Abstract
A compound feed preparation for enhancing body color and immunity of Chinese soft-shelled turtles comprises the following components in percentage by weight: 0.2-0.3% of bile acid, 0.5-0.6% of inositol, 0.8-1% of taurine, 0.15-0.18% of vitamin C, 0.4-0.5% of Chinese herbal medicine extract and 97.42-97.75% of shrimp shell powder. The compound feed preparation is added into the conventional feed of adult trionyx sinensis or young trionyx sinensis and young trionyx sinensis, can obviously improve the body color of the trionyx sinensis, improve the immunity of the trionyx sinensis, supplement calcium and phosphorus required by the growth of the trionyx sinensis to promote the growth, prevent the skirt edge of the trionyx sinensis from upwarping, enhance the finish of a dorsal scale, improve the body color and reduce the occurrence of diseases.
Description
Technical Field
The invention belongs to the field of feed preparations, particularly relates to a feed preparation for enhancing body color and immunity of Chinese soft-shelled turtles, and also relates to a feed containing the feed preparation and application thereof.
Background
The Chinese soft-shelled turtle is an important economic variety for aquiculture in China, and has rich nutritional value, medicinal value, ornamental value and economic value. With the rapid development of economy and the improvement of the living standard of consumers, the demand of people on the yield and the quality of the Chinese soft-shelled turtles is higher and higher, the increasing demand of consumption cannot be met only by wild soft-shelled turtle resources, and the high-quality demand cannot be met by the traditional breeding mode. Under the background of pursuing high yield, the domestic breeding mode of the Chinese softshell turtles, whether greenhouse breeding or external pond breeding, is mainly high-density; in addition, the quality consciousness and brand consciousness of the Chinese soft-shelled turtle culture production are thin, so that the quality of the product is mixed, the image of the Chinese soft-shelled turtle food is damaged, the price of the Chinese soft-shelled turtle is lowered, and the culture economic benefit is landslide. The method has the advantages of improving the body color of the Chinese soft-shelled turtle, improving the product quality of the Chinese soft-shelled turtle, strengthening the immune function of the Chinese soft-shelled turtle, reducing the application of medicaments for preventing and treating diseases, meeting the production requirements of organic agricultural products and being a research hotspot of the current aquaculture. Therefore, the research and development of the green, environment-friendly, efficient, pollution-free and residue-free feed immunopotentiator and body color enhancer for the Chinese soft-shelled turtles are particularly important, so that the harm of diseases to the Chinese soft-shelled turtles is reduced, and the healthy development of the breeding industry of the Chinese soft-shelled turtles is guaranteed.
The large processed products of shrimps in China are mostly frozen headless shrimps and frozen shrimp meat, so a large amount of leftovers such as shrimp heads, shrimp shells and the like are generated in the processing process, and due to the restriction of the processing industry, a large amount of leftovers cannot be effectively utilized in a value-added manner. The processing byproducts such as the shrimp heads and the like have comprehensive and high content of protein and amino acid, are rich in DHA, EPA, astaxanthin and various mineral elements, can be directly absorbed and quickly decomposed by partial components, and have high digestibility and bioavailability. Therefore, the shrimp processing by-product is a high-quality feed raw material after being processed and utilized. At present, common utilization ways of leftovers such as shrimp heads and the like mainly comprise: (1) as a raw material for preparing chitin; (2) processing to obtain shrimp shell powder feed.
Crustacean shells are the most important commercial source of chitin. At present, chitin has potential application in agriculture, food, medical treatment, biological medicine industry and the like. The leftovers such as the shrimp heads, the shrimp shells and the like are processed into the shrimp shell powder through the working procedures of high-temperature cooking, disinfection and sterilization, drying, sieving and the like, and the shrimp shell powder generally contains abundant chitin, astaxanthin cholesterol, calcium, phosphorus, iron, manganese and other beneficial elements. A large number of tests report that the shrimp shell powder can improve the palatability of the feed, provide protein for animals, not only has good food calling effect, but also can enhance the disease resistance of the animals, promote growth, and improve the body color and the product flavor.
The Chinese herbal medicine is treasure in China, has long-term source and has application history for thousands of years, and has a series of characteristics of wide source, various types, good effect, low cost, no drug resistance, no residue, no toxic or side effect, no environment pollution and the like. The Chinese herbal medicine not only has direct inhibition and killing effects on pathogenic microorganisms, but also can enhance the disease resistance of organisms by mobilizing the anti-stress capability of the organisms, improving the immunity and defense functions of the organisms, and in recent years, the Chinese herbal medicine is widely applied to aquatic animal culture as an additive, and the application of the Chinese herbal medicine in aspects of growth promotion, disease control and the like in Chinese soft-shelled turtle culture is reported.
In the disease prevention and treatment of the Chinese soft-shelled turtles, a method of adding chemicals into feed is generally adopted, which easily causes the diseases of the Chinese soft-shelled turtles, increases the death rate and causes the loss of cultivation. Therefore, there is a need to develop a green and healthy feed preparation for enhancing the body color and improving the immunity of the Chinese soft-shelled turtles.
Disclosure of Invention
The invention aims to provide a compound feed preparation for enhancing body color and immunity of the Chinese soft-shelled turtle, which is prepared by matching shrimp shell powder and a plurality of Chinese herbal medicine components.
In order to achieve the purpose, the technical scheme adopted by the invention is as follows: a compound feed preparation comprises the following components in percentage by weight: 0.2-0.3% of bile acid, 0.5-0.6% of inositol, 0.8-1% of taurine, 0.15-0.18% of vitamin C, 0.4-0.5% of Chinese herbal medicine extract and 97.42-97.75% of shrimp shell powder, wherein the sum of the components is 100%.
Preferably, the compound feed preparation of the invention comprises the following components by weight percent: 0.3 percent of bile acid, 0.6 percent of inositol, 1 percent of taurine, 0.18 percent of vitamin C, 0.5 percent of Chinese herbal medicine extract and 97.42 percent of shrimp shell powder, and the sum of the components is 100 percent.
The Chinese herbal medicine extract is prepared by uniformly mixing astragalus, acanthopanax, honeysuckle, wild chrysanthemum, liquorice, houttuynia, poplar flower, hawthorn, purslane and dandelion in the same weight ratio, decocting and drying. The preparation method comprises the following steps of taking radix astragali, acanthopanax, honeysuckle, wild chrysanthemum, liquorice, houttuynia cordata, poplar flower, hawthorn, purslane and dandelion according to the same weight ratio, mixing uniformly, adding water which is 8-10 times of the total weight of the medicines, soaking for 30-60 minutes until the water is soaked, covering and decocting, boiling with strong fire, decocting with slow fire for 60-80 minutes after boiling, cooling, removing medicine residues, and performing spray drying on the obtained medicine liquid by taking starch which is 3-4 times of the weight of the medicine liquid as a carrier to prepare the traditional Chinese medicine powder.
The shrimp shell powder is prepared by using leftovers of crawfish after processing as raw materials, cleaning, sterilizing, drying, and crushing by using a 500-mesh nano crusher. Preferably, the temperature for disinfection and sterilization is 120-135 ℃, and the time is 0.5-1 hour; the drying is to dry for 1.5 to 2 hours at the temperature of 50 to 80 ℃ by steam; the leftovers include shrimp head, shrimp shell, etc.
The obtained shrimp shell powder mainly contains: oligochitosan, astaxanthin, phospholipid, long-chain polyunsaturated fatty acid, 19 amino acids (threonine, valine, methionine, isoleucine, leucine, phenylalanine, lysine, histidine, arginine, glycine, cystine, tyrosine, aspartic acid, serine, glutamic acid, proline, alanine, tryptophan and taurine) and 8 trace elements (calcium, phosphorus, iron, zinc, sodium, potassium, magnesium and sulfur). Because the chitosan molecule has amino, hydroxyl and other groups, many metal ions can be chelated by hydrogen bonds, and therefore, the metal ions in the solution can be effectively adsorbed and separated. Not only can remove macromolecular substances in Chinese medicinal extract, but also can trap heavy metal ions, so that the heavy metal content can be reduced, and the quality of medicine and product can be improved. The oligosaccharide can be combined with effective components of Chinese medicinal materials to form chitosan composite medicinal film, and has antibacterial and antiviral effects.
The compound feed preparation is prepared by respectively mixing bile acid, inositol, taurine, vitamin C, Chinese herbal medicine extract and shrimp shell powder according to the proportion.
The invention also provides a feed for adult trionyx sinensis, which is obtained by adding the compound feed preparation into a conventional feed for adult trionyx sinensis and uniformly mixing, wherein the compound feed preparation accounts for 12-15% of the total weight of the feed for adult trionyx sinensis.
The invention also provides a feed for young and young trionyx sinensis, which is obtained by adding the compound feed preparation into the conventional feed for the young and young trionyx sinensis and mixing uniformly, wherein the compound feed preparation accounts for 10-12% of the total weight of the feed for the young and young trionyx sinensis.
The invention also provides application of the Chinese softshell turtle feed added with the compound feed preparation in aspects of enhancing body color and enhancing immunity.
The compound feed preparation provided by the invention is comprehensive and balanced in nutrition, contains 32-35% of animal protein, and is rich in 19 amino acids, 8 medium and trace amino acids and long-chain polyunsaturated fatty acid. The composition of the 19 amino acids is similar to that of the muscle amino acid model of the Chinese softshell turtle, and the restrictive amino acid requirement of the growth of the Chinese softshell turtle can be met; the long-chain polyunsaturated fatty acid can meet the requirements of the trionyx sinensis on unsaturated fatty acid in the feed; the ratio of calcium to phosphorus in the trace elements is 2:1, which can rapidly supplement calcium and phosphorus required by the growth of the Chinese softshell turtle, prevent the Chinese softshell turtle from upwarping, enhance the luster of the dorsal scale and improve the body color.
The following description will explain the influence of the compound feed preparation for enhancing body color and immunity added to the conventional feed of Chinese soft-shelled turtles on the survival rate, immunity and body color enhancement of the young Chinese soft-shelled turtles and adult Chinese soft-shelled turtles.
1. Cultivation experiment of young Chinese soft-shelled turtles
The experiment is carried out in aquaculture bases of Changde river continents in Hunan province, 1 test group and 1 control group are arranged, 3 repeated experiment groups are respectively arranged, and the area of each experiment pond is 3000m29200 young Chinese soft-shelled turtles are placed and cultured, the average initial weight is 158.5-160.2 g, and the specifications are uniform, healthy and active. The compound feed preparation is added into complete feed of young Chinese soft-shelled turtles to serve as test group feed (the feed preparation accounts for 12% of the weight of the complete feed of the young Chinese soft-shelled turtles), and the complete feed for feeding the young Chinese soft-shelled turtles serves as a control group. Feeding for 2 times every day (once in the morning and afternoon), wherein the bait amount accounts for about 2% of the body weight, adding water during feeding, kneading into dough, and placing on a feeding table. The culture time is 180 days. After the experiment is finished, the weight gain rate, the survival rate, the bait coefficient, the body color value and the like of the young Chinese softshell turtles of the control group and the test group are measured, and the results are shown in the following table 1; and the phagocytic activity of red blood cells, the activity of serum lysozyme and the serum agglutination titer of the young Chinese softshell turtle are measured, and the results are shown in tables 2 to 4.

As can be seen from table 1 above, by feeding the feed containing the compound feed preparation of the present invention, the growth rate of young trionyx sinensis is increased significantly, the average daily gain is higher by 0.3g, the survival rate is higher by 2.25%, and the feed coefficient is reduced by 0.22, compared with the control group. The test group had significantly lower dorsal L values than the control group (P <0.05) and significantly higher ventral L values than the control group (P < 0.05); the difference in values of a is not significant between the back and the abdomen (P > 0.05); the dorsal b values presented an increasing trend, whereas the ventral b values were not significantly different (P >0.05), all the test groups were higher than the control group.

Note: the phagocytic function of blood cells is determined by TTC reduction method, and the Phagocytic Percentage (PP) is the number of TTC cells phagocytosed per 100 red blood cells, and the Phagocytic Index (PI) is the ratio of the total number of bacteria in the cells to the number of cells participating in phagocytosis.
As can be seen from table 2 above, by feeding the feed containing the compound feed preparation of the present invention, the phagocytic percentage and phagocytic index of blood cells of the soft-shelled turtle can be increased, pathogens can be effectively removed, and the phagocytic effect of the nonspecific immune mechanism of young soft-shelled turtles of the soft-shelled turtle can be increased.

Note: the sensitive pathogenic bacteria staphylococcus aureus of the soft-shelled turtles are used as substrates to detect the activity of the serum lysozyme of each group of soft-shelled turtles.
As can be seen from table 3 above, by feeding the feed added with the compound feed preparation of the present invention, the activity of the serum lysozyme of the young trionyx sinensis can be significantly improved, and the bactericidal activity of the serum can be improved.

As can be seen from the above table 4, the serum agglutination titer of the test group is obviously higher than that of the control group without the compound feed preparation of the invention, and the serum agglutination titer of the test group is about 4.8 higher than that of the control group, so that the immunity of the young trionyx sinensis is obviously improved.
2. Adult Chinese soft-shelled turtle culture experiment
The experiment is carried out in aquaculture bases of Changde river continents in Hunan province, 1 test group and 1 control group are arranged, 3 repeated experiment groups are respectively arranged, and the area of each experiment pond is 1300m21600 mature trionyx sinensis are bred in each stocking, the average initial weight is 350 +/-13.8 (g), and the adult trionyx sinensis is healthy and active. The compound feed preparation is added into complete feed of adult trionyx sinensis to serve as test group feed (the compound feed preparation accounts for 15% of the weight of the complete feed of adult trionyx sinensis), and the complete feed of adult trionyx sinensis fed by the compound feed preparation serves as a control group. Feeding for 2 times every day (once in the morning and afternoon), wherein the bait amount accounts for about 2% of the body weight, adding water during feeding, kneading into dough, and placing on a feeding table. The culture time is 180 days. After the experiment, the weight, weight gain rate, survival rate, bait coefficient, body color value and the like of the trionyx sinensis in the control group and the trionyx sinensis in the experimental group are measured, and the results are shown in the following table 5.

As can be seen from Table 5, the growth indexes of the turtles formed in the test are obviously higher than those of the control group, the average daily gain is higher than 0.53g, the survival rate is higher than 3.73%, and the bait coefficient is reduced by 0.17. The test group had significantly lower dorsal L values than the control group (P <0.05) and significantly higher ventral L values than the control group (P < 0.05); the difference in values of a is not significant between the back and the abdomen (P > 0.05); the dorsal b values presented an increasing trend, whereas the ventral b values were not significantly different (P >0.05), all the test groups were higher than the control group.

Preparing the Chinese herbal medicine extract: mixing radix astragali, radix Acanthopanacis Senticosi, flos Lonicerae, flos Chrysanthemi Indici, Glycyrrhrizae radix, herba Houttuyniae, poplar flower, fructus crataegi, herba Portulacae and herba Taraxaci at the same weight ratio, adding 10 times of water, mixing, soaking for 60 min, decocting with cover, boiling with strong fire, decocting with slow fire for 60 min, cooling, removing residue, spray drying the obtained medicinal liquid with 4 times of starch as carrier, and making into Chinese medicinal powder.
Preparing shrimp shell powder: collecting leftovers of crawfish shells, crawfish heads and the like after crawfish processing as raw materials, cleaning the raw materials with clear water, sterilizing at 128 ℃ for 0.8 hour, drying with 70 ℃ steam for 1.8 hours, and crushing with a 500-mesh nanometer crusher.

Preparing shrimp shell powder: collecting leftovers of crawfish shell, crawfish head, etc. after crawfish processing as raw materials, cleaning the raw materials with clear water, sterilizing at high temperature of 135 deg.C for 0.5 hr, drying with 80 deg.C steam for 1.5 hr, and pulverizing with 500 mesh nanometer pulverizer.
0.3 percent of bile acid, 0.6 percent of inositol, 0.9 percent of taurine, 0.16 percent of vitamin C, 0.5 percent of Chinese herbal medicine extract and 97.54 percent of shrimp shell powder are mixed evenly according to the weight percentage to obtain the compound feed preparation of the invention.
Example 4
Adding the compound feed preparation prepared in the embodiment 1 into a conventional feed for adult trionyx sinensis, and uniformly mixing to obtain the adult trionyx sinensis feed containing the feed preparation, wherein the feed preparation accounts for 15% of the weight of the adult trionyx sinensis feed.
Example 5

The compound feed preparation as claimed in claim 1, wherein the Chinese herbal medicine extract is prepared by mixing radix astragali, radix Acanthopanacis Senticosi, flos Lonicerae, flos Chrysanthemi Indici, Glycyrrhrizae radix, herba Houttuyniae, Poplar flower, fructus crataegi, herba Portulacae and herba Taraxaci at the same weight ratio, decocting, and drying.

The compound feed preparation as claimed in claim 3, wherein the Chinese herbal medicine extract is prepared by taking radix astragali, radix Acanthopanacis Senticosi, flos Lonicerae, flos Chrysanthemi Indici, Glycyrrhrizae radix, herba Houttuyniae, Poplar flower, fructus crataegi, herba Portulacae and herba Taraxaci according to the same weight ratio, mixing well, adding 8-10 times of water to soak for 30-60 minutes, covering and decocting until the soaking, boiling with strong fire, decocting with slow fire for 60-80 minutes after boiling, cooling, removing the residue, and spray drying the obtained liquid medicine by using 3-4 times of starch as carrier.
