Abstract
本申请公开了一种纳米抗体药物偶联物。本申请的纳米抗体药物偶联物,包括偶联的抗体融合蛋白、连接子和小分子毒素,抗体融合蛋白为纳米抗体与多肽的融合蛋白,多肽为GALA多肽或者具有相同螺旋生成功能及两亲性结构的GALA变异多肽。本申请的纳米抗体药物偶联物,在纳米抗体上融合表达GALA多肽或其变异多肽,在靠近细胞膜时GALA多肽或其变异多肽被脂质诱导形成两亲性螺旋,促进内吞,增强纳米抗体药物偶联物活性,有效提高纳米抗体药物偶联物的药效、药动学性能和治疗指数。本申请研发的GALA多肽的新作用机制为提高纳米抗体药物偶联物药效提供了新思路和方法,也为其他内吞依赖的胞内高效递送提供了新思路和工具。

技术领域
本申请涉及抗体药物偶联物技术领域,特别是涉及一种纳米抗体药物偶联物。
背景技术
抗体药物偶联物(Antibody-drug conjugates,ADCs)是一类通过连接子(linker)将毒性极强(EC50:10-3-10-1nM)的小分子化疗药共价连接至单克隆抗体上的靶向治疗药物,希望以此实现更高的治疗指数。被获准上市的ADCs包括5个用于治疗血液肿瘤:gemtuzumabozogamicin(MYLOTARG;Pfizer)、brentuximab vedotin(ADCETRIS;Seattle Genetics)、inotuzumab ozogamicin(BESPONSA;Pfizer)、polatuzumab vedotin(POLIVY;Roche)、belantamab mafodotin(BLENREP;GSK),和4个用于治疗实体瘤:trastuzumab emtansine(KADCYLA;Roche)、enfortumab vedotin(PADCEV;Seattle Genetics)、trastuzumabderuxtecan(ENHERTU;Daiichi Sankyo)、sacituzumab govitecan(TRODELVY;Gilead),另有超过100个ADCs处在600项左右的临床试验当中。
纳米抗体是骆驼动物体内发现的重链抗体(heavy-chain antibody,HCAb)的抗原识别结构域,是迄今为止发现的最小的天然抗原识别片段。免疫动物后,分离淋巴细胞并构建cDNA文库,经筛选后得到理想的VHH克隆,对其进行大肠杆菌重组表达纯化后得到的抗体分子量一般在12-15kDa之间,直径约2.5nm,高度约4nm,故被称作纳米抗体(nanobody)或单域抗体(single-domain antibody,sdAb)。纳米抗体与人源抗体重链可变区的同源性达90%,本身具有极低的免疫原性,通过合适的人源化策略可使免疫原性进一步降低。
一般认为,抗体药物从血管内扩散渗入肿瘤组织内部的效率与抗体大小(size)成反比,传统抗体由于尺寸大、抗原亲和力强,常出现结合屏障效应(binding-site-barriereffect),导致其集中分布于肿瘤组织中靠近血管的位置。而纳米抗体具有显著高于传统抗体的肿瘤组织渗透效率及更加均匀的瘤内分布。
除此以外,纳米抗体相较于传统抗体的优势还包括:向外凸出并且柔软的互补决定区3(CDR3),更适合结合传统抗体无法进入的“山谷”或“裂隙”部位进行识别;容易制备;具有较强的水溶性,pH和热稳定性等。
虽然纳米抗体具有诸多优点,却并未作为制导分子被应用于药物偶联物开发中,最重要原因是药效低下。如何解决纳米抗体药物偶联药物刺激抗原内吞的能力不足而导致药效低下的问题,是纳米抗体药物偶联物研究的重点和难点。
发明内容
本申请的目的是提供一种新的纳米抗体药物偶联物。
本申请采用了以下技术方案:
本申请的一方面公开了一种纳米抗体药物偶联物,其包括偶联的抗体融合蛋白、连接子和小分子毒素,其中,抗体融合蛋白为纳米抗体与多肽的融合蛋白,其中多肽为GALA多肽或者在不影响GALA多肽螺旋生成及两亲性结构的基础上对其进行若干氨基酸修改形成的GALA变异多肽。本申请的纳米抗体药物偶联物(nanobody-drug conjugates,NDCs),即纳米抗体(nanobody)与连接子和小分子毒素偶联形成的抗体药物偶联物;因为本申请的纳米抗体药物偶联物中融合表达有GALA多肽,因此,本申请的新型纳米抗体药物偶联物又缩写为GALA-NDC。
需要说明的是,申请的纳米抗体药物偶联物,其关键在于,研究发现GALA多肽与纳米抗体融合后,不仅不会影响纳米抗体药物偶联物的药性和药效,而且还能增强纳米抗体药物偶联物的活性;并且,本申请的纳米抗体药物偶联物利用GALA多肽能够促进纳米抗体药物偶联物的内吞作用。本申请的一种实现方式中,纳米抗体药物偶联物的内吞能力增强和药效提高100倍以上。
还需要说明的是,虽然本申请的具体实现方式中采用的是GALA多肽与纳米抗体融合;但是,本领域技术人员熟知,在GALA多肽的基础上,还可以进行若干氨基酸的修改,只要不影响GALA多肽原本的螺旋生成及两亲性结构,这些GALA变异多肽都可以用于本申请的抗体药物偶联物,并起到相同的功能和相似的效果。例如,在GALA多肽中适当修改疏水性氨基酸A和L的位置,这样获得的GALA变异多肽,其不影响螺旋生成及两亲性能;与原本的GALA多肽功能和效果相同。当然,根据不同使用需求,还可以进行其他若干氨基酸修改,只要不影响GALA多肽螺旋生成及两亲性结构即可,在此不作具体限定。
本申请的一种实现方式中,纳米抗体为靶向CD38的纳米抗体。
优选的,纳米抗体为靶向CD38的纳米抗体1053。
需要说明的是,靶向CD38的纳米抗体1053只是本申请的一种实现方式中具体采用的纳米抗体;可以理解,本申请的关键在于利用GALA多肽能够促进纳米抗体药物偶联物的内吞作用,不排除还可以采用其它的纳米抗体制备获得不同靶向的纳米抗体药物偶联物。
本申请的一种实现方式中,小分子毒素为微管抑制剂类毒素。
优选的,小分子毒素为单甲基奥瑞他汀E。
需要说明的是,微管抑制剂类毒素或单甲基奥瑞他汀E,只是本申请的一种实现方式中具体采用的小分子毒素,不排除还可以采用其它的抗体药物偶联物中常规或不常规使用的小分子毒素。
本申请的一种实现方式中,连接子为可切断连接子。
优选的,连接子为缬氨酸-瓜氨酸二肽。
需要说明的是,缬氨酸-瓜氨酸二肽连接子只是本申请的一种实现方式中具体采用的连接子,根据使用需求,本申请的纳米抗体药物偶联物还可以采用其它的连接子,在此不作具体限定。
本申请的一种实现方式中,抗体融合蛋白由表达纳米抗体和多肽的编码基因构建到蛋白表达载体中,通过蛋白表达获得;蛋白表达载体为含有SUMO促溶标签的蛋白表达载体。
需要说明的是,将抗体融合蛋白的编码基因构建到蛋白表达载体中,然后采用蛋白表达系统进行表达,是获得融合蛋白的一种常规方式,本申请的关键在于,考虑到多肽中可能包含较多的疏水性氨基酸,为尽量避免其在大肠杆菌中表达时形成包涵体,并尽可能提高表达量,本申请的一种实现方式中具体采用了含有SUMO促溶标签的蛋白表达载体,SUMO标签可以在随后的蛋白纯化过程中被切除。可以理解,SUMO只是本申请的一种实现方式中具体采用的促溶标签,不排除还可以采用其它类似功能的促溶标签。
优选的,蛋白表达载体为原核表达载体pRHSUL2-SUMO。
需要说明的是,原核表达载体pRHSUL2-SUMO只是本申请的一种实现方式中具体采用蛋白表达载体,不排除还可以采用其它类似功能的蛋白表达载体。
本申请的一种实现方式中,多肽上设置有分拣酶A的识别标签LPETGG,利用分拣酶A介导的转肽反应进行位点特异性的药物偶联,将小分子毒素偶联到多肽上。
需要说明的是,本申请采用分拣酶A的识别标签LPETGG,主要是考虑到采用位点特异性药物偶联方式的ADCs,能够表现出更优的药效学、药动学性质和治疗指数。可以理解,一方面不排除还可以采用其它类似功能和作用的识别标签;另一方面不排除还可以采用其它方式实现药物偶联,在此不作具体限定。
本申请的另一方面公开了多肽在抗体药物偶联物中的应用,其中多肽为GALA多肽或者在不影响GALA多肽螺旋生成及两亲性结构的基础上对其进行若干氨基酸修改形成的GALA变异多肽,本申请的应用包括将多肽偶联在抗体药物偶联物的抗体上,利用抗体将GALA多肽或者GALA变异多肽带到细胞膜附近,使得GALA多肽或者GALA变异多肽被细胞膜上的脂质诱导生成两亲性螺旋,从而促进抗体药物偶联物的内吞。
需要说明的是,GALA多肽本身是已经存在的多肽结构,其序列如Seq ID No.1所示,GALA多肽被认为是迄今为止效应最强的pH敏感促融合肽。1987年Szoka等人将在酸性环境中可以被质子化的谷氨酸(Glu,E)间隔插入到流感病毒血凝素(hemagglutinin,HA)的HA2亚基N-末端的促融合肽段中得到GALA多肽。本申请的关键在于,创造性的发现了一种新的GALA多肽在中性条件下促进内吞的作用原理,即在GALA多肽靠近细胞膜时,可以被细胞膜上的脂质诱导生成两亲性螺旋,进一步与脂质相互作用;因此,本申请创造性的将其用于抗体药物偶联物,提出了GALA多肽或GALA变异多肽在制备抗体药物偶联物中的新用途。抗体药物偶联物中的抗体具有细胞靶向作用,能够准确有效的将GALA多肽或GALA变异多肽带到细胞膜附近,从而提高抗体药物偶联物的内吞和治疗效果;本申请的一种实现方式中,纳米抗体药物偶联物的内吞能力增强和药效提高100倍以上。
优选的,多肽上设置有分拣酶A的识别标签LPETGG。
需要说明的是,在多肽上设置分拣酶A识别标签,主要是为了方便小分子毒素偶联到多肽上;可以理解,根据不同的需求还可以在多肽上设置不同的标签,在此不作具体限定。
本申请的再一面公开了一种抗体药物偶联物,该抗体药物偶联物的抗体上融合有GALA多肽或者在不影响GALA多肽螺旋生成及两亲性结构的基础上对其进行若干氨基酸修改形成的GALA变异多肽。
需要说明的是,本申请的关键在于利用GALA多肽在靠近细胞膜的脂质时可以被诱导生成两亲性螺旋,进一步与脂质相互作用,从而促进内吞;因此,GALA多肽或GALA变异多肽不仅可以用于制备纳米抗体药物偶联物,也可以用于制备一般的抗体药物偶联物。至于抗体与GALA多肽或GALA变异多肽的融合方式,可以参考本申请的纳米抗体药物偶联物,在此不作具体限定。
本申请的再一面公开了一种偶联物,该偶联物包括偶联在一起的第一偶联部分和第二偶联部分,第一偶联部分为GALA多肽或者在不影响GALA多肽螺旋生成及两亲性结构的基础上对其进行若干氨基酸修改形成的GALA变异多肽,第二偶联部分为能够与细胞膜上的受体结合的物质。
优选的,本申请的偶联物中,能够与细胞膜上的受体为蛋白质、多肽、核酸、糖和化学小分子中的至少一种或至少一种的组合物。
需要说明的是,现有技术中,GALA多肽是在酸性pH条件下通过对细胞(器)膜进行打孔或融合促进逃逸;本申请关键在于研究发现了GALA多肽的一种新的内吞作用原理,即本申请创造性的发现,将GALA多肽与能够与细胞膜上的受体结合的物质(即第二偶联部分)偶联,利用第二偶联部分将GALA多肽带到细胞膜附近,在中性条件下,细胞膜上的脂质诱导GALA多肽形成两亲性螺旋,与脂质相互作用,从而促进内吞。因此,原则上,只要能够将GALA多肽带到细胞膜附近的物质都可以作为本申请偶联物的第二偶联部分,例如蛋白质、多肽、核酸、糖、化学小分子等,或者这些物质的组合物,在此不作具体限定。
本申请的再一面公开了一种用于抗体药物偶联物的多肽,该多肽为GALA多肽或在不影响GALA多肽螺旋生成及两亲性结构的基础上对其进行若干氨基酸修改形成的GALA变异多肽,且在多肽上设有分拣酶A的识别标签LPETGG。
需要说明的是,本申请的多肽实际上就是在GALA多肽或GALA变异多肽的基础上增加分拣酶A的识别标签LPETGG;本申请的多肽可以作为制备抗体药物偶联物原材料使用;当然,在进行具体的抗体药物偶联物制备时,也可以将本申请多肽的编码基因与抗体编码基因组合在一起,通过蛋白表达系统获得融合蛋白,用于制备抗体药物偶联物,在此不作具体限定。
本申请的有益效果在于:
本申请的纳米抗体药物偶联物,在纳米抗体的基础上融合表达GALA多肽或GALA变异多肽,GALA多肽或GALA变异多肽在靠近细胞膜时被脂质诱导形成两亲性螺旋,促进内吞,增强纳米抗体药物偶联物活性,有效提高了纳米抗体药物偶联物的药效、药动学性能和治疗指数。本申请通过纳米抗体与GALA多肽或GALA变异多肽的融合蛋白,使得纳米抗体药物偶联物内化的效率和程度显著增强,GALA多肽的新作用机制为提高纳米抗体药物偶联物的药效提供了新思路和新方法,为其他内吞依赖的胞内高效递送提供了新思路和新工具。

具体实施方式
下面通过具体实施例对本申请作进一步详细说明。以下实施例仅对本申请进行进一步说明,不应理解为对本申请的限制。
实施例
一、主要实验材料
菌株:分子克隆实验所采用的感受态菌株为DH5α;原核表达外源重组蛋白的菌株是Rosseta2(DE3)。
克隆载体:用于原核表达外源重组蛋白的载体为pRHSUL2,pET28a;所使用的pentamutant sortase A来源于质粒Addgene plasmid#51140。
细胞系:使用的悬浮细胞系有CD38阳性的骨髓瘤细胞系LP-1和CD38敲除的LP-1细胞系CD38-KO/LP-1;贴壁细胞系有:HEK293T,以及稳定表达CD38的HEK293T细胞系CD38+/HEK293T。
二、试验方法
1.蛋白表达载体的构建
将纳米抗体1053的基因序列通过分子克隆的方法克隆至pEHSUL2载体中,转化DH5α,进行序列鉴定,获得阳性质粒。1053的基因序列和蛋白表达载体构建参考专利201510808316.8,该专利中蛋白表达载体构建相关的技术内容全部援引到本申请中。
2.蛋白原核表达及纯化
本例采用含有SUMO标签的nanobody-GALA融合蛋白,其制备具体如下:
(1)将1μl带有目的基因的质粒转化入100μl Rosseta2(DE3)感受态细胞,涂相应抗性平板,37℃培养过夜。
(2)一次接种,刮取一小片菌落至5ml相应抗性LB液体培养基中,37℃摇至浑浊;二次接种,将5ml菌液接种到500ml相应抗性LB液体培养基中,37℃摇至OD600 0.6-0.8,将培养瓶置于4℃冰箱中,使菌液降温至16℃左右,加入0.25mM IPTG 16℃诱导重组蛋白表达16-24小时。
(3)检测蛋白表达情况:吸取2ml菌液,5000rpm离心5min后去除培养基上清,250μlPBS重悬菌体,超声破碎后18000rpm离心5min,分离上清和沉淀,沉淀用250μl PBS重悬,通过SDS-PAGE并进行考马斯亮蓝染色后初步检测重组蛋白表达情况,一般表达在上清组分中的为可溶性蛋白,表达在沉淀组分中的则为包涵体。确定目的蛋白在上清组分中有表达后继续进行后续步骤。
(4)5000rpm离心30min后弃上清。用40ml镍柱结合缓冲液(50mM Tris,500mMNaCl,pH 8.2)重悬菌体。
(5)将重悬液置于冰水混合物中,用超声破碎仪边搅拌边破碎菌体。使用φ10探头,40%功率超声(超3秒,停6秒)直至菌体重悬液从浑浊至清亮。然后4℃18000rpm离心40min,收集上清。
(6)镍柱纯化
1)柱平衡:用5倍柱体积的镍柱结合缓冲液平衡镍柱。
2)上样:将蛋白样品上入镍柱,流出液可重复上样一次。
3)冲洗:用25倍柱体积的镍柱洗涤缓冲液(50mM Tris,500mM NaCl,20mMImidazole,pH 8.2)冲洗柱子。
4)最后用洗脱液(50mM Tris,500mM NaCl,300mM Imidazole,pH 8.2)将目的蛋白从镍柱中洗脱下来。
(7)SUMO标签切除及透析
将镍柱中洗脱下的目的蛋白装入10K透析袋中,同时加入SUMO蛋白酶,透析袋放入装有镍柱结合缓冲液(50mM Tris,500mM NaCl,pH 8.2)的1L烧杯中,在4℃冰箱中酶切并透析12小时。
(8)第二次镍柱纯化
本次纯化的目的是从体系中去除被切掉的SUMO标签,由于纳米抗体-GALA融合蛋白的末端带有His标签可以吸附在镍柱上,而被切下的SUMO标签则不挂柱,因此可以得到不含SUMO标签的理想纳米抗体-GALA融合蛋白。
(9)若仍需进一步对目的蛋白进行纯化可以使用离子交换层析以及分子筛层析等。
(10)最终纯化完成的蛋白用10K超滤管浓缩至4-5mg/ml,分装后放-80℃保存。
3.连接子药物复合物的化学合成
(1)MC-vc-PAB-MMAE的合成
MC-vc-PAB-MMAE的合成路线如图1所示,合成步骤如下:
1)向一个放有磁力搅拌子和2ml DMF的圆底烧瓶中,加入MMAE(50mg,1equiv),MC-vc-PAB-PNP(61.65mg,1.2equiv),HOBt(1.9mg,0.2equiv)和DIPEA(25μl,2equiv),将该体系置于氩气氛围中,室温搅拌12小时。
2)用质谱监测反应进度,待MMAE反应完全后,反应液用反相HPLC(C18,5μm,150×10mm色谱柱,流动相A:乙腈,流动相B:0.05%TFA,流动相A5%→75%,25min,4ml/min)进行分离。
3)合并目标化合物,冻干,反应产率约为62%。产物的质谱表征结果如图2所示。
(2)化合物(Ⅱ)的合成
化合物(Ⅱ)-1的合成路线如图3所示,合成步骤如下:
1)向一个放有磁力搅拌子和1.5ml DMF的圆底烧瓶中,加入多肽原料NH2-GGGDTDTC-NH2(25.5mg,1equiv)和MC-vc-PAB-MMAE(46.2mg,1equiv),将该体系置于氩气氛围中,室温搅拌12小时。
2)用质谱监测反应进度,待MC-vc-PAB-MMAE反应完全后,反应液用反相HPLC(C18,5μm,150×10mm色谱柱,流动相A:乙腈,流动相B:0.05%TFA,流动相A 5%→75%,25min,4ml/min)进行分离。
3)合并目标化合物,冻干,反应产率约为69%。产物的高效液相色谱和质谱表征结果如图4所示,纯度约为97%。
化合物(Ⅱ)-2的多肽原料氨基酸序列为NH2-GGG-DTDTC-DTDTC-NH2,合成步骤同上。产物的高效液相色谱和质谱表征结果如图4所示,纯度约为95%。
4.转肽反应制备位点特异性修饰NDCs
(1)反应体系:包含LPETG的蛋白200μM、GGGDTDTC-MC-vc-PAB-MMAE 8mM、pentamutant sortase A 2μM、CaCl2 10mM、反应溶液为50mM Tris,500mM NaCl,pH 7.4-8.2;4℃孵育4小时。
(2)去除带有His-Tag的杂质
向体系中加入Ni-NTA琼脂糖,4℃孵育30min,目的是去除未反应的蛋白、sortaseA以及反应产生的副产物。4000rpm离心1min并收集上清。
(3)去除过量的GGGDTDTC-MC-vc-PAB-MMAE
用10K超滤管将上步收集的上清超滤6次,每次稀释比例为1:10,以免游离的小分子细胞毒药物对后续细胞实验造成干扰。
5.免疫印记(Western blot)
(1)样品制备
蛋白样品加入SDS loading buffer,沸水浴10min。
(2)SDS-PAGE
样品在浓缩胶中时,以70V低压进行电泳,待样品完全进入分离胶且能看到marker条带后,再以120V高压进行电泳。
(3)转膜
将胶片取出去除浓缩胶和分离胶前沿有溴酚蓝的部分后放入转膜液中。将NC膜或无水甲醇活化的PVDF膜放入转膜液中充分湿润。按照“黑面,海绵,滤纸,胶,膜,滤纸,海绵,白面”的顺序装好转膜夹。转膜过程的设置为300mA,60min,可根据蛋白条带的大小适当增减转膜时间。
(4)封闭
转膜完成后,将膜取出置于塑料盒中,TBST洗一次。用5%脱脂奶粉配成的牛奶室温封闭1小时。
(5)一抗孵育
一抗以1:2000稀释,室温孵育1小时。
(6)TBST洗3次,每次7min。
(7)二抗孵育
以1:10000稀释,室温孵育1小时。
(8)TBST洗3次,每次7min。
(9)显影
将ECL发光液中的A液和B液以1:1的比例混合均匀后,加到膜上。用凝胶成像仪收集发光信号。
6.制备荧光标记的重组蛋白
本试验所使用的荧光分子DyLight 650NHS Ester或NHS-Fluorescein标记的重组蛋白采用如下方法制备:
(1)将荧光分子用无水DMF配制成10mM溶液。
(2)用10K超滤管将蛋白样品用预冷的pH 8.2的0.2M NaHCO3溶液超滤3次,每次按1:10稀释。
(3)向100μM超滤后的蛋白样品中加入DyLight 650NHS Ester DMF溶液(10equiv),室温避光反应2小时。
(4)用10K超滤管将蛋白样品用预冷的PBS超滤3次,每次按1:10稀释,除去体系中过量未反应的小分子。
(5)超滤后得到的产物,加入甘油(50%v/v),储存于-20℃备用。
7.蛋白的质谱表征
(1)置换样品溶液
蛋白储存缓冲液中的盐会影响质谱测定,因此需要将体系置换为适合于高分辨质谱分析的环境。本研究用加入了0.1%甲酸和50%乙腈的Milli-Q水,将测试样品用10K超滤管超滤3次置换样品溶液,每次按1:10稀释。最后,将测试样品浓缩至1-2mg/ml。
(2)样品测试
用高分辨质谱仪QStar Elite(ESI-ToF MS)进行测试。
8.细胞培养
(1)贴壁细胞传代
1)吸去原培养基,PBS润洗一次除去残留培养基。
2)0.05%胰酶/EDTA消化,37℃消化5min。
3)镜下观察确认消化完成后,加入等体积培养基终止消化并将细胞吹匀。
4)根据细胞的生长速度确定传代比例,对于生长速度快的细胞按1:10的比例传代;对于生长速度较慢的细胞一般按1:5的比例传代。培养2~3天后需要再次传代。
(2)悬浮细胞传代
1)将需要传代的细胞放入无菌离心管中。
2)700rpm离心5min,去上清。
3)轻拍管尖使细胞松散,加入培养基重悬细胞。
4)根据细胞生长速度按1:5-1:10的比例传代。培养3天后需要再次传代。
9.细胞毒性实验
(1)细胞铺板
将细胞经计数后用相应培养基稀释至4.5×104/ml,使用排枪铺到96孔板中,90μl/孔,相当于4000个细胞/孔,铺好后放入37℃培养箱中静置2小时。
(2)加药处理
用相应培养基将NDCs配制成1μM溶液,用0.22μm滤膜过滤除菌,然后按1:4梯度稀释,形成8个药物浓度梯度:1000、25、6.25、1.563、0.391、0.098、0.024、0.006nM。以10μl/孔加入到铺好细胞的96孔板中,即处理细胞的药物终浓度为100、2.5、0.625、0.1563、0.0391、0.0098、0.0024、0.0006nM。37℃培养箱中孵育72小时。
(3)细胞活力检测
本试验通过resazurin测定细胞活力,resazurin本身无荧光,被活细胞代谢后产生有强荧光的resorufin。加入500mM resazurin 10μl/孔,37℃培养箱中孵育4小时后,用酶标仪检测530nm激发,600nm发射的荧光信号。
(4)细胞活力计算
Cell viability%=(FL样品-FL空白)/(FL未处理-FL空白)×100%,其中,FL样品为不同浓度药物处理孔的荧光强度,FL空白为培养基空白对照孔的荧光强度,FL未处理为不加药处理孔的荧光强度。
10.激光共聚焦活细胞显微成像
(1)细胞铺板
将玻璃底细胞培养皿(玻璃底直径20mm)用0.01%多聚赖氨酸(Poly-L-lysine,PLL)处理30min,吸去PLL后,培养皿用PBS润洗两次,完全晾干后备用。细胞用DMEM培养基重悬并计数后,铺4×105个细胞到上述玻璃底培养皿中,放入37℃培养箱中培养过夜。
(2)通过活细胞成像探究GALA的作用机制
将铺有CD38+/HEK293T细胞的玻璃底培养皿置于冰上预冷15min后分别加入终浓度20nM的1053-DL650、1053-GALA-DL650、1053-mutGALA-DL650,置于冰上孵育45min以便抗原抗体充分结合;去除上清,加入新的培养基并置于37℃培养箱中开启内吞,孵育相应时间(0min、2h、12h、24h)后,加入1×Hoechst 33342室温染5min;去除上清,细胞用HBSS润洗2次,加入新的培养基后用激光共聚焦显微镜的60倍油镜成像。
(3)内化的1053-GALA-DL650与溶酶体的共定位检测
向铺有CD38+/HEK293T细胞的玻璃底培养皿中加入终浓度为20nM的1053-GALA-DL650,置于37℃培养箱中孵育45min后去除上清,加入新的培养基后置于37℃培养箱中继续孵育8h;向培养基中加入终浓度为50nM的 Green DND-26,37℃孵育1h,最后加入1×Hoechst 33342室温染15min;去除上清,细胞用HBSS润洗2次,加入新的培养基后用激光共聚焦显微镜的100倍油镜成像。
11.LUVs的制备
向圆底烧瓶中加入POPG或POPC,对于表面装饰有DSPE-PEG2000-NHS的LUVs,还需加入2mol%的DSPE-PEG2000-NHS,随后加入氯仿使得脂质固体完全溶解得到透明溶液;将有机相旋干后使用油泵继续抽干3h,使得脂质在瓶底形成脂膜;向圆底烧瓶中加入10mM pH7.0的磷酸钾缓冲液,使得脂质的终浓度为20mM,在多管漩涡震荡器上室温震荡30min;最后用装有2张孔径100nm聚碳酸酯滤膜的迷你脂质体挤出器挤压脂质体,使得液体通过滤膜23次,得到的LUVs溶液立刻进行实验,剩下的保存于4℃冰箱。
12.圆二色谱测定
(1)纳米抗体-GALA融合蛋白中GALA对pH的敏感性试验
1)置换样品溶液
蛋白储存缓冲液中的盐会影响圆二色谱的信号,因此需要将体系置换为适合于圆二色谱测定的环境。本实验所使用的缓冲液为pH5.0的10mM醋酸-醋酸钾缓冲液和pH7.4的0.1%磷酸二氢钾-磷酸氢二钾缓冲液。使用上述缓冲液将测试样品用10K超滤管超滤3次除盐,每次按1:10稀释。最后,将测试样品浓缩至0.2mg/ml。
2)样品测试
使用光程为1mm的石英比色皿,测定温度设定为25℃。圆二色谱仪器参数为:波长扫描范围190nm到260nm,扫描分辨率为0.5nm,响应时间1s,扫描速度为每分钟20nm。每个样品进行5次重复扫描。
3)数据处理
将采集到的数据用Pro-Data Viewer软件进行平均、背景扣除和平滑处理(平滑窗口设置为15)得到最终谱图。圆二色谱数据用单位残基的平均椭圆率表示[θ],(deg×cm2×dmol-1)×10-3。
(2)圆二色谱实验
GALA多肽的终浓度为30μM,脂质终浓度约为2mM。样品测试和数据处理过程同上。
三、结果与讨论
1.纳米抗体-促融合肽融合蛋白的设计与表达纯化
本研究选择了功能上可能符合研究目的促融合肽GALA,与靶向CD38的纳米抗体1053融合,靶点CD38有被批准上市的单克隆抗体药物用于肿瘤治疗。考虑到所选择的促融合肽包含较多的疏水性氨基酸,为尽量避免其在大肠杆菌中表达时形成包涵体并尽可能提高表达量,本研究采用了含有SUMO促溶标签的蛋白表达载体,SUMO标签可以在随后的蛋白纯化过程中被切除。最后,加入了分拣酶A(Sortase A)识别标签LPETGG,利用Sortase A介导的转肽反应进行位点特异性的药物偶联。综上,将上述功能模块插入到纳米抗体远离抗原识别区的C-端尾部,就设计出了本例可以定点偶联小分子细胞毒药物的纳米抗体融合蛋白。
通过基因合成和分子克隆等手段,最终将编码上述融合蛋白的基因构建到原核表达载体pRHSUL2-SUMO中。载体构建完毕后,在大肠杆菌Rosetta中进行原核表达,并进行免疫印迹检测。
考虑到GALA多肽的序列中包含较多疏水性氨基酸,为避免Nb-GALA表达时形成包涵体并提高表达量,本研究采用了含有SUMO促溶标签的蛋白原核表达载体。为防止SUMO标签对后续试验造成干扰,菌体裂解液上清(ⅰ)经第一步镍柱纯化后的产物(ⅱ)用SUMO标签水解酶(SUMO protease)进行酶切(iii);在第二次镍柱纯化时,由于目的蛋白带有His标签可以吸附在镍柱上,而被切下的SUMO标签则不挂柱,因此可以得到不含SUMO标签的理想重组蛋白(iv),如图5所示。图5为重组蛋白表达纯化过程示意图及各主要步骤产物,菌体裂解液上清(ⅰ)经第一步镍柱纯化后的产物(ⅱ)用SUMO标签水解酶进行酶切(iii);在第二次镍柱纯化时,由于目的蛋白带有His标签可以吸附在镍柱上,而被切下的SUMO标签则不挂柱,最终得到不含SUMO标签的理想重组蛋白(iv)。
2.纳米抗体药物偶联物的构建方法、制备与表征
(1)纳米抗体药物偶联物的构建方法
本研究选用分拣酶A(Sortase A)的转肽反应进行MC-vc-PAB-MMAE连接子药物复合物的定点偶联,其原理如图6的A图所示:首先,分拣酶A识别纳米抗体底物(Ⅰ)尾部的LPETGG标签并切断其苏氨酸与甘氨酸之间的肽键(虚线所示),通过酶活中心的半胱氨酸残基与(Ⅰ)形成硫酯中间体;MC-vc-PAB-MMAE连接子药物复合物(图6的B图)被修饰在一条具有亲核攻击能力的多肽的半胱氨酸上(Ⅱ),(Ⅱ)通过其三个连续甘氨酸末端的氨基对硫酯中间体进行亲核进攻,与苏氨酸之间形成新的肽键,生成产物(Ⅳ)。待转肽完成后,向反应体系中加入Ni-NTA琼脂糖树脂,未反应的(Ⅰ)和带有His标签的分拣酶A可以被吸附并离心除去;得到的上清经超滤或分子筛纯化,除去体系中过量的(Ⅱ)和副产物(Ⅲ),得到纯净的纳米抗体药物偶联物(Ⅳ)。
图6为通过转肽反应构建位点特异性修饰的纳米抗体药物偶联物。A图为转肽反应原理示意图。首先,分拣酶A识别纳米抗体底物(Ⅰ)尾部的LPETGG标签并切断其苏氨酸与甘氨酸之间的肽键(虚线所示),通过酶活中心的半胱氨酸残基与(Ⅰ)形成硫酯中间体;连接子药物复合物(R)被修饰在一条具有亲核攻击能力的多肽的半胱氨酸上(Ⅱ),(Ⅱ)通过其三个连续甘氨酸末端的氨基对硫酯中间体进行亲核进攻,与苏氨酸之间形成新的肽键,生成产物(Ⅳ)。为了对比偶联药物的数量对纳米抗体药物偶联物药效的影响,本研究合成了两种化合物(Ⅱ),(Ⅱ)-1具有一个(n=1)半胱氨酸连接位点,可以偶联一个连接子药物复合物(R);(Ⅱ)-2具有两个(n=2)半胱氨酸连接位点,可以偶联两个R。B图为本研究中使用的MC-vc-PAB-MMAE连接子药物复合物的结构式。缬氨酸-瓜氨酸二肽(valine-citrulline,vc)可以被内吞体或溶酶体中酸性环境依赖的多种蛋白酶降解,随后PAB自动脱落将微管抑制剂MMAE释放出来。
(2)纳米抗体药物偶联物的制备与表征
首先,对图6的A图中的化合物(Ⅱ)进行化学合成。主要分为两步:
第一步,以MC-vc-PAB-PNP和MMAE为原料合成MC-vc-PAB-MMAE。合成路线如图1所示,产物的质谱表征结果如图2所示,该步反应产率约为62%。图2为MC-vc-PAB-MMAE的质谱表征,其中1316.9为产物的正离子峰,659.1为产物的双正电荷峰。
第二步,将合成的MC-vc-PAB-MMAE与多肽NH2-GGGDTDTC-NH2反应,(MC-vc-PAB-MMAE中的马来酰亚胺基团与多肽中半胱氨酸残基的巯基侧链反应,得到化合物(Ⅱ)-1,合成路线如图3所示。同时,为了在后续研究中比较GALA-NDC与偶联两个药物分子的NDC间的药效差异,还将MC-vc-PAB-MMAE与另一条具有两个半胱氨酸反应位点的多肽NH2-GGG-DTDTC-DTDTC-NH2反应得到化合物(Ⅱ)-2。产物的高效液相色谱和质谱表征结果如图4所示,该步反应产率约为69%。图4为化合物(Ⅱ)的表征,其中(A)为化合物(Ⅱ)-1的高效液相色谱图,产物纯度约为97%,分析使用C-18色谱柱;(B)为化合物(Ⅱ)-1的质谱图,其中1020.9为产物的双正电荷峰,681.0为产物的三正电荷峰;(C)为化合物(Ⅱ)-2的高效液相色谱图,产物纯度约为95%,分析使用C-18色谱柱;(D)为化合物(Ⅱ)-2的质谱图,其中3891.9483为产物的正离子峰。
随后,通过转肽反应,将合成的化合物(Ⅱ)定量定点转移到纳米抗体(Ⅰ)上,最终得到纳米抗体药物偶联物(Ⅳ)。考马斯亮蓝染色和免疫印迹检测结果表明:原本存在于纳米抗体尾部的His标签被取代为vcMMAE,初步证明通过转肽反应可以得到位点特异性修饰的纯净纳米抗体药物偶联物,结果如图7所示。图7为NDC的考马斯亮蓝染色和免疫印迹检测,图7的结果显示,原本存在于纳米抗体尾部的His标签被取代为vcMMAE,初步证明通过转肽反应可以得到位点特异性修饰的纯净纳米抗体药物偶联物。
由于蛋白凝胶电泳结合预染蛋白marker的方式难以对纳米抗体药物偶联物进行精确分子量鉴定以准确表征药物的偶联数量。因此,本研究还使用ESI-TOF高分辨质谱对NDCs产物及其对应的重组蛋白原料进行了精确分子量鉴定。本试验仅以研究中最重要的1053-vcMMAE和1053-GALA-vcMMAE的高分辨质谱表征结果举例说明:样品检出结果与根据氨基酸序列以及转肽反应原理计算出的理论值完全吻合,误差不超过1道尔顿。具体计算方法为:未连接药物的蛋白,分子量可以根据其氨基酸序列通过软件直接得到;偶联后的纳米抗体药物偶联物,根据转肽的原理,标签LPET后的GG-His6=954.9710被置换为GGGDTDTC-MC-vc-PAB-MMAE=2040.3620,因此分子量增加值应=2040.3620-954.9710=1085.391。
综上,本实验首先设计并表达纯化出了C-末端融合有促融合肽GALA的纳米抗体融合蛋白Nb-GALA。并通过转肽反应对其进行位点特异、数量可控的药物偶联,制备出了新型GALA-NDC。
(3)GALA-NDC的生物学活性测试
1)主要对照设计
为了验证融合GALA能否增强NDC对靶细胞的特异性毒性,以及药效的增强是否依赖于GALA发生螺旋化,本实验按照图8设计了三个主要阴性对照,即图8中的阴性对照(2)-(4),图8中(1)为主要研究对象,即本实验的新型GALA-NDC。阴性对照包括不含GALA的普通纳米抗体(2);将关键位置氨基酸进行突变的GALA突变体mutGALA(3),该突变体将GALA蛋白序列中第9、17和25位的三个亮氨酸突变为脯氨酸(WEAALAEAPAEALAEHPAEALAEAPEALAA),理论上在破坏了GALA形成稳定完整跨膜螺旋能力的同时最大限度地保持了整体结构的相似性;以及含GALA的无关纳米抗体GFPnb-GALA(4),其中,GFPnb是识别绿色荧光蛋白的纳米抗体,主要用于判断中性pH下GALA是否与细胞发生非特异结合。
图8中,第一个三角代表SUMO-Tag在蛋白纯化过程中将被切除,第二个三角标注的是转肽反应进行的位点。
2)GALA-NDC的细胞毒性测试
本研究接着比较了所制备的基于1053的NDCs在CD38阳性和阴性细胞系上的活性。实验中使用的所有NDCs的考马斯亮蓝染色和WB(anti-MMAE)检测结果如图9所示。
图9为细胞毒性测试实验中使用的基于纳米抗体1053并靶向CD38的NDCs的考马斯亮蓝染色(上图),和免疫印迹检测(anti-MMAE)(下图)。
细胞用不同浓度药物在37℃培养箱中处理72小时后使用刃天青(resazurin)测定细胞活力,结果如图10所示。实验结果表明:第一,1053-GALA-vcMMAE显示出对CD38阳性细胞系最强的抑制活性,EC50值分别为EC50(LP-1)=0.87nM和EC50(CD38+/HEK293T)=0.26nM,同时,在相同实验条件下并不影响CD38阴性细胞系的生长。第二,1053-vcMMAE和1053-mutGALA-vcMMAE显示出相仿的抑制效果,在浓度100nM时都未明显表现出对LP-1细胞的抑制效果,在阳性细胞系上的EC50值与1053-GALA-vcMMAE相差约两个数量级。以上两点验证了融合GALA能够增强NDCs活性的设想,并且通过GALA突变体的对照说明该现象依赖于GALA α-螺旋的形成。第三,无关纳米抗体对照GFPnb-GALA-vcMMAE并未显示出明显的细胞生长抑制作用,表明GALA在实验进行的生理pH条件下并不会造成非特异性毒性。第四,一般认为偶联药物的数量与ADCs的药效在一定范围内呈正相关,然而偶联药物数量过多会引发ADCs在体内被快速清除并导致脱靶毒性,因此偶联3.5-4个药物是ADCs领域为平衡药效学与药动学所普遍采取的策略。为了将本试验提出的提高药物利用率的思路与增加药物偶联数量的方法进行对比,本研究设置了偶联联两个vcMMAE的对照1053-(vcMMAE)2。实验结果表明增加GALA模块的效果显著优于增加药物偶联数量的方式。有效性方面,1053-GALA-vcMMAE对CD38阳性细胞系的EC50值比1053-(vcMMAE)2降低20-30倍;安全性方面,在本实验最高测试浓度100nM时,1053-(vcMMAE)2明显表现出对两株CD38阴性细胞系的非特异毒性,可能是vcMMAE在胞外脱落所导致的,而新型GALA-NDC在相同实验条件下则并未观察到明显的非特异毒性。
图10为靶向CD38的NDCs的细胞活性测试结果,LP-1为CD38阳性的骨髓瘤细胞系,CD38-KO/LP-1为CD38敲除的LP-1细胞系;CD38+/HEK293T为稳定表达CD38的HEK293T,HEK293T为CD38阴性细胞系;4000个细胞/孔,药物浓度梯度设置为100、2.5、0.625、0.1563、0.0391、0.0098、0.0024、0.0006nM,每个药物浓度重复3个孔;细胞在37℃培养箱中孵育72小时后使用resazurin测定细胞活力。
为了验证融合GALA的优化策略不仅局限于CD38这一个靶点,本研究用抗EGFR的纳米抗体7D12制备了靶向EGFR的NDCs,并进行了细胞活性测试。结果表明7D12-GALA-MMAE对EGFR阳性细胞系A431和EGFR+/HEK293T的抑制曲线相对于对照明显左移,证明该优化策略具备一定的普适性。
靶向EGFR的NDCs的细胞活性测试,4000个细胞/孔,药物浓度梯度为100、2.5、0.625、0.1563、0.0391、0.0098、0.0024、0.0006nM,每个药物浓度重复3个孔;细胞在37℃培养箱中孵育72小时后使用resazurin测定细胞活力。
(4)GALA-NDC的作用机制探究
上述研究结果表明,GALA-NDC的药效显著优于传统NDC,并且该现象依赖于GALAα-螺旋的形成。本实验进一步研究了GALA通过影响哪些环节提高NDC的药效,以及GALA形成螺旋的触发因素。
1)通过活细胞成像探究GALA的作用机制
为验证融合蛋白纳米抗体部分的靶向性,本研究制备了DyLight 650NHS Ester标记的(1)-(4),并对其中最具代表性的1053-DL650(图11的A图)和1053-GALA-DL650进行了高分辨质谱表征(图11的B图),结果表明每个抗体上标记有0或1个荧光分子。

本研究通过活细胞成像的方法,对GALA通过影响哪些环节提高了NDC的药效这一科学问题进行了探究。按照初始设计,GALA-NDC尾部的GALA多肽在溶酶体内被释放并在其酸性环境中形成α-螺旋,一定数量螺旋化的GALA在膜上聚集最终形成微孔,促进小分子细胞毒药物向细胞质逃逸,提升药物到达胞内靶点的有效浓度进而增强药效。为了验证以上猜想,本研究使用激光共聚焦显微镜分别观察了DyLight 650NHS Ester标记的纳米抗体或纳米抗体融合蛋白1053-DL650、1053-GALA-DL650、1053-mutGALA-DL650从与CD38+/HEK293T细胞结合到内化后24h的时空分布变化,结果如图12所示。
图12为活细胞激光共聚焦显微成像实验,CD38+/HEK293T细胞与20nM荧光探针在4℃共孵育45min,换入新的培养基后转移到37℃培养箱中开始内吞,分别在5min、2h、12h、24h后进行成像。标尺=20μm。
通过分析实验结果,可以得到两点重要信息:
第一,从第二行1053-GALA的结果来看,24h后荧光信号依然呈点状分布,并未观察到明显弥散在胞质中的荧光信号。DyLight 650荧光分子在胞质的中性pH环境中为带电离子,理论上若从内吞体或溶酶体逃逸后能够保留在胞质中,观察到弥散的荧光信号。本试验认为存在两种可能性:(1)虽然螺旋化的GALA膜穿孔能力极强,但形成的微孔仅允许分子量不超过800的小分子通过。由于荧光分子DyLight 650NHS Ester的相对分子质量为1066,并被标记于纳米抗体赖氨酸的氨基侧链上,即便被完全降解分子量也将超过800,难以通过GALA形成的微孔。(2)溶酶体中大量聚集的荧光分子,将对相对少量逃逸并广泛分布于细胞质中的荧光分子形成强烈干扰,使得后者不易被观察到。
第二,观察第二行1053-GALA的实验结果,5min时荧光信号全部存在于细胞膜上,2h时已经存在有较多内化的荧光信号,并且随着时间增加而显著增强,到12h时,几乎所有荧光信号从细胞膜上转移到细胞内,表明最初结合的1053-GALA几乎全部内化。与此形成鲜明对比的是,1053-DL650和1053-mutGALA-DL650处理的细胞在2h时仅存在极少数内化的荧光信号,即便在24h时,荧光信号仍然主要存在于细胞膜上。为了量化纳米抗体内化的程度,本研究用点状分布荧光信号的像素面积除以荧光信号的总像素面积得到“内化指数”(“internalization index”),结果如图13所示。内化指数的范围从0到1,0代表荧光信号完全膜分布,1则代表荧光信号完全点状分布。统计结果显示,1053-GALA的内化效率和程度远高于1053或1053-mutGALA,表明螺旋化的GALA能够显著促进融合蛋白的内化作用,进而增强了GALA-NDC药效。
图13为抗体内化指数随时间变化的统计图;其中,内化指数=点状分布荧光信号的像素面积÷荧光信号的总像素面积;内化指数的范围从0到1,0代表荧光信号完全膜分布,1则代表荧光信号完全点状分布;像素面积使用Fiji软件中的颗粒分析模块(particleanalyzer module)进行测量。误差线代表3个独立实验的标准差,每个独立实验包含5张照片。
接着,考虑到抗体药物偶联物内化后,需要进入溶酶体进行降解以释放小分子细胞毒药物。因此本研究进一步检测了内化的1053-GALA-DL650与溶酶体的共定位情况,结果如图14的A图所示。使用LysoTracker Green DND-26对溶酶体进行着色,对图14的A图中划线部分进行荧光强度统计分析,结果如图14的B图所示,结果显示内化后的1053-GALA与溶酶体存在明显的共定位关系,说明GALA-NDC将进入溶酶体中进行降解。同时,LysoTrackerGreen DND-26(MW 399)可以对有1053-GALA存在的溶酶体正常染色,表明溶酶体状态健康,可能说明1053-GALA的溶酶体膜微穿孔能力有限。
图14为内化的1053-GALA-DL650与溶酶体的共定位分析。图14中A图为CD38+/HEK293T细胞与20nM 1053-GALA-DL650在37℃培养箱中共孵育45min,换入新的培养基后在37℃继续孵育8h,最后1h分别用LysoTracker Green DND-26(即A图中的LysoTracker图)和Hoechst 33342(即A图中的Merge图)对溶酶体和细胞核进行着色。标尺=20μm。
此外,考虑到溶酶体中大量聚集的荧光分子,可能对相对少量逃逸并广泛分布于细胞质中的荧光分子形成强烈干扰,使得后者不易被观察到。本研究还尝试使用半乳糖凝集素斑点试验(galectin puncta assay)来探究在本试验设计构建的体系中GALA能否在内吞体溶酶体膜上形成微孔。半乳糖凝集素-3(galectin-3)是一种可以结合糖类修饰的可溶性小蛋白,可以被用作一种识别内吞体溶酶体膜破坏信号的传感器。当内吞体溶酶体膜完好无损的情况下,galectin-3均匀分布在细胞质以及细胞核中;当内吞体溶酶体膜发生破损时,galectin-3通过与其内膜上暴露的糖蛋白的糖链结合,被迅速招募到内吞体或溶酶体中,从而形成点状聚集。本研究构建的稳定表达EGFP-galectin-3的CD38+/HEK293T细胞系,用阳性药物60μM氯喹(chloroquine,CQ)处理10h后,可以明显观察到galectin-3聚集形成的斑点。而1053-GALA-DL650与1053-DL650处理24h后的结果并无显著差异,均未观察到galectin-3发生聚集的现象,表明1053-GALA不能引发半乳糖凝集素斑点试验能够检测到的内吞体溶酶体膜破坏现象。可能的原因是理论上GALA在膜上形成的微孔仅允许分子量不超过800的小分子通过,然而EGFP-galectin-3的相对分子质量约为54kDa,因此难以通过GALA形成的微孔进入溶酶体中形成聚集。
使用半乳糖凝集素斑点试验(galectin puncta assay)探究1053-GALA能否引起内吞体溶酶体膜穿孔,稳定表达EGFP-galectin-3的CD38+/HEK293T细胞系,与20nM 1053-DL650或1053-GALA-DL650在37℃共孵育24h后成像,并以60μM氯喹(chloroquine,CQ)在37℃处理10h为阳性对照。标尺=20μm。
2)通过生物膜模拟系统探究GALA形成螺旋的触发因素
突变体对照Nb-mutGALA的实验结果证明,抗体内化增强和NDC活性增强都与GALA形成稳定完整跨膜螺旋有关。本研究进一步借助生物膜模拟系统对GALA形成螺旋的触发因素是什么这一科学问题进行了探究。本试验推测纳米抗体通过对细胞膜表面抗原的特异性识别,将GALA拉至细胞膜附近,在膜的诱导作用下GALA能够形成α-螺旋并通过其疏水侧面与细胞膜发生“粘附”,使得GALA-NDC内化的效率和程度显著增强,进而提高了药效。为了初步验证这一猜想,本研究以20%(v/v)三氟乙醇(2,2,2-trifluoroethanol,TFE)水溶液和大单层囊泡(large unilamellar vesicles,LUVs)模拟细胞膜环境,用圆二色谱技术检测GALA多肽在不同环境中螺旋度的变化。
TFE的水溶液被认为是一种膜环境的模拟物,溶液中TFE分子能够取代作为氢键供体的水分子聚集在多肽周围,并营造一种低介电环境,因此促进了肽内氢键的形成。此外,TFE还能促进氨基酸侧链之间的疏水相互作用,辅助肽链二级结构形成。LUVs是一种直径约为50-200nM的单层囊泡,是生物膜与多肽相互作用研究中经常使用的生物膜模拟系统。圆二色谱技术(circular dichroism,CD)是一种用于大致评估多肽或蛋白二级结构的常用方法。α-螺旋的圆二色谱的典型特征是靠近193nm有一正的谱带,在222和208nm处有两个负的特征肩峰谱带;而无规卷曲则在200nm处有一负峰,212nm开始出现正的谱带。
本研究共制备了四种不同组分的LUVs并检测它们对化学合成的GALA多肽螺旋度的影响,分别为由阴离子型脂质1-palmitoyl-2-oleoyl-sn-glycero-3-phospho-(1’-rac-glycerol)(POPG)或两性离子型脂质1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine(POPC)组成的LUVs;为了控制GALA与膜的距离这一重要变量,本研究还制备了表面装饰有1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[carboxy(polyethylene glycol)-2000,NHS ester](DSPE-PEG2000-NHS)的LUVs,通过DSPE-PEG2000-NHS与GALA多肽N-末端的氨基共价交联,将GALA拉至LUVs表面,从而模拟了纳米抗体与细胞膜抗原特异性结合的情形。
圆二色谱实验结果如图15所示,图15的结果表明,在中性pH条件的磷酸钾缓冲液中,GALA以无规卷曲的状态存在;在模拟细胞膜环境的20%(v/v)TFE的磷酸钾溶液中,GALA发生明显的α-螺旋化,表明细胞膜可能具有诱导GALA形成α-螺旋的能力。从GALA与LUVs共孵育的结果来看,只有表面装饰有DSPE-PEG2000-NHS的POPG LUVs能够诱导GALA螺旋度的增加,这提示了两点重要信息:第一,膜表面的负电荷可能对诱导GALA形成α-螺旋有促进作用;第二,近距离接触是膜诱导GALA形成α-螺旋的必要条件,因此这种在生理pH下GALA与细胞膜的相互作用离不开抗体的靶向作用。这与生理pH下GFPnb-GALA-DL650并不会导致抗原阴性细胞的非特异性染色,以及GFPnb-GALA-vcMMAE并未表现出明显的非特异毒性等现象一致。也与文献中报道的:当GALA被包裹在由卵磷脂和胆固醇(摩尔比2:1)组成的脂质体内部时,圆二色谱实验结果表明其并不因脂质体外环境pH的变化而发生构象变化;而当GALA被共价修饰在脂质体表面时,在pH 7.4的环境中,本该以无规卷曲形式存在的GALA却以螺旋化的形式存在,说明当脂质体与GALA相距足够近时,能够诱导其形成螺旋。
图15为GALA多肽在中性溶液或与不同生物膜模拟系统共孵育的圆二色谱图。图中曲线分别为10mM pH 7.0的磷酸钾缓冲液,含20%(v/v)TFE的10mM pH7.0的磷酸钾缓冲液,POPG LUVs,表面装饰有2mol%DSPE-PEG2000-NHS的POPG LUVs,POPC LUVs以及表面装饰有2mol%DSPE-PEG2000-NHS的POPC LUVs。测试时GALA多肽的终浓度为30μM,脂质终浓度约为2mM。
四、总结
本实验首先证明了新型GALA-NDC相对于传统NDC以及偶联两个药物的NDC药效均显著增强,随后通过一系列机制研究阐明了该药效增强现象背后可能的工作原理,如图16所示,即纳米抗体通过对细胞膜表面抗原的特异性识别,将GALA拉至细胞膜附近,在膜的诱导作用下GALA能够形成α-螺旋并通过其疏水侧面与细胞膜发生“粘附”,使得GALA-NDC内化的效率和程度显著增强,进而提高了药效。本试验的研究成果揭示了促融合肽GALA在中性pH下发挥作用的一种新机制,为提高NDCs的药效提供了新思路和新方法,为胞内高效递送提供了新工具。
图16为本实验GALA-NDC的工作模型;生理pH下,融合表达在纳米抗体尾端的GALA以无规卷曲形式存在,当纳米抗体与抗原发生特异性识别后,GALA被拉至细胞膜表面并在膜诱导作用下形成α-螺旋,通过其疏水侧面与细胞膜发生“粘附”,使得GALA-NDC内化效率和程度显著增强,进而提高药效。
根据以上实验和分析,可以理解,只要是能够与细胞膜上的受体结合的物质,将其与GALA多肽偶联,形成偶联物,利用细胞膜受体结合物质将GALA多肽带到细胞膜表面,GALA多肽就可以促进偶联物的内吞。利用该原理,不仅可以用于制备抗体药物偶联物、纳米抗体药物偶联物,也可以用于制备其它需要将靶标带入细胞膜的偶联物。
此外,根据以上实验和分析可见,本例的关键在于GALA多肽在接近细胞膜时,细胞膜上的脂质会诱导GALA多肽形成两亲性螺旋,从而起到促进纳米抗体药物偶联物内吞的作用。因此,根据本领域的常规认识,在GALA多肽的基础上可以对其进行若干氨基酸的修改,只要修改后的GALA变异多肽不影响原本的螺旋生成及两亲性结构,就能够合理的预期GALA变异多肽同样能在接近细胞膜时,被脂质诱导形成两亲性螺旋,从而促进内吞。因此,在不影响GALA多肽螺旋生成及两亲性结构的基础上对其进行若干氨基酸修改形成的GALA变异多肽,同样可以用于本例的纳米抗体药物偶联物,与本例的GALA-NDC具有相同或相似的功能和效果。

Claims (7)
1.一种纳米抗体药物偶联物,其特征在于:包括偶联的抗体融合蛋白、连接子和小分子毒素,所述抗体融合蛋白为纳米抗体与多肽的融合蛋白,所述多肽为GALA多肽,所述抗体融合蛋白从N端到C端依序为纳米抗体-GALA多肽;
所述纳米抗体包括靶向CD38的纳米抗体1053或抗EGFR的纳米抗体7D12;
所述连接子为缬氨酸-瓜氨酸二肽;
所述小分子毒素为微管抑制剂类毒素。
2.根据权利要求1所述的纳米抗体药物偶联物,其特征在于:所述小分子毒素为单甲基奥瑞他汀E。
3.根据权利要求1或2所述的纳米抗体药物偶联物,其特征在于:所述抗体融合蛋白由表达所述纳米抗体和所述多肽的编码基因构建到蛋白表达载体中,通过蛋白表达获得;所述蛋白表达载体为含有SUMO促溶标签的蛋白表达载体。
4.根据权利要求3所述的纳米抗体药物偶联物,其特征在于:所述蛋白表达载体为原核表达载体pRHSUL2-SUMO。
5.根据权利要求1或2所述的纳米抗体药物偶联物,其特征在于:所述多肽上设置有分拣酶A的识别标签LPETGG,利用分拣酶A介导的转肽反应进行位点特异性的药物偶联,将所述小分子毒素偶联到所述多肽上。
6.多肽在制备纳米抗体药物偶联物中的应用,其特征在于:所述纳米抗体药物偶联物包括偶联的抗体融合蛋白、连接子和小分子毒素,所述多肽为GALA多肽,所述应用包括将所述多肽偶联在纳米抗体药物偶联物的纳米抗体上,形成从N端到C端依序为纳米抗体-GALA多肽的抗体融合蛋白,利用纳米抗体和GALA多肽的双重作用增强纳米抗体药物偶联物的细胞内吞,从而增强纳米抗体药物偶联物的药效,所述连接子为缬氨酸-瓜氨酸二肽,所述小分子毒素为微管抑制剂类毒素。
7.根据权利要求6所述的应用,其特征在于:所述多肽上设置有分拣酶A的识别标签LPETGG。
