Abstract
The invention provides a medicine or cosmetic composition for dry skin and a preparation method and application thereof, belonging to the field of traditional Chinese medicines. The composition is a pharmaceutical preparation or a cosmetic prepared from the following raw material medicines in parts by weight: 5-35 parts of rheum officinale, 5-25 parts of purslane, 5-35 parts of scutellaria baicalensis, 4-16 parts of soapberry, 1-10 parts of fructus cnidii, 1-10 parts of liquorice, 1-8 parts of sanguisorba officinalis and 1-8 parts of cortex toonae sinensis. The composition has the advantages of simple medicine and low cost; the compatibility of the medicines is precise and appropriate, the medicine has good improvement and treatment effects on dry skin, and can effectively improve and treat dry skin, especially symptoms of dry skin, redness, itching and the like of atopic dermatitis; compared with single medicine, the composition of the invention has synergistic effect. The weight ratio of the rhubarb to the astragalus to the purslane to the soapberry to the fructus cnidii to the liquorice to the garden burnet to the cortex toonae sinensis is 16:16:12:8:5:5:3, and the optimal effect is achieved. The composition can be used for preparing medicines or cosmetics for improving or treating dry skin, and has a good application prospect.
Description
Technical Field
The invention belongs to the field of traditional Chinese medicines, and particularly relates to a medicine or cosmetic composition for dry skin, and a preparation method and application thereof.
Background
Dry skin refers to a skin with less than 10% of the water content of the stratum corneum due to increased transepidermal water loss resulting from decreased sebaceous gland secretion and skin barrier damage. The skin of the skin is light and thin, and the skin is rarely provided with acne and pimples, and pores are not obvious. Dry skin is divided into two cases, water-deficient and oil-deficient. Dry skin is associated with genetics, age, sex, environment, season, skin care regimen, health condition, and the like. If ichthyosis is manifested as dry skin, the disease is one of hereditary diseases; children and the elderly are prone to dry skin; dry skin is easy to appear in autumn and winter; poor health conditions such as anemia and blood circulation can cause dry skin; the skin barrier can be damaged by not caring the skin or using the skin care product improperly, and the clinical manifestation is dry skin.
The dry skin is dry and rough, even has chaps, is seriously damaged and aggravated when being serious, has pruritus, causes dysphoria, and seriously influences the work and life of people. The most important point of dry skin care is to ensure that the skin is adequately moisturized. When selecting the skin cleaning and caring product, the skin cleaning product with strong alkalinity, fruit acid and scrub is used with caution so as to prevent the secretion of sebum and sweat from being inhibited, damage the skin barrier and make the skin drier; after the mild low-sensitivity facial cleanser is used for thoroughly cleaning the face, moisturizing lotion or emulsion and cream are immediately used for supplementing skin moisture; making one or two moisturizing masks every week to enhance moisturizing; cleaning skin before sleeping and massaging for 3-5 minutes to improve facial blood circulation, and properly using night cream; the next morning, after cleansing, the skin is kept moist by using lotion or nourishing cream. However, these methods are not effective for dry skin patients.
TCM refers to various normal fluids in the body as body fluids. The traditional Chinese medicine considers that the loss of body fluid is related to internal dryness damaging the body fluid and fire evil flourishing. Many natural plants have moisturizing effects, such as purslane, liquorice, garden burnet, soapberry and the like, and the medicaments can be used for moisturizing the skin, but the effect is not good when the medicaments are used alone. Patent CN110664635A discloses a moisturizing and hydrating mask composition, a mask and a preparation method thereof, which mainly uses extracts of plants such as purslane, and the like, has various raw materials and complex preparation, and is mainly used for moisturizing and moisturizing. However, the improvement of dry skin is not only moisturizing, but the composition does not improve the symptoms of redness and itching of dry skin well. The research on the composition which has simple raw materials and low cost, has the moisturizing effect and can improve and/or treat xerosis cutis is of great significance.

The present invention also provides a process for preparing the aforementioned pharmaceutical or cosmetic composition, comprising the steps of: directly pulverizing the raw materials in weight ratio, or extracting with water or organic solvent, and adding pharmaceutically or cosmetically acceptable adjuvants or auxiliary components.
Further, the aforementioned method comprises the steps of:
(1) weighing the raw material medicines in the weight ratio;
(2) drying the raw materials, pulverizing, adding water, and decocting to obtain extractive solution;
(3) centrifuging the extractive solution, collecting supernatant, filtering the supernatant with macroporous resin, eluting with 70% ethanol, collecting ethanol eluate, concentrating, and centrifuging to obtain active ingredient extract;
(4) adding pharmaceutically or cosmetically acceptable adjuvants or auxiliary components into the obtained active ingredient extract to obtain medicinal preparation or cosmetic;
preferably, the first and second electrodes are formed of a metal,
in the step (2), the drying temperature is 60-100 ℃; and/or in the step (2), the volume of the water is 8-10 times of the total weight of the raw material medicines (mL/g); and/or in the step (2), the decocting times are 1-3 times; and/or in the step (2), the decoction time is 30-60 min; and/or, in the step (3), the centrifugation is carried out at 4000 r/min; and/or, in the step (3), the macroporous resin is macroporous resin D101; and/or, in the step (3), the concentration is to concentrate the ethanol eluent to 60% of the weight of the raw material medicine.
The invention also provides the application of the medicine or the cosmetic composition in preparing medicines or cosmetics for improving or treating dry skin.
Further, the medicine or cosmetic is a medicine or cosmetic for improving or treating dry skin, redness, itching caused by dry skin;
preferably, the dry skin is atopic dermatitis.
The composition is prepared from eight raw material medicines of rhubarb, astragalus, purslane, soapberry, fructus cnidii, liquorice, garden burnet and ailanthus bark. Wherein, the purslane is a monarch drug, which can clear away heat and toxic material, cool blood and stop bleeding; the rhubarb and the scutellaria are used as ministerial drugs and are used for detoxifying and assisting monarch drugs; fructus Cnidii, fructus Sapindi Mukouossi, and cortex Ailanthi as adjuvant drugs for relieving itching and stopping bleeding; the sanguisorba officinalis is an adjuvant drug, so that the effects of removing dampness, cooling blood and stopping bleeding are achieved, and exudation bleeding of skin and other pathological changes are relieved; the liquorice is used as a guiding drug, clears away heat and toxic materials, coordinates the effects of the other drugs and plays a role in cooperation with other drugs.

All the technologies realized based on the above contents of the present invention belong to the scope of the present invention.
Detailed Description
The raw materials and equipment used in the embodiment of the present invention are known products and obtained by purchasing commercially available products.
Example 1 preparation of a composition according to the invention
1. The formula of the raw material medicine is as follows: 16g of rheum officinale, 12g of purslane, 16g of scutellaria baicalensis, 8g of Chinese soapberry, 5g of fructus cnidii, 5g of liquorice, 3g of garden burnet and 3g of ailanthus bark.
2. The preparation method comprises the following steps: weighing the raw materials according to the weight, drying the raw materials at 60-100 ℃, crushing, adding water with the volume being 8 times of the total weight of the raw materials (mL/g), and decocting for 3 times, wherein the decocting time is 30-60 min each time, so as to obtain an extracting solution; centrifuging the extract at 4000r/min, and collecting supernatant; filtering the supernatant with macroporous resin D101, washing with water to colorless, eluting with 70% ethanol to colorless, collecting ethanol eluate, and recovering ethanol. Concentrating the ethanol eluate to 60% of the weight of the raw materials, centrifuging, and collecting supernatant to obtain active ingredient extract. The cream of the composition of the present invention is prepared by adding the active ingredient extract to the base.
The matrix comprises the following components in percentage by weight: 35g of water, 5g of glycerol, 2g of mineral oil, 1g of dehydroacetic acid, 2g of goat glycerol, 1g of triglyceride, 1.5g of hexadecanoate, 1.331 g of cetearyl alcohol, 1.5g of hexadecanol, 201.5 g of polysorbate, 1.2g of polyisobutylene, 1g of isohexadecane, 801.2 g of tween, 3.5g of avocado oil, 1.3g of sodium acrylate, 3g of shea butter, 1g of xanthan gum, 1.2g of carbomer, 1.6g of phytosphingosine, 1.3g of cholesteryl ester, 2g of sodium laurate, 32.5 g of ceramide, 6-II 2.5g of ceramide, 2.5g of pentadecanolide, 3.2g of vitamin E, 1.5g of potassium sorbate, 1g of tripotassium citrate, 1.5g of butanediol, 1.5g of pentanediol, 1g of benzyl alcohol, 3g of glycyrrhizic acid dipotassium bisabolote, 1g of lactic acid, and 5g of hyaluronic acid.
Every 100g of matrix is added with 15g of active ingredient extract.
Example 2 preparation of the composition of the invention
1. The formula of the raw material medicine is as follows: 12g of rhubarb, 10g of purslane, 12g of scutellaria baicalensis, 8g of Chinese soapberry, 5g of fructus cnidii, 5g of liquorice, 3g of garden burnet and 3g of ailanthus bark.
2. The preparation method comprises the following steps: the active ingredient extract and cream were prepared in the same manner as in example 1.
Example 3 preparation of the composition of the invention
1. The formula of the raw material medicine is as follows: 20g of rheum officinale, 15g of purslane, 28g of scutellaria baicalensis, 8g of Chinese soapberry, 5g of fructus cnidii, 5g of liquorice, 3g of garden burnet and 3g of ailanthus bark.
2. The preparation method comprises the following steps: the active ingredient extract and cream were prepared in the same manner as in example 1.
The advantageous effects of the present invention are demonstrated by specific test examples below.
Test example 1 test for clinical Effect of the composition of the present invention
1. Clinical data
A total of 275 patients with dry skin (dry skin, redness, itching) were selected.
1.1 inclusion criteria
Firstly, patients diagnosed with dry skin have symptoms of dry skin, redness and itching;
② the patients are 18-45 years old.

Those not meeting the inclusion standard;
patients and family members refuse to cooperate with the treatment;

and fourthly, the patients are participating in other medicine clinical tests.
2. Method of treatment
Patients were randomly divided into 11 groups and cream prepared according to examples 1 to 3 of the present invention and control groups 1 to 8 were used for 15 days, respectively. The using method comprises the following steps: the cream is applied to affected part after washing face in the morning and evening every day.
Control 1 (rhubarb): weighing raw material medicines of rhubarb, drying the raw materials at 60-100 ℃, crushing, adding water with the volume being 8 times of the total weight of the raw material medicines (mL/g), and decocting for 3 times, wherein the decocting time is 30-60 min each time, so as to obtain an extracting solution; centrifuging the extract at 4000r/min, and collecting supernatant; filtering the supernatant with macroporous resin D101, washing with water to colorless, eluting with 70% ethanol to colorless, collecting ethanol eluate, and recovering ethanol. Concentrating the ethanol eluate to 60% of the weight of the raw materials, centrifuging, and collecting supernatant to obtain active ingredient extract. Adding the active ingredient extract into the matrix, and making into cream. The matrix comprises the following components in percentage by weight: 35g of water, 5g of glycerol, 2g of mineral oil, 1g of dehydroacetic acid, 2g of goat glycerol, 1g of triglyceride, 1.5g of hexadecanoate, 1.331 g of cetearyl alcohol, 1.5g of hexadecanol, 201.5 g of polysorbate, 1.2g of polyisobutylene, 1g of isohexadecane, 801.2 g of tween, 3.5g of avocado oil, 1.3g of sodium acrylate, 3g of shea butter, 1g of xanthan gum, 1.2g of carbomer, 1.6g of phytosphingosine, 1.3g of cholesteryl ester, 2g of sodium laurate, 32.5 g of ceramide, 6-II 2.5g of ceramide, 2.5g of pentadecanolide, 3.2g of vitamin E, 1.5g of potassium sorbate, 1g of tripotassium citrate, 1.5g of butanediol, 1.5g of pentanediol, 1g of benzyl alcohol, 3g of glycyrrhizic acid dipotassium bisabolote, 1g of lactic acid, and 5g of hyaluronic acid. Every 100g of matrix is added with 15g of active ingredient extract.
Control 2 (purslane): the preparation method of the active ingredient extract and cream is the same as that of the control group 1, and only the raw material medicine rhubarb is changed into purslane.
Control group 3 (scutellaria): the preparation method of the active ingredient extract and cream is the same as that of the control group 1, and only the raw material medicine rhubarb is changed into the baical skullcap root.
Control group 4 (soapberry): the preparation method of the active ingredient extract and cream is the same as that of the control group 1, and only the raw material medicine rhubarb is changed into soapberry.
Control 5 (cnidium fruit): the preparation method of the active ingredient extract and cream is the same as that of the control group 1, and only the raw material medicine rhubarb is changed into the common cnidium fruit.
Control 6 (licorice): the preparation method of the active ingredient extract and cream is the same as that of the control group 1, and only the raw material medicine rhubarb is changed into liquorice.
Control 7 (sanguisorba): the preparation method of the active ingredient extract and cream is the same as that of the control group 1, and only the raw material medicine rhubarb is changed into sanguisorba officinalis.
Control group 8 (ailanthus): the preparation method of the active ingredient extract and cream is the same as that of the control group 1, and only the raw material medicine rhubarb is changed into the toona sinensis skin.
3. Determination of efficacy and treatment outcome
The determination of the curative effect and the therapeutic effect of each example and the control group for improving dry skin are shown in tables 1 to 3. The total effective rate is (number of effective cases + number of effective cases)/the total number of cases multiplied by 100 percent
