Description

Composition for improving skin micro-ecology and preparation method and application thereof
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a composition for improving skin microecology and a preparation method and application thereof, in particular to a composition for improving skin barrier function and skin surface microbial population distribution and a preparation method and application thereof.
Background
It is well known that the skin, the largest organ of the human body, protects our body against external environmental insults. The stratum corneum in the epidermis is composed of a dense layer of cells, and plays an extremely important barrier role for the human body due to its compact structure.
With the development and improvement of the technology, the skin is layered more and more carefully, and in addition to needing to pay attention to the physical barrier of the skin which is often contacted, the newly introduced skin microbial barrier is not separated from the health of the skin in daily skin care. As a human body surface barrier-skin, there are often some microorganisms present, some of which may be parasitic on the skin of the human body for a longer period of time as a result of long-term adaptation, known as the normal skin microflora, also known as the normal skin flora. The normal flora and the host macroorganisms and the external environment form a habitat, microorganisms in the habitat colonize certain organs of the macroorganisms to form microbial localization, and the skin is also one of the localizations. This interdependence constitutes an ecosystem. The skin has 'good flora' which is the crucial point of the good state of our skin, and the microbial barrier and the skin physical barrier play the role of protecting our skin at the same time.
However, with the development of modern society, the daily working and rest time of people is not regular, the environmental conditions are worse and worse, and the adverse effects on the skin are worse and worse due to the mental pressure from all aspects and the frequent cosmetic and makeup removing habits. The physical barrier and the microbial barrier of the skin are damaged, so that the skin is subjected to various troubles, the skin becomes fragile, the current skin barrier repair method mainly focuses on the repair of the physical barrier, and moisture is retained on the skin by externally coating composite components of humectants such as grease blocking moisturizers, hydrophilic matrix moisture absorbing moisturizers, hydrating moisturizers and the like, so that the method cannot fundamentally solve the problem of repairing the skin barrier dysfunction, and has little effect particularly on the problem of the damage of the microbial barrier.
CN111265468A discloses a skin double-barrier repair composition and application thereof, wherein prebiotics, probiotics and microbial metabolites in skin microbial components are simulated by mutual matching of yeast bifida fermentation product filtrate, oat fermentation filtrate and skin-like lipid complex, so that the skin microbial barrier is effectively stabilized and balanced; the skin lipid complex and the oat kernel oil mainly repair a skin brick wall structure and improve the composition of stratum corneum lipid so as to achieve the aim of repairing a physical barrier of the skin, and the skin microbial barrier and the physical barrier of the skin are repaired simultaneously through the mutual matching of the two barrier repairing components, so that the skin is helped to reestablish the stratum corneum with a complete structure and establish a healthy and good microbial colony ecosphere.
CN110934804A discloses a skin micro-ecological nursing facial mask added with distiller's grains, which comprises the following substances: the alcohol dreg facial mask provided by the invention is based on skin microbiology, improves the skin microorganism problem, and can effectively balance the skin microorganism population and the quantity, thereby leading the skin to return to a healthy state.
However, there are few strategies in the prior art for better improving both the microbial barrier and the physical barrier of the skin, and therefore, it is of great interest to develop a product for improving the skin barrier function and the skin surface microbial flora.

Evaluation test:
(1) testing of skin Water loss (TEWL)
1.1 test Instrument
Skin moisture loss (TEWL) Tewameter TM300 test probe manufactured by Courage + Khazaka, Germany (CK, Germany).
1.2 test methods
160 volunteers are selected, wherein the age is 20-45 years, and the volunteers are half of the male and the female. The groups were randomized into 16 groups of 5 males and 5 females. The using method comprises the following steps: 0.4g of the product (example 1, application examples 1-9 and comparative application examples 1-6) was applied evenly to the face 2 times a day for 14 consecutive days. Prior to the test, subjects were asked to wash their face and to enter a climate controlled room (22. + -. 1 ℃ C., 50% relative humidity) 2 hours after washing and sit still for more than 20min and remain relaxed. The use was continued for 14 days and the subjects were asked to return visits on day 7 (d7), day 14 (d14) of the trial.
1.3 test results are shown in Table 1 (data are presented as mean values):
TABLE 1
Figure BDA0002841021170000141
Figure BDA0002841021170000151
From the data results in table 1, it can be seen that: the composition of the invention can reduce the percutaneous water loss of the skin, so that the barrier of the skin is better.
(2) Testing of diversity and abundance of skin microorganism shannon
160 volunteers are selected, wherein the age is 20-45 years, and the volunteers are half of the male and the female. The groups were randomized into 16 groups of 5 males and 5 females. The using method comprises the following steps: 0.4g of the product (example 1, application examples 1-9 and comparative application examples 1-6) was applied evenly to the face 2 times a day for 7 consecutive days.
Analyzing a DNA sequencing result of a 16S rNA gene V3 region and a Shannon diversity index through a microorganism detector and bioinformatics software; the relative abundance of different classes was further evaluated by analyzing the sequencing results of 7 hypervariable regions on the 16S rNA gene. The test results are shown in table 2 (data are presented as mean values):
TABLE 2
Figure BDA0002841021170000152
Figure BDA0002841021170000161
Wherein a lower shannon diversity index value indicates a lower microbial diversity, which is often associated with skin unhealthy. Wherein the staphylococcaceae family is associated with skin diseases such as atopic dermatitis and psoriasis; moraxeaceae belongs to the phylum Proteobacteria, gamma Proteobacteria, and has been shown to have potential anti-inflammatory properties, which are associated with immune homeostasis.
From the data results in table 2, it can be seen that: the composition can increase the microbial diversity of the skin, promote the generation of beneficial bacteria and reduce harmful bacteria.
