Performance testing
The compositions of examples 1-4 and comparative examples 1-4 were tested for performance as follows:
first, repair ability test (HaCaT cell migration activity test)
1. Test materials:
the compositions obtained in examples 1 to 4 and comparative examples 1 to 4 were filtered through a 0.22 μm filter.
2. The test method comprises the following steps:
HaCaT cell migration was tested using 24-well culture plates and transwell chambers (8 μm wells; Corning, U.S. A.). HaCaT cells were cultured in culture flasks. When the bottle bottom is full, the cell density of the digested cells is 2 multiplied by 105Single cell suspension of individual cells, suspended in DMEM medium (without serum), was added to each upper chamber (100. mu.L/well). Then, the cells were divided into 4 groups of 3 wells, a control group and an experimental group. Each composition was tested at a concentration of 80% and 40% (V/V, volume fraction), physiological saline and each composition at different concentrations were added to the corresponding lower chamber (600. mu.L/well), and incubated at 37 ℃ for 24 hours. On the upper surface of the chamber, non-migrating cells are carefully removed. Fixing the cells with methanol for 20min, staining with 0.1% crystal violet for 20min, eluting the stained cells, measuring the absorbance by enzyme labeling, and measuring the OD at 570 nm.
The test results are shown in fig. 1 and 2. The results show that: compared with a normal saline control, the examples 1 to 4 obviously increase the migration number of HaCaT cells, and have certain concentration dependence, which shows that the composition can promote the migration capacity of keratinocytes. After the composition of comparative examples 1 to 4 is added, the keratinocyte migration capacity under high concentration is equivalent to that of normal saline, and is obviously lower than the test results of examples 1 to 4, which shows that the arginine/lysine polypeptide, the marshmallow extract, the meyerba extract, the industrial hemp extract, the ectoine, the millettia extract, the milfoil extract and the carboxymethyl chitosan of the invention can play a role in promoting keratinocyte migration only by being matched with various raw materials, and the effect cannot be achieved by the lack of any one of the raw materials.
Moreover, in examples 1 to 4, example 1 exhibited the best performance and had the best keratinocyte migration promoting ability, indicating that arginine/lysine polypeptide, marshmallow extract, meyerba extract, industrial hemp extract, ectoine, millettia extract, yarrow extract and carboxymethyl chitosan exhibited the best synergistic effect under the formulation of example 1. As can be seen from the formulations of examples 1 to 4, the formulation of example 1 slightly increased the amount of the illipe, millettia and yarrow extracts and slightly decreased the amount of ectoine compared to example 4, resulting in an increase in the keratinocyte migration-promoting ability of about 11% at 80% addition and about 13% at 40% addition. Although the amount of the extract of yarrow is significantly increased in example 3 compared to example 4, the result is not changed compared to example 4. Similarly, example 1 increased marshmallow extract and milfoil extract and decreased millettia extract compared to example 2, with a significant change in the ability to promote keratinocyte migration; in examples 3 and 4, the amount of the plant extract was increased or decreased compared to example 2, and as a result, the ability to promote the migration of keratinocytes was not changed. The proportion of the raw materials has important influence on the performance of the composition, and the raw materials have the most effective synergistic promotion effect only under the specific proportion, so that the keratinocyte migration is effectively promoted, and the skin barrier is repaired. It is also assumed that, when the formulation of example 1 is finely adjusted to a small extent, for example, in the proportions of 0.05 to 0.1 part of arginine/lysine polypeptide, 2 to 4 parts of marshmallow extract, 2 to 4 parts of maidenhair extract, 0.5 to 1 part of industrial hemp extract, 0.5 to 1 part of ectoin, 4 to 4.5 parts of millettia extract, 4 to 4.5 parts of yarrow extract, and 0.15 to 2 parts of carboxymethyl chitosan, the keratinocyte migration-promoting ability similar to that of example 1 can be obtained, and the skin-repairing ability can be enhanced.
Second, moisture restoration experiment (human body evaluation test)
1. Experimenter and arrangement:
100 volunteers were recruited, aged 20-55 years, and randomly divided into 4 groups of 25 persons each. Volunteers were all persons with skin lesions or sensitive skin or impaired skin barrier or dry, itchy skin.
