Abstract
The invention provides skin lotion containing an anti-aging composition and a preparation method thereof. The preparation raw materials of the emollient water containing the anti-aging composition comprise the anti-aging composition, a skin conditioner, a humectant and water; the anti-aging composition comprises a ginger root extract, a scutellaria root extract and a cacao seed extract; the skin conditioning agent comprises lactobacillus fermentation lysate, carrageen crispus extract and acetyl chitosamine. In a second aspect, the present invention provides a method of preparing a lotion comprising an anti-aging composition according to the first aspect, the method comprising the steps of:
(1) mixing the humectant with water, heating and stirring to obtain a water phase mixed solution;
(2) and (2) cooling the water phase mixed liquor obtained in the step (1), and mixing with the anti-premature composition and the skin conditioner to obtain the skin lotion containing the anti-premature composition.
Preferably, in the step (1), the temperature for heating and stirring is 80 to 85 ℃, for example, 80 ℃, 81 ℃, 82 ℃, 83 ℃, 84 ℃, 85 ℃ and the like.
Preferably, in step (1), a thickener and a chelating agent are also added.

(1) the components in the anti-aging composition and the skin conditioner are synergistic, so that the fragile and sensitive state of the skin caused by the external environment can be effectively reduced, the resistance of the skin is enhanced, the elasticity of the skin is improved, wrinkles are reduced, the skin is reddened, the moisture of the skin is increased, and the skin is kept in a healthy state of gloss and fineness;
(2) the skin lotion provided by the invention regulates the metabolism of skin through mutual matching and synergistic interaction of all components, can promote the synthesis of extracellular matrix, improve the skin hydration degree, reduce the percutaneous water loss, reduce skin fine lines, improve the skin roughness, inhibit the activity of tyrosinase, reduce the generation of melanin, improve the uneven skin color and brighten the skin color.
Detailed Description
The technical solution of the present invention is further explained by the following embodiments. It should be understood by those skilled in the art that the examples are only for the understanding of the present invention and should not be construed as the specific limitations of the present invention.
Each of the components in the following preparation examples is commercially available.
Preparation example 1
The preparation example provides a curcuma alismatifolia root extract, which is prepared by the following preparation method:
(1) freezing root of Zingiber officinale Roscoe at-15 deg.C for 18h, pulverizing at-2 deg.C, and sieving to obtain 150 mesh Zingiber officinale Roscoe root powder;
(2) mixing the ginger flower root powder obtained in the step (1) with n-butanol, leaching for 2 hours at 25 ℃, and concentrating the solution obtained by leaching to obtain a concentrated solution; mixing the concentrated solution with ethanol, leaching at 40 deg.C for 1 hr, and filtering to obtain crude extract of rhizoma Zingiberis recens;
wherein the mass ratio of the ginger flower root powder to the n-butanol to the ethanol is 1:10: 8;
(3) and (3) passing the crude extract of the ginger flower roots obtained in the step (2) through macroporous adsorption resin AB-8, washing with deionized water, removing water solution, washing with 60 wt% ethanol water solution, collecting the washed ethanol water solution part, and concentrating to obtain the ginger flower root extract.
Preparation example 2
The preparation example provides a radix scutellariae extract, which is prepared by the following preparation method:
(a) pulverizing radix Scutellariae, and sieving to obtain 200 mesh radix Scutellariae powder;
(b) mixing the radix scutellariae powder obtained in the step (a) with an ethanol water solution with the concentration of 60 wt% according to the mass ratio of 1:10, performing microwave extraction for 8min at the power of 600W, and concentrating to obtain a radix scutellariae crude extract;
(c) dissolving the crude extract of radix scutellariae obtained in the step (b) by using 10 wt% of ethanol water solution, purifying by using a macroporous resin gel column D101, eluting by using 60 wt% of ethanol water solution, and concentrating to obtain the radix scutellariae extract.
Preparation example 3
The present preparation example provides a cocoa seed extract prepared by the following preparation method:
(A) pulverizing cacao seed to particle size of 100 mesh, mixing with n-hexane at a mass ratio of 1:10, soaking at 30 deg.C for 3 hr, filtering, and drying to obtain defatted cacao seed powder;
(B) mixing the defatted cocoa seed powder obtained in the step (A) with water, heating to 45 ℃, mixing with biological enzyme and acid, wherein the mass ratio of the defatted cocoa seed powder to the water to the cellulase is 1:15:0.006, carrying out enzymolysis reaction for 1.5h at 45 ℃ and pH of 5, and then inactivating to obtain a cocoa seed enzymolysis product;
(C) mixing the cocoa seed enzymolysis product obtained in the step (B) with 60 wt% ethanol water solution according to the mass ratio of 1:10, performing ultrasonic extraction for 50min at the power of 300W, filtering, and concentrating to obtain the crude extract of cocoa seeds.
Except that the ginger flower root extract, the scutellaria root extract and the cacao seed extract were prepared by the above-mentioned preparation methods, the remaining components in the following examples and comparative examples were all commercially available.

Test example 1
Safety performance testing
Safety performance tests were performed on the skin lotions provided in application examples 1-9 and on the skin lotions provided in comparative application examples 1-6, as follows:
(1) haemolysis test of erythrocytes
Preparation of erythrocyte suspension: selecting healthy rabbits, taking 9mL of blood from heart, adding 1mL of 2% potassium oxalate solution, centrifuging, discarding supernatant, diluting the precipitate to 20mL with 20mmol/L PBS solution, and storing at 4 ℃ for later use. As shown in the test data in Table 3, the moisturizing rate of the lotion containing the anti-aging composition is more than 88% after 4 hours; the change rate of percutaneous water loss is maintained below 14 percent; the water content of skin is above 45% after 4 hr. Therefore, the skin moisturizer disclosed by the invention can promote the synthesis of extracellular matrix, stabilize the self moisture and lipid barrier of skin, keep the cell moist, improve the skin hydration degree, reduce the loss of percutaneous moisture, ensure better skin absorbability, be beneficial to the damage repair of skin and nourish the skin.
Test example 3
Anti-wrinkle efficacy test
The anti-wrinkle efficacy test was performed on the emollient water containing the anti-aging composition provided in application examples 1-9 and on the emollient water provided in application examples 1-6, and the method was as follows:
(1) elastin production promotion test: inoculating human fibroblasts on a 96-well plate according to the density of 20000 cells per well, adding a culture medium, culturing for 24h in an incubator at 37 ℃, replacing a new culture medium after the cells adhere to the wall, respectively adding each test sample composition (controlling the final concentration of the sample to be 50mg/mL), taking a PBS solution of 20mmol/L as a blank control, and then culturing for 2 days in the incubator at 37 ℃;
(2) clinical anti-wrinkle efficacy testing: 150 healthy 18-55 year old volunteers were selected and randomized into 15 groups of 5 males and 5 females, and the wrinkles on the face of the subjects were blotted with silicone resin and applied to the wrinkles on the face of the subjects 2 times a day for 60 consecutive days. Wrinkle imprints were taken from the test sites of the subjects 30 days and 60 days after treatment, respectively, and wrinkle depth was measured with a confocal microscope. Calculating the percentage of wrinkle depth reduction after 30 and 60 days of treatment;
the specific test results are shown in table 4:

As shown in the test data in Table 4, the emollient water containing the anti-aging composition can promote the expression of elastin, and the expression level of elastin is more than 4.5; the wrinkle depth reduction value reaches more than 14% after 30 days of use, and the wrinkle depth reduction value reaches more than 18% after 60 days of use, which shows that the skin lotion containing the anti-aging composition supplements each other, has a synergistic effect, can adjust the oxidation-reduction balance of cells, adjust autophagy of cells, promote the growth of cells, promote the synthesis and repair of collagen and fibronectin, improve the symptoms of cracks, and further lighten fine lines; can quickly consume older protein and unstable peptide chain, eliminate sunken wrinkles and tighten skin, thereby essentially repairing fine lines of the skin.
Test example 4
Anti-aging and brightening effect test
The anti-aging and brightening effect test is carried out on the emollient water containing the anti-aging composition provided in the application examples 1-9 and the emollient water provided in the comparative application examples 1-6, and the method comprises the following steps:
150 healthy 18-55-year-old volunteers were selected and randomly divided into 15 groups of 5 males and 5 females, each group using the samples of each group on the face, 1 time each day, morning and evening, for a total of 4 weeks.
(1) The skin melanin content changes before and after application of the mask composition were evaluated by a skin melanin tester (Hexameter MX 18). The measurement range of the used instrument is 0-999, and the higher the measurement value, the higher the melanin content in the skin is. The amount of melanin reduction on the skin of the front and back of a subject using the mask composition prepared according to the present invention;
(2) VISIA skin assay evaluation: after the face of the tested volunteer is cleaned, a VISIA facial image analyzer is used for carrying out facial shooting, the detection data of facial pigment spots, textures and wrinkles are analyzed, and the phenomenon that the pigment spots and the textures are more obvious when the numerical value is higher is represented. After the treatment course is finished, comparing the information obtained by the VISIA facial image analyzer with the information before treatment, and analyzing the improvement condition of facial pigmented spots, textures and wrinkles of the tested volunteers, wherein p is less than 0.02, and the statistical significance is achieved;
(3) the brightening effect is as follows: the skin color difference tester (probe CL400 and multi-probe skin test system MPA10) was used to test the skin color of the face of the test volunteers before and after treatment, respectively, and the ITA was calculated. The test conditions are a constant temperature and humidity environment with the temperature of 22 ℃ and the humidity of 50 percent.
The specific test results are shown in table 5:

As can be seen from the test data in table 5, by comparing the results of the VISIA test before and after treatment, the melanin reduction of the subject is up to 54% or more, the pigmented spots are reduced by 19% or more, the texture reduction is up to 23% or more, and the brightening efficacy is up to 24% or more after the skin moisturizing lotion provided in application examples 1 to 9 is used. The reduction of melanin of a tested subject is obvious, the skin color spots and the texture of the tested volunteer are obviously improved, the brightening effect of the facial skin color is obvious, and the curative effect evaluation is effective. Therefore, the skin lotion provided by the invention has reasonable compatibility of components, has a synergistic effect, can promote the synthesis of extracellular matrix, improve the skin hydration degree, reduce the percutaneous water loss, reduce the skin fine lines, improve the skin roughness, inhibit the activity of tyrosinase, reduce the generation of melanin, improve the uneven skin color and brighten the skin color.
The applicants state that the present invention is illustrated by the above examples to provide a lotion containing an anti-aging composition and a method of making the same, but the present invention is not limited to the above examples, which is not meant to imply that the present invention must be practiced by the above examples. It should be understood by those skilled in the art that any modification of the present invention, equivalent substitutions of the raw materials of the product of the present invention, addition of auxiliary components, selection of specific modes, etc., are within the scope and disclosure of the present invention.
Claims (10)
1. The lotion containing the anti-aging composition is characterized in that the lotion containing the anti-aging composition is prepared from the raw materials of the anti-aging composition, a skin conditioner, a humectant and water;
the anti-aging composition comprises a ginger root extract, a scutellaria root extract and a cacao seed extract;
the skin conditioning agent comprises lactobacillus fermentation lysate, carrageen crispus extract and acetyl chitosamine.
2. The emollient water containing anti-aging composition according to claim 1, wherein in said anti-aging composition, the mass ratio of said ginger root extract, scutellaria root extract and cacao seed extract is (3-8): (5-15): (2-7);
preferably, in the skin conditioner, the mass ratio of the lactobacillus fermentation lysate, the carrageen crispus extract and the acetyl chitosamine is (1-5): (0.1-1).
3. The lotion containing the anti-aging composition according to claim 1 or 2, wherein the lotion containing the anti-aging composition is prepared from 0.01 to 5% by mass of the anti-aging composition, 0.01 to 10% by mass of a skin conditioner, 5 to 25% by mass of a humectant, and the balance of water, based on 100% by mass of the raw materials for preparing the lotion containing the anti-aging composition.
4. The emollient water containing anti-aging composition according to any of claims 1-3, wherein said skin conditioner further comprises any one or a combination of at least two of Sophora flavescens root extract, Glycyrrhiza inflate root extract, niacinamide, alpha-arbutin, yeast/Sichuan millet fermentation product filtrate, yeast dehiscence product lysate, animal umbilical cord extract, animal placental protein, animal placental enzyme or cajeput oil;
preferably, the skin conditioner comprises 1-5% of lactobacillus fermentation lysate, 0.1-1% of carrageen crispus extract, 0.1-1% of acetyl chitosamine, 0.01-0.1% of sophora flavescens root extract, 0.01-0.1% of glycyrrhiza inflate root extract, 0.1-1% of nicotinamide, 0.05-0.3% of alpha-arbutin, 0.01-0.1% of yeast/Sichuan millet seed fermentation product filtrate, 0.01-0.3% of schizosaccharomyces cerevisiae fermentation product lysate, 0.01-0.1% of animal umbilical cord extract, 0.01-0.1% of animal placental protein, 0.01-0.1% of animal placental enzyme and 0.01-0.1% of melaleuca oil by mass of 100% of the raw materials for preparing the emollient water containing the anti-premature aging composition.
5. The emollient water containing anti-aging composition according to any of claims 1 to 4, wherein said humectant comprises any one or combination of at least two of butylene glycol, glyceryl polyether-26, trehalose, sodium hyaluronate or 1, 2-hexanediol;
preferably, the moisturizer comprises 5-10% of butanediol, 261-5% of glyceryl polyether, 0.1-2% of trehalose, 0.05-0.2% of sodium hyaluronate and 0.1-1% of 1, 2-hexanediol by mass of 100% of raw materials for preparing the emollient water containing the anti-aging composition.
6. The emollient water containing the anti-aging composition according to any of claims 1 to 5, wherein the raw materials for preparing the emollient water further comprise any one or a combination of at least two of a thickener, a pH regulator, a chelating agent, an antioxidant, a solubilizer or a fragrance;
preferably, the raw materials for preparing the emollient water containing the anti-aging composition further comprise 0.01-0.2% of thickening agent, 0.05-0.15% of pH regulator, 0.01-0.05% of chelating agent, 0.1-1% of antioxidant, 0.01-0.05% of solubilizer and 0.01-0.05% of aromatic, wherein the mass of the raw materials is 100%.
7. The emollient water containing anti-aging composition according to claim 6, wherein said thickener comprises hydroxyethylcellulose and/or carbomer;
preferably, the pH adjusting agent is arginine;
preferably, the chelating agent is disodium EDTA;
preferably, the antioxidant is p-hydroxyacetophenone;
preferably, the solubilizer is PEG-40 hydrogenated castor oil;
preferably, the fragrance is a perfume.
8. A method of preparing a lotion comprising an anti-aging composition according to any of claims 1-7, wherein the lotion is prepared by a method comprising the steps of:
(1) mixing the humectant with water, heating and stirring to obtain a water phase mixed solution;
(2) and (2) cooling the water phase mixed liquor obtained in the step (1), and mixing with the anti-premature composition and the skin conditioner to obtain the skin lotion containing the anti-premature composition.
9. The method for preparing the anti-aging composition-containing emollient water according to claim 8, wherein in step (1), the temperature of the heating and stirring is 80-85 ℃;
preferably, in the step (1), a thickening agent and a chelating agent are also added;
preferably, in the step (2), the mixing is specifically: cooling the water phase mixed liquor obtained in the step (1) to below 60 ℃, adding a pH regulator and an antioxidant, uniformly stirring, cooling to below 45 ℃, adding an anti-aging composition, a skin conditioner, a solubilizer and an aromatic, and uniformly stirring.
10. The method for preparing the anti-aging composition-containing emollient water according to claim 8 or 9, wherein said anti-aging composition-containing emollient water is prepared by the steps of:
(1) mixing humectant, water, thickener and chelating agent, heating and stirring at 80-85 deg.C to obtain water phase mixed solution;
(2) cooling the water phase mixed liquor obtained in the step (1) to below 60 ℃, adding a pH regulator and an antioxidant, uniformly stirring, cooling to below 45 ℃, adding an anti-aging composition, a skin conditioner, a solubilizer, a preservative and an aromatic, and uniformly stirring.
