Abstract
The application of the chlorella extract and the lactobacillus fermentation product in the care product comprises the application of the chlorella extract and the lactobacillus fermentation product in balancing skin micro-ecology, relieving skin and/or reducing the dosage of a preservative of a skin care product, and the application in the corresponding care product and the preparation. The chlorella extract and the lactobacillus fermentation product can adjust unbalanced skin micro-ecology to normal skin micro-ecology, recover skin barrier and improve skin state; can also be applied to skin for relieving, effectively eliminating red and relieving skin. Besides being applied to skin care products to improve the skin care effect, the chlorella extract and the lactobacillus fermentation product are also found to reduce the dosage of preservative and ensure the preservative effect after being added into the skin care products. Based on the new application provided by the invention, the nursing product with the functions of balancing skin micro-ecology, relieving skin and low preservative content is convenient to prepare, meets more skin nursing requirements, and has high safety and wide applicable population.
Description
Technical Field
The invention relates to the technical field of skin care products, and particularly relates to an application of chlorella extract and lactobacillus fermentation products in a care product.
Background
Chlorella (Chlorella) belongs to a green alga widely distributed on the earth from the frigid zone to the tropical zone, and is an approximately spherical unicellular organism with a diameter of 2-10 μm. Chlorella is rich in protein, lipid, edible fiber, biotin, cellulose, unsaturated fatty acid, bioactive polysaccharide, nucleic acid, vitamins, microelements, minerals, folic acid and chlorophyll. The chlorella extract is widely applied to various cosmetics, generally exists as a skin conditioner, and has the effects of promoting the synthesis of collagen and elastin and the like.
Lactobacillus the lactobacillus family, named for the large amount of lactic acid produced by fermenting sugars. The lactobacillus fermentation product contains lactic acid, peptide, thalli polysaccharide, hydrogen peroxide, bacteriostat, flavonoid, microorganism, mineral substances and the like, and has the effects of softening cutin, promoting collagen regeneration and the like.
Although chlorella extract and lactobacillus fermentation products are applied to cosmetics in the prior art, the chlorella extract and lactobacillus fermentation products are generally used as auxiliary components for promoting collagen synthesis, and whether the chlorella extract and lactobacillus fermentation products have other applications or not is not researched yet. Meanwhile, the chlorella extract and the lactobacillus fermentation product are natural components with high safety and can be widely added into various daily chemical products, so that the application development of the chlorella extract and the lactobacillus fermentation product is very important; the method is beneficial to providing a safe nursing product with more efficacies on the basis of high safety of chlorella extract and lactobacillus fermentation products so as to meet more nursing requirements.
Disclosure of Invention
The invention aims to overcome at least one defect of the prior art, provides the application of the chlorella extract and the lactobacillus fermentation product in the nursing product, and the nursing product with the functions of balancing skin micro-ecology, relieving skin and reducing preservative content is conveniently prepared by the compound application of the chlorella extract and the lactobacillus, so that more skin nursing requirements are met, the safety is high, and the application range is wide.
It is an object of the present invention to provide the use of a chlorella extract in combination with a lactobacillus fermentation product for the preparation of a care product for balancing skin micro-ecology, soothing the skin and/or reducing the amount of preservatives used. The effects of balancing skin micro-ecology, relieving skin and reducing the using amount of preservative are realized through the chlorella extract and lactobacillus compound composition.
The normal skin micro-ecosystem has competition and restriction relationship, and when the skin flora is abnormally increased, reduced or even disappeared, the skin micro-ecology is destroyed. Changes in the skin's micro-ecology are associated with a variety of inflammatory reactions and infectious diseases on the skin, such as inflammatory reactions like atopic dermatitis, acne and chronic wounds. Therefore, the balance of skin microbial flora is the basis of skin normal, and the skin environment is maintained to be stable.
However, when the skin is excessively clean, chemical substances are irritated, medicines (especially antibiotics) and skin free radicals are increased, so that sebum peroxidation and the like destroy the flora balance, pathogenic bacteria are explanted and invaded, and when resident flora is disordered, the skin barrier function is damaged, so that the problems of water and oil imbalance, sensitivity, inflammation and the like of the skin are easily caused.
In one or more embodiments of the present invention, the inventors have found that the addition of chlorella extract and lactobacillus fermentation product can effectively adjust the unbalanced skin micro-ecology to the normal skin micro-ecology, i.e., balance the skin microbial flora and restore the skin to the normal state. The corresponding nursing product is prepared by the chlorella extract and the lactobacillus fermentation product, and the problem of deficiency of the prior art in balancing skin microecological products can be solved. Meanwhile, the inventor also finds that the chlorella extract and the lactobacillus fermentation product are combined to provide remarkable effects of moisturizing and relieving the skin. The chlorella extract and the lactobacillus fermentation product can obviously improve the corresponding skin care effect and improve the skin care effect from multiple aspects.
In addition, the chlorella extract and lactobacillus fermentation product are found to have the effect of reducing the amount of preservatives. Specifically, the preservative means a substance having a sustained inhibitory effect on the growth of microorganisms. The preservative has the important characteristic of inhibiting the putrefaction under different conditions, and particularly has a persistent preservative and bacteriostatic effect when the general sterilization effect is insufficient. The chemical synthetic preservative is widely applied to the cosmetic industry due to the advantages of low cost, good preservative effect, convenient application and the like, but the chemical preservative has toxic and side effects of different degrees and has irritation risk to skin when the dosage is large. In one or more embodiments of the invention, the invention was tested for preservative efficacy and verified that chlorella extract and lactobacillus fermentation product also have utility in reducing the amount of preservative used. At least one of chlorella extract and lactobacillus is added, so that the dosage of the preservative can be reduced on the premise of ensuring the preservative effect.
More than one embodiment of the invention discovers that the chlorella extract and the lactobacillus fermentation product are compounded and used, and the chlorella extract and the lactobacillus fermentation product at least have the purposes of balancing microbial colonies of skin, soothing skin and reducing the using amount of preservatives. Based on the application, the preparation of corresponding care products is facilitated, so that the requirements of the prior art on skin micro-ecology balance, skin soothing and low-preservative care products are met.
Further, balancing skin micro-ecology includes regulating the abundance of flora of propionibacterium, acinetobacter, corynebacterium, staphylococcus, bacteroides, prevotella, weissella, bifidobacterium, lactobacillus, and/or waterborne bacteria on skin. In one or more embodiments of the present invention, it was verified that at least chlorella extract and lactobacillus fermentation product have a regulating effect on the above microorganisms and are regulated to a normal skin micro-ecological level.
Further, the preservative comprises phenoxyethanol and/or ethylhexyl glycerin. In more than one embodiment of the invention, the chlorella extract and the lactobacillus fermentation product are added, so that the use amount of the preservatives phenoxyethanol and ethylhexyl glycerol can be effectively reduced, and meanwhile, good preservative effect is kept.
Further, the care products comprise skin-care moisturizing lotion, skin-care essence, skin-care cream, skin-care emulsion, skin-care soothing lotion and skin-care facial mask.
The invention also aims to provide an application method for reducing the using amount of the preservative in the skin care product by combining the chlorella extract and the lactobacillus fermentation product. Namely, the application method for reducing the using amount of the preservative in the skin care product by providing the chlorella extract and lactobacillus fermentation product composition comprises the following steps: adding chlorella extract and lactobacillus fermentation product composition into skin care product. As described above, in one or more embodiments of the present invention, the amount of preservative in the skin care product is reduced by adding the chlorella extract and the lactobacillus fermentation product, while the preservative efficacy is ensured.
Further, the preservative comprises phenoxyethanol and/or ethylhexyl glycerin.
Still further, preservatives include phenoxyethanol and ethylhexylglycerin.
It is still another object of the present invention to provide a skin care composition comprising chlorella extract and lactobacillus fermentation product.
Still another object of the present invention is to provide a moisturizing skin care product comprising:
phase A: 2-4 parts of glycerol; 0.03-0.06 part of EDTA-2 Na; 0.03-0.06 part of sodium hyaluronate; 0.3-0.6 part of panthenol; 0.05-0.15 part of PEG-40 hydrogenated castor oil;
phase B: 0.7-1 part of preservative; and at least one selected from chlorella extract and lactobacillus fermentation product;
the preservative is a mixture of phenoxyethanol and ethylhexyl glycerin.
In more than one embodiment of the invention, the moisturizing skin care product using the chlorella extract and the lactobacillus fermentation product can obviously improve the water content of the skin cuticle and has an obvious moisturizing effect. The chlorella extract and the lactobacillus fermentation product are compounded for use, so that the synergistic effect is achieved, and the water content of the skin cuticle is obviously improved.

Still another object of the present invention is to provide a method for preparing the moisturizing skin care product, comprising the steps of:
s1, adding the phase A materials into a main pot, heating to 80-85 ℃, stirring for dissolving, and keeping the temperature for 10-15 min;
s2, cooling to 40-45 ℃, adding the phase B materials into the main pot, and uniformly stirring;
s3, cooling to 38 ℃, performing quality inspection, and discharging to obtain the moisturizing skin care product.
The preparation method of the moisturizing skin care product is simple, easy to operate and suitable for large-scale production.
Compared with the prior art, the invention has the beneficial effects that: the chlorella extract and the lactobacillus fermentation product have the function of balancing microbial colonies of the skin, regulate unbalanced skin micro-ecology to normal skin micro-ecology, recover skin barriers and improve the skin state. Meanwhile, the inventor also finds that the chlorella extract and the lactobacillus fermentation product can be applied to skin soothing and can effectively remove red and soothe skin. Besides the chlorella extract and the lactobacillus fermentation product are applied to the skin care product to improve the skin care effect, the inventor also finds that the dosage of the preservative can be reduced and the preservative effect is ensured after the chlorella extract and the lactobacillus fermentation product are added into the skin care product; based on the safety of chlorella extract and lactobacillus fermentation products, the damage of components such as preservative and the like to the skin surface can be obviously reduced, and the adverse reaction of skin care products is reduced. The chlorella extract and the lactobacillus fermentation product composition are matched for use, so that the dosage of the chemical preservative can be reduced while the skin microbial flora is balanced, the skin irritation effect of the chemical preservative under the condition of large dosage is reduced, the problem of large toxic and side effects of the existing chemical preservative is solved, and the chlorella extract and the lactobacillus fermentation product composition have good preservative capability, wide antibacterial spectrum and wide antibacterial range. Therefore, the chlorella extract and the lactobacillus fermentation product can be applied to skin care products to provide the skin care products with the functions of balancing the microbial colonies of the skin, relieving the skin and reducing the preservative; also can provide a product which aims at promoting the balance of skin microbial flora, enhancing the skin soothing effect and reducing the dosage of the preservative by utilizing the chlorella extract and the lactobacillus fermentation product. Based on the new application provided by the invention, a nursing product with the functions of balancing microbial colonies of the skin, relieving the skin and reducing the content of preservatives can be conveniently prepared, more skin nursing requirements are met, the safety is high, and the application population is wide.

The embodiment provides a moisturizing skin care product which comprises the following components in parts by weight:

The embodiment also provides a moisturizing skin care product, and the phase B further comprises at least one selected from chlorella extract and lactobacillus fermentation products. The preparation method comprises the following steps:
s1, adding the phase A materials into a main pot, heating to 80-85 ℃, stirring for dissolving, and keeping the temperature for 10-15 min;

Example 1
In the embodiment, according to the component proportion of the formula 1-10, the moisturizing skin care product is prepared according to the following steps:
s1, adding the phase A materials into a main pot, heating to 80-85 ℃, stirring for dissolving, and keeping the temperature for 10-15 min;
s2, cooling to 40-45 ℃, adding the phase B materials into the main pot, and uniformly stirring;
s3, cooling to 38 ℃, checking to be qualified, and discharging to obtain the moisturizing skin care product.

Experimental example 1
Chlorella extract and lactobacillus fermentation product composition for balancing skin micro-ecology, keeping moisture and relieving skin
120 subjects aged 18-45 years were tested in this study, all subjects had a predisposition to inflammatory reactions in their skin type, and subjects were divided into 4 groups of 30 on average, each group participating in a double-blind randomized clinical study in accordance with the ethical principles of medical research. The subjects in each group used 1ml of formula 4, 5, 6, 7 samples each day, morning and evening, respectively, on the face. Before starting the test on the same day D 0 And day 28 after testing D 28 The skin microflora (non-invasive skin patch collection, 16S rRNA sequencing of 120 skin sample microorganisms, analysis of flora structure, diversity and gene function), skin stratum corneum water content, facial skin erythema number and erythema area value were tested separately.

1. 16S rRNA sequencing assay
The 16SrRNA sequencing is a technology for carrying out PCR amplification and high-throughput sequencing on a hypermutation region of a gene (namely 16SrDNA) of 16S ribosomal RNA of an environmental sample, and can effectively identify the types and abundance of bacteria and archaea under specific environments. The 16S sequence consists of 9 hypervariable regions (V1-V9) interspersed with conserved regions, and is the most commonly used bacterial classification standard. Extracting DNA of an environmental sample, and amplifying a 16SrDNA gene; and detecting the sequence variation and abundance of the 16SrRNA gene, and reflecting the classification and abundance of bacteria in the environmental sample. The 16S sequence can be used to identify most bacteria. Based on an Illumina Miseq sequencing platform, parallel sequencing of a plurality of samples can be completed at one time, and a plurality of information such as environmental sample species classification, species abundance, population structure, system evolution, community comparison and the like can be provided. The 16S third region (V3) and the fourth region (V4) are sequenced, the total length is about 459bp, and most of human microorganism species can be identified (the accuracy reaches the class I).
In this example, the detection of skin microbiota was performed by 16s rrna sequencing. Specifically, facial sampling was performed for skin microbiota analysis (16S-rDNA sequence, metagenomic analysis); and obtaining diversity index, bacterial composition, abundance of biological flora and stable diversity state.
The standard operation flow of the 16S rRNA sequencing comprises the following steps:
(1) extracting by using a QIAamp DNA extraction kit, and detecting the extracted DNA;
(2) detecting the concentration of the DNA by a fluorescence spectrophotometer, and detecting the quality of the DNA by electrophoresis of 1% agarose gel;
(3) adjusting the concentration of the DNA solution, storing the DNA working solution at 4 ℃ and storing the storage solution at-20 ℃;
(4) carrying out PCR amplification on the V3-V4 region of the 16S rRNA gene of the sample;
(5) and (3) glue recovery and purification: tapping and recovering the target strip to obtain a purified sample;
(6) quantification of each sample: quantifying each sample by using a Qubit fluorescence quantifier;
(7) constructing an on-machine library by adopting a standard IlluminaTruSeq DNA library Preparation experimental process (IlluminaTruSeq DNA Sample Preparation Guide); in-silico sequencing was performed using illumina miseq PE 300.
2. And (3) test results:
species composition analysis was performed in this example. In order to obtain species classification information corresponding to each OTU, performing taxonomic analysis on OTU representative sequences with 97% similarity level by using an RDP classifier Bayesian algorithm, and performing taxonomic analysis on OTU representative sequences at each taxonomic level: domain, kingdom, phylum, class, order, family, genus, and species.
All sequences were subjected to otu (operational taxomic units) partition according to 97% similarity and statistical analysis of the bioinformatics. According to the result of the taxonomic analysis, the taxonomic comparison condition of one or more samples at each classification level can be known. The data is now presented by the relative abundance changes of the species of the genus flora structure.
Relative abundance of the flora structural species in healthy facial skin is shown in table 2.

The test result shows that the moisturizing water containing the chlorella extract and the lactobacillus fermentation product composition can restore the disordered microbial flora of the damaged skin to a more stable and balanced skin microbial flora state close to that of healthy skin after being used for 28 days in the morning and evening.
Second, the function of the chlorella extract and lactobacillus fermentation product composition in skin moisture preservation
1. Stratum corneum moisture test
Testing an instrument: skin Water tester Corneometer CM825 (higher value represents higher water content of stratum corneum, unit C.U.)
The measurement of skin moisture is based on the internationally recognized Corneometer method, i.e. the capacitance method. The measurements are based on the completely different dielectric constants of water and other substances. The skin is a weak conductive medium, and the conductivity coefficient of the skin can be increased along with the moisture content of the skin, so that when the test probe is applied to the skin, the measurement capacitor shows the change of capacitance according to the moisture content of the skin.
2. The result of the detection
In this example, the test results of the test groups of formulas 4, 5, 6 and 7 are shown in table 4.

The results shown in table 4 show that the moisture retention water using the chlorella extract or the lactobacillus fermentation product can improve the moisture content of the skin cuticle, and that when the chlorella extract and the lactobacillus fermentation product are used in combination, the synergistic effect is achieved, and the moisture content of the skin cuticle is obviously improved.
Thirdly, the function of the chlorella extract and lactobacillus fermentation product composition in relieving skin
1. And (3) red removing test:
the skin inflammation index was measured using the American VISIA microanalyzer skin image analyzer (VISIA. TM. Complexion Analysis) at D 0 、D 28 The number and area of erythema on the face of the test person were analyzed separately.
2. The result of the detection
In this example, the test groups of formulas 4, 5, 6 and 7 were tested, specifically, the number of erythema and the area of erythema were tested, and the results shown in tables 5 and 6 were obtained.

After 28 days of moisturizing water containing the chlorella extract and the lactobacillus fermentation product composition is used, the change rate of the number and the area of facial erythema is shown, and the combination of the chlorella extract and the lactobacillus fermentation product composition can effectively reduce the number and the area of redness of skin, effectively remove redness and relieve the skin.
Experimental example 2
Chlorella extract and lactobacillus fermentation product composition with reduced preservative use effect
In this example, the test was performed according to the water-soluble cosmetic preservation efficacy test method CTFA M-3(2007), and the following steps were included:
(1) preparation of a bacterial suspension: respectively inoculating staphylococcus aureus ATCC6538, pseudomonas aeruginosa ATCC9027 and escherichia coli ATCC8739 on a soybean casein agar slant culture medium, and culturing at 37 ℃ for 24 h. Washing thallus Porphyrae with 5ml sterilized 0.85% physiological saline, and determining bacterial colony number according to total bacterial colony number determination method to ensure that bacterial colony number is 10 8 CFU/ml, mixing the above 3 kinds of bacteria suspension in equal volume, and keeping at 4 deg.C.
(2) Preparation of a fungus suspension: respectively inoculating Candida albicans ATCC10231 and Aspergillus niger ATCC16404 on potato glucose agar slant culture medium, culturing Candida albicans at 28 deg.C for 48 hr, washing thallus Porphyrae with 5ml sterilized 0.85% physiological saline, and testing colony number of fungi according to colony total number determination method to ensure that colony number is 10 7 CFU/ml is about; culturing Aspergillus niger at 28 deg.C for 5-7 days, washing thallus Porphyrae with 5ml sterilized 0.85% normal saline containing 0.05% Tween, and testing colony count of fungi according to colony count test method to ensure that colony count is 10 7 CFU/ml. Mixing the suspension of 2 kinds of fungi in the same volume and keeping at 4 deg.C.
(3) Sample inoculation:
taking 2 parts (30 g of each part) of the weighed samples of the formulas 1, 2, 3, 8, 9 and 10 respectively, adding 0.3ml of mixed bacterium suspension and 0.3ml of mixed fungus suspension respectively, and mixing uniformly. The samples are stored at room temperature (22.5 +/-2.5) DEG C, and the colony counts in the cosmetics are detected on 0, 7, 14, 21 and 28 days respectively to judge the preservative efficacy of the cosmetics.
(4) Sample detection:
weighing 1g or 1ml of sample to be detected, adding the sample to a 9ml sterilized normal saline dilution bottle, fully and uniformly shaking to prepare a 1:10 sample diluent, adding 1ml of the diluent to 9ml normal saline, fully and uniformly mixing and diluting to prepare a 1:100 sample diluent, and sequentially carrying out the steps of preparing 1:1000, 1:10000 and the like, and diluting to a proper multiple. 2ml of diluent with proper dilution is selected according to the experimental result of each time, and the diluent is added into 2 sterilized culture dishes, and 1ml of diluent is added into each culture dish. Wherein one of the bacteria and the fungi which can not reach the above standard is judged as unqualified antiseptic effect of the cosmetic.
In this example, the test groups of formulas 1, 2, 3, 8, 9, and 10 were tested, and the test results are shown in table 7 below.

The results are shown in table 7, and the preservation challenge test shows that the chlorella extract and lactobacillus fermentation product composition can reduce the addition amount of the preservative and maintain good preservation effect. Taking formula 9 as an example, after 2 parts of chlorella extract is added and 2 parts of lactobacillus fermentation product is added, the antiseptic performance of the skin care product can be achieved even better than that of other skin care products without chlorella extract and lactobacillus fermentation product and with more antiseptic under the condition of only adding 0.8 part of antiseptic (phenoxyethanol: ethylhexyl glycerol: 9: 1).

Claims (10)
1. Use of a chlorella extract in combination with a lactobacillus fermentation product for the preparation of a care product for balancing skin micro-ecology, soothing skin and/or reducing the amount of preservative used.
2. Use according to claim 1, wherein balancing skin micro-ecology comprises regulating the abundance of propionibacterium, acinetobacter, corynebacterium, staphylococcus, bacteroides, prevotella, weissella, bifidobacterium, lactobacillus and/or aquaticum on skin.
3. Use according to claim 1, wherein the preservative comprises phenoxyethanol and/or ethylhexyl glycerol.
4. The use according to any one of claims 1 to 3, wherein the care product comprises skin-care moisturizer, skin-care essence, skin-care cream, skin-care emulsion, skin-care soothing liquid and skin-care mask.
5. An application method of combining chlorella extract and lactobacillus fermentation product for reducing the using amount of preservative in skin care products is characterized in that the chlorella extract and lactobacillus fermentation product are added into the skin care products.
6. A skin care composition comprising chlorella extract and lactobacillus fermentation product.
7. A moisturizing skin care product, comprising:
phase A: 2-4 parts of glycerol; 0.03-0.06 part of EDTA-2 Na; 0.03-0.06 part of sodium hyaluronate; 0.3-0.6 part of panthenol; 0.05-0.15 part of PEG-40 hydrogenated castor oil;
phase B: 0.7-1 part of preservative; and at least one selected from chlorella extract and lactobacillus fermentation product;
the preservative is a mixture of phenoxyethanol and ethylhexyl glycerin.
8. The moisturizing skin care product of claim 7, wherein the ratio of phenoxyethanol to ethylhexylglycerin is 9: 1.
9. The moisturizing skin care product of claim 8, comprising:
phase A: 3 parts of glycerol; 0.05 part of EDTA-2 Na; 0.05 part of sodium hyaluronate; 0.5 part of panthenol; 0.1 part of PEG-40 hydrogenated castor oil;
phase B: 0.8 part of preservative; 2 parts of chlorella extract; 2 parts of lactobacillus fermentation product.
10. A process for producing a moisturizing skin care product according to any one of claims 7 to 9, comprising the steps of:
s1, adding the phase A materials into a main pot, heating to 80-85 ℃, stirring for dissolving, and keeping the temperature for 10-15 min;
s2, cooling to 40-45 ℃, adding the phase B materials into the main pot, and uniformly stirring;
s3, cooling to 38 ℃, performing quality inspection, and discharging to obtain the moisturizing skin care product.
