Abstract
本发明属于中药应用技术领域,具体公开一种太子参须提取物在制备抑制鸭H9N2亚型禽流感病毒药物中的应用。本发明首次发现太子参须提取物对鸭H9N2亚型禽流感病毒在MDCK细胞上的体外增殖具有一定抑制作用,且呈相应的剂量关系,为探究太子参须在AIV疫苗佐剂的开发提供一定的基础。
Description
技术领域
本发明属于中药应用技术领域,具体涉及一种太子参须提取物在制备抑制鸭H9N2亚型禽流感病毒药物中的应用。
背景技术
H9N2亚型禽流感病毒(AIV)起源于野鸟与水禽,自1966年首次分离后便开启全球化的蔓延与爆发,禽流感属禽类接触性传染病,随着现代养禽集约化与规模化的推进,H9N2亚型 AIV已成制约养禽业健康可持续发展的重要因素之一。目前,H9N2亚型 AIV在国内家禽中分布广泛,且在新型禽流感的演化中角色重要,而当前疫苗接种是公认的控制AIV流行的最有效方法。
随着国内养殖业 “减抗”、“无抗”模式的推行,中草药及其提取物已引起行业专家和学者的足够重视。于潼等人研究发现黄芪多糖纯品与灭活H9N2亚型禽流感病毒混合时具有协同作用,可提高小鼠外周血B淋巴细胞和CD3+CD4+T淋巴细胞亚群比例,并对小鼠外周血T淋巴细胞亚群有调节作用;黄劲等人研究发现野菊花、穿心莲、柴胡等10味中药提取的中药制剂对H9N2亚型 AIV攻毒鸡具有有效抵抗效力,减少发病率和死亡率,但截止目前,经文献检索未见有针对植物根须部位组分具有抑制鸭H9N2亚型禽流感病毒的作用报道,更未见太子参植株体及其根须具有类似或相似的功能研究结果。
发明内容
为克服现有技术中存在的不足之处,本发明的目的旨在提供一种太子参须提取物在制备抑制鸭H9N2亚型禽流感病毒药物中的应用。
为实现上述目的,本发明采取的技术方案如下:
一种太子参须提取物在制备抑制鸭H9N2亚型禽流感病毒药物中的应用。
所述太子参须提取物可按现有技术制备获得或通过市购获得,较好地,所述太子参须提取物优选按照中国专利CN 201610758305.8中公开的制备方法获得。
有益效果:本发明首次发现太子参须提取物对鸭H9N2亚型禽流感病毒在MDCK细胞上的体外增殖具有一定抑制作用,且呈相应的剂量关系,为探究太子参须在AIV疫苗佐剂的开发提供一定的基础。

以下结合具体实施例,对本发明做进一步说明。应理解,以下实施例仅用于说明本发明而非用于限制本发明的范围。
实施例1
太子参须提取物在对抑制鸭H9N2亚型禽流感病毒的抑制实验:
1 材料和方法:
1.1 材料
太子参须提取物干粉:按照中国专利CN 201610758305.8中实施例1公开的制备方法获得太子参须提取液,以g/mL计,向子参须提取液中加入6%的可溶性淀粉,搅拌混匀,然后经喷雾干燥获得太子参须提取物干粉。
鸭H9N2亚型禽流感病毒和MDCK细胞(犬肾传代细胞)均保藏于福建省农业科学院畜牧兽医研究所。
实验方法
实验中,鸭H9N2亚型禽流感病毒的扩增与MDCK细胞的培养均参考文献1:张彬. 鸭H9N2亚型禽流感病毒在鹧鸪和鸡体内复制能力的比较[D]. 南宁:广西医科大学, 2016.;鸭H9N2亚型禽流感病毒毒力检测方法采用文献1公开的TCID50实验法。
实验分为太子参须提取物高、低剂量组和空白对照组,不同组别条件下使得103 TCID50 鸭H9N2亚型禽流感病毒进行感染MDCK细胞,具体用药剂量如下:A、太子参须提取物低剂量组每孔0.1g/3mL(每孔添加3mL MDCK细胞培养液+0.1g的太子参须提取物干粉);B、太子参须提取物高剂量组每孔0.25g/3mL(每孔添加3mL MDCK细胞培养液+0.25g的太子参须提取物干粉);C、空白对照组采用每孔3mL生理盐水;每组3个复孔,48 h后测定后感染孔TCID50值,结果用平均值±标准差值(M±S)表示,数据均采用SPSS统计处理软件进行单因素方差分析。
实验结果
实验结果见表1。由表1可知,较空白对照组,太子参须提取物对鸭H9N2亚型禽流感病毒在MDCK细胞上的体外增殖具有一定抑制作用,且太子参须提取物高剂量组的抑制作用最优,统计学差异极显著(P<0.01);同时对比太子参须提取物高低剂量组的H9N2亚型禽流感病毒Log10 TCID50值,可知:太子参须提取物对鸭H9N2亚型禽流感病毒体外生长的抑制作用呈一定的剂量关系。
结论与讨论
实验中采用TCID50实验法,TCID50可检测病毒毒力大小,比较空白对照组和药物处理组的TCID50值可利于直观判断鸭H9N2亚型禽流感病毒在MDCK细胞上的体外增殖情况和药物抑制效果,结果发现:太子参须提取物对鸭H9N2亚型禽流感病毒在MDCK细胞上的体外增殖具有一定抑制作用,且呈相应的剂量关系。
