Description

Repairing and relieving skin care product containing metazoan components and preparation method thereof
Technical Field
The invention relates to the technical field of cosmetics, in particular to a repairing and relieving skin care product containing metazoan components and a preparation method thereof.
Background
The skin has mainly a triple barrier: stratum corneum barrier, microecological barrier and immune barrier, and secondly, skin microecology, which is the subject of current enthusiastic research in the dermatology community, have been increasingly studied for the treatment of skin diseases (atopic dermatitis, acne, rosacea, dandruff, chloasma, sensitive skin, etc.) by correcting the dysbiosis. For example, acne problems and Propionibacterium acnes hyperproliferation have a positive correlation, dandruff is associated with abnormal proliferation of Malassezia, and drugs used to treat these skin problems often have a mechanism of action that inhibits the excessive proliferation of certain harmful bacteria. Therefore, the skin state stability and the skin micro-ecological balance have important relation.
Under normal conditions, skin microorganisms, the external environment and a host are in interaction in dynamic balance, the ecological balance of the skin is maintained, the skin is stimulated when the ecological balance of the facial skin is broken, a series of problems are aggravated or derived if the skin cannot be repaired in time, inflammation is further generated, color spot aging, seborrheic dermatitis, acne, folliculitis and the like are caused, allergic dermatitis can be caused when staphylococcus aureus excessively occupies the surface of the skin, and skin pruritus and stabbing pain are caused. The key point of nursing microecological balance lies in balance, the proportion of any kind of bacteria is not increased, the use of postbiotic bacteria avoids the problems of low bioavailability and unstable effect of the probiotic viable bacteria, the capacity of inhibiting lipid peroxidation and scavenging free radicals for skin cells is obviously improved by adding postbiotic filtrate generated by fermentation and metabolism of various probiotic viable bacteria, and the skin barrier is repaired by reasonably selecting bacteria in a multi-dimensional manner to enhance the skin immunity. The metazoan is different from prebiotics, can directly generate beneficial effect on skin, and refers to inactivated thallus components or metabolites of specific strains in the in vitro culture process.
Chinese patent document CN 110772433A (201911120976.1) is a compound multiple post-growth vitality-activating essence, which comprises the following components in percentage by weight: 8.0-13.0% of butanediol, 2.0-3.0% of caprylic acid/capric acid/succinic acid triglyceride, 2.0-3.0% of polymonosodium glutamate, 0.5-1.0% of Japanese ardisia herb extract, 0.5-1.0% of yeast/rice fermentation product filtrate, 0.5-1.0% of fission yeast mycelium fermentation product filtrate, 0.5-1.0% of lactobacillus/mung bean seed extract/sodium glutamate fermentation product filtrate, 0.5-1.0% of lactobacillus fermentation product, 0.5-1.0% of bacillus/rice bran extract/soybean extract fermentation product filtrate, 0.5-1.0% of streptococcus thermophilus fermentation product, 0.1-0.2% of colloidal silver and 65.3-79.4% of deionized water; the essence is placed in a sealed container, and hydrogen is injected into the essence. The implementation scheme is as follows:
screening was performed by the investigator, meeting enrollment and exclusion criteria, and after signing the volunteer protocol book, the test was attended, similar efficacy cosmetics were discontinued 48h before the test began, and any other activities affecting the screening assessments were performed.
Subjects met inclusion criteria and were enrolled in the study. Similar efficacy cosmetics are not used for 48 hours before the test is started, and on the test day, the test part is cleaned by clear water, and data collection is carried out after the test part is statically seated for 30min in a constant temperature and humidity environment (the temperature is 21 +/-1 ℃ and the relative humidity is 50% +/-10% RH). Collecting facial skin moisture content, transdermal moisture loss, skin elasticity, skin hemoglobin and facial VISIA image, taking data, filling in consumer questionnaire, and distributing sample.
The using method comprises the following steps: after cleaning the face before sleeping, 5g of the facial mask is uniformly smeared on the face for standing for 15-20 minutes, and then the redundant facial mask is wiped off. The sample wafers of example 1, comparative example 2, comparative example 3 and comparative example 4 were selected for efficacy test in consideration of feasibility factors such as product safety, effect and economy, the service cycle was 4 weeks, and skin test was performed according to the return visit cycle. Testing indexes are as follows:
water content of skin stratum corneum: the test was performed 3 times using Corneometer CM825 and the average was taken.
Parameter interpretation: the moisture content of the skin can influence the formation of a water and grease mixed film on the surface of the skin, the barrier function of the skin can be directly influenced, and the larger the measured value is, the higher the moisture content of the skin is.
Transdermal water dispersion loss: the measurements were performed 3 times using a TewameterTM300 and averaged.
Parameter interpretation: the transdermal water loss is generally related to the skin barrier, and a larger value indicates a greater skin water loss, a lower water-locking effect, and a poorer skin barrier.
Facial VISIA image analysis software: VISIA was used to photograph the left, middle and right sides of the face for image analysis.
Parameter interpretation: and analyzing the index data score, wherein the improvement is better when the score is smaller.
Skin elasticity: the skin elasticity R2, Q1 index was tested using a Cutometer MPA 580.
Parameter interpretation: the smaller the measurement value, the poorer the skin elasticity is.
Skin heme test: skin care products can be evaluated for improvement in skin inflammation using mexametrmx 18 to test for skin hemoglobin.
Parameter interpretation: the larger the measurement, the larger the amount of skin erythema and the more severe the inflammation.
And (3) data analysis:
the SPSS software is used to make descriptive statistics on various measurements, including quantity, mean, standard deviation, etc.
Performing pairing T test on all index data, analyzing whether the skin parameters have significant differences before and after using the product, and adopting a significance labeling method: "n.s" indicates no statistical difference, P > 0.05; "+" indicates significant difference, P is more than or equal to 0.01 and less than 0.05; "+" indicates significant difference, 0.001 ≦ P < 0.01, "+" indicates very significant difference, P < 0.001.

Table 4 example 1 sample significant comparison analysis of skin indicators at different periods
Figure BDA0002958081180000162
Note: significance notation method "n.s" indicates no statistical difference, P > 0.05; "+" indicates significant difference, P is more than or equal to 0.01 and less than 0.05; "+" indicates significant difference, 0.001 ≦ P < 0.01, "+" indicates very significant difference, P < 0.001.
The test results for the sample of example 1 were analyzed as follows:
the change in water content of the stratum corneum over time during the test period is shown in table 3, table 4 and fig. 2 a. The moisture content of the skin can influence the formation of a water and grease mixed film on the surface of the skin, can directly influence the barrier function of the skin and participate in the pathogenesis of aged skin and sensitive skin. The subjects showed an upward trend in facial moisture content throughout the test period, with a significant difference of P ≦ 0.001 at 14D and a 28.90% increase in the average gloss value at 28D, indicating that the sample of test example 1 was significantly effective in increasing skin moisture content.
The change in facial transdermal water dispersion over time during the test period is shown in table 3, table 4 and figure 2 b. Compared with the background value, the face cheek roughness shows a remarkable descending trend at 14D and 28D, and the descending degree is maximum at 14D; on day 28, the mean value of the percutaneous water dispersion was reduced by 36.44%. Statistical significance analysis shows that p is less than or equal to 0.001 (shown in Table 2) at day 14, and the difference is very significant.
The change in the red zone data measured by VISIA during the test period is shown in tables 3 and 4, fig. 2c and fig. 3, the red zone reflects the condition of capillary vessels, generally associated with skin inflammation and impaired stratum corneum skin barrier, and the lower score indicates a healthier skin barrier. The improvement in the red zone was significant by 18.28% over the 28 day time range using the sample of example 1. Statistical significance analysis shows that the significance difference is obvious when P is more than or equal to 0.01 and less than 0.05 at 14D and 28D (shown in Table 2).
The time course of hemoglobin during the test period is shown in Table 3, Table 4 and FIG. 2 d. The hemoglobin can be used for analyzing skin erythema change before and after skin care product. The smaller the heme value, the less inflammatory erythema of the skin and the better the skin barrier status. After the sample of example 1 is used, the skin heme shows a remarkable reduction trend in the 28-day time range of the experimental period, and the average value of the heme is reduced by 12.33% (P is more than or equal to 0.001 and less than 0.01) on the 28 th day, and the remarkable difference is remarkable.
In the same test evaluation method, the samples of comparative examples 1 to 4 were tested for 28-day change rate% of skin index, and the results are shown in table 5:
TABLE 5 28-day Change in skin index (%). of comparative examples 1 to 4
Figure BDA0002958081180000171
Figure BDA0002958081180000181
The results of example 1 and comparative examples 1,2 illustrate that: from the change of water content, the change of heme, the change of red area and the change rate of water loss, the improvement effect (moisturizing, red fading and inflammation diminishing repair) of the skin of the embodiment 1 is far higher than that of the comparative example 1 and the comparative example 2, and the red fading and inflammation diminishing repair effect is far better than that of the comparative example 1 and the comparative example 2. The reason is that the selected metazoan component in the embodiment 1 has good ion resistance and wide applicable pH range, and a small amount of low-cost components are added for compounding, so that a good skin improvement effect can be achieved. Compared with the commonly used skin conditioner ingredient, the post-biotic ingredient of the invention is the yeast fermentation lysate of fission yeast, and has the advantages of low cost and good effect.
As can be seen from the example 1 and the comparative examples 3 and 4, the yogurt extract, the calendula extract and the metazoan component are compounded and added into the formula together, so that the synergistic effect is achieved, the skin moisturizing effect is better, the moisturizing performance is improved, and the red fading, inflammation diminishing and repairing effects are better.
The VISIA facial image analysis system and the MPA580 elastic measuring probe are used for smoothly collecting the skin data of the volunteer and analyzing indexes to verify the effects of the product in the aspects of improving the skin elasticity and reducing wrinkles.

Note: significance notation method "n.s" indicates no statistical difference, P > 0.05; "+" indicates significant difference, P is more than or equal to 0.01 and less than 0.05; "x" indicates significant difference, 0.001 ≦ P < 0.01, and "x" indicates very significant difference, P < 0.001.
The test results for the sample of example 1 were analyzed as follows:
the facial wrinkle measurements during the test period are shown in table 6, table 7, fig. 4a and fig. 5. Compared with the initial value, the wrinkles show a descending trend, the average value of the facial wrinkles is reduced by 11.77% on the 28 th day, and the statistical analysis shows that P is more than or equal to 0.01 and less than 0.05, and the remarkable difference is obvious. As can be seen in fig. 5, the fine lines are significantly reduced in the test subjects after the example 1 sample is used, indicating that the example 1 sample has some effect in ameliorating wrinkles, especially fine lines.
The results of the skin elasticity measurements during the test period are shown in table 6, table 7 and figure 4 b. The R2 and Q1 values tended to rise during the test period compared to the initial values. Higher R2 and Q1 values indicate better skin elasticity. When the elastic R2 index is used to 28D, the average value of the elastic R2 index is improved by 16.07 percent (P is more than or equal to 0.01 and less than 0.05), and the obvious difference is achieved.
TABLE 8 comparative examples 1-4 sample skin elasticity and 28 day wrinkle Change Rate%
Skin wrinkles change% Skin elasticity R2 Change% Rate of change in skin elasticity Q1%
Comparative example 1 -7.36% 9.28% 9.23%
Comparative example 2 -7.8% 9.17% 9.09%
Comparative example 3 -6.2% 5.42% 8.25%
Comparative example 4 -3.6% 3.86% 6.36%
From the results of example 1 and comparative examples 1 and 2, it can be seen that: example 1 is shown by skin wrinkle change, skin elasticity R2 change and skin elasticity Q1 change rate, which are all much higher than those of comparative example 1 and comparative example 2, namely, the wrinkle removal effect and the skin elasticity enhancing effect of example 1 are much better than those of comparative example 1 and comparative example 2. The single strain fermentation product can not solve the problem of multidimensional skin and has limited exertion effect.
As can be seen from the example 1 and the comparative examples 3 and 4, the yogurt extract, the calendula extract and the metazoan component are compounded and added into the formula together, so that the synergistic effect can be achieved, the coarse lines are changed into fine lines, the facial wrinkles are changed more obviously, the skin elasticity is improved, and the skin aging is delayed, which shows that the efficacy after compounding is superior to that of a single component.
Through testing on the moisture content of the stratum corneum, the transdermal water dispersion amount, the heme content, the skin elasticity and the facial VISIA analysis system, the sample in the test example 1 has different degrees of improvement effects on volunteers in a test period, so that skin wrinkles and inflammations are reduced, the elasticity and the moisture content are increased, and the test sample has certain effects of repairing, relieving and tendering the skin.
As can be seen from the analysis of the water content of the facial cuticle and the transdermal water dispersion loss data, the water content of the cuticle is in an increasing trend and the transdermal water dispersion loss is in a decreasing trend compared with the initial value in the testing period. The test example 1 shows that the moisture content of stratum corneum is obviously increased and the transdermal water loss is obviously reduced after the sample is used, and the test sample has certain effects of repairing and tendering skin.
The analysis of the heme data and the skin red zone data shows that the skin heme content and the red zone show obvious descending trend compared with the initial value in the test period. After the test example 1 sample is used, the content of heme and the red area index are obviously reduced, which indicates that the test sample has a certain relieving and repairing effect on skin.
Through VISIA facial image analysis and skin elasticity numerical analysis, facial wrinkles are in a descending trend and skin elasticity is in an ascending trend compared with an initial value in a test period. After the sample of the test example 1 is used, fine lines of the volunteers are obviously reduced, and the elasticity of the skin is increased, which shows that the test sample has certain effects of improving the elasticity of the skin, reducing wrinkles and delaying aging.
