Abstract
本发明公开了一种提高断奶仔猪免疫力的饲料添加剂及其制备方法,该饲料添加剂由组份A和组份B通过固态发酵工艺制得,发酵时,所述组份A和所述组份B的质量比为1:15~1:30;其中,所述组份A为包括至少两种益生菌种子液的混合菌种,且每种益生菌菌落数大于108CFU/ml;所述组份B至少包括党参和黄芪中的一种或两种混合。本发明提供的一种提高断奶仔猪免疫力的饲料添加剂,是经益生菌发酵的中草药添加剂,能促进免疫活性物质多糖的释放及小肽的合成,多糖发酵前后含量分别是2.35%和2.98%,小肽含量从4.97%提高到6.42%,有效提高机体免疫力和抗病能力,是饲料完全禁抗后理想的抗生素替代产品。
Description
技术领域
本发明涉及一种饲料添加剂,特别是一种提高断奶仔猪免疫力的饲料添 加剂。本发明还涉及该饲料添加剂的制备方法。
背景技术
因环境、应激和日粮形态的改变,仔猪断奶时极易引起消化不良,造成 仔猪腹泻、生长速度缓慢,甚至发生死亡。据有关资料报道,在我国中小规 模的猪场,仔猪断奶时腹泻率高达50%~60%,死亡率高达15%~20%,断 奶仔猪腹泻给养猪生产造成了巨大的经济损失。
过去,人们普遍做法是在饲料中添加抗生素或化学合成药物来预防或减 轻断奶仔猪腹泻,但这些药物会使病原微生物产生抗药性,并导致畜产品中 抗生素残留,给食品安全带来巨大隐患。农业农村部发布第194号公告,自 2020年1月1日起,退出除中药外所有促生长类饲料添加剂品种,自2020 年7月1日起饲料完全“禁抗”,寻找新型、安全的抗生素替代产品已成为近 年来世界各国动物营养学研究的热点和趋势。
例如,在目前一些应用中,仔猪生病后已可采用中药散剂和注射剂来替 代抗生素进行治疗。但是中药散剂因中药有效成分在细胞浆中,不易透过细 胞壁而发挥作用,而注射剂也容易造成个体强烈应激反应。另外,中药有效 成分多为大分子物质,不易透过血脑屏障被吸收;并且传统炮制方法煎、煮、 炼、浸等方法难以适应养殖业规模化生产需要,这意味着中草药在规模化养 殖业中的应用仍然存在障碍。
也有研究是通过在饲料中添加经发酵的中药组合物构成的添加剂来预防 仔猪腹泻的方法,具体地,例如在中国专利申请CN106472840A中公开了一 种预防巴克夏与民猪杂交断奶仔猪腹泻的发酵液及其制备方法,所述发酵液 由党参、黄芪、桔梗、白术、麦芽、山楂、蒲公英、白头翁、水和乳酸菌制 备而成,其中,以重量份数计,含有党参200~300份、黄芪100~200份、 桔梗100~300份、白术100~300份、麦芽200~300份、山楂100~200份、 蒲公英200~300份、白头翁200~300份,乳酸菌的含量为104~106个/g, 乳酸菌8~10份,水的加入量为党参、黄芪、桔梗、白术、麦芽、山楂、蒲 公英和白头翁总重量的8~10倍。但是,乳酸菌(lactic acid bacteria,LAB) 是一类能利用可发酵碳水化合物产生大量乳酸的细菌的统称。这类细菌在自 然界分布极为广泛,具有丰富的物种多样性,至少包含18个属,共200多种。 这其中大多数的乳酸菌,活力极弱,它们只能在相对受限制的环境中存活, 一但脱离这些环境,其自身就会遭到灭亡。而且对于乳酸菌类的微生物饲料 添加剂来说,菌落数必须具备数量多、活力强才能发挥生物功效。因此,在 实际应用中,往往仍需结合抗生素进行疾病预防。
发明内容
鉴于现有技术存在的上述问题,本发明的目的在于提供一种提高断奶仔 猪免疫力的饲料添加剂,该饲料添加剂能有效提高仔猪机体免疫力和抗病能 力,可作为饲料完全禁抗后理想的抗生素替代产品。
为了实现上述目的,本发明一方面提供的一种提高断奶仔猪免疫力的饲 料添加剂,由组份A和组份B通过固态发酵工艺制得,发酵时,所述组份A 和所述组份B的质量比为1:15~1:30;其中,所述组份A为包括至少两种益 生菌种子液的混合菌种,且每种益生菌菌落数大于108CFU/ml;所述组份B 至少包括党参和黄芪中的一种或两种混合。
作为优选,所述组份B为党参和黄芪的混合料,且以质量百分比计,党 参和黄芪之比为2:1~1:2。
作为优选,所述组份A的混合菌种由选自嗜酸乳杆菌、干酪乳杆菌、詹 氏乳杆菌、拉曼乳杆菌、长双歧杆菌、短双歧杆菌、卵形双歧杆菌、嗜热双 歧杆菌、粪链球菌、屎肠球菌、乳球菌、中介链球菌及地衣芽孢杆菌中的两 种或多种益生菌种子液混合制成。
作为优选,所述组份A的混合菌种由屎肠球菌和地衣芽孢杆菌的种子液 按照2:1~1:2的比例混合制成,其中每个菌种的菌落数大于108CFU/ml。
作为优选,所述组份A中屎肠球菌种子液和地衣芽孢杆菌种子液的混合 比例为1:1,且所述组份B中党参和黄芪的混合比例为1:1。
本发明的另一方面,还提供了一种上述饲料添加剂的制备方法,具体地, 该方法包括:
1)组份B制备:按比例选取党参和黄芪中的一种或两种混合,粉碎成 粗粉,过24目筛,备用;
2)固态发酵:将已活化的至少两种益生菌种子液的混合,制成组份A 的混合菌种,按组份A和组份B的质量比为1:15~1:30,将组份A加入组份 B中,充分均匀混合,设定发酵温度为30℃~37℃±1℃,相对湿度40~60%, 固态发酵150~240小时,制得混合型无抗中草药发酵饲料添加剂,发酵呼吸 袋装袋,阴凉保存。
作为优选,在将组份A加入组份B中,充分均匀混合后,加入无菌水适 量,使含水量在保持在30~40%之间。
作为优选,作为优选,所述组份B为党参和黄芪的混合料,且以质量百 分比计,党参和黄芪之比为2:1~1:2。
作为优选,所述组份A的混合菌种由选自嗜酸乳杆菌、干酪乳杆菌、詹 氏乳杆菌、拉曼乳杆菌、长双歧杆菌、短双歧杆菌、卵形双歧杆菌、嗜热双 歧杆菌、粪链球菌、屎肠球菌、乳球菌、中介链球菌及地衣芽孢杆菌中的两 种或多种益生菌种子液混合制成;且所述组份A的混合菌种由屎肠球菌和地 衣芽孢杆菌的种子液按照2:1~1:2的比例混合制成,其中每个菌种的菌落数 大于108CFU/ml。
作为优选,所述组份A中屎肠球菌种子液和地衣芽孢杆菌种子液的混合 比例为1:1,且所述组份B中党参和黄芪的混合比例为1:1。
与现有技术比较,本发明提供的一种提高断奶仔猪免疫力的饲料添加剂, 是经益生菌发酵的中草药添加剂,能促进免疫活性物质多糖的释放及小肽的 合成,多糖发酵前后含量分别是2.35%和2.98%,小肽含量从4.97%提高到 6.42%,有效提高机体免疫力和抗病能力,是饲料完全禁抗后理想的抗生素 替代产品。临床试验表明,仔猪日粮中添加1.0%,平均日增重提高了16.58% (P<0.05),耗料增重比降低了8.77%(P<0.05),因益生菌活菌数高,有效 改善断奶仔猪肠道菌群,解决断奶仔猪因环境、应激和日粮形态的改变引起 肠道菌群紊乱导致生长缓慢、腹泻的发生,提高生产性能。

具体实施方式
为了使得本公开实施例的目的、技术方案和优点更加清楚,下面将结合 本公开实施例,对本公开实施例的技术方案进行清楚、完整地描述。显然, 所描述的实施例是本公开的一部分实施例,而不是全部的实施例。基于所描 述的本公开的实施例,本领域普通技术人员在无需创造性劳动的前提下所获 得的所有其他实施例,都属于本公开保护的范围。
除非另外定义,本公开使用的技术术语或者科学术语应当为本公开所属 领域内具有一般技能的人士所理解的通常意义。
实施例1
以党参、黄芪为中药添加剂制备组份B。
以屎肠球菌、地衣芽孢杆菌为益生菌发酵菌种组份A。
在该实施例中,饲料添加剂益生菌发酵菌种比例为屎肠球菌:地衣芽孢 杆菌=1:1,补中益气中药基质质量比为党参:黄芪=1:1,按中药基质质量的5% 加入菌种,进行固态发酵,制备而成提高断奶仔猪免疫力的混合型无抗中草 药发酵饲料添加剂。
制备方法,步骤包括:
1)中药基质制备(组份B):优选道地中药材党参、黄芪,按照党参: 黄芪=1:1比例配比后,粉碎成粗粉,过24目筛,反复搅拌,混合均匀,备 用。
2)固态发酵:将已活化的屎肠球菌和地衣芽孢杆菌种子液(组份A)(每 ml菌落数不少于1~5*108CFU)按菌种比例为=1:1混合,按组份A与组份B 质量比1:20的比例,在中药基质中加入混合菌种,再加入无菌水适量,使含 水量在35%左右,充分均匀混合,设定温度为37℃+1℃,相对湿度50%,固 态发酵192小时,制得混合型无抗中草药发酵饲料添加剂,发酵呼吸袋装袋, 阴凉保存。
具体地,上述制备过程中,益生菌发酵菌种屎肠球菌、地衣芽孢杆菌用 量为每克中药接种菌落数不少于5*108CFU。
临床效果试验
1、试验设计:将仔猪按体重相近的原则随机分为两组,每组3个重复, 每个重复15头猪。对照组饲喂无中药添加的饲粮,试验组饲喂添加0.1%的 发酵中草药发酵饲料添加剂饲粮。仔猪断奶后,原圈饲喂7d作为预试期,然 后转入保育舍,正试期28d,共计35d。试验猪以圈为单位饲养,饲喂试验饲 粮,每日饲喂4次,分别在7:00、11:00、15:00和19:00进行饲喂,自由饮 水。试验过程中按照猪场的日常操作程序进行免疫、消毒和管理。
试验饲粮参照NRC(2012)猪饲养标准进行配制,饲粮配方及营养水平见 表1。

注:①预混料为每千克日粮提供:维生素A 12 000IU,维生素D3 2 000IU,维生素E60IU,维 生素K3 3.0mg,生物素0.3mg,叶酸1.5mg,尼克酸30.0mg,泛酸20.0mg,维生素B12.0mg,维 生素B2 6.0mg,维生素B6 2.0mg,维生素B12 0.03mg,氯化胆碱1 000.0mg,抗氧化剂200.0mg; 铁120.0mg,锌100.0mg,锰35.0mg,铜120.0mg,硒0.30mg,碘0.5mg。②消化能根据原料 组成计算所得,其余为实测值。
2、测定指标与方法
2.1生产性能:分别于试验的第1d、第29d早上6:00对仔猪进行空腹 称重,试验期间每天记录饲料饲喂量,根据初重和末重计算平均日增重,并 根据饲料饲喂量计算耗料增重比。
2.2免疫指标的测定:试验结束后(即第29d)早晨6:00,分别从每个 处理中选择5头空腹仔猪(共25头),经前腔静脉采血5ml,在4℃冰箱 中静置6h,然后3000r/min离心15min,制备血清,-20℃保存,待作血 清免疫指标测定。
3、结果
3.1中草药发酵饲料添加剂对断奶仔猪生长性能的影响

由表2可知,与对照组相比,试验B组仔猪头均末重提高了7.84%(P <0.05),平均日增重提高了16.58%(P<0.05),耗料增重比降低了8.77%(P <0.05)。
3.2中草药发酵饲料添加剂对断奶仔猪免疫指标的影响

由表3可知,与对照组相比,试验B组血清中IgA含量提高了47.06% (P<0.05)(P<0.05);血清中IgG有增高的趋势,但两组之间差异不显著 (P>0.05)。
结论:本试验研究结果表明,在断奶仔猪日粮中添加0.1%中草药发酵饲 料添加剂,能提高断奶仔猪的生长性能,对仔猪的免疫功能具有促进和改善 作用。
当然,以上所述是本发明的优选实施方式,应当指出,对于本技术领域 的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进 和润饰,这些改进和润饰也视为本发明的保护范围。
Claims (10)
1.一种提高断奶仔猪免疫力的饲料添加剂,由组份A和组份B通过固态发酵工艺制得,发酵时,所述组份A和所述组份B的质量比为1:15~1:30;其中,所述组份A为包括至少两种益生菌种子液的混合菌种,且每种益生菌菌落数大于108CFU/ml;所述组份B至少包括党参和黄芪中的一种或两种混合。
2.如权利要求1所述的饲料添加剂,所述组份B为党参和黄芪的混合料,且以质量百分比计,党参和黄芪之比为2:1~1:2。
3.如权利要求1所述的饲料添加剂,所述组份A的混合菌种由选自嗜酸乳杆菌、干酪乳杆菌、詹氏乳杆菌、拉曼乳杆菌、长双歧杆菌、短双歧杆菌、卵形双歧杆菌、嗜热双歧杆菌、粪链球菌、屎肠球菌、乳球菌、中介链球菌及地衣芽孢杆菌中的两种或多种益生菌种子液混合制成。
4.如权利要求3所述的饲料添加剂,所述组份A的混合菌种由屎肠球菌和地衣芽孢杆菌的种子液按照2:1~1:2的比例混合制成,其中每个菌种的菌落数大于108CFU/ml。
5.如权利要求4所述的饲料添加剂,所述组份A中屎肠球菌种子液和地衣芽孢杆菌种子液的混合比例为1:1,且所述组份B中党参和黄芪的混合比例为1:1。
6.一种提高断奶仔猪免疫力的饲料添加剂的制备方法,该方法包括:
1)组份B制备:按比例选取党参和黄芪中的一种或两种混合,粉碎成粗粉,过24目筛,备用;
2)固态发酵:将已活化的至少两种益生菌种子液的混合,制成组份A的混合菌种,按组份A和组份B的质量比为1:15~1:30,将组份A加入组份B中,充分均匀混合,设定发酵温度为30℃~37℃+1℃,相对湿度40~60%,固态发酵150~240小时,制得混合型无抗中草药发酵饲料添加剂,发酵呼吸袋装袋,阴凉保存。
7.如权利要求6所述的制备方法,在将组份A加入组份B中,充分均匀混合后,加入无菌水适量,使含水量在保持在30~40%之间。
8.如权利要求6所述的制备方法,所述组份B为党参和黄芪的混合料,且以质量百分比计,党参和黄芪之比为2:1~1:2。
9.如权利要求6所述的制备方法,所述组份A的混合菌种由选自嗜酸乳杆菌、干酪乳杆菌、詹氏乳杆菌、拉曼乳杆菌、长双歧杆菌、短双歧杆菌、卵形双歧杆菌、嗜热双歧杆菌、粪链球菌、屎肠球菌、乳球菌、中介链球菌及地衣芽孢杆菌中的两种或多种益生菌种子液混合制成;且所述组份A的混合菌种由屎肠球菌和地衣芽孢杆菌的种子液按照2:1~1:2的比例混合制成,其中每个菌种的菌落数大于108CFU/ml。
10.如权利要求9所述的制备方法,所述组份A中屎肠球菌种子液和地衣芽孢杆菌种子液的混合比例为1:1,且所述组份B中党参和黄芪的混合比例为1:1。
