Description

Composition for repairing sensitive skin, preparation method and application thereof
Technical Field
The invention relates to the technical field of cosmetics, in particular to a composition for repairing sensitive skin, and a preparation method and application thereof.
Background
With the development of society and the improvement of living standard of people, the skin care problem is more and more paid attention by consumers. The need for skin care has shifted from previous prevention and delay to current restorative aspects. Particularly, the modern living rhythm is accelerated, the working pressure is increased, and the night is increased, so that the skin state of a consumer is poorer and poorer, and the trend of changing to sensitive skin is accompanied. The sensitive skin means that the skin generates various reactions which are not tolerant to the skin under the external stimulation, and various problems such as redness, peeling, tightness and the like occur, and the sensitive skin is essentially characterized in that the skin cuticle is thinned to cause the skin moistening degree to be insufficient, and finally, the barrier function of the skin is too weak to resist the external stimulation.
Sensitive skin, due to its special intolerance characteristics, not only responds significantly to external stimuli, but also makes healing more difficult. From the composition, most of the skin care products for repairing sensitive skin in the market are composed of various plant extracts. However, different natural plant extracts may bring about synergistic and antagonistic effects during the compounding process. Adding multiple plant extracts without hasty while the additive effects of efficacy and effects have not been analyzed and verified is not necessarily a sensible option. In addition, the addition of various plant extracts adds extra burden to sensitive skin and has opposite effects on skin repair.
Therefore, the development of a composition which has simple components and has the effects of skin repair, inflammation resistance and soothing is urgent.

Compared with the traditional repairing composition, the skin care composition provided by the invention focuses on skin repair, and meanwhile, the effects of the skin care composition are extended to the fields of water replenishing, moisture preserving, anti-aging, wrinkle removing and the like. While multiple effects impart more functionality to a composition to some extent, diversification of effects is often accompanied by a reduction in the effect of the effects or a complication of the composition. Either the decline in efficacy or the complication of the composition is not conducive to the repair of sensitive skin, and the complicated efficacy composition can cause additional skin burden. The method focuses on the problem of repairing the sensitive skin, starts from the root of the skin problem, and solves the problem of repairing the sensitive skin in multiple dimensions. The composition has more specific efficacy and obviously improves the effect of repairing skin.
The traditional repairing composition achieves the purpose of skin repair mainly by promoting the proliferation of skin cells. However, such compositions only address the existing skin repair issues and do not address the issues of skin inflammation and soothing. The invention combines skin repair and anti-inflammatory relief into a whole and cooperates together, thereby realizing the comprehensive repair of the skin. The repair of the sensitive skin can be effectively realized, the skin inflammation can be reduced, and the problem of the repair of the sensitive skin is fundamentally solved.
The functional composition consists of ultramicro hyaluronic acid, acetylated hyaluronic acid, ectoin and madecassoside, the three polysaccharides have high biocompatibility, and the madecassoside is the main component in the plant extract, so that the composition has high biocompatibility and safety and cannot cause the problem of stimulation to sensitive skin.

Efficacy evaluation
1 Water content of stratum corneum
1) Principle of testing
The invention adopts CK German skin moisture tester CM825 to test the moisture content of the stratum corneum. The water content of the horny layer not only reflects the physiological function of the horny layer of human skin, but also represents the physiological function of the whole skin and even the whole body on the skin. The increase and decrease of the water content of the horny layer are closely related to the damage and repair degree of the skin. When the skin is damaged to a high degree, the moisture content of the stratum corneum of the skin is reduced, and when the damaged skin is repaired, the moisture content of the stratum corneum of the skin is increased. Measuring the water content of the stratum corneum is of great significance to understanding the physiological functions of the body, particularly the repair state of the skin. The present invention is used to measure the efficacy of the efficacy composition in repairing an organ.
2) Test method
-a test environment: the testing environment temperature is 22 +/-1 ℃, the humidity is 50 +/-5%, and real-time dynamic detection is carried out;
-test volunteers: age between 16-60 years (excluding pregnant or lactating women); patients without severe systemic disease, immunodeficiency or autoimmune disease; those who have not had a history of severe allergy to skin care cosmetics in the past; hormone drugs and immunosuppressants have not been used in the last month; those who did not participate in other clinical trials; the tested drugs are used according to the regulations and the data is complete; all volunteers should fill out informed consent before testing.
-a testing step:
preparation before testing: any product (cosmetic or external medicine) cannot be used for 2-3 days before the tested part. Before the test, the skin moisture tester CM825 evaluation was performed by drawing a sample application area on each cheek side of the tester.
-a test procedure: the skin stratum corneum water content of the area demarcated by the cheek of the subject was measured as the original value after the subject was rested in a constant temperature and humidity room for 30 minutes. Then, the same amount of experimental samples are respectively used in the test areas of the testees, the same thickness and time are coated, and the moisture content of the stratum corneum of the skin is respectively measured aiming at the test areas after the skin is washed clean and dried by air. The test of the same volunteer was performed by the same measurement staff.
3) Test results
The calculation formula of the water content increase rate of the horny layer is as follows: c% ((C))2-C1)/C1*100%
Note: c is the moisture content increase rate,%;
C2skin moisture content for continued use of the sample after 28 days;
C1the original value of the skin moisture content before using the sample;
the data are collated in Table 3 below:
TABLE 3
Figure BDA0003289118210000091
FIG. 1 shows a graph of the rate of change of the moisture content of the stratum corneum of the skin, and FIG. 1 shows a graph of the rate of change of the moisture content of the stratum corneum of the skin according to the examples and comparative examples of the present invention; it can be seen from fig. 1 and table 3 that the composition for repairing sensitive skin provided by the present invention increases the moisture content of the stratum corneum of the skin to some extent, and also laterally feeds back the damaged skin to some extent to be repaired. By comparing the increase rate of the moisture content of the stratum corneum between comparative examples 3 and 4 and example 2, it can be seen that the compositions of the present invention have a synergistic effect between the components, and when some of the components are missing, the efficacy of the compositions is significantly reduced.
2. Percutaneous water loss of skin
1) Principle of testing
The present invention uses a Delfin finland skin moisture loss tester VapoMeter to test the transdermal moisture loss. Transdermal moisture loss refers to the transport of moisture through the skin by passive diffusion, the most common use being the assessment of the skin barrier function and the condition of the skin. The invention is used for measuring the skin state and barrier function after using the efficacy composition and feeding back the repairing effect of the composition.
2) Test method
-a test environment: the testing environment temperature is 22 +/-1 ℃, the humidity is 50 +/-5%, and real-time dynamic detection is carried out;
-test volunteers: age between 16-60 years (excluding pregnant or lactating women); patients without severe systemic disease, immunodeficiency or autoimmune disease; those who have not had a history of severe allergy to skin care cosmetics in the past; hormone drugs and immunosuppressants have not been used in the last month; those who did not participate in other clinical trials; the tested drugs are used according to the regulations and the data is complete; all volunteers should fill out informed consent before testing.
-a testing step:
preparation before testing: any product (cosmetic or external medicine) cannot be used for 2-3 days before the tested part. Before testing, the using area of the sample is drawn on one side of the cheek of the tester respectively, and the evaluation of the skin percutaneous water loss test is carried out.
-a test procedure: the subjects rested in a constant temperature and humidity room for 30 minutes, and then the skin water loss in the area demarcated by the cheek was measured as the original value. Then, the same amount of experimental samples are respectively used in the test areas of the testees, the test areas are coated with the same thickness and time, and the test areas are respectively subjected to percutaneous moisture loss measurement after being washed clean and dried in the air. The test of the same volunteer was performed by the same measurement staff.
3) Test results
The long-acting skin percutaneous water loss improvement rate calculation formula: v% ((V))1-V2)/V1*100%
Note: v is the improvement rate of skin percutaneous water loss,%;
V2the amount of water dispersed through the skin after 28 days of continuous use of the sample;
V1the original value of the skin transdermal water dispersion loss before using the sample;
the experimental data are shown in table 4:
TABLE 4
Figure BDA0003289118210000101
A change of the skin moisture loss through skin is shown in figure 2, and figure 2 is a change chart of the change of the skin moisture loss through water of the embodiment and the comparative example of the invention.
From the results, the composition for repairing sensitive skin provided by the invention improves the percutaneous water loss of the skin to a certain degree, and indicates that the barrier function of the skin is repaired to a certain degree. The composition of the present invention has a synergistic effect between the components by comparing the skin transdermal moisture improvement rates between comparative examples 3 and 4 and example 2, and the efficacy effect of the composition is remarkably reduced when some components are missing.
3. Elasticity of skin
1) Principle of testing
The invention adopts CK Germany skin elasticity tester Cutomer MPA580 to test the skin elasticity. Skin elasticity is the ability of the skin to stretch and recover its original shape, and is generally related to the content of skin collagen, elastin and natural fats. The invention is used for measuring the recovery condition of skin collagen and elastin, the health condition of skin and the like after the functional composition is used, and is used for feeding back the repairing effect of the composition.
2) Test method
-a test environment: the testing environment temperature is 22 +/-1 ℃, the humidity is 50 +/-5%, and real-time dynamic detection is carried out;
-test volunteers: age between 16-60 years (excluding pregnant or lactating women); patients without severe systemic disease, immunodeficiency or autoimmune disease; those who have not had a history of severe allergy to skin care cosmetics in the past; hormone drugs and immunosuppressants have not been used in the last month; those who did not participate in other clinical trials; the tested drugs are used according to the regulations and the data is complete; all volunteers should fill out informed consent before testing.
-a testing step:
preparation before testing: any product (cosmetic or external medicine) cannot be used for 2-3 days before the tested part. Before the test, the areas of application of the samples were drawn on the cheek sides of the subjects, respectively, and skin elasticity was evaluated.
-a test procedure: the skin elasticity of the area demarcated by the cheek of the subject was measured as the original value after the subject was rested in a constant temperature and humidity chamber for 30 minutes. Then, the same amount of test samples were applied to the test areas of the subjects, respectively, with the same thickness and time, and skin elasticity measurements were performed on the test areas, respectively, after rinsing, and air drying. The test of the same volunteer was performed by the same measurement staff.
3) Test results
Skin elasticity improvement rate calculation formula: n% (N)2-N1)/N1*100%
Note: n is the skin elasticity improvement rate,%;
N2skin elasticity for the continued use of the sample after 28 days;
N1the original value of the skin elasticity before using the sample;
the experimental data are as follows in table 5:
TABLE 5
Figure BDA0003289118210000111
The change of the skin elasticity improvement rate is obtained as shown in fig. 3, and fig. 3 is a graph showing the change of the skin elasticity improvement rate of the examples and comparative examples of the present invention.
The above results demonstrate that the composition for repairing sensitive skin provided by the invention has the effect of improving the skin elasticity, and the repairing effect of the composition of the invention is also laterally confirmed. The synergistic effect between the components of the composition of the present invention is shown by comparing the skin elasticity improvement rate between comparative examples 3 and 4 and example 2, and the efficacy effect of the composition is remarkably reduced when some components are missing.
4. Skin hemoglobin levels
1) Principle of testing
The present invention uses a skin pigment tester, andrera 3D, to test skin hemoglobin levels. The red blood streak is mainly caused by the fact that the position of a capillary vessel is easier to contact and sense the change of the external environment due to the weakness of the stratum corneum of the face, and therefore the phenomenon of the face is caused by the expansion of the capillary vessel. The method is used for measuring the skin heme level after the use of the efficacy composition, and is used for feeding back the effect of the composition on the restoration of the red blood silk.
2) Test method
-a test environment: the testing environment temperature is 22 +/-1 ℃, the humidity is 50 +/-5%, and real-time dynamic detection is carried out;
-test volunteers: age between 16-60 years (excluding pregnant or lactating women); patients without severe systemic disease, immunodeficiency or autoimmune disease; those who have not had a history of severe allergy to skin care cosmetics in the past; hormone drugs and immunosuppressants have not been used in the last month; those who did not participate in other clinical trials; the tested drugs are used according to the regulations and the data is complete; all volunteers should fill out informed consent before testing.
-a testing step:
preparation before testing: any product (cosmetic or external medicine) cannot be used for 2-3 days before the tested part. Before the test, the sample application area was drawn on the cheek side of the test subject, and skin hemoglobin evaluation was performed.
-a test procedure: after the subject was rested in a constant temperature and humidity room for 30 minutes, the skin red blood streak in the area demarcated by the cheek was measured as the original value. Then, the same amount of test samples were applied to the test areas of the subjects, respectively, and the skin hemoglobin measurement was performed on the test areas after rinsing, air drying, and the like. The test of the same volunteer was performed by the same measurement staff.
3) Test results
Long-acting calculation formula: m% ((M))1-M2)/M1*100%
Note: m is improvement rate of red blood streak,%;
M2skin hemoglobin levels 28 days after application of the samples;
M1is the skin hemoglobin level prior to use of the sample;
the experimental data are shown in table 6:
TABLE 6
Figure BDA0003289118210000121
FIG. 4 is a graph showing the change of the improvement rate of red blood cells in the skin, and FIG. 4 is a graph showing the change of the improvement rate of red blood cells in the skin according to the example of the present invention and the comparative example.
As can be seen from FIG. 4, the composition of the present invention has the effect of repairing red blood cells, and the skin hemoglobin is significantly reduced after the composition of the present invention is applied. By comparing the improvement rate of the red blood cells of the skin between the comparative examples 3 and 4 and the example 2, the composition of the invention has synergistic effect, and when partial components are absent, the efficacy effect of the composition is obviously reduced.
5. Evaluation of lactic acid stinging
1) Principle of testing
It is thought that the development of skin sensitization is a complex process involving the skin barrier-cutaneous nervous system-skin immune cells. Under the combined action of internal and external factors, the barrier function of the skin is damaged, so that the sensory nerve afferent signals are increased, the response of the skin to external stimulation is enhanced, and the skin immune inflammatory response is initiated. The score of lactic acid stabbing pain tests before and after using the product can directly reflect the repairing effect of the product on the skin barrier function. The present invention tests the skin barrier repair efficacy of the composition by assessing lactic acid stinging before and after use of the product.
2) Test method
-a test environment: the testing environment temperature is 22 +/-1 ℃, the humidity is 50 +/-5%, and real-time dynamic detection is carried out;
-test volunteers: age between 16-60 years (excluding pregnant or lactating women); patients without severe systemic disease, immunodeficiency or autoimmune disease; those who have not had a history of severe allergy to skin care cosmetics in the past; hormone drugs and immunosuppressants have not been used in the last month; those who did not participate in other clinical trials; the tested drugs are used according to the regulations and the data is complete; all volunteers should fill out informed consent before testing.
-a testing step:
preparation before testing: any product (cosmetic or external medicine) cannot be used for 2-3 days before the tested part.
-a test procedure: the subject sits statically in a constant temperature and humidity room for 30 minutes, then uses 10% lactic acid water solution and distilled water with equal amount in the left and right nasolabial folds of the subject respectively, and inquires the self skin feeling symptoms of the subject at 0.5, 2 and 5min, and scores according to a quartering method: no prickling sensation is 0 point, slight prickling sensation is 1 point, moderate prickling sensation is 2 points, and severe prickling sensation is 3 points. The test of the same volunteer was performed by the same measurement staff.
3) Test results
The lactic acid stabbing pain improvement rate calculation formula is as follows: r ═ R1-R2)/R1×100%
R: lactic acid sting improvement rate;
R1: pre-sample lactate sting scores;
R2: lactate sting score 28 days after using the sample;
lactic sting score 0.5min +2min +5min sting score;
the experimental data are shown in table 7:
TABLE 7
Figure BDA0003289118210000141
Fig. 5 is a graph showing the change of the lactic acid stinging improvement rate, and fig. 5 is a graph showing the change of the lactic acid stinging improvement rate of the example of the present invention and the comparative example.
The results show significant differences, which indicate that the composition for repairing sensitive skin provided by the invention has the efficacy of repairing skin barriers. By comparing the improvement rates of comparative examples 3 and 4 with example 2, it can be seen that the efficacy composition of the present invention has a synergistic effect, and the efficacy effect of the composition is remarkably decreased when some components are absent. The lactic acid sting test result shows that the composition has polar irritation, and the skin sensitivity can be obviously reduced and the external irritation can be relieved by using the composition.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.
