Description
Technical Field
The invention belongs to the field of cosmetics, and particularly relates to a cosmetic composition containing glycerol glucoside, and a preparation method and application thereof.
Background
The information in this background section is only for enhancement of understanding of the general background of the invention and is not necessarily to be construed as an admission or any form of suggestion that this information forms the prior art that is already known to a person of ordinary skill in the art.
The dermis layer 75% of the skin is composed of collagen, and the fibrous collagen forms a net structure to provide tension and elasticity to the skin; at the same time, collagen in the dermis is also the nutrient supply site for the epidermis layer and epidermal appendages (hair), and delivers moisture to the epidermis. The important role of collagen on skin is therefore apparent, and skin growth, repair and nutrition are not well separated from collagen. The collagen can make the cells plump, so as to make the skin plump, keep the skin elastic and moist, and maintain the skin fine and smooth. Anti-wrinkle and moisturizing of skin are both associated with collagen.
The spirulina has wide application value in cosmetics. Because the spirulina contains vitamin mineral group, the spirulina can repair the skin, supplement the skin nutrition and make the skin glowing. Such as algal polysaccharides, glycerol glucosides, gamma-linolenic acid, and the like. Glycerol Glucoside (glycocide) is a glycoside compound formed by glycerol and a glucose molecule through a glycosidic bond. The glycerol glucoside can promote the synthesis of aquaporins, maintain osmotic balance, bring required moisture to skin cells, prevent inflammation, and also has obvious effects on improving skin elasticity, reducing skin redness, repairing barriers and reducing crow's feet. Glycerol glucosides have been used as cosmetic raw materials for health and beauty purposes.
The current glycerol glucoside is applied to moisturizing, anti-aging and anti-inflammatory cosmetic products, but only can achieve single effect. Therefore, the glycerol glucoside cosmetic composition can simultaneously play the roles of promoting collagen synthesis, delaying senility, reducing skin inflammation, moisturizing and the like in a short time.

Example 1
Table 1 formulation 1 composition
A toning lotion was prepared according to the formulation in table 1, and the preparation method of the toning lotion comprises the steps of:
1) sequentially adding the phase A raw materials into a main container, and heating to 80 ℃;
2) stirring until no insoluble substance is present, keeping the temperature for 10min, and cooling;
3) cooling to 20 deg.C, adding phase B, stirring until there is no insoluble matter
4) Mixing with the phase C raw material, stirring to be uniform and transparent, slowly adding into the main container, and stirring to be uniform;
5) cooling to room temperature, filtering with 400 mesh filter cloth.
Testing of the properties of the lotions:
1. type I collagen content detection test
Cells in logarithmic growth phase were collected at a cell density of 1.0X 105One/well was inoculated into 12-well plates. In an incubator (37 ℃, 5% CO)2) After 24h of medium incubation, the drug to be tested (formula 1 lotion) was added to it, with 30 μmol/L ascorbic acid (VC) as a positive control, untreated cells as a Negative Control (NC), and reaction system reagents and solvents as solvent controls, with 4 replicate wells per set.
Adding the drug into an incubator (37 ℃, 5% CO)2) Culturing for 72h, and collecting the culture solution. And detecting the content of the type I collagen in the culture solution by adopting a Col IELISA kit.
TABLE 2
The negative control is the type I collagen content in the cells themselves without any component. The positive control is a control which can increase the collagen type I content in the cells after adding a component, which means an effective control. Formula 1 is the formula in table 1, and the content of type I collagen in cells can be much higher than that of a positive control, which indicates that the effect is better.
As can be seen from Table 2, the lotion of formula 1 in Table 1 was effective in achieving anti-aging benefits.
2. Test for quick repair and wrinkle reduction of fine lines in dermis
Human body test conditions:
after cleaning forehead skin of 15 people and 30-50 years old women, applying the toning lotion with the formula in the table 1 on the surface of the forehead skin, and carrying out wrinkle detection by using a skin diagnosis instrument SPIMI (SpIMI), wherein the analysis principle is as follows: light wave parameter images of different levels of the skin are obtained through light wave parameter microscopic imaging, and the deep skin images are subjected to super-resolution imaging analysis through advanced software algorithm processing.
And (3) testing results:
1. directly detecting after smearing, wherein fine lines of the dermis layer are unchanged;
2. after 5 hours of application, the effect reached the dermis and fine lines 1 mm from the skin surface (dermis) disappeared, as shown in fig. 2.
3. In vitro anti-inflammatory and allergy-relieving efficacy assessment (IL-1. beta.)
The test method comprises the following steps: the experiment is divided into 4 groups in total,
1. negative control group (NC);
2. LPS (lipopolysaccharide stimulated cells to produce inflammatory factors) model group: only LPS is added for stimulation without sample pretreatment;
3. sample set (formulation 1): LPS stimulation is carried out after sample pretreatment;
4. positive control group (PC): LPS stimulation was performed after 0.1mg/mL dexamethasone pretreatment.
Collection logarithmCells in the growth phase at a cell density of 4X 105One/well was inoculated into 12-well plates. In an incubator (37 ℃, 5% CO)2) After 24h of medium culture, the drug to be tested (formula 1 lotion and positive control) was added and the mixture was incubated in an incubator (37 ℃ C., 5% CO)2) After the culture is continued for 2h, 1 mu g/mL LPS is added into the model group, the sample group and the positive control group, and the model group, the sample group and the positive control group are put into a cell incubator to be continued for 24 h. And (3) detecting the content of IL-1 beta in the culture solution by adopting a mouse IL-1 beta ELISA kit and taking the supernatant according to the instruction.
TABLE 2
Compared with a control group, the release amount of 1L-1 beta secreted by cells induced by LPS is respectively reduced by 72% and 23%, and the effect is obvious. The cosmetic water of the formula 1 is shown to be capable of effectively reducing the release amount of inflammatory cytokines and has the effects of improving skin inflammation, removing acne, relieving skin redness and allergy and the like.
4. Guaranteed skin elasticity test
Subject: qualified test subject by screening (0-50 years old 15 persons)
Experiment time: 2 hours
The experimental group was formula 1 lotion, and the control group contained 0.5% Hyaluronic Acid (HA). And (3) coating in different areas, measuring the water content value and the water loss value after coating until no visible water exists on the surface, cleaning the arms, and measuring the water content and the water loss value again after 20 minutes in a constant temperature and humidity environment. The results of the experiment are shown in fig. 3 and 4.
