Description

发明背景
脂质氧化是来自不饱和脂肪酸的自由基链式反应的在食品体系(food system)中发生的过程。脂肪酸中不饱和的存在使脂肪酸易受经由这样的自由基反应的氧化。这样的反应是链式反应,包括引发、增殖和终止的步骤。食品中氧化的存在对食品的保质期具有很大的影响,并且可以引起若干异常特征(off note),并且甚至出现有毒产品。
氧化的影响已经被很好地研究。已经发现抗氧化剂可以有效地控制脂质氧化和/或减慢氧化的速率。包括温度、pH、金属的存在、不饱和的水平和/或其他因素的因素可以导致增加的氧化速率。因此,避免这样的因素可以帮助控制氧化,但是控制这样的因素可能是有问题的。因此,抗氧化剂的使用是控制氧化的另一种解决方案。
微量金属是食品体系中的氧化的良好引发剂。螯合剂通过限制体系中存在的催化金属的活性,从而消除最初的氧化步骤,而起到预防性抗氧化剂(preventiveantioxidant)的作用。铁、铜和锌是在许多食品中最常遇到的,并且总是催化氧化中关注的原因。

对脂质氧化具有影响的其他非传统的金属螯合剂是植酸、磷脂和类黄酮。

发明详述
本发明公开了一种包含基于植物的抗氧化剂、有机酸、水、至少一种表面活性剂和非极性溶剂的微乳液。基于植物的抗氧化剂可以是水溶性的或油溶性的。基于植物的抗氧化剂可以是绿茶提取物(green tea extract)、薄荷提取物、迷迭香、苹果、金虎尾、橄榄、姜黄素、柠檬烯、酚酸、酚二萜、类黄酮、挥发油及其任何组合。
至少一种表面活性剂可以是卵磷脂,并且可能包含助表面活性剂。助表面活性剂可以是单酸甘油酯和甘油二酯、聚山梨醇酯、脱水山梨糖醇酯、聚甘油酯(polyglyceril)、蔗糖酯、烷基聚葡糖苷、烷基多糖苷、泊洛沙姆、普郎尼克类(pluronics)、乙氧基化脂肪酸、乙氧基化单酸甘油酯、丙二醇、聚乙二醇酯、十二烷基硫酸钠(SDS)、月桂基硫酸钠(SLS)、皂树皮(quillaja)、鼠李糖脂、槐糖脂、蔗糖酯、聚甘油酯(polyglyceril ester)、皂苷、它们的任何衍生物以及它们的任何组合。

抗氧化剂可以是水溶性的基于植物的抗氧化剂或油溶性的基于植物的抗氧化剂。
有机酸可以是抗坏血酸、柠檬酸、乳酸及其任何组合。
至少一种表面活性剂可以是卵磷脂和助表面活性剂。
非极性溶剂可以是植物油、中链甘油三酯及其组合。
抗氧化剂的功效来自于抗氧化剂在油中被递送的方式。在相似的抗氧化剂浓度,乳化剂共混的抗氧化剂与包含抗氧化剂的纳米分散体行为不同。差异可能来自于颗粒尺寸和表面积的显著变化。
制剂中水的存在还对抗氧化剂的稳定性和功能性具有影响。水溶性的基于植物的抗氧化剂,诸如鼠尾草酸、鼠尾草酚、绿茶提取物、羟基酪醇(hydroxyl tyrosol)和其他抗氧化剂,在水分、空气和温度存在的情况下可能随时间失去它们的功效,这对产品的保质期可以具有很大的影响。
本发明的包含植物提取物的微乳液不仅为生产功能性增加了显著的价值,而且还对于延长并入这样的微乳液的产品的保质期增加了显著的价值。因此,在包封活性成分(即,抗氧化剂、维生素、矿物质、益生菌、颜料、香料和其他功能性营养制品)的产品中,包含基于植物的提取物的微乳液提供良好的优势。
可以被并入到本文描述的微乳液中的抗氧化剂包括但不限于绿茶提取物、薄荷提取物、迷迭香、苹果、金虎尾、橄榄、姜黄素、柠檬烯、酚酸(即,没食子酸、原儿茶酸、咖啡酸、迷迭香酸)、酚二萜(即,鼠尾草酚、鼠尾草酸、迷迭香酚、迷迭香双醛(rosmadial))、类黄酮(即,槲皮素、儿茶素、柚皮素、山奈酚(kaepferol))、挥发性油(即,丁香酚、香芹酚、麝香草酚、薄荷醇)及其任何组合。
微乳液是水、油和表面活性剂的自发形成的体系,并且是光学各向同性和热力学稳定的低粘度溶液。取决于微乳液的组成,微乳液可以形成油包水或水包油的双连续体系。表面活性剂还可以包括助表面活性剂,诸如小链醇(small chain alcohol)或酯。这样的包含自乳化抗氧化剂的微乳液还可以在食品应用中被用于携带生物活性物质(bioactive)。
包含基于植物的抗氧化剂的水溶性的微乳液还可以被配制成是可溶于油的并且保护油免受脂质氧化。
包含基于植物的抗氧化剂的微乳液可以包含来自中链甘油三酯的单酸甘油酯/甘油二酯、卵磷脂、抗坏血酸、绿茶提取物和混合的生育酚。这样的制剂在被包封时在一年内是非常贮存稳定的,其中抗氧化剂的功效没有变化。在有卵磷脂和没有卵磷脂的情况下评估基于植物的提取物微乳液,并且在有卵磷脂的情况下基于植物的提取物微乳液的功能性显著较高。绿茶提取物可以用其他水溶性提取物补充或替代,所述其他水溶性提取物诸如活性多酚,包括但不限于迷迭香提取物或薄荷提取物。这样的基于植物的提取物微乳液在中链甘油三酯中是无限可溶的,这使得活性成分的浓度能够被改变为低剂量。
包含基于植物的抗氧化剂的微乳液允许至少一种水溶性成分在反相微乳液(reverse microemulsion)中容易地溶解在水的核心中。多于一种水溶性成分可以被并入到包含基于植物的抗氧化剂的微乳液中。在包含基于植物的抗氧化剂的微乳液中,可以用最少的水实现水溶性成分的非常高的负载量。这样的包含基于植物的抗氧化剂的微乳液还能够并入具有极端pH和/或盐条件的活性物质。
包含基于植物的抗氧化剂的微乳液还可以充当通用溶剂,其在水体系和油体系中可以具有双重溶解性,这有助于限制不同应用中所需的制剂的数目。包含基于植物的抗氧化剂的微乳液还可以同时保持水溶性成分和水不溶性成分,诸如例如水溶性抗氧化剂和水不溶性抗微生物组分。
含有乳酸钠的、包含基于植物的抗氧化剂的微乳液可以提供抗微生物效果,以及有助于降低由氧化引起的肉制品的变黑。
包含基于植物的抗氧化剂的微乳液也可以不含乙醇、丙二醇和/或聚山梨醇酯。

实施例
实施例1.
制备绿茶提取物微乳液。绿茶提取物(可获自Archer-Daniels-Midland Company,Decatur,IL)是自由流动的粉末,其被标准化为90%的多酚和50%的EGCG(表没食子儿茶素没食子酸酯(Epigallocatechin Gallate))。该产品水溶性非常好。将20%-30%的GTE连同2%-10%的抗坏血酸或柠檬酸溶解在水中,产生微乳液的水相。采用可获自Abitec Corp.的30%-70%的CAPMUL471(一种C8/C10椰子单酸甘油酯/甘油二酯)来制备油-乳化剂相。油相可以包含混合的生育酚(DECANOX MTS 90,可获自Archer-Daniels-Midland Company,Decatur,IL)和大豆卵磷脂(BEAKIN LV1,可获自Archer-Daniels-Midland Company,Decatur,IL)。不同的制剂(A-F)在表1中示出。

测定氧化稳定性指数(OSI)。对于样品B和样品E,110℃时的OSI是30.2,并且与样品A相比,这两个样品具有少5%-15%的GTE,这表明OSI的效力来自于制剂的稳定性。
样品G-样品K采用更高的抗坏血酸浓度来配制,以改进GTE的功效并且增强氧化稳定性。

通过OSI时间,比较了抗氧化剂制剂在菜籽油中以3000ppm剂量的功能性。结果在图1中示出。
对GTE微乳液进行EPR测量,以评估菜籽油在80持续3小时的氧化稳定性。结果在图2中示出。
EPR测量使用Bruker Bio-Spin Corp.(可获自Billerica,MA,USA)台式微型ESR光谱仪进行。分析以2个阶段完成。作为第一步,采用TEMPO(2,2,6,6-四甲基哌啶-1-氧基),一种稳定的自由基,完成校准曲线。通过在室温搅拌将TEMPO溶解在菜籽油中,并且被用作外标。由TEMPO溶液的储备溶液制备在油中从2.5μM-20μM的范围的TEMPO浓度。将这些溶液置于ESR管中,并且测量强度。EPR强度和对应的TEMPO浓度被用于获得校准曲线。已知油自由基+PBN加合物的强度,使用来自Bruker’sMicroESR的ELBA软件得出自由基浓度。
为了评估不同的抗氧化剂在菜籽油中的作用,在使用自旋捕捉剂(spin trap)的情况下遵循以下EPR方法。菜籽油样品添加有以3000ppm的四种不同的抗氧化剂。将40μL的自旋捕捉剂PBN(正叔丁基-α-苯基硝酮)加入到1ml的添加有抗氧化剂的菜籽油中,包括未添加抗氧化剂的对照样品。将每个脂质样品填充在ESR管中,并且放置在80℃的加热块中。每隔15分钟,将每个ESR管放置在MicroESR光谱仪中并且测量强度。继续分析持续3小时的时间段,并且绘制EPR强度/自由基浓度(μM)相对于时间的图,以获得含有抗氧化剂的菜籽油和不含抗氧化剂的菜籽油两者的滞后时间。在T 175(175分钟)时自由基浓度的量的比较提供了研究中抗氧化剂的功效。
图3示出了来自EPR研究的成分的协同作用的效果,所述EPR研究测量在80℃时的菜籽油的自由基浓度。在不存在卵磷脂和混合的生育酚的情况下(样品I),抗坏血酸相对于样品H的少量增加确实有助于降低油中的自由基浓度。然而,EPR和OSI研究两者确实示出了采用卵磷脂、生育酚、绿茶和抗坏血酸获得的协同作用。
实施例2.
将抗氧化剂的两个商业领先的工业标准物与本文公开的包含抗氧化剂的微乳液进行比较。被评估的工业标准物是迷迭香和迷迭香/绿茶提取物抗氧化剂。图4和图5示出了EPR和OSI数据的比较,其示出了工业标准抗氧化剂与本文公开的包含抗氧化剂的微乳液相比的功能性。
实施例3.
在本实施例中,将本文公开的包含抗氧化剂的微乳液与市售抗氧化剂进行比较,以评估大豆油、菜籽油和NuSun油的氧化稳定性。油储存评估根据Schaal烘箱测试在烘箱中进行。这些油在强制通风烘箱中在565℃被储存在半封闭的琥珀色Qorpak玻璃罐中。在固定的时间间隔,从玻璃罐中取出油样品,并且根据AOCS官方方法Cd 8-53测定过氧化值(PV)。在取出之后,样品用氮气覆盖并冷冻,直到准备好用于分析。对过氧化值、脂肪酸、对茴香胺值和OSI进行分析。
用菜籽油获得的OSI值在图6中示出。菜籽油的过氧化值在图7中示出。脂肪酸值在图8中示出。
取决于浓度,绿茶提取物在油中表现出强烈的红色。包含抗氧化剂的微乳液的较高负载能力允许油中非常小的样品夹杂物以保护油。这使得对油实现很好的保护,不受低剂量的影响,但对油没有任何添加的颜色。关于这样的颜色的数据在图9中示出。
用大豆油、菜籽油和Nusun油获得的OSI值在图10中示出。来自对大豆油、菜籽油和Nusun油的Schaal烘箱研究的过氧化值在图11中示出。来自对大豆油、菜籽油和Nusun油的Schaal烘箱评估的对茴香胺值在图12中示出。
总的来说,对于包含抗氧化剂的微乳液,获得良好的稳定性,其中评估了三种油。与市售抗氧化剂相比,本文所述的包含绿茶的微乳液表现出相同或更好的性能,尽管包含绿茶的微乳液是基于水溶性绿茶提取物。
实施例4.
还评估了与在棕榈油和椰子油中的市售迷迭香产品和迷迭香/脂溶性绿茶共混物相比的本文所描述的包含抗氧化剂的微乳液的能力。图13示出了在椰子油中的氧化稳定性,并且图14示出了在棕榈油和棕榈/大豆共混物中的氧化稳定性。
在棕榈油中,包含绿茶的微乳液表现明显好于市售产品。在190ppm的TBHQ(一种合成抗氧化剂)的存在下,棕榈油OSI是63.8。在180ppm的TBHQ的存在下,棕榈油/大豆油共混物OSI是39。图14的包含绿茶的微乳液的这些结果与合成的TBHQ非常相似。
实施例5.
将包含绿茶的微乳液(实施例1的样品A)与迷迭香在载体油(carrier oil)中共混,如图15中所示。图15示出,在包含绿茶的微乳液中并入少量的迷迭香没有示出协同效果,但OSI的总体增加表明它们这些共混物工作良好。
实施例6.
在表3的制剂中,用最小的加热制备包含绿茶的微乳液的澄清溶液。这些溶液被用作储备溶液来制备包含抗坏血酸、卵磷脂和混合的生育酚的抗氧化剂制剂。

如此高浓度的微乳液提供降低油中功能性抗氧化剂的剂量水平的优点。不仅活性成分(例如抗氧化剂)可以被最小化,而且油中的功能非活性成分诸如乳化剂也可以被最小化。
对于具有25%的GTE浓度的样品在菜籽油中的OSI数据在110℃是37小时。在2000ppm剂量的微乳液中的GTE的有效浓度相当于约500ppm的有效GTE。在其他成分的类似的制剂条件和浓度下,与对应于284ppmGTE浓度的样品A相比,在菜籽油中在2000ppm剂量的OSI是27小时。随着较高负载量的10小时增加允许油中抗氧化剂的较低剂量水平。这不仅在生产制剂的经济性上提供了若干益处,因为基于植物的提取物可能非常昂贵。另外的益处是基于用水溶性绿茶为油添加颜色的性质,较低的颜色贡献。
实施例7.
在油中的迷迭香被用来制作微乳液。表5示出了包含迷迭香的微乳液。

实施例8.
矿物质是很好的助氧化剂,并且在氧化过程中可以充当催化剂。通常,金属螯合有机酸诸如抗坏血酸和柠檬酸与其他抗氧化剂组合使用,以便减轻负面影响。用制备成胶束溶液的添加有25μm的铁和氯化锌的菜籽油进行对EPR的评估。抗氧化剂以3000ppm被添加到具有Fe或Zn的菜籽油中,并且PBN被用作自旋捕捉剂。在80℃测量作为时间的函数的自由基浓度持续3小时的时间段,以加速氧化过程。图18和图19示出了本文描述的包含抗氧化剂的含有绿茶的微乳液的效果。与市售迷迭香产品相比,包含绿茶的微乳液在较大程度上有效地控制Fe和Zn的影响。
实施例9.
制备包含在油中的迷迭香和薄荷提取物的微乳液,其具有如表7所示的配方。

实施例10.
抗氧化剂制剂通常被设计用于水溶性体系和油溶性体系两者。为了实现这个,多种制剂被开发,并且基于具体应用交付给市场。然而,理想地,将适用于水基体系和油基体系两者的通用制剂可以提供不仅基于通常所需的多种库存,还基于对抗氧化剂功能性的稳健性的理解的广泛的益处。
水基应用体系具有多种组分,诸如pH、离子强度的变化,以及可以影响脂质组分、蛋白质等的氧化稳定性的添加剂。被设计用于水溶性体系的抗氧化剂制剂通常包含用于抗氧化活性组分的载体体系,诸如丙二醇、中链甘油三酯、植物油等。这样的体系遇到的最常见的问题之一是分散体的稳定性。当油基表面活性剂体系在极端的pH和高盐(盐水)条件下分散时,分散体可以容易地相分离,并且导致对脂质的差的保护。此外,基于食品乳化剂的HLB,存在可以适合食品和饲料类别的有限数目的乳化剂。为了提高制剂的稳定性,需要纳米乳化的体系,其可以耐受高盐和极端的pH条件,但仍形成稳定的分散体。微乳液在改变乳化剂的表面特性的电解质的存在下提供了这样的制剂稳定性并且在水基体系中提供了良好的稳定性。下图示出了微乳液相对于包含迷迭香的纯乳化剂体系的比较。
将具有在油中的20%-40%的迷迭香的包含迷迭香的微乳液与70%的乳化剂基质混合,所述乳化剂基质具有以下配方:20%-70%的YELKIN SS品牌卵磷脂(可获自Archer-Daniels-Midland Company,Decatur,IL);5%-30%的聚山梨醇酯80;2%-10%的抗坏血酸;和10%-30%的乳酸钠。
与为15.6小时的市售的迷迭香/乳化剂共混物相比,本实施例的3000ppm的包含迷迭香的微乳液在110℃在菜籽油中的氧化稳定性是19.8小时。乳酸钠改进了抗微生物功能,并且改进了肉制品的变黑效果。图21示出了与市售迷迭香/乳化剂共混物相比,本实施例(标记为实施例9)的包含迷迭香的微乳液在盐水溶液中分散的能力。
实施例11.
为了证明包括抗坏血酸的不同抗氧化剂的效果,在微量金属铁和锌的存在下进行EPR研究。菜籽油被添加有100μm的Fe和100μm的Zn用于EPR研究。油样品被添加有40μL的PBN,并且运行样品以测量在70℃的强度/自由基浓度。图22和图23示出了在菜籽油中的铁和锌的效果。与市售迷迭香共混物相比,本发明的包含绿茶提取物的微乳液对铁和锌更有效。
实施例12.
为了证明金属在抗氧化剂体系中的分配,制备包含抗氧化剂的微乳液(AOX共混物2),诸如本发明的包含抗氧化剂的微乳液,并且与传统的乳化剂共混物(AOX共混物1)进行比较。在向菜籽油中添加6ppm铁的菜籽油中,使用EPR和OSI方法。抗氧化剂被选择为具有与表8中所示的相同量的活性物质。不存在另外的螯合剂。在微乳液组合物中,甘油是极性溶剂,以便限制另外的水分的影响。

16.权利要求1-11中任一项所述的微乳液在预防或减慢食品/饲料产品或食品/饲料成分中的氧化中的用途。
17.一种使微乳液负载有抗氧化剂的工艺,所述工艺包括:
将抗氧化剂与水混合,从而产生水相;
将非极性溶剂与至少一种表面活性剂混合,从而产生非极性相;以及
将所述水相与所述非极性相混合,从而产生所述微乳液。
18.根据权利要求17所述的方法,其中所述抗氧化剂选自由以下组成的组:绿茶提取物、薄荷提取物、迷迭香、苹果、金虎尾、橄榄、姜黄素、柠檬烯、酚酸、酚二萜、类黄酮、挥发油及其任何组合。
19.根据权利要求17所述的方法,其中所述抗氧化剂是水溶性的基于植物的抗氧化剂。
20.根据权利要求17所述的方法,其中所述抗氧化剂是油溶性的基于植物的抗氧化剂。
21.根据权利要求17-20中任一项所述的方法,还包括将有机酸与所述抗氧化剂和所述水混合。
22.根据权利要求17-21中任一项所述的方法,其中所述至少一种表面活性剂包括卵磷脂和助表面活性剂。
23.根据权利要求17-22中任一项所述的方法,其中所述非极性溶剂选自由植物油、中链甘油三酯及其组合组成的组。
