The composition is administered by gavage at 10ml/kg body weight 1 time per day for 2 weeks. The low dryness of the instant ginseng for strengthening the spleen and tonifying the kidney is verified by taking the anal temperature of rats, the water intake and the content of 17-hydroxycorticosteroid in urine as evaluation indexes.
2.1 determination of anal temperature:
anal temperature was measured with an anal thermometer before dosing and 1 hour after the last dose. The results are shown in Table 3.
2.2 measurement of Water intake
The water supply quantity is fixed, the residual quantity is measured, and the difference value is the water drinking quantity. The change of the water intake before and after the administration is recorded,
and differences between groups were compared. Urine was collected before and after administration using a home-made metabolism cage, and the amount of collected urine was measured using a measuring cylinder for comparison. The results are shown in Table 3.
2.3 determination of the content of 17-Hydrocorticosteroides in urine
Two days before the experiment, 24-hour urine of rats was collected twice in metabolic cages. Finally, the urine was mixed 2 times per rat and the content was measured. Urine is tested with pH paper, and the pH should be below 2. The test tubes with plugs are numbered according to the order of administration. 0.5ml of urine was sequentially taken up by a pipette and put into a test tube, and 0.5ml of distilled water was added to make the volume 1 ml. Adding 0.3g anhydrous sodium sulfate, adding stopper, ultrasonic treating for 2min, and adding 4ml dealdehydized n-butanol. Performing ultrasonic treatment for 10min, centrifuging at 2000rpm for 10min, sucking out supernatant (n-butanol), placing into another small test tube, adding 30mg activated carbon, shaking, standing for 3min, and removing pigment. Centrifuging at 4000rpm for 10min, and sucking the supernatant to prepare color comparison. A large number of tubes were selected and A, B, C, D, E, F were noted according to the table. Wherein A, B tube is blank of color developing agent and test tube, C tube is blank of cortisone standard, and D tube is blank of cortisone standard containing 10 μ g. 0.8ml of dealdehydized n-butanol was added to each of the above 4 tubes, and 0.8ml of the n-butanol extract of urine was added to each of the tubes E as a sample blank tube, and the compatibility of the reagents is shown in Table 2.
