Background
Alopecia is a common scalp problem of modern people and is mainly divided into androgenetic alopecia, alopecia areata, cicatricial alopecia and other types, and also other rare types such as infectious alopecia and congenital alopecia. The androgenic alopecia, also called seborrheic alopecia, is the most common type, most of the anti-alopecia products on the market are products aiming at the alopecia, the anti-alopecia effect is realized by inhibiting the activity of 5 alpha reductase, lysine hydroxylase and other ways, and the importance of scalp health is ignored.

Further provided is the use of the scalp care composition in scalp pre-milk.
The invention provides a composition for preventing hair loss, fixing hair, removing dandruff and controlling oil, which comprises calamus, houttuynia cordata and aloe extracts, 17 type collagen and a solvent acceptable in the field of cosmetics; wherein the extracts of the calamus, the houttuynia cordata and the aloe are ethanol extracts of the bulk drugs of the calamus, the houttuynia cordata and the aloe;
wherein the mass ratio of the 17-type collagen to the raw medicines of the calamus, the houttuynia cordata and the aloe is (0.0001-5): 10-50);
the cosmetically acceptable solvent includes polyhydric alcohols; the polyalcohol comprises one or more of 1, 3-butanediol, 1, 2-propylene glycol, dipropylene glycol and glycerol.
Preferably, the mass ratio of the 17-type collagen to the raw medicines of the calamus, the houttuynia cordata and the aloe is (0.001-3): 10-30, and preferably 0.002:30:30 or 0.002:20:30 or 0.002:30:20: 20; the polyol comprises 30% of 1, 2-propylene glycol and 70% of glycerol by mass.
In certain embodiments, the composition is obtained by:
1) pulverizing rhizoma Acori Calami, herba Houttuyniae and Aloe respectively, drying, sieving, weighing at a certain ratio, and mixing;
2) adding ethanol, extracting for 1-3 times at 45-60 deg.C, and collecting extractive solution;
3) cooling the extracting solution obtained in the step 2) to 20-30 ℃, and filtering to obtain primary filtrate;
4) centrifuging the primary filtrate obtained in the step 3), and filtering to obtain secondary filtrate;
5) concentrating the secondary filtrate obtained in the step 4) under reduced pressure to obtain an extract, adding the polyalcohol and the 17-type collagen powder, uniformly mixing, and filtering to obtain the composition.
Further preferably, the drying in step 1) is low-temperature drying, including but not limited to one or a combination of oven drying, freeze drying and vacuum drying.
More preferably, in the step 2), the mass fraction of the ethanol is 30-70%, and more preferably 35%; the mass ratio of the total mass of the raw medicines of the calamus, the houttuynia cordata and the aloe to the ethanol is 1:10-1:30, and the adopted extraction method is the combination of microwave and ultrasonic; the microwave time is 1-3min, and the ultrasonic time is 20-30 min.
Further preferably, the time of centrifugation in the step 4) is 20-60min, and the rotation speed is 5000-10000 rpm.
Further preferably, in the step 5), the addition amount of the polyhydric alcohol is 10-30 times of the mass of the extract.
Further preferably, the step 1) is carried out by sieving with a 60-mesh sieve; filtering by using 100-200-mesh gauze in the step 3); filtering by using a 0.5 mu m microfiltration membrane in the step 4); filtering by using a 0.5 mu m microfiltration membrane in the step 5); the mass fraction of the ethanol is 60%.
The present invention also provides a cosmetic comprising a composition according to any one of claims 1 to 9; the cosmetics include but are not limited to shampoo, hair conditioner, scalp essence, scalp lotion.
Further preferably, the cosmetic further comprises the following components by mass: 0.5-5% of the composition, 0.02% of EDTA-2Na, 4-8% of polyhydric alcohol, 2-4% of fatty alcohol, 0.5-2% of fatty acid, 3-5% of an emulsifier, 35-40% of grease and 0.2-0.4% of a thickening agent.
The cosmetic may also be scalp pre-wash milk, comprising the above composition.
The polyhydric alcohol can be one or more of glycerol, 1,2 pentanediol and 1,3 butanediol, the fatty alcohol can be one or more of cetyl alcohol, stearyl alcohol, behenyl alcohol and myristyl alcohol, the fatty acid can be one or more of stearic acid, myristic acid and 16 acid, the emulsifier can be one or more of glyceryl stearate, PEG-100 stearate, polysorbate-60 and sorbitan sesquioleate, the thickening agent can be carbomer series, and the grease can be one or more of cetyl ethyl hexanoate, isopropyl myristate and octyl dodecanol.
The calamus extract, the houttuynia cordata extract, the aloe extract and the mint extract are all natural herbal medicines, have good antibacterial effect, are synergistic in synergy and are green and safe, and can effectively remove dandruff and oil and improve scalp ecology through scientific proportioning.
Kyoumi Shirai et al found hair follicle associated pluripotent (HAP) stem cells to be present in the hair follicle. The differentiation of the cells is directly related to hair growth. Type 17 collagen plays an important role in differentiation of HAP stem cells. The DNA damage response of HAP hair follicle stem cells induces proteolysis of type 17 collagen (COL17a1/BP180), stimulating aging of hair follicle stem cells, stem cell loss, hair follicle miniaturization associated with androgenic alopecia, and ultimately hair loss. The 17-type collagen can promote the differentiation of hair follicle stem cells again, activate hair follicles fundamentally and promote the regrowth of hairs.
The calamus is also called as acorus calamus, is a traditional Chinese medicine, and is found through literature investigation to contain main chemical components such as sesquiterpene, phenylpropanoid, flavone and the like, and the extract of the calamus has good antioxidant and antibacterial activities. And can inhibit the expression of proinflammatory factors such as interleukin-8, interleukin-6 and the like through various channels, and has better improvement effect on skin health.
Herba Houttuyniae, also called herba Houttuyniae, has main effective component of flavonoid, and most of glycosides existing in glycoside form, mainly comprises quercetin such as rutin, quercitrin, isoquercitrin, renoside, hyperoside, etc., and flavonoid compounds with quercetin as aglycon, and has good biological activity of resisting bacteria and oxidation
The aloe contains barbaloin, alomycin, aloesin, aloarbutin and the like, has good sterilization and anti-inflammatory effects, and polysaccharide substances in the aloe barbaloin have very good moistening and beautifying effects, can effectively reduce the loss of water, and improve the barrier function of the scalp.
The composition also has the following effects that the three extracts are all natural herbal medicines, the antibacterial effect is better, the components are synergistic, green and safe, and the composition can effectively remove dandruff and oil and improve the scalp ecology by scientific proportioning.

Detailed Description
Hereinafter, the present invention is described in more detail and specifically with reference to examples, but the following examples are not intended to limit the present invention.
In the present invention, all the equipment and materials are commercially available or commonly used in the art, and the methods in the following examples are conventional in the art unless otherwise specified.
Example (b): preparation of scalp Care compositions
In the examples and comparative examples of the present invention described below, scalp care compositions were prepared using the following processes.
(1) Respectively crushing the plant raw material medicines, sieving the crushed plant raw material medicines by a 60-mesh sieve, weighing the crushed plant raw material medicines according to the proportion, and uniformly mixing the weighed plant raw material medicines;
(2) adding ethanol for extraction at 50 deg.C for 2min with 2 times of ultrasonic treatment, wherein the mass ratio of the raw materials to ethanol (35%) is 1: 30;
(3) cooling the extracting solution obtained in the step (2) to 25 ℃, and filtering the extracting solution by using 200-mesh gauze to obtain primary filtrate;
(4) centrifuging the primary filtrate obtained in the step (3) for 40min, collecting filtrate at the rotating speed of 6000rpm, filtering with a microfiltration membrane with the pore diameter of 0.5 mu m, and collecting filtrate to obtain secondary filtrate;
(5) concentrating the secondary filtrate obtained in the step (4) under reduced pressure to obtain an extract, adding 1, 2-propylene glycol 2 times the mass of the extract, adding 17-type collagen powder according to a proportion, and stirring or ultrasonically mixing uniformly; the resulting mixture was filtered through a microfiltration membrane having a pore size of 0.5. mu.m, to obtain a composition.
Compositions 1-19 comparative examples 1-4 each of the extract compositions 1-19 was prepared according to the above-described method in the proportions shown in table 1 and the compositions obtained in the proportions were tested for their bacteriostatic effect and safety according to the test methods described below.

Experimental example 1: detection of bacteriostatic effects
Selecting strains: staphylococcus aureus, escherichia coli, pseudomonas aeruginosa, bacillus subtilis and malassezia furfur; the preparation of the bacterial liquid selects pseudomonas aeruginosa, staphylococcus aureus, escherichia coli, bacillus subtilis and malassezia furfur as the strains. Preparation: it was diluted to about 1.0x10 with sterile physiological saline5cfu/ml~1.0x106cfu/ml bacterial suspension. The prepared bacterial suspension can be used within 2h, or can be stored at 2-8 ℃ for no more than 24 h.
And (3) detecting the antibacterial effect: a sterile dried filter paper sheet was used as the composition obtained in the step (5) of example, and 20. mu.L of an aqueous solution (10mg/mL) of the composition at a concentration practically used was dropped onto each sheet. Test bacterium inoculation: taking 1mL of each test bacterial suspension, and uniformly smearing the test bacterial suspension on the surface of a nutrient agar culture medium; the bacteriostatic test samples are attached, one bacterial staining flat plate is attached to each test, 4 filter paper sheets are attached to each dish, 3 filter paper sheets are test sample sheets, and 1 filter paper sheet is blank. A sample piece is attached to the surface of the flat plate by using a sterile forceps, the flat plate is placed in a constant temperature incubator at 36 +/-1 ℃, the result is observed after the flat plate is cultured for 18 hours, and the diameter of the antibacterial ring is measured by using a vernier caliper. The results are the average of 3 measurements.
The experimental results are as follows: the results are shown in Table 2, and when compositions 1-19 are compared to comparative examples 1-3, it was found that:
1) the bacteriostatic activity of the plant compound combination is superior to that of a single plant;
the antibacterial activities corresponding to different proportions of the 3 plant compound combinations are different, and preferably 17 type collagen: calamus: houttuynia cordata: aloe ((0.001-3): 1-30), more preferably 17 type collagen, acorus, houttuynia cordata and aloe 1:30:30:20 or 1:30:20:10 or 1:20:30: 30.

Experimental example 2: promotion of dermal papilla cell viability assay
The dermal papilla cell viability test was performed according to the following test method. The proliferation effect of the composition on hair papilla cells is detected by adopting an MTT colorimetric method.
The specific experimental method is as follows 1X10 in 96well multi plate (corning)4cells/well were inoculated at a density of 100. mu.L each with DMEM medium containing 10% bovine serum and keratinocytes (HHDPC), and the medium was changed to serum-free medium after 24 hours of culture. Samples of composition 5, composition 6, composition 13, composition 17, composition 18, composition 19, aqueous composition solution and 5 μ M minoxidil solution, each at a mass fraction of 1%, were added to serum-free medium and cultured for 24 hours after treatment. Thereafter, the medium was removed, treated with 20. mu.L of MTT solution, and allowed to react at 37 ℃ for 2 hours. To the cells from which the MTT solution was removed, 200 μ L of isopropanol was added, gently shaken for 30min to completely dissolve the crystalline formazan, and the absorbance at 570nm was measured, and the relative cell number was calculated according to the following formula.
The test was performed without adding the sample to the control. The results relating to cell proliferation are shown in FIG. 1.
As shown in figure 1, the composition has the effect of promoting the proliferation of human hair papilla cells, the effect is better than that of the commonly used anti-hair loss pilatory minoxidil in the market, and the effect is in a quantitative relation with the content of 17 type collagen, but the composition is preferably composition 5, composition 6 and composition 13 due to the factors of high price of 17 type collagen, comprehensive cost and the like.
Experimental example 3: security patch test
Dripping 20 μ L of the solution to be detected into a spot tester, wherein the control hole is blank control (pure water); the spot tester with the tested object is attached to the bent side of the forearm of the tested person, and the tested person is lightly pressed by the palm to be uniformly attached to the skin for 24 hours; the skin irritation and sensitization were observed as in table 3 after 30min, 24 hours, and 48 hours, respectively, after removal of the test article plaque test device, and the observation results were recorded.

The experimental results are as follows: the results of the human skin patch tests on compositions 5, 9, 13 are shown in table 4. The result of the skin patch test of the human body shows that 20 people have no adverse reaction to the skin.

Experimental example 4:
A-H was prepared according to the formulation of Table 5 by a conventional method and tested for stability, anti-dandruff antipruritic properties, and anti-alopecia ability.

The anti-dandruff performance test adopts a mode of volunteer sensory evaluation, namely please refer to two groups of 20 volunteers, the volunteers wash hair according to the same method when using the pre-washing milk, and the anti-dandruff effect after using is evaluated in a sensory mode according to the amount and the size of dandruff. The evaluation period is 2 weeks, the evaluation is divided into 6 points, the 6 points are the highest points, the 1 point is the lowest point, and the higher the point is, the better the effect is. The results are shown in Table 6.

Sensory evaluation results show that the scalp care composition prepared by the invention has an obvious anti-dandruff function, and the effect is improved to a certain extent compared with that of the composition which is independently added with plant extracts or common anti-dandruff agents.
Evaluation of stability
The formulations a and D were placed in an environment of-7, 4,25,27,45 ℃ and light for 3 months and the appearance was observed, and the results are shown in table 7, with the degree of change indicated by 0-3, 0 being no significant change, 1 being an acceptable slight change, 2 being an unacceptable change, and 3 being a severe change.

Evaluation of anticreep Effect
Selecting 50 volunteers, wherein 40 male patients and 10 female patients are aged 30-55 years, severe alopecia is 10, 25 moderate patients and 15 mild patients, randomly dividing the 50 patients into 5 groups including blank control groups and experimental groups, respectively using the formulas A, B, D, E and H, and each group has 10 patients, and the ages and alopecia conditions have no significant difference.
The experimental method comprises the following steps: before using the shampoo to clean hair, the volunteers apply the sample on the scalp, slightly massage the scalp for 5min, then wash the scalp with clear water, then use the shampoo to clean the hair once a day, and after insisting on using the shampoo for one month, evaluate the hair loss amount in a sensory mode. And (5) scoring by 1-5 points. The results are shown in Table 8, in which 1 is no effect, 2 is slight effect, 3 is effective, 4 is significant effect, 5 is excellent

Sensory evaluation results show that the scalp care composition prepared by the invention has obvious functions of preventing hair loss and strengthening hair.

Claims (10)
1. A composition for preventing hair loss, fixing hair, removing dandruff and controlling oil is characterized by comprising extracts of calamus, houttuynia cordata and aloe, 17 type collagen and a solvent acceptable in the field of cosmetics;
wherein the extracts of the calamus, the houttuynia cordata and the aloe are ethanol extracts of the bulk drugs of the calamus, the houttuynia cordata and the aloe;
the mass ratio of the 17 type collagen to the raw medicines of the calamus, the houttuynia cordata and the aloe is (0.0001-5): 10-50);
the cosmetically acceptable solvent includes polyhydric alcohols; the polyalcohol comprises one or more of 1, 3-butanediol, 1, 2-propylene glycol, dipropylene glycol and glycerol.
2. The composition of claim 1, wherein the mass ratio of the 17 type collagen to the drug substance of acorus calamus, houttuynia cordata and aloe is (0.001-3): 10-30: (10-30): 10-30), preferably 0.002:30:30:30 or 0.002:20:30:30 or 0.002:30:20: 20; the polyol comprises 30% of 1, 2-propylene glycol and 70% of glycerol by mass.
3. The composition of claim 2, wherein the composition is obtained by:
1) pulverizing rhizoma Acori Calami, herba Houttuyniae and Aloe respectively, drying, sieving, weighing at a certain ratio, and mixing;
2) adding ethanol, extracting for 1-3 times at 45-60 deg.C, and collecting extractive solution;
3) cooling the extracting solution obtained in the step 2) to 20-30 ℃, and filtering to obtain primary filtrate;
4) centrifuging the primary filtrate obtained in the step 3), and filtering to obtain secondary filtrate;
5) concentrating the secondary filtrate obtained in the step 4) under reduced pressure to obtain an extract, adding the polyalcohol and the 17-type collagen powder, uniformly mixing, and filtering to obtain the composition.
4. The composition of claim 3, wherein the drying of step 1) is low temperature drying, including but not limited to one or a combination of oven drying, freeze drying, and vacuum drying.
5. The composition according to claim 3, wherein in step 2), the mass fraction of ethanol is 30-70%, more preferably 35%; the mass ratio of the total mass of the raw medicines of the calamus, the houttuynia cordata and the aloe to the ethanol is 1:10-1:30, and the adopted extraction method is the combination of microwave and ultrasonic; the microwave time is 1-3min, and the ultrasonic time is 20-30 min.
6. The composition as claimed in claim 3, wherein the centrifugation time in step 4) is 20-60min and the rotation speed is 5000-10000 rpm.
7. The composition of claim 3, wherein in step 5), the amount of the polyhydric alcohol added is 10-30 times of the mass of the extract.
8. The composition of claim 3, wherein step 1) is performed by passing through a 60 mesh screen; filtering by using 100-200-mesh gauze in the step 3); filtering by using a 0.5 mu m microfiltration membrane in the step 4); filtering with a 0.5 mu m microfiltration membrane in the step 5).
9. A cosmetic comprising the composition according to any one of claims 1 to 9; the cosmetics include but are not limited to shampoo, hair conditioner, scalp essence, scalp lotion.
10. The cosmetic according to claim 9, comprising the following components, all by mass: 0.5-5% of the composition, 0.02% of EDTA-2Na, 4-8% of polyhydric alcohol, 2-4% of fatty alcohol, 0.5-2% of fatty acid, 3-5% of an emulsifier, 35-40% of grease and 0.2-0.4% of a thickening agent.
