂盒,适用于阴道分泌物标本;
③找到适合念珠菌、加德纳菌和毛滴虫这三种病原的荧光PCR反应条件,制作荧光定量PCR检测试剂;
④组装成完整试剂盒。
2.根据权利要求1所述的快速检测阴道病原微生物的多重PCR试剂盒制作方法,其特征在于:

SEQ ID NO:3所示核苷酸序列的探针的5’端标记有荧光报告基团CY5,3’端标记有荧光淬灭基团BHQ3;
SEQ ID NO:6所示核苷酸序列的探针的5’端标记有荧光报告基团FAM,3’端标记有荧光淬灭基团BHQ1;
SEQ ID NO:9所示核苷酸序列的探针的5’端标记有荧光报告基团ROX,3’端标记有荧光淬灭基团MGB。
