Abstract
The invention discloses a moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate and a preparation method thereof, aiming at providing an essence which is easy to absorb, can improve the water shortage, darkness and dryness of skin, does not block pores, does not cause secondary damage to skin, and has the moisturizing skin-brightening repair effect; the technical scheme comprises the following components in percentage by weight: 1.0-20.0% of yeast/rice fermentation product filtrate, 0.01-0.3% of poly gamma-sodium glutamate, 0.02-0.1% of tremella polysaccharide, 0.1-1.0% of lactobacillus plantarum fermented rice, 0.05-0.3% of Ceracarte T450, 0.1-1.0% of skin brightening agent, 0.1-3.0% of composite fullerene stock solution, 0.3-3.0% of ionic humectant, 1, 3-propylene glycol 1.0-5.0%, 0.05-0.5% of carrageen crispus extract, 0.1-0.5% of diisostearyl malate, 0.05-0.3% of oligomeric sodium hyaluronate, 1.0-5.0% of yeast fermentation product filtrate mixture, 0.05-0.3% of beta-glucan, 0.2-1.0% of phytosphingosine, and 100% of deionized water; belongs to the technical field of cosmetics.

Technical Field
The invention relates to essence, in particular to moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate and a preparation method thereof, and belongs to the technical field of cosmetics.
Background
The existing formula technology is mainly based on chemical basic raw materials, such as carbomer TEA salts, hydroxyethyl cellulose, diatomite and other thickeners, the green degree is low, the ingredients are strong in residue, the effective ingredients are mainly based on chemical skin placebo and cannot be effective for a long time, and the commonly used ingredients such as phenethyl resorcinol, kojic acid, and methoxy potassium salicylate have the defects of large residue and large irritation, can cause an allergic phenomenon, are short in effect and need to be maintained for a long time.

The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate comprises the following components in percentage by weight: 1.0-20.0% of yeast/rice fermentation product filtrate, 0.01-0.3% of poly gamma-sodium glutamate, 0.02-0.1% of tremella polysaccharide, 0.1-1.0% of lactobacillus plantarum fermented rice, 0.05-0.3% of Ceracarte T450, 0.1-1.0% of skin brightening agent, 0.1-3.0% of composite fullerene stock solution, 0.3-3.0% of ionic humectant, 1, 3-propylene glycol 1.0-5.0%, 0.05-0.5% of carrageen crispus extract, 0.1-0.5% of diisostearyl malate, 0.05-0.3% of oligomeric sodium hyaluronate, 1.0-5.0% of yeast fermentation product filtrate mixture, 0.05-0.3% of beta-glucan, 0.2-1.0% of phytosphingosine and 100% of deionized water.
Further, the moisturizing skin-brightening repair essence containing the yeast rice fermentation product filtrate comprises the following components in parts by weight: 1.0-5.0 parts of birch bark extract, 1.0-5.0 parts of ampelopsis japonica root extract, 1.0-5.0 parts of bighead atractylodes rhizome root extract, 1.0-5.0 parts of dictamnus dasycarpus root extract, 1.0-5.0 parts of bletilla striata root extract, 1.0-5.0 parts of oldenlandia diffusa root extract and 3.0-10.0 parts of water lily root extract.
Further, the moisturizing skin-brightening repair essence containing the yeast rice fermentation product filtrate also comprises 1.0-5.0 parts of alkanna tinctoria root extract and 1.0-5.0 parts of lily bulbus extract.
Further, the moisturizing skin-brightening repair essence containing the yeast rice fermentation product filtrate comprises the following components and contents thereof, wherein the total weight of the composite fullerene raw liquid is calculated according to 100 percent: 60.0-75.0% of dipropylene glycol, 15.0-22.0% of nicotinamide, 8.0-12.0% of tranexamic acid, 600.1-1.0% of fullerene C and 0.1-1.0% of polyvinylpyrrolidone.
Further, the moisturizing and skin-brightening repair essence containing the yeast rice fermentation product filtrate comprises the following components and contents thereof, wherein the total weight of the ionic moisturizing agent is calculated according to 100 percent: 0.1-3.0% of sodium lactate, 0.1-3.0% of lactic acid, 0.1-3.0% of lysine, 0.1-2.5% of histidine, 0.1-3.0% of arginine, 0.1-3.0% of aspartic acid, 0.05-0.5% of threonine, 0.1-1.0% of serine, 0.1-3.0% of glutamic acid, 0.1-3.0% of proline, 0.1-1.0% of glycine, 0.1-1.0% of alanine, 0.1-2.0% of valine, 0.1-1.0% of isoleucine, 0.1-1.0% of leucine, 0.1-1.0% of tyrosine, 0.1-1.0% of phenylalanine, 8.0-15.0% of 1, 3-propanediol, and 100% of deionized water.
Further, the moisturizing skin-brightening repair essence containing the yeast rice fermentation product filtrate comprises the following components and contents thereof, wherein the total mass of the yeast fermentation product filtrate mixture is 100 percent: 20-30.0% of lactobacillus/roselle flower fermentation product filtrate, 20.0-30% of lactobacillus/mung bean seed extract/sodium glutamate fermentation product filtrate, 10.0-20% of yeast/rice seed fermentation product filtrate, 8-12% of lactobacillus/soybean fermentation product extract, 15-25% of rice fermentation product filtrate and 4.0-6.0% of ergothioneine.
Further, the moisturizing skin-brightening repair essence containing the yeast rice fermentation product filtrate is prepared by the following steps of:
1) activation and multiplication of bacterial strains
Inoculating the saccharomyces cerevisiae to a sterilized skim milk culture medium, culturing for 48-72 h at 30 ℃, inoculating the saccharomyces cerevisiae to a fresh skim milk culture medium for culturing after peptonization of the skim milk culture medium, inoculating the saccharomyces cerevisiae to an MRS liquid culture medium after the growth of thalli is stable, and culturing for 48h at 30 ℃; taking out the culture medium, selecting a saccharomyces cerevisiae single colony by using an inoculating loop under an aseptic condition, inoculating the saccharomyces cerevisiae single colony into an MRS liquid culture medium, and culturing for 24-48 h at 30 ℃; inoculating the activated and proliferated saccharomyces cerevisiae into an MRS liquid culture medium, and culturing for 24-48 h at 30 ℃ until the concentration of the saccharomyces cerevisiae reaches 1 multiplied by 105cfu/mL is used as bacterial suspension;
2) weighing 200g of rice, and soaking in 400ml of deionized water for 1-3 h; after cleaning, deionized water and rice are mixed according to a mass ratio of 1: 2, cooking for 20-40min to sticky rice pulp, stopping cooking, placing into a glass wide-mouth bottle, and naturally cooling to 28-32 ℃;
3) adding 4-6 wt% of bacterial suspension into the step 2), sealing, culturing and fermenting for 60-80h at 38 +/-1 ℃ and controlling the pH value to be 6.8-7.2;
4) standing the fermentation liquid, filtering and collecting clear liquid, namely the yeast/rice fermentation product filtrate.

1) activation and multiplication of bacterial strains
Inoculating the saccharomyces cerevisiae into the sterilized skim milk culture medium, culturing for 48-72 h at 30 ℃, and then conducting peptonization on the skim milk culture mediumInoculating saccharomyces cerevisiae 336058 into a fresh skim milk culture medium for culture, inoculating the saccharomyces cerevisiae into an MRS liquid culture medium after the thalli grow stably, and culturing for 48h at 30 ℃; taking out the culture medium, selecting a saccharomyces cerevisiae single colony by using an inoculating loop under an aseptic condition, inoculating the saccharomyces cerevisiae single colony into an MRS liquid culture medium, and culturing for 24-48 h at 30 ℃; inoculating the activated and proliferated saccharomyces cerevisiae into an MRS liquid culture medium, and culturing for 24-48 h at 30 ℃ until the concentration of the saccharomyces cerevisiae reaches 1 multiplied by 105cfu/mL is used as bacterial suspension;
4) weighing 200g of rice, and soaking in 400ml of deionized water for 1-3 h; after cleaning, deionized water and rice are mixed according to a mass ratio of 1: 2, cooking for 20-40min to sticky rice pulp, stopping cooking, placing into a glass wide-mouth bottle, and naturally cooling to 28-32 ℃;
5) adding 4-6 wt% of bacterial suspension into the step 2), sealing, culturing and fermenting for 60-80h at 38 +/-1 ℃ and controlling the pH value to be 6.8-7.2;
4) standing the fermentation liquid, filtering and collecting clear liquid, namely the yeast/rice fermentation product filtrate.

5) respectively weighing the raw materials in the group C, adding the raw materials into the mixture obtained in the step 4), stirring the mixture at a rotating speed of 25r/min until the mixture is semitransparent liquid, controlling the pH value to be 5.5-6.5, and filtering and discharging the filtered mixture after the inspection is qualified.

The components of the repair essence are divided into A, B groups, and the materials are fed according to the following items:
group A: 1, 3-propylene glycol, Chondrus crispus extract, oligomeric sodium hyaluronate and deionized water.

The specific implementation production process is as follows:
weighing the raw materials of the group A, sequentially adding the raw materials into a beaker container, heating the raw materials to 83-85 ℃ at a speed of 25r/min by a stirrer to dissolve the raw materials for transparency, keeping the temperature to 83-85 ℃, cooling the raw materials after 20 minutes, cooling the raw materials to 45 ℃, adding the raw materials of the group B into the group A, stirring the raw materials for 20 minutes at a speed of 25r/min to obtain transparent liquid, controlling the ph to be 5.5-6.5, filtering the transparent liquid after the inspection is qualified, and discharging the filtered liquid.
Efficacy tests of the moisturizing and skin-lightening repair essence provided in comparative example 1 and example 1 were compared as follows:

Skin moisture content tester (Corneometer CM825)
The skin moisture content tester Corneometer (CM 825, Courage and Khazaka, Germany) measures on the basis of the principle of capacitance. Water has a dielectric constant of 81, other substances generally have a dielectric constant of less than 7, and water is the substance with the highest dielectric constant on the skin. When the moisture content changes, the capacitance of the skin also changes, so the moisture content on the skin surface can be analyzed by measuring the capacitance of the skin. The greater the skin moisture content value, the higher the skin moisture content.
Skin mirror system (medical 800hD)
The dermoscopy system (medical 800hD, FotoFinder systems Gmbh, Birbach, Germany) is a non-invasive examination device that amplifies skin lesions and allows the superficial layers of the epidermis and dermis to be visualized through the stratum corneum layer of the epidermis, with microscopic structures such as pigments and vascular structures that are not visible to the naked eye. In the test, a skin photo is collected at a magnification of 50 times, and the fine lines and dandruff condition of the skin are observed. The dry skin is often accompanied by raised tiny scaly dandruff in the grain gaps, the fine lines of the skin are more obvious due to the dandruff among the grains, and the skin looks dry and rough. Excessive dryness of the skin causes more severe dandruff, white flakes are observed in the image, and raised dandruff causes the skin to feel stroked and feel large in frictional resistance and poor in moisturized feel.

the results of the skin moisture content test before the sample was used and 1h, 4h, 8h and 24h after the sample was used are shown in table 1 and fig. 1 and 2. The greater the skin moisture content value, the greater the skin moisture content.

As can be seen from Table 1 and FIGS. 1 and 2, example 1 showed a significant increase in skin moisture content at each time point after use compared to before use (1h, 4h and 8 h: P < 0.05).
2. Skin image acquisition (medical 800hD)
Using example 1 and comparative example 1 on the flexed side of the forearm, before use of the sample, 5min and 2h after use of the sample, a 50-fold magnification image of the skin in the test area was collected using a skin mirror system, and the effects of example 1 on instantly smoothing fine lines and dandruff and lasting nourishing and moistening dry skin were visually evaluated. The test results are shown in FIG. 3. After 15 min using the blank control, i.e. comparative example, the lines on the skin become lighter and the scales disappear, and after 2h using, the fine lines on the skin are more obvious than before using, and the scales are more serious. After the samples of the embodiment 1 are used for 5min and 2h, the skin lines become light and the dandruff is weakened, and the states of the skin fine lines and the dandruff after 2h are not obviously poor compared with the states after 5min of use.
3. Sensory evaluation of skin
The skin sensory evaluation was performed by the panelists on the skin after application of the samples, with a score of 0-5, and the test results were averaged and shown in table 2, with the higher the score, the more obvious the sensation.

And (4) conclusion: the examples show that the sensory effect experienced in each index is higher than that experienced in comparative example 1, indicating that the moisture retention of example 1 is stronger.
4. Skin lightening efficacy evaluation:
skin lightening efficacy within 4 weeks of example 1 and comparative example 1 was picked out of 16 dark skin volunteers in moisturizing program. Testing facial skin, controlling the temperature at 20-22 ℃, and controlling the relative humidity: 40-60%, test cycle 4 weeks (visit weeks 0, 2, 4).
Test protocol
Selecting 16 suitable volunteers to participate in the test (the selection standard is 25-35 years old, the skin color is dark, the skin color is classified into middle skin color people (namely the ITA degree value is between 28-41). The volunteers respectively use the repair essence of the embodiment 1 on 8 full faces, use the repair essence of the comparative example 1 on 8 full faces, measure the change of skin state indexes before use, after use for 2 weeks and after use for 4 weeks through trial for 4 weeks continuously, and evaluate the skin brightening effect of the repair essence through comparison of the face condition of the volunteers before use and after use.
The data are respectively expressed as: 0W, 2W and 4W.
Evaluation method
The 1.5.2.2 dermoscope system (medical 800hD) for the test instrument provides safe, stable standard lighting conditions, and all camera settings and imaging parameter settings are controlled by software to ensure stability and standardization of imaging. The instrument has multiple light source modes: standard light, blue light, orange light, cross polarized light and parallel polarized light can be used for carrying out multi-angle imaging on facial skin, and the imaging quality can reach up to 2000 ten thousand pixels. The face of the subject is shot in the test, and the following evaluation parameters are analyzed by selecting a standard 2 light source picture:
skin brightness L: degree of skin lightening. Skin brightness L of selected areas (smeared in any area);
skin yellowness b: degree of skin yellowing. Skin yellowness b of selected areas (applied to either area);
skin whiteness ITA °: degree of skin whitening. L, b from the above analysis are calculated by the following formula:

the results of the skin brightness L, skin yellowness b, skin whiteness ITA ° analysis and statistics for any area painted before use (initial value), after 2 weeks, 4 weeks of use (medical 800hD) are shown in tables 3 and 4 below.
Skin brightness L: the greater the skin brightness L, the brighter the skin;
skin yellowness b; the greater the skin yellowness b, the darker the skin;
skin whiteness ITA °: the greater the skin whiteness ITA °, the whiter the skin.

And (4) conclusion: analysis of the data from table 3, example 1, shows that the facial skin tone enhancement effect is more pronounced with 4 weeks of continuous use.

And (4) conclusion: analysis of the data from table 3, example 1, shows that the facial skin tone enhancement effect is more pronounced with 4 weeks of continuous use.
Table 5 shows the image analysis-color analysis and statistics (n-8) before (initial value), after 2 weeks and 4 weeks of use in comparative example 1.

And (4) conclusion: the facial skin tone enhancement effect was not apparent from the data analysis of comparative example 1 of table 5 for 4 weeks of continuous use.
5. And (5) testing the repairing efficacy.
30 female skin sensitive muscle volunteers aged 35-45 years are collected in a head office to evaluate the repair effect of any one of the groups of example 1, example 5 and comparative example 1 within 4 weeks; the main manifestations of the sensitive skin include bad feelings of itch, red swelling, tightness, stabbing pain, etc., three groups of people test for improving the sensitive skin problem, 10 test examples 1, 10 test examples 5, 10 test comparative examples 1, requiring wiping once every day in the morning and evening at home, continuously testing for 4 weeks, filling in sample trial questionnaires, and feeding back the using feeling and effect of the product.
After 4 weeks of use, volunteers' use feedback was collected on-line and analyzed for best relief, improvement and results as shown in table 3 and fig. 1. The larger the score is, the better the soothing and improving effects on the corresponding dimension of the sensitive skin are.

Conclusion
The human trial results of 4 weeks reflect that the examples 1 and 5 can relieve and repair the sensitive state of the skin in five dimensions of itching relieving, swelling reducing, tingling, redness removing and tightening, and the relieving and repairing effects in five dimensions are superior to those of the comparative example 1.
While the preferred embodiments of the present invention have been illustrated and described in detail, it should be understood that modifications and variations can be made by persons skilled in the art in light of the above teachings without inventive faculty. Therefore, any technical solutions that can be obtained by a person skilled in the art through logical analysis, reasoning or limited experiments based on the prior art according to the present inventive concept should be within the scope of protection defined by the present claims.
Claims (10)
1. The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate is characterized by comprising the following components in percentage by weight: 1.0-20.0% of yeast/rice fermentation product filtrate, 0.01-0.3% of poly gamma-sodium glutamate, 0.02-0.1% of tremella polysaccharide, 0.1-1.0% of lactobacillus plantarum fermented rice, 0.05-0.3% of Ceracarte T450, 0.1-1.0% of skin brightening agent, 0.1-3.0% of composite fullerene stock solution, 0.3-3.0% of ionic humectant, 1, 3-propylene glycol 1.0-5.0%, 0.05-0.5% of carrageen crispus extract, 0.1-0.5% of diisostearyl malate, 0.05-0.3% of oligomeric sodium hyaluronate, 1.0-5.0% of yeast fermentation product filtrate mixture, 0.05-0.3% of beta-glucan, 0.2-1.0% of phytosphingosine and 100% of deionized water.
2. The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate according to claim 1, wherein the skin-brightening agent comprises the following components in parts by weight: 1.0-5.0 parts of birch bark extract, 1.0-5.0 parts of ampelopsis japonica root extract, 1.0-5.0 parts of bighead atractylodes rhizome root extract, 1.0-5.0 parts of dictamnus dasycarpus root extract, 1.0-5.0 parts of bletilla striata root extract, 1.0-5.0 parts of oldenlandia diffusa root extract and 3.0-10.0 parts of water lily root extract.
3. The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate as claimed in claim 2, wherein the skin-brightening agent further comprises 1.0-5.0 parts of alkanna tinctoria root extract and 1.0-5.0 parts of lily bulbus extract.
4. The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate according to claim 1, wherein the composite fullerene stoste comprises the following components and contents thereof, calculated by 100% in total: 60.0-75.0% of dipropylene glycol, 15.0-22.0% of nicotinamide, 8.0-12.0% of tranexamic acid, 600.1-1.0% of fullerene C and 0.1-1.0% of polyvinylpyrrolidone.
5. The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate as claimed in claim 1, wherein the ionic moisturizing agent comprises the following components and contents thereof by 100% of the total weight: 0.1-3.0% of sodium lactate, 0.1-3.0% of lactic acid, 0.1-3.0% of lysine, 0.1-2.5% of histidine, 0.1-3.0% of arginine, 0.1-3.0% of aspartic acid, 0.05-0.5% of threonine, 0.1-1.0% of serine, 0.1-3.0% of glutamic acid, 0.1-3.0% of proline, 0.1-1.0% of glycine, 0.1-1.0% of alanine, 0.1-2.0% of valine, 0.1-1.0% of isoleucine, 0.1-1.0% of leucine, 0.1-1.0% of tyrosine, 0.1-1.0% of phenylalanine, 8.0-15.0% of 1, 3-propanediol, and 100% of deionized water.
6. The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate according to claim 1, wherein the yeast fermentation product filtrate mixture comprises the following components and contents thereof in a total mass of 100%: 20-30.0% of lactobacillus/roselle flower fermentation product filtrate, 20.0-30% of lactobacillus/mung bean seed extract/sodium glutamate fermentation product filtrate, 10.0-20% of yeast/rice seed fermentation product filtrate, 8-12% of lactobacillus/soybean fermentation product extract, 15-25% of rice fermentation product filtrate and 4.0-6.0% of ergothioneine.
7. The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate as claimed in claim 1, wherein the yeast/rice fermentation product filtrate is sequentially prepared by the following steps:
1) activation and multiplication of bacterial strains
Inoculating the saccharomyces cerevisiae to a sterilized skim milk culture medium, culturing for 48-72 h at 30 ℃, inoculating the saccharomyces cerevisiae to a fresh skim milk culture medium for culturing after peptonization of the skim milk culture medium, inoculating the saccharomyces cerevisiae to an MRS liquid culture medium after the growth of thalli is stable, and culturing for 48h at 30 ℃; taking out the culture medium, selecting a saccharomyces cerevisiae single colony by using an inoculating loop under an aseptic condition, inoculating the saccharomyces cerevisiae single colony into an MRS liquid culture medium, and culturing for 24-48 h at 30 ℃; inoculating the activated and proliferated saccharomyces cerevisiae into an MRS liquid culture medium, and culturing for 24-48 h at 30 ℃ until the concentration of the saccharomyces cerevisiae reaches 1 multiplied by 105cfu/mL is used as bacterial suspension;
2) weighing 200g of rice, and soaking in 400ml of deionized water for 1-3 h; after cleaning, deionized water and rice are mixed according to a mass ratio of 1: 2, cooking for 20-40min to sticky rice pulp, stopping cooking, placing into a glass wide-mouth bottle, and naturally cooling to 28-32 ℃;
3) adding 4-6 wt% of bacterial suspension into the step 2), sealing, culturing and fermenting for 60-80h at 38 +/-1 ℃ and controlling the pH value to be 6.8-7.2;
4) standing the fermentation liquid, filtering and collecting clear liquid, namely the yeast/rice fermentation product filtrate.
8. The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate as claimed in claim 7, wherein the skim milk medium is prepared by dissolving skim milk powder in distilled water to obtain a mixture with a concentration of 120g/L, packaging, and steam sterilizing at 115 ℃ for 30 min.

The moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate according to claim 7, wherein the MRS agar culture medium has a pH of 6.2 +/-0.2, and comprises the following components in percentage by weight: 5.0g of yeast extract, 3.0g of malt extract, 10.0g of glucose, 5.0g of peptone, 20.0g of agar and 1000ml of distilled water.
10. The method for preparing the moisturizing skin-brightening repair essence containing yeast rice fermentation product filtrate according to claim 1, is characterized by sequentially comprising the following steps:
1) weighing the components according to the weight percentage of claim 1, and dividing the components into a group A, a group B and a group C, wherein:
group A: lactobacillus plantarum fermented rice, diisostearyl malate, phytosphingosine, CeracareT 45;
group B: deionized water, poly gamma-sodium glutamate, tremella polysaccharide, oligomeric sodium hyaluronate, Chondrus crispus extract and 1, 3-propylene glycol;
group C: yeast/rice fermentation product filtrate, composite fullerene stock solution, an ionic humectant and beta-glucan;
2) adding the component A into a beaker, uniformly mixing, and heating to 78-83 ℃ while stirring;
3) adding the component B into another beaker, and heating to 83-85 ℃ while stirring to dissolve the component B into a transparent state;
4) slowly adding the dissolved group A into the group B, stirring and homogenizing for 5 minutes until a semitransparent emulsion is formed; heating to 83-85 ℃, keeping the temperature for 20 minutes, and then cooling to below 45 ℃;
5) respectively weighing the group C raw materials, adding the group C raw materials into the step 4), stirring the mixture until the mixture is semitransparent liquid, and controlling the pH value to be 5.5-6.5; filtering and discharging after the inspection is qualified.
