Abstract
本发明为一种光热协同化疗的靶向长循环纳米药物载体的制备方法。一种光热协同化疗的靶向长循环纳米药物载体的制备方法,包括:(1)制备介孔二氧化硅;(2)用聚多巴胺包覆介孔二氧化硅;(3)负载抗癌药物;(4)采用生物素修饰红细胞膜;(5)用生物素修饰后的红细胞膜包裹负载抗癌药物的介孔二氧化硅。本发明通过PDA涂层实现癌症组织局部的光热/化疗协同治疗,提升治疗效率;通过红细胞膜的包裹使其合成的纳米粒子可有效躲避生物屏障,进行全身的血液循环,实现纳米粒子在肿瘤中的高效蓄积;通过修饰生物素来进行主动的靶向进一步提高了其在肿瘤部位的聚集,极大的提高了药物载体的生物相容性、在人体中的循环时间。

技术领域
本发明属于纳米药物载体材料领域,具体涉及一种光热协同化疗的靶 向长循环纳米药物载体的制备方法。
背景技术
世界卫生组织发布的《2018全球癌症报告》显示,我国新增病例数380.4 万例、死亡病例数229.6万例,均高居全球第一,癌症的防控与治疗已上 升为国家战略,具有重大需求。寻找更加有效的癌症治疗方法已经成为需 要攻坚的主要课题。目前,中化疗和放疗是现阶段的两种主流治疗方式, 但其因低效率和严重的副作用限制了它们的进一步发展。
而对纳米药物载体的研究有望解决低效率和严重副作用的问题,它具 有可设计的物理、化学和生物学性能,从而提高药物利用度、改变药物组 织器官分布、提高药物生物安全性。美国2004年就宣布启动“肿瘤纳米技 术”,建立了8个专门从事纳米医学研究的纳米中心。纳米肿瘤治疗通过 化学方法合成纳米材料作为药物载体,降低了药物对人体的副作用,可实 现高效杀死肿瘤细胞的目的。但纳米载体药物进入体内后免疫系统会对其 进行识别,巨噬细胞的吞噬消除使得治疗效率降低,而载体的表面修饰是 实现长循环和提升治疗效率的关键。因此,仿生纳米药物载体得到越来越 多的关注。天然细胞如红细胞、白细胞、血小板等具有的长循环性和靶向 特异性,使其自身在人体内有效循环的同时,在正常条件下不被免疫系统 清除。
大量研究表明,肿瘤细胞的外表面pH显弱酸性,可以利用这种微环境 的特性接入靶向物质,比如带有-NH-的生物素、叶酸等物质。此类物质对 pH的特殊相应性,使得其对肿瘤进行识别,从而达到精准、高效、主动靶 向的目的。
介孔二氧化硅具有较高的孔隙率、高比表面积、热稳定性,载药量大、 结构稳定、生物相容性好、无毒副作用的特点,但其在体内循环时间较短, 不能充分发挥作用,且单一介孔二氧化硅负载后进入人体,经过免疫系统 的清除,只有极少量的纳米粒子在肿瘤组织积累。
长循环纳米载体结合化疗/光热协同治疗是近年来研究的热点,设计一 种高效的纳米载体,满足长循环、缓释、靶向、化疗和光热治疗协同的要 求,提升效率,减少副作用,这对肿瘤治疗具有重要意义。因此,本发明 提出一种新的光热协同化疗的靶向长循环纳米药物载体的制备方法,该药 物载体具有高的生物相容性、长循环以及靶向特性,是提高肿瘤治疗效果 的有效途径。
发明内容
本发明的目的在于提供一种光热协同化疗的靶向长循环纳米药物载体 的制备方法,利用层级化结构设计,获得一种多功能、光热/化疗协同治疗 肿瘤的纳米药物载体,弥补了传统药物利用率低,毒副作用强的缺陷,解 决了药物精准释放的重要问题,通过仿生细胞膜实现了纳米载体的长循环, 达到高效治疗癌症的目的。
为了实现上述目的,所采用的技术方案为:
一种光热协同化疗的靶向长循环纳米药物载体的制备方法,包括以下 步骤:
(1)制备介孔二氧化硅;
(2)采用聚多巴胺包覆介孔二氧化硅;
(3)负载抗癌药物:将所述的步骤(2)得到的包覆后的介孔二氧化硅 和抗癌药物加入到水中,并搅拌12h后,得负载抗癌药物的介孔二氧化硅;
(4)修饰红细胞膜:将二硬脂酰基磷脂酰乙醇胺-聚乙二醇-生物素与 破碎的红细胞膜,在4℃下搅拌12h后,用微型挤出机通过200nm滤膜连续 挤出,得生物素修饰后的红细胞膜;
(5)将所述的生物素修饰后的红细胞膜和所述的负载抗癌药物的介孔 二氧化硅加入到PBS溶液中,充分分散后,用微型挤出机过滤,得到具有 生物素修饰的红细胞膜包裹聚多巴胺包覆的介孔二氧化硅纳米粒子,即所 述的光热协同化疗的靶向长循环纳米药物载体。
进一步地,所述的制备介孔二氧化硅的方法为:
将正戊醇、TEOS加入到环己烷中,搅拌均匀后,加入CTAB和去离子水 并搅拌,再在120℃下反应2h,然后用去离子水洗涤、干燥、煅烧,得白 色粉末的介孔二氧化硅;
所述的采用聚多巴胺包覆介孔二氧化硅的方法为:将碱性Tris HCl溶 液和多巴胺粉末充分混合后,加入介孔二氧化硅,搅拌下反应4小时,离 心洗涤。
再进一步地,所述的正戊醇、TEOS、环己烷、CTAB和去离子水得质量 比为2:4:30:2.44:30;
所述的碱性Tris HCl溶液、多巴胺和介孔二氧化硅的质量比为5:2:1。
再进一步地,所述的干燥温度为70℃,煅烧温度为550℃,煅烧时间 为6h。
进一步地,所述的步骤(3)中,介孔二氧化硅、抗癌药物和水的质量 比为5:1:20;
所述的步骤(4)中,二硬脂酰基磷脂酰乙醇胺-聚乙二醇-生物素与红 细胞膜的质量比为1:5。
进一步地,所述的抗癌药物为抗癌药物DOX。
进一步地,所述的红细胞膜的制备:将红细胞分散在pH为7.4的PBS 中,胀破后,离心分离,得破碎的红细胞膜。
进一步地,所述的步骤(5)中,所述的生物素修饰后的红细胞膜、所 述的负载抗癌药物的介孔二氧化硅和PBS溶液的质量比为1:1:40;
所述的步骤(5)中,采用超声分散处理0.5h。
进一步地,所述的步骤(5)中,用脂质体挤出器过滤后,进行冷冻干 燥。
本发明的另一个目的在于提供一种光热协同化疗的靶向长循环纳米药 物载体,采用上述的制备方法制备而成,是一种多功能,光热/化疗协同治 疗肿瘤的纳米药物载体,红细胞膜的包裹使其合成的纳米粒子可以有效的 躲避生物屏障,进行全身的血液循环,实现纳米粒子在肿瘤中的高效蓄积; 在通过修饰生物素来进行主动的靶向进一步提高了其在肿瘤部位的聚集, 从而极大的提高了药物载体的生物相容性、在人体中的循环时间,靶向作 用提高载体在肿瘤区域的聚集效果,可用于癌细胞中药物的递送。
与现有技术相比,本发明的有益效果在于:
本发明技术方案制得生物素修饰红细胞膜包裹的载药介孔二氧化硅纳 米粒子,具有以下的优点:(1)采用的介孔二氧化硅由正硅酸乙酯和十六 烷基三甲基溴化铵,再通过高温煅烧制得,其具有大的比表面积以及丰富 孔道结构,主要用于提高药物载体的载药量。(2)红细胞膜的包裹使其合 成的纳米粒子可以有效的躲避生物屏障,进行全身的血液循环,实现纳米 粒子在肿瘤中的高效蓄积。(3)生物素的修饰使得药物载体进行主动靶向,进一步提高了其在肿瘤部位的聚集。本发明展现了提高载体生物相容性、 长循环以及在肿瘤部位的聚集,可用于肿瘤部位药物的递送。

具体实施方式
为了进一步阐述本发明一种光热协同化疗的靶向长循环纳米药物载体 的制备方法,达到预期发明目的,以下结合较佳实施例,对依据本发明提 出的一种光热协同化疗的靶向长循环纳米药物载体的制备方法,其具体实 施方式、结构、特征及其功效,详细说明如后。在下述说明中,不同的“一 实施例”或“实施例”指的不一定是同一实施例。此外,一或多个实施例 中的特定特征、结构或特点可由任何合适形式组合。
下面将结合具体的实施例,对本发明一种光热协同化疗的靶向长循环 纳米药物载体的制备方法做进一步的详细介绍:
本发明针对目前临床药物的有效利用率低的问题,致力于设计一种具 有靶向长循环多级仿生纳米药物载体来提高药物有效利用率。目前癌症治 疗中,临床药物面临低效率和副作用严重的问题,纳米药物载体的研究有 望解决此类问题,它具有可设计的物理、化学和生物学性能,从而提高药 物利用度、改变药物组织器官分布、提高药物生物安全性。针对纳米药物 载体承载能力差、免疫清除率高和抗肿瘤效率低的问题,本发明以多级结构为研究对象,利用介孔二氧化硅结构稳定、孔道可调和生物相容性好的 特点,去负载抗癌药物实现缓释,然后使用纳米膜技术将聚多巴胺涂层和 红细胞膜包覆在介孔二氧化硅上,获得光热/化疗协同治疗的同时,减少免 疫系统对介孔二氧化硅的攻击,实现长循环性能。修饰靶向基团后,实现 载体对肿瘤部位的靶向聚集并释药,高效杀死肿瘤细胞,提升效率,减少 副作用。该方法制备工艺简单,具有个性化纳米载体的特点,将为设计具 有实际应用价值的纳米药物载体提供研究思路和依据,主要用于纳米医疗 领域。
聚多巴胺(PDA)含有邻苯二酚基和氨基,使其可以附着在不同类型的 材料表面,并且还具有稳定性良好、厚度可控的性质。结构中的氨基、酚 羟基等活性官能团能够充当活性位点,螯合金属离子或共价修饰分子,进 行光热。在室温、弱碱性条件下,多巴胺及其衍生物会被溶解氧氧化,自 发聚合形成PDA。实验表明,PDA可以黏附在几乎所有材料表面,形成均 匀的纳米薄膜。根据PDA的反应性还可以进一步对材料分子进行修饰,已 有研究证实仿生PDA膜具有优良的生物相容性。因此,可以利用PDA对癌 细胞进行光热治疗,在化疗的协同作用下,提升治疗效率。
具体的技术方案为:
一种光热协同化疗的靶向长循环纳米药物载体的制备方法,包括以下 步骤:
(1)制备介孔二氧化硅;
(2)采用聚多巴胺包覆介孔二氧化硅;
(3)负载抗癌药物:将所述的步骤(2)得到的包覆后的介孔二氧化硅 和抗癌药物加入到水中,并搅拌12h后,得负载抗癌药物的介孔二氧化硅;
(4)修饰红细胞膜:将二硬脂酰基磷脂酰乙醇胺-聚乙二醇-生物素与 破碎的红细胞膜,在4℃下搅拌12h后,用微型挤出机通过200nm滤膜连续 挤出,得生物素修饰后的红细胞膜;
(5)将所述的生物素修饰后的红细胞膜和所述的负载抗癌药物的介孔 二氧化硅加入到PBS溶液中,充分分散后,用微型挤出机过滤,得到具有 生物素修饰的红细胞膜包裹聚多巴胺包覆的介孔二氧化硅纳米粒子,即所 述的光热协同化疗的靶向长循环纳米药物载体。
优选地,所述的制备介孔二氧化硅的方法为:
将正戊醇、TEOS加入到环己烷中,搅拌均匀后,加入CTAB和去离子水 并搅拌,再在120℃下反应2h,然后用去离子水洗涤、干燥、煅烧,得白 色粉末的介孔二氧化硅;
所述的采用聚多巴胺包覆介孔二氧化硅的方法为:将碱性Tris HCl溶 液和多巴胺粉末充分混合后,加入介孔二氧化硅,搅拌下反应4小时,离 心洗涤。
介孔二氧化硅是以十六烷基三甲基溴化铵为模板,在模板的表面沉积 硅源正硅酸乙酯,再通过高温煅烧制得介孔二氧化硅纳米粒子。介孔二氧 化硅具有大的比表面积以及丰富孔道结构,主要用于提高药物载体的载药 量。
进一步优选地,所述的正戊醇、TEOS、环己烷、CTAB和去离子水得质 量比为2:4:30:2.44:30;
所述的碱性Tris HCl溶液、多巴胺和介孔二氧化硅的质量比为5:2:1。
进一步优选地,所述的干燥温度为70℃,煅烧温度为550℃,煅烧时 间为6h。
优选地,所述的步骤(3)中,介孔二氧化硅、抗癌药物和水的质量比 为5:1:20;
所述的步骤(4)中,二硬脂酰基磷脂酰乙醇胺-聚乙二醇-生物素与红 细胞膜的质量比为1:5。

优选地,所述的步骤(5)中,用脂质体挤出器过滤后,进行冷冻干燥。 通过冷冻干燥的方式,可以提高产品的保质时间。
实施例1.
具体操作步骤如下:(所取的物量“份”为重量份)
(1)制备介孔二氧化硅:
将2份正戊醇、4份TEOS(正硅酸乙酯)加入到30份环己烷中搅拌均 匀,加入2.44份CTAB(十六烷基三甲基溴化铵)和30份去离子水,搅拌后 转移到聚四氟乙烯反应釜中。置于120℃烘箱里反应2小时,用去离子水洗 涤三次,再置于70℃空气中干燥一天,得到白色粉末,然后在550℃马弗 炉烧结6h,即可得白色粉末的介孔二氧化硅(MSN)。
(2)采用聚多巴胺包覆介孔二氧化硅:
将5份配置的碱性Tris HCl溶液及2份多巴胺粉末充分混合溶解后, 加入制备好的1份介孔二氧化硅纳米粒子,在高速搅拌下反应4小时,离 心洗涤三次,得到包覆聚多巴胺的介孔二氧化硅纳米粒子(PDA@MSN)。
(3)制备负载抗癌药物DOX的介孔二氧化硅:
将包覆聚多巴胺的介孔二氧化硅纳米粒子和抗癌药物DOX加入到水中 (包覆聚多巴胺的介孔二氧化硅纳米粒子、抗癌药物DOX和水的质量比为 5:1:20),搅拌12h,通过静电作用负载上抗癌药物,得负载抗癌药物的介 孔二氧化硅(PDA@MSN-DOX)。
通过对上清液吸光度的测定,进一步定量负载的DOX在纳米颗粒上的 吸附量。
(4)制备红细胞膜:
全血采自雄性Balb/c小鼠。将采集的红细胞立即分散在PBS(pH=7.4) 中,胀破,离心分离,得到红细胞膜碎片(RBCm)。
(5)对红细胞膜进行生物素修饰:
将1份二硬脂酰基磷脂酰乙醇胺-聚乙二醇-生物素(Bio-PEG-DSPE) 与5份破碎的红细胞膜,在4℃下搅拌过夜,用微型挤出机通过200nm滤膜 连续挤出,得生物素修饰后的红细胞膜(Bio-RBCm)。
(6)生物素修饰的红细胞膜包裹聚多巴胺包覆的介孔二氧化硅负载的 抗癌药物DOX(Bio-RBCm@PDA@MSN-DOX)
将生物素修饰后的红细胞膜(Bio-RBCm)和负载抗癌药物的介孔二氧 化硅(PDA@MSN-DOX)加入到PBS溶液中(生物素修饰后的红细胞膜、负载 抗癌药物的介孔二氧化硅和PBS溶液的质量比为1:1:40),在超声浴中处理 30min,充分分散后,用脂质体挤出器过滤,得到生物素修饰的红细胞膜包 裹聚多巴胺包覆的介孔二氧化硅纳米粒子,即所述的光热协同化疗的靶向 长循环纳米药物载体。生物素修饰红细胞膜是通过脂质体挤出器将膜包裹 与载体的表面,使得纳米粒子分散性良好,同时具有较好的稳定性。
为了提高产品的保质期,可以进行冷冻干燥。
实施例2.
采用实施例1中步骤(1)的方法,在调节多巴胺聚合所需时间及反应 物比例的基础上,制备了包覆聚多巴胺的介孔二氧化硅纳米粒子。
对制备的包覆聚多巴胺的介孔二氧化硅纳米粒子进行载药性能测试, 结果如图1所示,载药量最高可以达到约11%。由于包膜的原因,大孔径的 介孔硅比表面积及孔体积降低,相比于未包膜的介孔硅,载药量有所降低, 在仍具有较高的载药量。调控不同浓度的抗癌药物DOX,最终得到载药量最 大时,DOX的浓度为350μg/mL,药物负载效果好,为以后的药物或其他大 分子、蛋白质等负载提供了可能性。
实施例3.
将实施例1制备的生物素修饰的红细胞膜包裹聚多巴胺包覆的介孔二 氧化硅纳米粒子,即所述的光热协同化疗的靶向长循环纳米药物载体,在 808纳米红外光下进行光热性能的测定。
结果如图2的Bio-RBCm@PDA@MSN-DOX的光热效果曲线所示,在808纳 米红外光下,随着照射时间的延长,纳米粒子浓度越高,溶液升温越迅速, 且计算得出光热转换效率约为40%,说明以该方法得到的纳米粒子具有较强 的光热转换能力,且红外光照射,减少了对生物体的伤害。
在近红外光照下(808纳米),随着时间增长,浓度越高,升温效果越 明显,具有良好的光热性能,可以有效抑制肿瘤生长。
实施例4:测定Bio-RBCm@PDA@MSN-DOX纳米粒子的稳定性。
(1)对实施例1制备的生物素修饰的红细胞膜包裹聚多巴胺包覆的介 孔二氧化硅纳米粒子,即所述的光热协同化疗的靶向长循环纳米药物载体 (Bio-RBCm@PDA@MSN-DOX)进行透射电镜扫描,得到图3,比例尺为200纳 米。
(2)将实施例1制备的Bio-RBCm@PDA@MSN-DOX,在10%FBS溶液下将 纳米粒子充分分散,在制备完成及制备完成后的24小时两个时间点测定纳 米粒子的粒径分布。
结果如图4的Bio-RBCm@PDA@MSN-DOX的Bio-RBCm@PDA@MSN-DOX的纳 米粒子稳定性及粒径分布图所示,粒径分布并没有明显变换,且粒径曲线 呈正态分布,在200纳米以内,符合高效利用纳米粒子的粒径大小,且纳 米粒子可以在体液状态下实现稳定分布存在。
(3)对Bio-RBCm@PDA@MSN-DOX、PDA@MSN-DOX在不同pH的PBS溶液 中进行释药的实验测试,结果如图5所示。左图为Bio-RBCm@PDA@MSN-DOX、 PDA@MSN-DOX在pH=5的PBS溶液中随时间变化的释药情况;右图为 Bio-RBCm@PDA@MSN-DOX、PDA@MSN-DOX在pH=7.4的PBS溶液中随时间变化 的释药情况。由图5可知,本发明制备的Bio-RBCm@MSN-DOX在不同pH下都可以具有释药效果,且释药效果缓慢、长效。
实施例5:测定该核壳结构纳米粒子细胞毒性。
采用不同浓度的实施例1制备的PDA@MSN-DOX、DOX、 Bio-RBCm@PDA@MSN-DOX分别与Hela细胞共孵育时,检测细胞毒性,结果如 图6所示。
由图6可知,对Hela细胞用不同浓度的纳米粒子载药后,随 Bio-RBCm@PDA@MSN-DOX浓度的增加具有较好的杀死肿瘤细胞的效果。因 Bio-RBCm@PDA@MSN-DOX具有靶向效果,细胞存活率(9.48%)低于 PDA@MSN-DOX(12.46%),具有更强的细胞毒性,有效提高药物载体的生物 利用率。
由本发明的实施例可知,本发明利用红细胞膜上CD47蛋白的抗巨噬细 胞识别的能力,制备模仿红细胞的长循环纳米载体。通过调控所加正硅酸 四乙酯及表面活性剂十六烷基三甲基溴化铵的比例,制备大孔介孔硅,实 现纳米粒子的高载药性能。通过引入生物素,利用肿瘤附近酸性微环境实 现纳米粒子的精准靶向。通过以上物质的结合及包覆,锚定,形成宏观尺 度下具有多层级化结构的仿生纳米粒子,使得载药纳米载体具有靶向长循环的特性,同时可以实现药物载体的个性化,降低患者对纳米药物载体的 非特异性免疫。其层级化的结构具有很大的可操作性和特异性性,可以实 现不同患者特定的个性化需求。

Claims (10)
1.一种光热协同化疗的靶向长循环纳米药物载体的制备方法,其特征在于,包括以下步骤:
(1)制备介孔二氧化硅;
(2)采用聚多巴胺包覆介孔二氧化硅;
(3)负载抗癌药物:将所述的步骤(2)得到的包覆后的介孔二氧化硅和抗癌药物加入到水中,并搅拌12h后,得负载抗癌药物的介孔二氧化硅;
(4)修饰红细胞膜:将二硬脂酰基磷脂酰乙醇胺-聚乙二醇-生物素与破碎的红细胞膜,在4℃下搅拌12h后,用微型挤出机通过200nm滤膜连续挤出,得生物素修饰后的红细胞膜;
(5)将所述的生物素修饰后的红细胞膜和所述的负载抗癌药物的介孔二氧化硅加入到PBS溶液中,充分分散后,用微型挤出机过滤,得到具有生物素修饰的红细胞膜包裹聚多巴胺包覆的介孔二氧化硅纳米粒子,即所述的光热协同化疗的靶向长循环纳米药物载体。

所述的制备介孔二氧化硅的方法为:
将正戊醇、TEOS加入到环己烷中,搅拌均匀后,加入CTAB和去离子水并搅拌,再在120℃下反应2h,然后用去离子水洗涤、干燥、煅烧,得白色粉末的介孔二氧化硅;
所述的采用聚多巴胺包覆介孔二氧化硅的方法为:将碱性Tris HCl溶液和多巴胺粉末充分混合后,加入介孔二氧化硅,搅拌下反应4小时,离心洗涤。

所述的步骤(5)中,所述的生物素修饰后的红细胞膜、所述的负载抗癌药物的介孔二氧化硅和PBS溶液的质量比为1:1:40;
所述的步骤(5)中,采用超声分散处理0.5h。
9.根据权利要求1所述的制备方法,其特征在于,
所述的步骤(5)中,用脂质体挤出器过滤后,进行冷冻干燥。
10.一种光热协同化疗的靶向长循环纳米药物载体,其特征在于,所述的靶向长循环纳米药物载体采用权利要求1-9任一项所述的制备方法制备而成。
