Application experiments:
the traditional Chinese medicine formula is used for muscle atrophy resistance experiments in a chronic stress environment, 50 normal male rats are selected, and the average composition is 5 groups: blank control group, stress model group, low dosage group of traditional Chinese medicine, middle dosage group of traditional Chinese medicine and high dosage group of traditional Chinese medicine; except for the blank control group, all the groups are bound for 12 hours every day, stimulated by noise sound for 2 hours, stimulated by ice water and tail clamping pain for 10 minutes, and the method of reversing day and night lasts for 6 weeks to establish a chronic stress rat model. After maintaining the model stimulation and adjusting for 6 weeks, the Chinese medicinal formula is respectively administered as follows: blank control group (not edible), stress model group (not edible), traditional Chinese medicine low dose group (once a day), traditional Chinese medicine medium dose group (twice a day with 12-hour interval), and traditional Chinese medicine high dose group (three times a day with 8-hour interval); after 7 days of continuous dosing, the behavioral and muscle inflammatory responses of each group of model animals were examined. The behavioral experiments included: open field behavior experiments, mechanical pain threshold test and sugar water preference test.
Open field behavior experiments: in a relatively closed indoor environment, placing an experimental model rat from the middle of an open field, and recording the moving distance (m) and the moving speed (m/s) of the rat in the open field; and the activity time(s) of the central zone. The test time per rat was 15 min.
Hanging tail struggle test: hanging various rats on a cross bar 60cm away from the ground for 6min, wherein the fixing point of the rat is a position slightly 10cm away from the tail, observing and recording the motionless latency of the rat within the first 2min and the total motionless time of the rat within the last 4min, and the motionless state is defined as that the rat stops struggling and the body is in a relaxed state.
Mechanical pain threshold test: under silent environment, the mechanical foot-contracting threshold of each group of rats is measured by a Von-Frey cellosilk method, the strength of the cellosilk is started from 4.0g, the left hind paw pelma of the rat is sequentially stimulated, positive reactions of foot contraction, foot swinging, foot lifting and foot licking of the rat are firstly occurred, and the strength of the cellosilk in the positive reactions is recorded as the pain threshold.
And (4) testing syrup preference: the groups of experimental rats were tested for their preference for a 1% sucrose solution. Adaptive feeding is carried out 3 days before the experiment, rats in each experimental group are fasted and water is forbidden for 24 hours on the 4 th day, then 1% sucrose water and a bottle of normal drinking purified water are placed at the same time, the consumption of the sucrose water and the purified water in 1 hour is recorded, and the proportion of the consumption of the sucrose water to the total consumed liquid is calculated as the preference degree of the sucrose water.
After anesthesia, rats in each group were killed by cervical dislocation, gastrocnemius tissue was taken, HE stained to observe pathological changes, and the mitochondrial state of muscle and fluorescent staining of fast and slow muscle fibers were observed under an electron microscope. The levels of HMGB-1, IL-1 beta, IL-6 and TNF-alpha in the muscle tissues of rats in each group are detected by an Elisa kit method.
And (4) analyzing results:
1. open-field behavior experiments are shown in figures 1-5, wherein figure 1 shows the open-field behavior distribution of the blank control group, and it can be seen that rats move uniformly in the open field and move frequently in the central area;
FIG. 2 shows the open field behavior distribution of the stress model group, showing that the total distance of movement of the rat is small in the open field and the rat has no activity basically in the central area;
the traditional Chinese medicine formula has the advantages that the traditional Chinese medicine low-dose group, the traditional Chinese medicine medium-dose group and the traditional Chinese medicine high-dose group are distributed in the open field behavior, the total exercise mileage of a rat in the open field is obviously increased compared with that of a stressed group, and the rat also has obvious activities in a central area.
2. The results of the suspended tail struggle test, the mechanical pain threshold test and the sugar water preference test are shown in fig. 6, and it can be seen that the application of the drug of the invention can improve the struggle time of a stress model, reduce the pain threshold, reduce the pain feeling, significantly improve the sugar water preference of an experimental animal model and improve the stress state of the model.
3. The pathological result and electron microscope detection of the traditional Chinese medicine for resisting rat muscle stress injury are as follows:
the HE staining patterns are shown in FIGS. 7-9. FIG. 7 is a HE staining pattern of a blank control group, FIG. 8 is a HE staining pattern of a stress model group, FIG. 9 is a HE staining pattern of a low dosage group of Chinese medicinal materials, FIG. 10 is a HE staining pattern of a medium dosage group of Chinese medicinal materials, and FIG. 11 is a HE staining pattern of a high dosage group of Chinese medicinal materials;
HE staining chart shows that compared with the blank control group, the stress model has obvious inflammatory reaction in the muscle tissue gaps, the traditional Chinese medicine low-dose group, the traditional Chinese medicine medium-dose group and the traditional Chinese medicine high-dose group have obviously reduced inflammatory reaction even have no inflammatory reaction;
as shown in fig. 12-21, mitochondrial electromicroscopic examination. Fig. 12 and 13 are mitochondrial electron microscopic examination images of a blank control group, fig. 14 and 15 are mitochondrial electron microscopic examination images of a stress model group, fig. 16 and 17 are mitochondrial electron microscopic examination images of a low dosage group of a traditional Chinese medicine, fig. 18 and 19 are mitochondrial electron microscopic examination images of a medium dosage group of a traditional Chinese medicine, and fig. 20 and 21 are mitochondrial electron microscopic examination images of a high dosage group of a traditional Chinese medicine;
mitochondrial electron microscopy images show that the structure of the mitochondrial outer membrane of the blank control group is complete, and the structure of the mitochondrial inner ridge is clear; secondly, the outer membrane structure of mitochondria under a mitochondrial electron microscope of the stress model group is partially incomplete, the inner structure of mitochondria basically disappears, and the mitochondria have serious inner air bubbles and oxidative damage; the outer mitochondrial membrane of the electron microscope of the low-dose group, the middle-dose group and the high-dose group of the traditional Chinese medicine has complete structure, the inner mitochondrial structure is partially damaged, and the oxidation of mitochondria is less along with the increase of the administration dose.
4. The results of fluorescence staining detection are shown in FIGS. 22-26. FIG. 22 is a result of fluorescence staining test of blank control group, FIG. 23 is a result of fluorescence staining test of stress model group, FIG. 24 is a result of fluorescence staining test of low dosage group of Chinese medicine, FIG. 25 is a result of fluorescence staining test of medium dosage group of Chinese medicine, and FIG. 26 is a result of fluorescence staining test of high dosage group of Chinese medicine;
the fluorescent staining detection result shows that compared with a blank control group, red fast muscle tissue in a stress model group is seriously lost, and green slow muscle fibers are obviously increased. Compared with stress model group, the traditional Chinese medicine formula of the invention, the traditional Chinese medicine low dose group, the traditional Chinese medicine medium dose group and the traditional Chinese medicine high dose group can obviously reduce muscle loss, and the muscle fiber level of the muscle is improved.
5. As shown in FIG. 27, the results of measuring the levels of HMGB-1, IL-1. beta., IL-6, and TNF-. alpha.in each experimental group: compared with a blank control group, a stress model group, a traditional Chinese medicine low-dose group, a traditional Chinese medicine medium-dose group and a traditional Chinese medicine high-dose group, the levels of HMGB-1, IL-1 beta, IL-6 and TNF-alpha in muscle tissues are remarkably increased. Compared with the stress model group, the traditional Chinese medicine low-dose group, the traditional Chinese medicine medium-dose group and the traditional Chinese medicine high-dose group can obviously reduce the levels of HMGB-1, IL-1 beta, IL-6 and TNF-alpha, and the low, medium and high-dose groups have obvious effects; the more significant the effect with increasing dose, the dose-related trend.
Analyzing the map and detecting the conclusion: the traditional Chinese medicine low-dose group, the traditional Chinese medicine medium-dose group and the traditional Chinese medicine high-dose group have obvious effect of resisting inflammatory reaction of muscle tissues, can obviously increase the autonomous movement of model animals, reduce tissue pain, inhibit the apoptosis of mitochondria in the muscle tissues of the model animals, inhibit the increase of inflammatory reaction related factors in the muscle tissues, and have more obvious effect of inhibiting the inflammatory reaction along with the increase of the dose and show the dose-related trend.

The traditional Chinese medicine formula for treating the muscle reduction atrophy caused by the psychosomatic stress according to claim 1, is characterized by comprising the following raw materials in parts by weight: 5-30 parts of turmeric, 5-30 parts of ligusticum wallichii, 5-30 parts of acorus calamus, 5-30 parts of liquorice and 100-400 parts of cocoa butter.

The traditional Chinese medicine formula for treating the muscle reduction atrophy caused by the psychosomatic stress according to claim 1, is characterized by comprising the following raw materials in parts by weight: 10-20 parts of turmeric, 10-20 parts of ligusticum wallichii, 10-20 parts of acorus calamus, 10-20 parts of liquorice and 1-100 parts of cocoa butter.
