Description

一种枯草芽孢杆菌及其在制备抗菌肽、饲料中的应用
技术领域
本发明涉及一株枯草芽孢杆菌及其代谢产物,属于微生物领域。
技术背景
魏氏梭菌又称产气荚膜梭菌(Clostridium perfringens),是临床上气性坏疽病原菌中最多见的一种梭菌。它广泛分布于自然界,存在于很多健康动物的肠道中,当饲养和环境条件改变时,可引起畜禽的产气荚膜梭菌病。在家禽和母猪养殖领域,产气荚膜梭菌是重要的危害动物健康的病原菌,其分为三种等级:当肠道中的产气荚膜梭菌<104CFU/g,不影响动物的生产性能和健康水平,当处于104-106CFU/g,动物处于亚临床状态,没有明显的病理症状,但其生产性能下降,表现为饲料转化率降低、产蛋率降低、母猪繁殖性能下降;当处于>106CFU/g,其表现为临床状态,如精神倦怠、粪便带血、肠道出血和胀气,死亡率高等症状,所以预防肠道中产气荚膜梭菌的过度增殖,对动物肠道健康及提高养殖效益具有重要意义。而传统的方法是在饲料中添加抗生素如恩拉霉素、维吉尼亚霉素等,效果较好,但极容易造成抗生素的残留影响食品安全。近些年,随着规模化养殖的普及和扩大,抗生素的泛滥用和超量添加,给人类健康造成潜在威胁。中华人民共和国农业农村部公告第194号要求,自2020年7月1日起,饲料生产企业停止生产含有促生长类药物饲料添加剂(中药类除外)的商品饲料。开发替代抗生素类新型促生长饲料添加剂成为研究的热点。
枯草芽孢杆菌作为公认安全的益生菌,具有促进动物生长,无药残、绿色环保的饲料添加剂。而一般的枯草芽孢杆菌不产生抗菌肽,无法直接针对产气荚膜梭菌起抑制所用,从而难以达到理想的效果。而近期许多重要研究发现,能够产生抗菌肽的枯草芽孢杆菌具有改善动物健康,降低肠道病原菌数目以及抗感染的作用,是潜在的抗生素替代品(Pathogen elimination by probiotic Bacillus via signaling interference,Nature,2018,562:532–537)。比如,表面活性素(Surfactin)是一种环状脂肽生物表面活性剂,是由β-羟基脂肪酸和7个氨基酸组成的环状脂肽组成,可介导单价和二价阳离子(如钙离子)穿过脂质双层膜的通量。Surfactin可以作为抗微生物佐剂,具有抗菌,抗真菌,抗支原体和溶血作用。iturin通过破坏靶细胞的细胞膜而具有抗真菌感染的作用,同时对革兰氏阳性病原菌具有一定的抑制作用。Mersacidin是上世纪90年代发现的一类具有很强抑菌作用的抗菌多肽,其由21个氨基酸组成,多种革兰氏阳性病原菌具有抑制作用(Gabriele,FEMS Microbiology Letters 127(1995)121-126)。
现有技术中枯草芽孢杆菌产生的抗菌肽单一,大部分对产气荚膜梭菌没有抑制作用或抑制作用较弱,无法对坏死性肠炎起到预防和治疗作用。
发明内容
本发明的目的是提供一种可以分泌特定抗菌肽配方的枯草芽孢杆菌。所述枯草芽孢杆菌及其代谢产物具有抗菌和抗炎症的双重效果。
本发明的目的是通过以下措施实现的:
一种枯草芽孢杆菌,其特征在于:该菌保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏地址是中国北京,保藏日期是2020年11月03日,保藏编号为CGMCCNo.20997。
本发明提供了上述枯草芽孢杆菌在制备抗菌和/或抗炎生物兽药或功能性饲料添加剂中的新用途。
优选的,所述枯草芽孢杆菌在制备抗菌肽中的用途。
本发明所述枯草芽孢杆菌,可以产生3种特定抗菌肽,对产气荚膜梭菌具有很强的抑菌作用,对促进动物生长和降低肠道产气荚膜梭菌数量具有明显的作用,同时还具有抗炎效果。
本发明另一目的在于提供上述枯草芽孢杆菌的代谢产物。
一种抗菌肽组合物,包括表面活性素(Surfactin)、伊枯草菌素(iturin)、Mersacidin。优选的,所述表面活性素(Surfactin)、伊枯草菌素(iturin)、Mersacidin的质量比为15~20:5~10:10~15。具体的,所述枯草芽孢杆菌可至少产生三种抗菌肽Surfactin、iturin、Mersacidin,产量为1.5g/L、0.8g/L和1.4g/L。
上述枯草芽孢杆菌或其发酵产物和/或上述抗菌肽组合物在饲料中的应用。
本发明另一目的在于提供一种饲料。
一种饲料,包含所述枯草芽孢杆菌或其发酵产物和/或所述抗菌肽组合物,所述枯草芽孢杆菌添加量为100-200g/T。可明显降低家禽粪便中产气荚膜梭菌数量,提高饲料转化率。
采用上述的枯草芽孢杆菌制备所述的抗菌肽组合物的制备方法,包括以下步骤:
步骤1:挑取枯草芽孢杆菌(CGMCC No.20997)单菌落3-5个,接种于5mL营养肉汤培养基,培养11-12小时,再转接于100mL营养肉汤培养基,继续培养8小时后,接种于50L发酵罐中。
步骤2:发酵罐培养基为:淀粉3g/L,豆粕粉2g/L,硫酸铵为0.5g/L,磷酸氢二钾0.1g/L,调节pH=6.5-7.0。在发酵罐培养12小时,通过补碱液(20%NaOH)和柠檬酸(25%),保持pH=6.5-7.0,溶氧>85%。
步骤3:将发酵好的培养基放入均质仪中,加入可溶性淀粉20%,麦芽糊精5%、硫酸钠0.5%,均质30min;
步骤4:将均质好的发酵液,进行喷雾干燥,采用仪器为YC-2100中试型真空喷雾干燥机(上海雅程),条件为:进口温度150℃,出口温度90℃,真空度-0.05~-0.08MPa。
有益效果
1.本发明公开了一种枯草芽孢杆菌及其含有三种抗菌肽的代谢产物的制备方法,其可以作为饲料添加剂,可以替代抗生素,促进动物生长,减少坏死性肠炎发病率,提高饲料转化率。在饲料添加剂、生物兽药、食品添加剂方面具有广阔的应用前景。
2、根据目前养殖现场的情况,本发明提供一株枯草芽孢杆菌,可以同时产生3种抗菌肽,对产气荚膜梭菌具有很强的抑菌作用,对促进动物生长和降低肠道产气荚膜梭菌数量具有明显的作用,对坏死性肠炎起到预防和治疗作用,同时还具有抗炎症的作用。
3、本发明提供的枯草芽孢杆菌及其代谢产物的抑菌稳定性十分优秀,高温耐受性好,肠液、胃液中的抑菌活性留存率高。
4、本发明具有提高生产性能并治疗和预防坏死性肠炎等作用,所述枯草芽孢杆菌及代谢产物能提高日增重、降低肉鸡料肉比;在攻毒状态可以抑制家禽肠道产气荚膜梭菌,降低炎症反应,能够改善家禽肠道健康。
5、本发明所述制备抗菌肽组合物的方法简便易操作,工序少劳动强度小,降低了资源和人力成本,适合工业化应用。

下文将结合具体实施方式和实施例,具体阐述本发明,本发明的优点和各种效果将由此更加清楚地呈现。本领域技术人员应理解,这些具体实施方式和实施例是用于说明本发明,而非限制本发明。
在整个说明书中,除非另有特别说明,本文使用的术语应理解为如本领域中通常所使用的含义。因此,除非另有定义,本文使用的所有技术和科学术语具有与本发明所属技术人员的一本理解相同的含义。若存在矛盾,本说明书优先。
下面将结合实施例对本发明的实施方式进行详细描述,实施例中未注明具体条件的,按照常规条件或制造商建议的条件进行。所用试剂或仪器未注明生产厂商的,为可以通过商购获得的常规产品。
实施例
一、抗梭菌枯草芽孢杆菌的筛选:
(1)收集发酵食品、微生态制剂等样品,按照1:100置于营养肉汤培养基中摇床震荡培养12h
(2)将培养好的营养肉汤进行梯度稀释并涂布,涂布营养琼脂平皿长出单菌落30-300个之间,选取差异较大的单菌落,用灭菌牙签将差异较大的单菌落培养于96孔板(孔板中每个孔放置100μL营养肉汤培养基),继续培养12小时。
(3)将培养好的96孔板离心,取上清液,备用
(4)将产气荚膜梭菌CVCC2027在BHI培养基中厌氧培养12小时
(5)将TSC培养基灭菌后,55℃保温,然后将培养好的产气荚膜梭菌按照1:100加入TSC培养基,摇匀,将培养基倾倒于培养皿中
(6)培养皿凝固后,用打孔器打孔,将上清液取50μL滴入孔中,厌氧培养12-16小时,观察抑菌圈情况,能产生明显抑菌圈的小孔的上清液所对应的菌即为可抑制产气荚膜梭菌的目的菌。
(7)试验从2000株菌种,筛选到5株能产生明显抑菌圈的菌,选取产生抑菌圈最大的目的菌进行进一步验证
(8)验证后,其产生抑菌效果稳定,抑菌效果明显
(9)菌株进行全基因组测序和分析,鉴定其为枯草芽孢杆菌。
二、枯草芽孢杆菌及其代谢产物的制备
步骤1:挑取枯草芽孢杆菌(CGMCC No.20997)单菌落3-5个,接种于5mL营养肉汤培养基,培养11-12小时,再转接于100mL营养肉汤培养基,继续培养10小时后,接种于50L发酵罐中。
步骤2:发酵罐培养基为:淀粉(80目)3g/L,豆粕粉(80目)2g/L,硫酸铵为0.5g/L,磷酸氢二钾0.1g/L,调节pH=6.5-7.0。在发酵罐培养12小时,通过补碱液(20%NaOH)和柠檬酸(50%),保持pH=6.5-7.0,溶氧>85%。
步骤3:将发酵好的培养基中加入可溶性淀粉20%,麦芽糊精5%、硫酸钠0.5%,均质30min;
步骤4:将均质好的发酵液,采用仪器为中试型真空喷雾干燥机(上海雅程),条件为:进口温度150℃,出口温度90℃,真空度-0.05~-0.08MPa。
三、全基因组测序和数据挖掘预测抗菌物质
菌株测序在北京诺禾致源科技有限公司进行,细菌基因组二代de novo建库(详细步骤请参阅北京全式金生物技术有限公司产品手册KP201_2021-03-12),HiSeq 4000测序平台,测序策略PE150。测序数据分析:Fastp软件对测序数据执行质量控制、接头删除和质量过滤操做;采用Unicycler软件对上述数据进行组装;组装序列采用Dfast,抗生素和次级代谢产物基因簇分析采用Antismash。
序列分析后,获得surfactin、iturin和mersacidin的完整基因簇(表1)。同时还得到细菌素Bacilleane和Bacillibactin合成相关的部分基因簇,但其通路不完整,故不予考虑。

四、高效液相色谱对抗菌物质的检测
三种抗菌肽氨基酸序列结构如图7。
(1)surfactin:(CAS No.:24730-31-2)
(2)Iturin:(CAS No.:52229-90-0)
(3)Mersacidin:(CAS No.:128104-18-7)
检测方法:将发酵液离心(8000rpm),收集上清液,用HCl调节pH=2.0,置于4℃冰箱8~10小时,离心去上清液,等体积甲醇抽提。根据所参考文献,对surfactin、iturin、mersacidin进行高效液相检测。
Surfactin色谱条件为:色谱柱:COSMOSIL 5AR-II(4.6×250mm);进样量:10μl;柱温:35℃;检测波长:210nm;流速:0.84ml/min:溶剂:A:乙腈(含0.1%TFA)和B:水(含0.1%TFA)。梯度变化条件为:0~9min,A 60%~93%,B 40%~7%;9~20mia,A 93%,B 7%(参考文献:食品科学,2009,30(06):188-190)。
iturin色谱条件为:色谱柱:COSMOSIL 5AR-II(4.6×250mm);进样量:10μl;柱温:30℃;检测波长:210nm;流速:1.0ml/min:溶剂:A:乙腈(含0.1%TFA)和B:水(含0.1%TFA)。梯度变化条件为:0~5min,A 60%,B 40%;5~15min,A 60%~90%,B 40%~10%;15~25min,A 90%,B 10%(参考文献:微生物学通报,2016,43(8):1715-1724)。
Mersacidin色谱条件为:色谱柱:COSMOSIL 5AR-II(4.6×250mm);进样量:10μl;柱温:30℃;检测波长:220nm;流速:1.0ml/min:溶剂:A:乙腈(含0.1%TFA)和B:水(含0.1%TFA)。梯度变化条件为:0~12min,A 95~70%,B 5%~30%;12~16min,A 70%~67%,B30%~33%;16~18min,A 67%~0%,B 33%~100%(参考文献:FEMS MicrobiologyLetters,1995,127:121-126)。
如图1,其提取物的图谱与surfactin对照品(sigma)的图谱保留时间和特征吸收峰一致,说明其代谢产物为surfactin,通过计算,其代谢产物中surfactin含量为1.5g/L。如图2,其提取物的图谱与iturin对照品(sigma)的图谱保留时间和特征吸收峰一致,说明其代谢产物为iturin,通过计算,其代谢产物中iturin含量为0.8g/L。如图3,其提取物的图谱与mersacidin对照品(sigma)的图谱保留时间和特征吸收峰一致,说明其代谢产物为mersacidin,通过计算,其代谢产物中mersacidin含量为1.4g/L。
五、枯草芽孢杆菌及其代谢产物的抑菌稳定性

将枯草芽孢杆菌发酵提取物分别在65、75和85℃处理10min后,与对照(室温)比较抑菌圈直径,计算活性留存率,发现不同温度处理,其活性保持在90%以上,可以耐受高温。

将枯草芽孢杆菌发酵提取物分别人工肠液(人工肠液按照《中华人民共和国药典》(2015年版Ⅳ部)配制:取磷酸二氢钾6.8g,加水500mL使溶解,用0.1mol/L氢氧化钠溶液调节pH至6.8;另取胰酶10g,加水适量使溶解,将两液混合后,加水稀释至1 000mL)处理不同时间后后,与对照(0min)比较抑菌圈直径,计算活性留存率,发现经过90min,其活性留存率95%,说明其能够耐受肠液。

将枯草芽孢杆菌发酵提取物分别人工胃液(人工胃液按照《中华人民共和国药典》(2015年版Ⅳ部)配制:取稀盐酸16.4mL,加水约800mL与胃蛋白酶10g,摇匀后,加水稀释成1000mL)处理不同时间后后,与对照(0min)比较抑菌圈直径,计算活性留存率,发现经过90min,其活性留存率大于90%,说明其能够耐受胃液。
六、枯草芽孢杆菌及其代谢产物在动物生产方面的应用
6.1枯草芽孢杆菌及其代谢产物对肉鸡生产性能和粪便梭菌数的影响
选取1000只1日龄AA肉鸡,随机分为5个处理,每个处理200只鸡。肉鸡饲喂不含抗生素的玉米-豆粕型粉料。试验处理组分为:空白对照组,基础日粮;阳性对照抗生素组,基础日粮+10ppm恩拉霉素,试验组组分别添加100、200、300g/T枯草芽孢杆菌及其代谢产物。

试验发现,不同添加量的枯草芽孢杆菌及代谢产物都能提高日增重、降低肉鸡料肉比,其中,300g/T组与对照组、抗生素组差异明显(P<0.05)。与对照组相比,抗生素组和不同剂量枯草芽孢杆菌及其代谢产物都能显著降低肠道产气荚膜梭菌数量(P<0.05)。试验说明,枯草芽孢杆菌及其代谢产物能降低肉鸡肠道产气荚膜梭菌数量,提高动物健康,从而改善生产性能。
6.2枯草芽孢杆菌及其代谢产物对产气荚膜梭菌攻毒肉鸡生产性能和肠道
选取3000只1日龄ROSS 308肉鸡,随机分为5个处理,每个处理600只鸡。肉鸡饲喂不含抗生素的玉米-豆粕型粉料。试验处理组分为:
空白对照(阴性对照):无任何处理,即正常饲喂,不攻毒,不饮药物;
试验组1:产气荚膜梭菌饮水攻毒(106CFU/mL,攻毒7-14d,下同);
试验组2:产气荚膜梭菌攻毒,日粮中添加亚甲基水杨酸杆菌肽(BND)10ppm;
试验组3:产气荚膜梭菌攻毒,日粮中添加枯草芽孢杆菌及其代谢产物100g/T;
试验组4:产气荚膜梭菌攻毒,日粮中添加枯草芽孢杆菌及其代谢产物200g/T;
产气荚膜梭菌CVCC 2027接种于BHI培养基,37℃厌氧条件下培养18h;然后将发酵液接种于巯基乙酸盐培养基,37℃厌氧条件下培养24h。于7-14日龄,攻读组饮水中添加106CFU/mL,连续饮水7d,空白对照组的肉鸡灌服等体积的0.75%生理盐水。于试验1、35d,每个重复随机抽取50只鸡,测定初重、末重。于试验1、21d,每个重复随机抽取50只鸡,收集粪便,测定粪便产气荚膜梭菌数量,采集血清,测定血清中细胞因子(IL-1β、IL-6、IL-10、TNF-α等):

试验发现,攻毒后,末重、日采食量明显降低(P<0.05),粪便产气荚膜梭菌数量明显增加(P<0.05),而添加抗生素BND和100、200g/T枯草芽孢杆菌及其代谢产物都能明显降低攻毒后的粪便产气荚膜梭菌数量(P<0.05),添加枯草芽孢杆菌及其代谢产物的料肉比明显低于对照组及攻毒组(P<0.05),同时发现,抗生素BND和100、200g/T枯草芽孢杆菌及其代谢产物都能降低炎症因子IL-1β、IL-6和TNF-α的表达量,同时,100、200g/T枯草芽孢杆菌及其代谢产物能明显提高抑炎因子IL-10的表达量,而抗生素BND与攻毒组无差异,这说明枯草芽孢杆菌及其代谢产物能明显抑制肠道梭菌数量,调节肠道健康,同时,具有降低炎症反应,从而改善生产性能。
综上所述,枯草芽孢杆菌(CGMCC No.20997)具有抑制家禽肠道产气荚膜梭菌、改善家禽肠道健康,从而减少坏死性肠炎的发生和提高家禽生产性能。
