本发明提供了一种降低菌株胆固醇能力的方法,属于微生物领域。本发明提供的降低菌株胆固醇能力的方法包括如下步骤:将菌株加入到培养基中,并加入胆固醇和\或牛胆汁进行培养。因为乳酸脱氢酶是催化丙酮酸转化为乳酸的关键酶,如果阻断这一转化途径,培养基的pH值会发生变化,本发明基于这一假设通过敲除ldh关键酶基因,再结合敲除bsh基因的菌株进行一系列实验验证,从而得出培养基中的pH值会对菌株的降胆固醇能力产生一定的影响。所以本发明通过对比野生型菌株L.plantarum AR113和敲除菌株的生长特性、培养基pH值及其降胆固醇能力的差异,来佐证BSH和培养基pH值对益生菌L.plantarum AR113降解胆固醇的影响。

技术领域
本发明涉及一种微生物领域,具体涉及一种降低菌株胆固醇能力的方法。
背景技术
植物乳杆菌体外降胆固醇的能力受到多种因素的影响,并且不同菌株间表现出差异性。近年来,在体外高效降解胆固醇的益生菌菌株的研究中,主要作用是源于其产生多量或高活性的BSH,即BSH在体外降胆固醇方面发挥重要作用,如乳酸乳球菌亚种KS4和P1、嗜热链球菌Tx、植物乳杆菌FB003、植物乳杆菌H13和植物乳杆菌Lp91等。
乳酸脱氢酶(LDH)是催化丙酮酸转化为乳酸的关键酶,如果阻断这一转化途径,培养基的pH值会发生变化,基于这一假设,还没有实验对这一假设进行验证。胆固醇摄入量过高,会引起冠心病、动脉粥样硬化等心脑血管疾病,严重威胁人体的健康,增加病亡率。研究表明益生菌对人体健康的有益作用之一是降低胆固醇。

本发明是为了解决上述问题而进行的,目的在于提供一种降低菌株胆固醇能力的方法。
本发明提供了一种降低菌株胆固醇能力的方法,具有这样的特征,包括如下步骤:将菌株加入到培养基中,并加入胆固醇和\或牛胆汁进行培养。
在本发明提供的降低菌株胆固醇能力的方法中,还具有这样的特征:其中,菌株为敲除ldh基因的菌株。
在本发明提供的降低菌株胆固醇能力的方法中,还具有这样的特征:其中,敲除ldh基因的菌株的制备方法包括:步骤1,制备菌株感受态细胞;步骤2,将质粒导入菌株感受态细胞中,得含质粒的菌株感受态细胞;以及步骤3,构建ldh敲除质粒并电转入含质粒的菌株感受态细胞中,涂布、培养24-48h,得敲除ldh基因的菌株。
在本发明提供的降低菌株胆固醇能力的方法中,还具有这样的特征:其中,涂布在红霉素和氯霉素双抗性的平板上进行。
在本发明提供的降低菌株胆固醇能力的方法中,其特征在于:其中,培养基为MRS培养基、含有胆固醇的MRS培养基、含有牛胆汁的MRS培养基或含有胆固醇和牛胆汁的MRS培养基中的任意一种。

发明的作用与效果

为了使本发明实现的技术手段、创作特征、达成目的与功效易于明白了解,以下结合实施例及附图对本发明作具体阐述。
下述实施例中的菌株来源如下:
植物乳杆菌(Lactobacillus plantarum)AR113菌株已于2017年03月22日保藏于中国普通微生物保藏中心(地址:北京市朝阳区北辰西路1号院3号),其编号为CGMCCNo.13909。
下述实施例中四种培养基的配方如下:
MRS培养基:蛋白胨10.0g、磷酸氢二钾2.0g、牛肉浸出粉10.0g、酵母膏5.0g、硫酸锰0.25g、无水乙酸钠5.0g、葡萄糖20.0g、硫酸镁0.58g、Tween-801.0mL,加去离子水定容1000mL。固体培养基需要在此基础上加20g的细菌琼脂粉。115℃,灭菌20min。
MC培养基:精确称取胆固醇0.5g,用少量无水乙醇溶解并定容50mL,终浓度为10mg/mL。用孔径为0.45μm微孔滤膜过滤除菌,按照1%(v/v)的量加入到已灭菌的MRS液体培养基中,使MRS液体培养基中胆固醇终浓度为0.1mg/ml。
MB培养基:MRS培养基中,加入0.1%(w/v)牛胆粉。
MCB培养基:MC培养基中,加入0.1%(w/v)牛胆粉。

<实施例1>
在AR113-Δbsh13的基础上对ldh基因的敲除实验
实验方法:质粒pLCP11-0467和pLCP11-1778分别电转到植物乳杆菌AR113-Δbsh13(gba)感受态细胞中,MRS+Cm(氯霉素)+Em(红霉素)平板,37℃厌氧培养48~60h,长出转化子后,进行菌落PCR验证,并将PCR产物通过1%(w/v)琼脂糖凝胶电泳分析,绘制图像。
其中,pLCP11-1778质粒构建的构建方法如下:1778up(F:CTTTTTCTAAACTAGGGCCCGCGAAGGGTGCTAAGAGCCAGATT;R:ATTTTCAGTGATAATCGAGATACAAAAATTCCTCCTATAT)、1778down(F:ATATAGGAGGAATTTTTGTATCTCGATTATCACTGAAAATGG;R:ACCGAGTCGGTGCTTTTTTTCATCATTTTTGACCTTGACG)以植物乳杆菌AR113基因组为模板,1778gRNA(F:CGTCAAGGTCAAAAATGATGAAAAAAAGCACCGACTCGGTR:GGACATACTATGATATATTCTAGAGTTCCTACTGTTAAAGCACGGTTTTAGAGCTAG)以质粒pHSP01为模板,然后使用FX KOD酶将三者进行Overlap PCR,割胶回收获得1778up-down-gRNA片段,再与经ApaI和XbaI酶切回收的pHSP01载体(本实验室保藏)进行无缝克隆,连接产物转化(方法:将质粒转入感受态中,冰上静置30min,然后置于42℃恒温水浴锅热击90s,冰上静置2min,加入900μL LB液体培养基(胰蛋白胨10.0g,酵母浸出粉5.0g,NaCl 10.0g,121℃灭菌15min)至E.coli Top10感受态细胞中,摇床培养1h后,涂布于含卡那霉素抗性的平板中。长出的单菌落用引物pLCP11-yz(F:ATCATTTCGTTTTTCTTTTGCGCT;R:GTAGAACAACTGTTCACCGT)进行验证,获得的正确质粒命名命名为pLCP11-1778。质粒pLCP11-0467与质粒pLCP11-1778的制备方法相同,在此不再赘述。
实验结果如图1所示。图1(a)为AR113-Δbsh13-Δldhl。图1(b)为AR113-Δbsh13-Δldhd。
根据图1(a)可知,标号1-6是AR113-Δbsh13-Δldhl的6个单菌落,用引物Delete-0467-YZ(F:GTGACTCTGGTGAACTTGTC:R:AACCACTGGTGATTGGTG)验证。根据图1(b)可知,标号1-12是AR113-Δbsh13-Δldhd的12个单菌落,用引物Delete-1778-YZ(F:GACGAATTGAATGGAGAG;R:ATACAATCTCGCATAGGG)验证。AR113_0467和AR113_1778菌落PCR验证获得预期条带在大约2300bp,野生型(阳性对照)条带约为3000bp。结果表明ldh基因单敲除菌株L.plantarum AR113-Δbsh13-Δldhl(简称Δbsh13-Δldhl)和L.plant arum AR113-Δbsh13-Δldhd(简称Δbsh13-Δldhd)成功获得。对敲除带进行测序,比对结果与预期完全吻合。
<实施例2>
敲除ldh基因的菌株的制备方法
步骤1,AR113感受态的制备:
具体步骤:将植物乳杆菌AR113划线于MRS固体平板后于厌氧培养至长出1~2mm大小的单菌落。挑取单菌落接于5mLMRS液体培养基中,37℃静止培养12~16h。按3%接种量转接至100mL SGMRS(MRS+0.3M蔗糖+1%甘氨酸。115℃灭菌20min)液体,37℃静置培养至OD600为0.3~0.5,在4℃、4500rpm/min的条件下离心10min,弃上清,用预冷的SM(952mM蔗糖+5mMMgCl2。115℃灭菌20min)缓冲液洗涤两次,在4℃、5500rpm/min的条件下离心8min。用适量预冷的SM缓冲液重悬,每100μL进行分装至1.5mLEP管中,-80℃保存备用;
步骤2,AR113(pLH01)感受态的制备:
具体步骤:当OD600达到0.3左右时,加入终浓度为100ng/mL的诱导肽(氨基酸序列为MAGNSSNFIHKIKQIFTHR)以诱导RecE/T的表达,将质粒pLH01电转到AR113感受态细胞中,划线于MRS固体平板后于厌氧培养至长出1~2mm大小的单菌落。挑取单菌落接于5mLMRS液体培养基中,37℃静止培养12~16h。按3%接种量转接至100mL SGMRS(MRS+0.3M蔗糖+1%甘氨酸。115℃灭菌20min)液体,37℃静置培养至OD600为0.3~0.5,在4℃、4500rpm/min的条件下离心10min,弃上清,用预冷的SM(952mM蔗糖+5mM MgCl2。115℃灭菌20min)缓冲液洗涤两次,在4℃、5500rpm/min的条件下离心8min。用适量预冷的SM缓冲液重悬,每100μL进行分装至1.5mL EP管中,-80℃保存备用,得AR113(pLH01)感受态细胞;

<实施例3>
胆固醇对敲除菌株自身生长特性的影响实验
实验方法:对野生型菌株AR113和12株敲除菌株在MRS、MC、MB和MCB液体培养基中进行培养,并测定生长曲线。
实验结果如图2所示。图2(a)为敲除菌株在MRS培养基中的生长曲线。图2(b)为敲除菌株在MC培养基中的生长曲线。图2(c)为敲除菌株在MB培养基中的生长曲线。图2(d)为敲除菌株在MCB培养基中的生长曲线。
根据图2(a)可知,在MRS培养基中,除了菌株Δldh外,其余菌株的生长趋势相似,生长速率相近,15h进入稳定期,总生物量达1.6左右。根据图2(b)可知,在仅添加胆固醇的MC培养基中,Δldhl、Δldhd和Δldh的生长速率变小,到20h才趋于稳定,总生物量减少至1.0左右,其余菌株在15h进入稳定期,生长趋势相似,总生物量达1.6左右。根据图2(c)可知,在仅添加牛胆汁的MB培养基中,各菌株的生长速率与生长趋势略有差异,总生物量最高达1.4左右,其中Δldhl、Δldhd和Δldh的生长缓慢,菌株Δldh的总生物量最低,仅为0.92。根据图2(d)可知,在添加胆固醇和牛胆汁的MCB培养基中,大部分菌株的生长受到了抑制,菌株的总生物量从1.6降低至1.4,最低仅为0.8,其中菌株Δldh、Δldhl和Δldhd在25h左右才进入稳定期,生长最缓慢。综上,表明胆固醇和牛胆汁的添加会抑制菌株的生长,当二者共同作用时抑制效果更明显。
<实施例4>
胆固醇对敲除菌株生长中pH值的影响实验
实验方法:对野生型菌株AR113和12株敲除菌株在MRS、MC、MB和MCB液体培养基中分别培养0h、5h、15h和24h,并对4种培养基的pH值进行测定,绘制图像。
实验结果如图3所示。图3(a)为敲除菌株在MRS培养基中的pH值。图3(b)为敲除菌株在MC培养基中的pH值。图3(c)为敲除菌株在MB培养基中的pH值。图3(d)为敲除菌株在MCB培养基中的pH值。
根据图3(a)可知,在MRS培养基中,除了菌株Δldh和Δbsh13-Δldhd外,其他菌株的pH值在培养24h时均降至4.0,菌株之间无显著变化。根据图3(b)可知,在MC培养基中,菌株Δldh的pH值在24h时提高至6.11,Δldhl也达到4.28,其余菌株也略有提高。。根据图3(c)可知,在MB培养基中,菌株Δldh的pH值在24h时显著提高至6.13,Δldhd和Δldhl在24h也分别达到4.49和4.20,其余菌株pH值的波动不明显。根据图3(d)可知,在MCB培养基中,菌株Δldh的pH值在24h时提高至6.15,Δldhd和Δldhl在24h也分别达到4.74和4.34。总体来看,与MRS培养基相比,添加胆固醇和牛胆汁对大多数菌株的培养基pH值没有显著影响,双敲除l型和d型的ldh基因的菌株Δldh在4种培养基中培养24h时培养基pH值还高于初始培养基的pH值6.04,菌株的pH值呈现显著增加的趋势。根据图3可知,菌株Δldh在4种培养基中培养24h时培养基pH值还高于初始培养基的pH值6.04,说明双敲除l型和d型的ldh基因的作用显著,在菌株Δldh生长过程中能够有效地抑制丙酮酸转化为乳酸的过程。
<实施例5>
胆固醇对敲除菌株降胆固醇能力的影响实验
实验方法:用改良的OPA法测定野生型菌株AR113和敲除菌株在MC、MCB培养基中的胆固醇去除率。
实验结果如图4所示。图4(a)为敲除菌株在MC培养基中培养24h的胆固醇去除率。图4(b)为敲除菌株在MCB培养基中培养24h的胆固醇去除率。
根据图4(a)可知,在MC培养基中,除了菌株Δbsh1和Δbsh2外,其余菌株都与AR113相比都具有显著性差异(p<0.05)。Δbsh13的胆固醇去除率降至约10%,与Δbsh3、Δbsh134、Δldh以及Δbsh13-Δldhd四株菌有显著性差异,而Δbsh3、Δldh和Δldh之间无显著性差异,表明基因bsh13对菌株降胆固醇贡献较大。根据图4(b)可知,添加牛胆汁后,与AR113相比,所有菌株的胆固醇去除率都显著降低了,Δbsh1、Δbsh2、Δbsh3、Δbsh132、Δbsh134、Δldhl、Δldhd、Δbsh13-Δldhl和Δbsh13-Δldhd之间无显著差异性,但Δbsh13,Δldh的作用效果较为明显,胆固醇去除率仅为6%和5%,但是二者差异不太明显,说明bsh13和ldh在降胆固醇机制中发挥重要作用,作用效果相似。相比于Δldhl、Δldhd的降胆固醇能力,Δldh的胆固醇去除率显著性降低,表明敲除ldh基因对菌株降胆固醇能力有明显的抑制,进而表明pH值对胆固醇去除的作用影响较大。
实施例的作用与效果
根据本实施例所涉及的降低菌株胆固醇能力的方法,因为包括如下步骤:将菌株加入到培养基中,并加入胆固醇和\或牛胆汁进行培养。因为乳酸脱氢酶是催化丙酮酸转化为乳酸的关键酶,如果阻断这一转化途径,培养基的pH值会发生变化,基于这一假设通过敲除ldh关键酶基因,再结合敲除bsh基因的菌株进行一系列实验验证,从而得出培养基中的pH值会对菌株的降胆固醇能力产生一定的影响。所以本实施例通过对比野生型菌株L.plantarum AR113和敲除菌株的生长特性、培养基pH值及其降胆固醇能力的差异,来佐证BSH和培养基pH值对益生菌L.plantarumAR113降解胆固醇的影响。
上述实施方式为本发明的优选案例,并不用来限制本发明的保护范围。

5.根据权利要求1所述的降低菌株胆固醇能力的方法,其特征在于:
其中,所述培养基为MRS培养基、含有胆固醇的MRS培养基、含有牛胆汁的MRS培养基或含有胆固醇和牛胆汁的MRS培养基中的任意一种。
6.根据权利要求1所述的降低菌株胆固醇能力的方法,其特征在于:
其中,所述菌株为植物乳杆菌AR113。
