Abstract
The invention discloses a composition for improving microcirculation and a preparation method thereof, wherein the composition for improving microcirculation comprises the following components: marine organism complex peptides; chinese medicine fermentation liquor; extracting mushroom liquid; perilla seed oil; astaxanthin and lycopene. The invention is based on the theory of meridian circulation and the theory of health preservation of traditional Chinese medicine, combines the biological extraction technology and the fermentation technology, coordinates three positions of 'dredging, discharging and tonifying', and nourishes qi and blood together, thereby integrally improving the function of a human body circulatory system. Chinese angelica, curcuma zedoary, dendrobium officinale, radix pseudostellariae, roselle and other traditional Chinese medicines are used for preparing fermentation liquor, so that the blood circulation is promoted, the blood stasis is removed, and the normal running of qi and blood is promoted; the antioxidation activity of the mushroom is applied to dissolve fat and clear blood, and the metabolism is accelerated to discharge vivotoxin and blood garbage; the application of the marine organism composite peptide can activate cell regeneration, improve cell activity, repair blood vessel injury, recover blood vessel elasticity, generate blood and nourish blood. (1) pretreatment of raw materials:
sea cucumber: cutting open fresh sea cucumber along abdomen from anus, taking out sea cucumber intestine, squeezing out residue in intestine, cleaning sea cucumber body wall and sea cucumber intestine with clear water, soaking in ice water of 0-4 deg.C 5 times volume twice for 1 hr each time; taking out, draining, weighing, adding distilled water with equal mass, and beating into sea cucumber pulp in a tissue mashing and homogenizing machine;
jellyfish: soaking jellyfish in warm water, washing to remove Alumen and salt, soaking in 50 wt% salt solution, rubbing, repeating for 2-3 times, washing sand out, taking out, draining, cutting into strips, soaking in purified water for 12 hr, taking out, draining, weighing, adding equal mass of distilled water, and pulping in tissue mashing and homogenizing machine to obtain jellyfish pulp;

A method for preparing the composition for improving microcirculation,
mixing and stirring 90-200 parts of traditional Chinese medicine fermentation liquor, 100-360 parts of mushroom extract and 5-13 parts of perilla seed oil uniformly to obtain mixed liquor, preserving the heat at 95 ℃ for 20 minutes, and filtering and sterilizing 500 parts of marine organism composite peptide with a sterile 0.22 mu m microporous filter membrane; adding the marine organism composite peptide subjected to filtration sterilization into the mixed solution, adding 2-6 parts of sterile astaxanthin and 3-5 parts of lycopene, and uniformly stirring and mixing to obtain the composition for improving microcirculation.
Compared with the prior art, the invention improves the functions of the human body circulatory system integrally by coordinating three positions of 'dredging, discharging and tonifying' and nourishing qi and blood by combining the biological extraction technology and the fermentation technology according to the traditional Chinese medicine meridian circulation theory and the health preserving theory. The fermentation liquor is prepared from Chinese medicinal herbs such as angelica sinensis, curcuma zedoary, dendrobium officinale, radix pseudostellariae, roselle and the like, and has the effects of promoting blood circulation, removing blood stasis, clearing and activating the channels and collaterals, nourishing viscera and promoting normal circulation of qi and blood; the antioxidation activity of the mushroom is applied to dissolve fat and clear blood, and the metabolism is accelerated to discharge vivotoxin and blood garbage; the sea cucumber, the sipunculus nudus and the jellyfish are used for preparing the marine organism composite peptide, are rich in various amino acids, various mineral substances and active substances, and can activate cell regeneration, improve cell activity, repair blood vessel injury, recover blood vessel elasticity, generate blood and nourish blood. Step 3) mushroom extract
Smashing and uniformly mixing the fragrant pleurotus eryngii, the honey mushroom and the rhodobacter giganteus in a mass ratio of 1:1: 1; adding 30 times of distilled water, ultrasonic extracting at 50 deg.C and 240W for 30min, filtering under reduced pressure to obtain filtrate, heating the filtrate, evaporating, concentrating to obtain fungus extract with concentration of 100mg/ml and pH of 6-7, and refrigerating at 4-8 deg.C for use.
Step 4)
Mixing Chinese medicinal fermentation liquor, mushroom extract and perilla seed oil at the weight ratio shown in table 1, stirring uniformly to obtain a mixed solution, keeping the temperature at 95 ℃ for 20 minutes, and filtering and sterilizing the marine organism composite peptide by using a sterile 0.22 mu m microporous filter membrane; adding the marine organism composite peptide subjected to filtration sterilization into the mixed solution, adding sterile astaxanthin and lycopene, and uniformly stirring and mixing to obtain the composition for improving microcirculation.

Rat thrombus simulation experiment
1. Experimental Material
Carrageenan, normal saline, 35 healthy Kunming rats, 14 females, 21 males, and 200g + -10 g body weight.
2. Experimental methods
The rats were divided on average into 7 groups of 5 rats each. A carrageenan solution was prepared with physiological saline, and 5mg/kg of carrageenan was injected subcutaneously into the plantar area of hind limb of each rat, and each group of rats was normally fed with the basal diet and observed for 24 hours, during which the process of thrombus formation (length, color) was recorded.
After 24h, group A is a control group, and 0.5ml of normal saline is injected into the abdominal cavity every day; group B is fermentation broth group, and 0.5ml of the fermentation broth of the traditional Chinese medicine of the embodiment 2 is injected into the abdominal cavity every day; group C is marine peptide group, and 0.5ml of marine organism composite peptide of example 2 is intraperitoneally injected every day; group D is mushroom group, and 0.5ml of mushroom extract of example 2 is intraperitoneally injected every day; group E is group 1 of example, i.p. with 0.5ml of the composition of example 1 per day; group F was group 2, i.p. with 0.5ml of the composition of example 2 per day; group G was group 3, i.p. with 0.5ml of the composition of example 3 per day; the injection of each group of rats is continuously carried out for 7 days, during which the rats are normally fed, freely drunk and freely moved, and continuously observed for 14 days, including visual observation of thrombus color, digital display caliper measurement of thrombus length and observation of rat state.

The tail part of each group of rats appears dark red thrombus area after injecting carrageenan for 6 hours, and the thrombus area becomes purplish red after 24 hours, so that the length of each group of rats tends to be stable; starting to inject the solution, and after 7 days, comparing the thrombus color of the A control group and the thrombus color of the B-G group, the thrombus length is shortened, and each sample has a certain repairing effect; the effect of each embodiment is better than that of singly using the traditional Chinese medicine fermentation liquor, the marine compound peptide and the mushroom extracting solution, and the components can be synergistic. The group of example 3 had the best repairing effect, but the rats had an adverse reaction of too fast microcirculation, and considering that the group of example 2 had a repairing effect equivalent to that of the group of example 3, the ratio of the components of the group of example 2 was more suitable.
Rabbit hyperlipidemia and hyperglycemia simulation experiment
1. Experimental Material
A blood glucose test kit, a triglyceride test kit, a cholesterol test kit; the healthy weight of 48 New Zealand white rabbits (female and male) is 2000g +/-100 g.
2. Experimental methods
The first stage is as follows:
after the rabbit is fed adaptively for one week, blood is taken from ear vein in fasting state, and the values of blood sugar, blood cholesterol and blood triglyceride of the rabbit are measured. Randomly 8 (4 males and 4 females) were selected as a normal blank group A, and each was fed with 100g of basal diet each day, with free drinking and free movement. The rest 40 rabbits are fed with 100g of high sugar and high fat feed (25% white sugar, 8% lard, 5% egg yolk powder, and 62% basal feed) per rabbit per day, and fed with free water and free activity for five weeks. After the fifth week of the experiment and fasting, blood was collected from the ear vein, and the values of blood sugar, total cholesterol and triglyceride in the rabbit were measured.
And a second stage:
the rabbits with successful modeling are divided into 5 groups, 8 rabbits in each group are continuously fed with the normal blank group A for two weeks, water is freely drunk, the rabbits freely move, the feeding condition is as follows every day, and the values of blood sugar, blood total cholesterol and blood triglyceride are measured in an empty stomach after the two weeks are over.

The contents of blood sugar, total cholesterol and triglyceride in the blood before modeling and in the normal group are obviously increased by comparing the model group, which shows that the modeling is successful.
Compared with the model group, after C, D, E, F groups of samples are applied for feeding, the blood sugar value, the blood total cholesterol value and the blood triglyceride value are all obviously reduced, and the F embodiment 2 group has the best effect and is obviously superior to the effect of singly using a certain component; the components in the composition, such as traditional Chinese medicine fermentation liquor, marine composite peptide, mushroom extract and the like, have synergistic effect when combined.

Before the test is started, a microcirculation microscopic observer is used for observing the microcirculation capillary vessels of the nail folds of the sub-health volunteers, so that the thickness of the capillaries of the sub-health volunteers is uneven, deformed capillaries can be seen by part of people, the arrangement is disordered, the perfusion is insufficient, the tube loops are blurred, and the blood flow is relatively slow.
Trial process:
(1) volunteers with elevated blood glucose: taking 50ml of microcirculation polypeptide drink in the morning and evening, continuously taking for 14 days, measuring blood sugar value in the morning on empty stomach at 2, 3, 4, 8 and 15 days, and collecting feedback for trial use.
(2) Volunteers with high blood fat: taking 50ml of microcirculation polypeptide beverage in the morning and evening, continuously taking for 14 days, measuring blood cholesterol value and triglyceride value on the empty stomach in the morning on 2, 3, 4, 8 and 15 days, and collecting trial feedback.
(3) Volunteers with stroke sequelae: the microcirculation polypeptide drink is 25ml taken in the morning and evening, and the trial feedback is collected after the microcirculation polypeptide drink is continuously taken for 14 days.
(4) Other volunteers: the microcirculation polypeptide drink is taken 50ml in the morning and evening, and the trial feedback is collected after the microcirculation polypeptide drink is continuously taken for 14 days.
And (4) trial results:
(1) the blood sugar of 10 volunteers with high blood sugar does not feel uncomfortable during the trial period, and the blood sugar of all the volunteers gradually decreases in 3 rd and 4 th days; measuring blood sugar on an empty stomach on day 8, and reducing the blood sugar value of 6 persons to a normal range; on day 15, blood glucose was measured on empty stomach and blood glucose value of 10 persons was reduced to the normal range.
(2) The volunteers with higher blood fat have no uncomfortable feeling during the trial period of 10 persons, and the blood fat level of 10 persons is obviously reduced; on day 8, the cholesterol value of 4 persons was reduced to the normal range, and the triglyceride value of 4 persons was reduced to the normal range; sky 15 abdominal measures 7 human cholesterol was reduced to the normal range and 8 human triglycerides were reduced to the normal range.
(3) Volunteers with stroke sequelae: during use, the pillow has no discomfort, improves sleep quality, and improves symptoms of headache and dizziness; after being taken for 14 days, 6 volunteers with leg and foot fatigue can improve the fatigue symptoms.
(4)30 sub-healthy volunteers: 9 female volunteers felt improvement of qi and blood, and 7 reflected that the menstrual period after administration was smoother than before; the sleep of 10 volunteers staying up all night or insomnia is improved, and the mental state becomes better; the volunteers with body pain, 10 people felt relaxed body after taking the medicine, and 7 people felt reduced pain. A microcirculation microscopic observer is used for observing the microcirculation capillary vessels of the nail folds of 30 sub-healthy volunteers, so that the vessel loops of the visible capillaries are clearer, the perfusion volume is increased, the blood flow is quicker than before, the arrangement disorder condition is improved, and the vascular malformation proportion is reduced.

1. A composition for improving microcirculation comprising the following ingredients:
marine organism complex peptides;
chinese medicine fermentation liquor;
extracting mushroom liquid;
perilla seed oil;
astaxanthin;
lycopene.
2. The composition for improving microcirculation according to claim 1, which comprises the following components in parts by weight:
150 portions of marine organism composite peptide-500 portions;
90-200 parts of traditional Chinese medicine fermentation liquor;
100-360 parts of mushroom extracting solution;
5-13 parts of perilla seed oil;
2-6 parts of astaxanthin;
3-5 parts of lycopene.
3. The composition for improving microcirculation according to claim 2, wherein said marine organism complex peptide is an enzymolytic complex peptide of sea cucumber, jellyfish and Sipunculus nudus;
the marine organism composite peptide is prepared by the following method:
(1) pretreatment of raw materials:
sea cucumber: cutting open fresh sea cucumber along abdomen from anus, taking out sea cucumber intestine, squeezing out residue in intestine, cleaning sea cucumber body wall and sea cucumber intestine with clear water, soaking in ice water of 0-4 deg.C 5 times volume twice for 1 hr each time; taking out, draining, weighing, adding distilled water with equal mass, and beating into sea cucumber pulp in a tissue mashing and homogenizing machine;
jellyfish: soaking jellyfish in warm water, washing to remove Alumen and salt, soaking in 50 wt% salt solution, rubbing, repeating for 2-3 times, washing sand out, taking out, draining, cutting into strips, soaking in purified water for 12 hr, taking out, draining, weighing, adding equal mass of distilled water, and pulping in tissue mashing and homogenizing machine to obtain jellyfish pulp;
sipunculus nudus: thawing frozen Sipunculus nudus, soaking in 50% salt solution, scrubbing, washing out residual sand, taking out, washing with clear water, draining, weighing, adding equal mass of distilled water, and pulping into Sipunculus nudus pulp in a tissue mashing and homogenizing machine;
(2) method for extracting marine organism composite peptide by enzymolysis method
Weighing sea cucumber pulp, Sipunculus nudus pulp and jellyfish pulp in a weight ratio of 3:2:1, adding neutral protease at a pH of 7.0 and a substrate ratio of 5000U/g and a temperature of 50 ℃ for enzymolysis for 5h, and heating and boiling for 5min to inactivate enzyme; centrifuging the zymolyte at 8000r/min for 15min, collecting supernatant, and filtering with 5 μm microporous membrane to obtain large granular tissue; adding 5% active carbon powder, stirring at 60 deg.C for 1 hr, decolorizing, removing fishy smell, centrifuging at 8000r/min for 10min to remove part of active carbon, filtering with microporous membrane, purifying, and fine filtering with ultrafiltration membrane to obtain marine biological compound peptide, and refrigerating at 4-8 deg.C.
4. The composition for improving microcirculation according to claim 3, wherein said fermented Chinese herbal medicine liquid is prepared by the following steps:
(1) preparing the Chinese medicinal powder
3-10 parts of curcuma zedoary, 6-12 parts of angelica sinensis, 3-5 parts of frankincense, 9-30 parts of radix pseudostellariae, 3-9 parts of radix curcumae, 10-15 parts of rhizoma polygonati, 3-10 parts of dendrobium officinale, 10-20 parts of adenophora tetraphylla, 15-30 parts of Chinese yam, 6-12 parts of radix ophiopogonis, 6-15 parts of red-skinned lotus seed, 6-9 parts of andrographis paniculata and 9-15 parts of roselle are placed in a constant-temperature drying oven at 60 ℃ to be dried and crushed, and sieved by a 80-mesh sieve;
(2) culture and activation of cells
Inoculating yeast strain to YPD liquid culture medium with inoculum size of 10%, shake culturing at 28 deg.C and 200r/min for 24 hr, continuously activating for 2 times, and preparing 1 × 109CFU/mL yeast seed liquid;
inoculating lactobacillus strain in MRS culture medium with inoculum size of 5%, standing and culturing in 37 deg.C incubator for 24 hr, continuously activating for 2 times, and preparing 1 × 109CFU/mL lactobacillus seed solution;
(3) preparation of Chinese medicinal fermentation liquor
Taking the traditional Chinese medicine1 part of the powder is added with 3 parts of distilled water and inoculated with 1X 109CFU/mL lactobacillus seed solution and 1 × 109Culturing 5 wt% of each of CFU/mL yeast seed liquid in 30 deg.C and 120r/min constant temperature shaking table for 48h, maintaining the temperature at 80 deg.C for 5min to terminate fermentation, cooling to room temperature, centrifuging at 4 deg.C and 8000r/min for 15min, collecting supernatant, filtering with 0.45 μm microporous membrane to obtain Chinese medicinal fermentation liquid, and refrigerating at 4-8 deg.C for use.
5. The composition for improving microcirculation according to claim 4, wherein said mushroom extract is prepared by the following steps:
smashing and uniformly mixing the fragrant pleurotus eryngii, the honey mushroom and the rhodobacter giganteus in a mass ratio of 1:1: 1; adding 30 times of distilled water, ultrasonic extracting at 50 deg.C and 240W for 30min, filtering under reduced pressure to obtain filtrate, heating the filtrate, evaporating, concentrating to obtain fungus extract with concentration of 100mg/ml and pH of 6-7, and refrigerating at 4-8 deg.C for use.
6. A method of preparing the composition for improving microcirculation according to any of claims 1 to 5, wherein:
mixing and stirring 90-200 parts of traditional Chinese medicine fermentation liquor, 100-360 parts of mushroom extract and 5-13 parts of perilla seed oil uniformly to obtain mixed liquor, preserving the heat at 95 ℃ for 20 minutes, and filtering and sterilizing 500 parts of marine organism composite peptide with a sterile 0.22 mu m microporous filter membrane; adding the marine organism composite peptide subjected to filtration sterilization into the mixed solution, adding 2-6 parts of sterile astaxanthin and 3-5 parts of lycopene, and uniformly stirring and mixing to obtain the composition for improving microcirculation.
