背景技术
丹参是我国重要的传统中药,属于40种常用大宗中药材品种之一,在临床上广泛用于治疗心脑血管疾病。我国的山东、四川、陕西和河南等省份是丹参的主产区,其中山东、四川两省的丹参产量即可达1.6万吨,预计全国丹参药材产量在2万吨左右。丹参药材加工提取有效成分后,可产生万吨级的丹参药渣。近年来,针对中药渣的开发利用研究成为热点,包括生物炼制、食用菌栽培基质、双向发酵以及动物饲料添加剂等应用。
丹参中成药制备常采用水浸丹参,其水溶性浸出物和其它药物混合制成丹参中成药。经水浸提加工后的丹参药渣中,仍存在部分脂溶性的功能物质未得到充分利用,导致丹参资源性化学物质的浪费。因此,对丹参药渣开展功能组分和精深加工的研究,可以促进丹参药材物尽其用,提升综合经济效益。

发明目的:本发明的目的是为了克服现有技术的不足,对丹参水提取后的药渣进行深入研究,开发出具有抗氧化活性的丹参提取物。

作为优选方案,以上所述的一种具有抗氧化活性的丹参药渣提取物,它是由以下方法制备得到:取丹参药渣或灭菌处理后的丹参药渣,按质量比1:10加蒸馏水或75%乙醇,60℃超声提取2h,放冷,再称定,用蒸馏水或75%乙醇补足损失量,抽取滤液,过0.45μm有机膜,既得。

下面通过实施例的方式进一步说明本发明,但并不因此将本发明限制在所述的实施例范围之中。在阅读了本发明之后,本领域技术人员对本发明的各种非实质性的修改均落于本申请所附权利要求所限定的范围。
实施例1
1材料与方法
1.1材料
1.1.1丹参药渣
丹参经水提加工后的药渣,由江苏海昇药业有限公司提供,药渣粉碎过0.425mm筛,烘干备用。
1.1.2化学试剂
中药化学对照品丹参酮I、丹参酮IIA、丹参酮IIA-磺酸钠、隐丹参酮以及二氢丹参酮,四川维克奇生物科技有限公司。2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS·)、1,1-二苯基-2-苦肼基(自由基)(DPPH·)、邻二氮菲、铁氰化钾等试剂均为分析纯,国药集团化学试剂有限公司。
1.2方法
1.2.1丹参药渣中纤维素、半纤维素、木质素组分检测
采用美国国家可再生能源实验室(NREL)方法对其处理后,采用HPLC进行检测,计算药渣中纤维素、半纤维素及木质素的含量。
1.2.2丹参药渣中可溶性还原糖和蛋白质
称取药渣20g/L,121℃保温20min,抽滤,取滤液为待测样品。采用3,5-二硝基水杨酸(DNS)显色法测滤液中还原糖含量,以葡萄糖制作标准曲线。采用Brandford法测药渣中水溶性蛋白的含量,以牛血清蛋白制作标准曲线。
1.2.3丹参药渣中丹参酮组分检测
药渣处理方法参照文献,略有改动。取丹参药渣5g于圆底烧瓶中,加入100mL甲醇,称定质量,加热回流1h,放冷,再称定,用甲醇补足损失量,取滤液,过0.45μm有机膜,于4℃避光备用。
检测条件:色谱分离柱为Amethyst C18(4.6mm×250mm,5μm),岛津公司LC-20AB高效液相色谱仪及紫外检测器。检测波长为270nm,流动相为体积分数80%甲醇,进样体积为10μL,流速为1.0mL/min,柱温为30℃。
分别精密称取适量各种丹参酮标准品,用甲醇溶解,配制浓度5~50mg/L,过0.45μm有机膜避光保存备用。分别以丹参酮标准品的浓度为横坐标,峰面积为纵坐标作标准曲线。1.2.4丹参药渣体外抗氧化活性分析
丹参药渣水浸提物或乙醇浸提物的制备:取丹参药渣或灭菌处理后的丹参药渣(含水量65%,经121℃灭菌处理1h)的样品,按干物质含量1:10(质量比)分别加蒸馏水或75%乙醇,称定质量,60℃超声提取2h,放冷,再称定,用蒸馏水或75%乙醇补足损失量,抽取滤液,过0.45μm有机膜,于4℃避光备用。对水或醇提取物用蒸馏水进行体积比1:20稀释后,进行抗氧化活性分析。
ABTS·清除:用蒸馏水配制含7mmol/LABTS和2.45mmol/L过硫酸钾的混合溶液,将溶液放置于25℃避光孵育12~16h,即为ABTS·溶液。使用时用蒸馏水将该溶液稀释至其在波长734nm处吸光度为0.700±0.005,即得ABTS·工作液。测量时取0.1mL受试物溶液,加入3.9mLABTS·工作液,混合摇匀后置于25℃孵育6min,测量其在波长734nm处的吸光度,并计算清除率(以纯溶剂做空白对照)。ABTS·清除率=(1-实验组吸光值/空白组吸光值)×100%。
DPPH·清除:将0.1mmol/L的DPPH溶于乙醇溶液,即为DPPH·溶液。测量时取2.0mL受试物溶液,加入2.0mL DPPH溶液混合摇匀,置于25℃避光孵育30min,测其在波长517nm处的吸光度,并计算清除率(以95%乙醇做空白对照。)。DPPH清除率=(1-实验组吸光值/空白组吸光值)×100%。
还原能力测定:测量时取2.5mL受试品溶液,加入2.5mL磷酸缓冲液(0.2mmol/L,pH为6.6)和2.5mL1%(质量分数)铁氰化钾溶液混匀,置于50℃孵育20min,迅速冷却,迅速加入2.5mL10%(质量分数)三氯乙酸溶液并混匀,在3000r/min下离心10min,取2.5mL上清溶液,再加入2.5mL纯水和0.5mL 0.1%(质量分数)三氯化铁溶液,混匀后测定其在波长700nm处的吸光度。吸光度值越大,表示受试物的还原能力越强。
·OH的生成及清除:在10mL试管中依次加入1.5mL 5mmol/L的邻二氮菲溶液,3mL0.5mol/L、pH7.4的磷酸缓冲液,1ml 7.5mmol/L的FeSO4,立即摇匀,迅速加入待测溶液3mL,1mL 0.1%的H2O2,然后用蒸馏水将反应体系体积补充至10mL,迅速摇匀,然后将混合溶液置于37℃水浴锅保温60min,测量其在波长510nm处的吸光值。清除率计算公式为:E=(AS-A0)/(A1-A0)×100%。其中AS为含样品及H2O2的反应体系吸光值,A1为蒸馏水代替样品及H2O2的反应体系吸光值,A0为蒸馏水代替样品的反应体系吸光值。
2结果与讨论
2.1丹参药渣中的“三大素”及可溶性还原糖、蛋白质组分
植物源的药渣中含有丰富的纤维素、半纤维素、木质素、多糖、蛋白和多种微量元素。丹参药渣体量达万吨级,深入分析药渣组分中的纤维素、半纤维素和木质素等组分含量,有助于从生物炼制角度去探讨药渣的利用潜力。
采用美国国家可再生能源实验室(NREL)方法对丹参药渣的“三大素”组分进行了分析,并对药渣的可溶性还原糖和蛋白质进行了检测,结果如表1所示。由表1可知,丹参药渣中可溶性还原糖和蛋白质含量较低,难以作为微生物代谢的速效碳源或氮源。丹参药渣中的纤维素、半纤维素和木质素的占比分别为19.5%、20.8%和16.8%。值得关注的是,丹参药渣中的纤维素含量相比玉米秸秆中的(37.3%)显著偏低,使得通过纤维素酶解丹参药渣制备可发酵糖的可行性存在较大局限。半纤维素和木质素等组分可以较好地被白腐真菌利用。因此,丹参药渣可用于白腐真菌的培养基质。

2.2丹参药渣中的丹参酮组分检测
丹参酮是丹参中的脂溶性松香烷型二萜类化合物,又称为总丹参酮,是丹参根部的主要提取物之一,具有广泛的药理作用,受到医药学家的高度关注。采用水浸提工艺对丹参药材进行加工后,脂溶性的丹参酮组分仍残留在药渣中,并未得到充分利用。
根据标准曲线计算各组分峰面积对应的含量,测得丹参药渣中的丹参酮各组分含量:二氢丹参酮0.47mg/g,隐丹参酮0.61mg/g,丹参酮I1.44mg/g,丹参酮IIA2.38mg/g,总丹参酮含量为4.90mg/g。
2.3丹参药渣的体外抗氧化活性
以ABTS·清除、DPPH·清除、还原能力测定以及·OH清除等为评价方法,将丹参药渣的水提取液和醇提取液分别进行了体外抗氧化活性研究,结果如表2所示。由表2可知,丹参药渣的乙醇提取物较水提取物具有更好的抗氧化活性。尤其值得关注的是,对丹参药渣进行高温(121℃)处理1h后,水/醇提取物的抗氧化活性均显著降低;这一结果有两个方面的指导作用:一是丹参中的抗氧化活性物质不耐受高温,在提取处理过程中有必要关注高温对抗氧化活性物质的降效作用;二是以丹参药渣为培养基质进行“双向发酵”时,高温灭菌处理药渣后,菌质的抗氧化活性会有较大损失。

由以上表2实验结果表明,本发明提供的丹参药渣提取物具有显著的抗氧化活性。可以变费为宝,充分利用丹参水提后的药渣资源,可开发成抗氧化、抗衰老的保健品或药品。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (7)
1.一种具有抗氧化活性的丹参药渣提取物,其特征在于,它是由以下方法制备得到:取丹参药渣或灭菌处理后的丹参药渣,加蒸馏水或乙醇,超声提取,放冷,再称定,用蒸馏水或乙醇补足损失量,抽取滤液,过0.45 μm有机膜,既得。
2.根据权利要求1所述的一种具有抗氧化活性的丹参药渣提取物,其特征在于,它是由以下方法制备得到:取丹参药渣或灭菌处理后的丹参药渣,按质量比1:10加蒸馏水或75%乙醇,60 ℃超声提取2h,放冷,再称定,用蒸馏水或75%乙醇补足损失量,抽取滤液,过0.45 μm有机膜,既得。
3.根据权利要求2所述的具有抗氧化活性的丹参药渣提取物,其特征在于,丹参药渣提取物包含丹参酮、纤维素、半纤维素和木质素成分。
4.根据权利要求3所述的具有抗氧化活性的丹参药渣提取物,其特征在于,纤维素、半纤维素和木质素成分的百分含量分别为19.5%、20.8%和16.8%。
5.根据权利要求3所述的具有抗氧化活性的丹参药渣提取物,其特征在于,丹参酮的含量为4.90 mg/g。
6.根据权利要求5所述的具有抗氧化活性的丹参药渣提取物,其特征在于,丹参酮包含二氢丹参酮0.47mg/g,隐丹参酮0.61mg/g,丹参酮I 1.44mg/g,丹参酮IIA 2.38mg/g,总丹参酮含量为4.90mg/g。
7.权利要求1~6任一项所述的具有抗氧化活性的丹参药渣提取物在制备抗氧化、抗衰老的保健品或药品中的应用。
