Abstract
The invention belongs to the technical field of animal immunopotentiator pharmacy, and particularly relates to application of taurine in preparation of a medicine for improving the capacity of Chinese softshell turtle to resist viral parotitis. Application of taurine in preparing medicine for treating and preventing viral parotitis of Chinese softshell turtle is disclosed. The Chinese soft-shelled turtle is young soft-shelled turtle, and the injection dose of 120-600 mu g of taurine is used for each young soft-shelled turtle with the weight of 20 g. The invention proves that the taurine has the capacity of resisting the TSHSV virus of the Chinese softshell turtle, provides a treatment method, can obviously improve the survival rate after virus infection, and provides a candidate substance for the development of the antiviral immunopotentiator of the Chinese softshell turtle.
Description
Technical Field
The invention belongs to the technical field of animal immunopotentiator pharmacy, and particularly relates to application of taurine in preparation of a medicine for improving the capacity of Chinese softshell turtle to resist viral parotitis.
Background
Chinese soft-shelled turtle (Trionyxsinensis) Belongs to the class of reptiles, the order of terrapin, the family of turtles, the genus of turtles, commonly known as turtles, and is an important famous aquaculture species in China. In recent years, the breeding industry of Chinese soft-shelled turtles is rapidly developed.
With the rise of industrial culture, natural life habits of the Chinese soft-shelled turtles are greatly challenged due to high-density culture, seed quality degradation and the like, so that the disease resistance is rapidly reduced, various diseases frequently occur, great risks are brought to culture production, and the development of healthy culture of the Chinese soft-shelled turtles is seriously hindered. According to the national Chinese soft-shelled turtle disease monitoring report, the red bottom plate disease, the rotten skin disease, the gastroenteritis disease, the branchitis disease, the ulcer syndrome and other 5 diseases are most common in the culture process, wherein the mortality rate of the branchitis disease is highest, and the influence on culture production is greatest. In recent years, gill gland diseases are developed in a plurality of Chinese softshell turtle breeding farms in Zhejiang province, severe bleeding symptoms appear in all organs and tissues of the diseased softshell turtles, the disease is acute, the death rate is extremely high, and once the disease occurs, the disease is almost completely covered within one week of the same breeding pond. The applicant proves and separates for the first time that the pathogen of the branchitis disease is a high lethal virus, and is named as Chinese softshell turtle hemorrhagic syndrome virus (Trionyx Sinensis Hemorrhagic Syndrome Virμs,TSHSV) And the whole gene sequence was determined.
The viral disease of the Chinese soft shell turtle is the result of the interaction among pathogenic pathogens, environmental conditions and autoimmune capacity, and no specific medicine aiming at the viral disease exists at present. Immunoprophylaxis is an important measure for preventing and controlling viral diseases, and comprises specific immunity of vaccination and non-specific immunity mainly comprising immunopotentiators. The safe and effective vaccine has long development period, specific pathogen, difficult immunization mode of aquatic animals and the like, thus causing the limitation of application and popularization. The immunopotentiator has the advantage of non-specific immunity, can comprehensively improve the disease prevention capability of animal individuals, has stable general components, can be fed in a feed adding mode, and can achieve a good disease prevention effect by standard use. However, the basic research related to the immunity of the Chinese softshell turtles is still relatively deficient at present, and the development and the application of an immunopotentiator are hindered to a certain extent.
Taurine is a free beta-sulfur-containing amino acid in the organism, is named after being separated from the bile of bulls for the first time, is widely distributed in various tissues and organs of mammals, birds, fishes and aquatic invertebrates, and accounts for 25 percent of the free amino acid in the liver. The research shows that taurine is an active substance for regulating the normal physiological activities of the organism, and has the wide biological functions of diminishing inflammation, easing pain, maintaining the osmotic pressure balance of the organism, regulating nerve conduction, participating in endocrine activities, regulating lipid digestion and absorption, increasing the heart contraction capacity, improving the immunity of the organism, enhancing the oxidation resistance of cell membranes and the like. In the production of aquatic animals, taurine can be used as a phagostimulant to improve the food intake and promote the growth and development of animals; can improve the stability of erythrocyte membrane and increase the content of hemoglobin in erythrocyte, thereby enhancing the anti-hypoxia capability of animals; the immunity of the organism can be enhanced by improving the activity of superoxide dismutase and lysozyme; can improve the oxidation resistance of cell membranes, and simultaneously can be combined with a strong oxidant and free radicals in vivo to prevent redox reaction, thereby avoiding oxidative damage; can also be used as a neuromodulator to influence the level of hypothalamic-pituitary hormone secretion. Research has proved that taurine can obviously improve the weight gain rate and specific growth rate of young Chinese soft-shelled turtles, but no published report that taurine has the capacity of resisting TSHSV virus of the Chinese soft-shelled turtles exists.
Disclosure of Invention
In order to solve the technical problems, the invention aims to provide application of taurine or taurine derivatives for replacing taurine in preparing a medicament for resisting TSHSV virus. Preferably, the dosage form of the medicine is one of injection, tablets, capsules and powder; alternatively, taurine or a taurine derivative for replacing taurine is added to the feed.
Furthermore, the invention also discloses application of taurine or taurine derivatives for replacing taurine in preparing a medicament for treating and preventing the viral parotitis of the Chinese softshell turtle. Preferably, the Chinese soft-shelled turtle is a young soft-shelled turtle. The injection dose of 120-600 mug taurine is taken as the injection dose for each young turtle with the weight of 20 g. Preferably, the dosage form of the medicine is one of injection, tablets, capsules and powder; alternatively, taurine or a taurine derivative acid for substituting taurine is added to the feed.
The invention proves that the taurine has the capacity of resisting the TSHSV virus of the Chinese softshell turtle, provides a treatment method, can obviously improve the survival rate after the virus infection, and provides a candidate substance for the development of the antiviral immunopotentiator of the Chinese softshell turtle.

Detailed Description
1. Taurine solution is prepared and injected, and the method is as follows:
1) Weighing taurine 600 mg, dissolving in 1 XPBS of 100 ml, mixing well, and storing at 4 deg.C for use;
2) 100 mul of taurine is sucked by a syringe of 1 ml and injected into the abdominal cavity of the Chinese softshell turtle.
2. Injection of virus (TSHSV), the procedure used was as follows:
1) Aspirate 100 μ l of TSHSV virus fluid (containing 100 copies) with a 1 ml syringe;
2) 100 μ l of the virus solution was injected into the hind leg muscle of Chinese soft-shelled turtles (20 g of young soft-shelled turtles).
3. The method for breeding management after challenge is as follows: placing the Chinese soft shell turtle in a room with the temperature of 28 ℃ after toxin attacking, feeding the Chinese soft shell turtle for 2 times at fixed time at 8 am and 5 pm every day for 7 days, and observing survival conditions every day.
4. The method for observing and judging the antiviral ability is as follows:
1) Counting the death situation 168 hours after the toxin is attacked, and calculating the death rate;
2) Dissecting livers of live turtles and dead turtles, adding 10 ml stationary liquid into 1 g of liver tissue, and conveying the liver tissue to a biotechnology company for slicing, HE dyeing and observation;
3) Comparing whether the injection of taurine has the protection effect on the liver of the young Chinese softshell turtle.
The content of taurine is found to be obviously reduced after TSHSV infection by utilizing metabonomics, and if 600 mu g/soft-shelled turtle (20 g of young soft-shelled turtle) of taurine is supplemented in advance before infection, the disease resistance of the soft-shelled turtle to the TSHSV can be improved. As shown in fig. 1, survival rate increased by 43.4% after 5 days of infection. As shown in figure 2, the results of pathological liver sections show that the addition of taurine can protect the integrity of liver cell nuclei, reduce intercellular vacuoles, maintain the normal state of liver cells, reduce liver bleeding and reduce the occurrence of blood coagulation, and thus, the liver tissue has an important protective effect in the TSHSV (Chinese softshell turtle) resistant process.

2. The use of claim 1, wherein the medicament is in the form of one of an injection, a tablet, a capsule, and a powder; alternatively, taurine or a taurine derivative for replacing taurine is added to the feed.
3. Application of taurine or taurine derivatives for replacing taurine in preparing medicine for treating and preventing viral parotitis of Chinese softshell turtle is provided.
4. The use according to claim 3, wherein the trionyx sinensis is a juvenile trionyx sinensis.
5. The use as claimed in claim 4, wherein the injection dose of 120-600 μ g taurine is used for each 20 g young turtle.
6. The use of claim 3, wherein the medicament is in the form of one of an injection, a tablet, a capsule, and a powder; alternatively, taurine or a taurine derivative for replacing taurine is added to the feed.
7. A medicine for treating and preventing viral parotitis of Chinese softshell turtle is characterized in that the active component of the medicine contains taurine or taurine derivatives for replacing taurine.
