Abstract
本发明涉及一种提高大口黑鲈糖耐受性的配合饲料及其制备方法和应用,按重量份计,由以下制成:鱼粉45‑55份、鸡肉粉4‑6份、猪肉粉3‑5份、大米蛋白粉2‑4份、玉米蛋白粉6‑8份、豆粕6‑8份、豆油3‑5份、大豆磷脂油2‑3份、啤酒酵母1‑3份、豌豆淀粉7‑10份、谷朊粉2‑4份、普鲁兰酶0.5‑0.8份、磷酸二氢钙1‑2份、复合维生素0.4‑0.6份、复合矿物质1‑2份、中药复合剂0.5‑2份;中药复合剂包括重量份3‑5:3‑5:1‑3的葛根、熟地黄和玄参。本发明显著提高大口黑鲈生长性能和饲料利用率,血糖水平的波动较小,稳定餐后血糖,降低糖原积累,有利于鱼体健康,有利于水产养殖业的可持续发展。

技术领域
本发明涉及大口黑鲈糖配合饲料,具体涉及一种提高大口黑鲈糖耐受性的配合饲料及其制备方法和应用。
背景技术
大口黑鲈(Micropterus salmoides),分类学上隶属于鲈形目、太阳鱼科、黑鲈属。因其肉嫩味美深受消费者和广大钓鱼者的喜爱,国际市场甚为畅销,素有“淡水石斑”之称。由于其生长快、饲料转化率高、市场价值高,在中国已经广泛养殖2019年总产量达到60万吨。
肉食性鱼类,对饲料中碳水化合物的利用能力十分有限,而大口黑鲈为典型肉食性鱼类,长期摄入过多的碳水化合物会导致餐后血糖升高、肝糖原积累异常等症状,不仅严重影响生长性能,甚至危害到养殖鱼类的健康。但是,由于淀粉是谷物的主要碳水化合物成分,在饲料中能发挥粘合剂的作用,且利于膨化饲料的制作,而过低含量的淀粉又不利于饲料的膨化。
因此,有必要开发一种对大口黑鲈血糖水平的影响较小,稳定餐后血糖、降低糖原积累的大口黑鲈配合饲料。
发明内容
针对以上不足,本发明提供一种采用非常用淀粉源的大口黑鲈配合饲料,提高大口黑鲈糖耐受性,稳定餐后血糖,降低糖原积累,有利于鱼体健康。
本发明通过以下方案达到上述目的:
第一方面,本发明提供一种提高大口黑鲈糖耐受性的配合饲料,按重量份计,由包括以下组分的原料制成:

进一步优选的,所述配合饲料中将豌豆淀粉以采用普鲁兰酶酶解的方式使用。
淀粉按其在消化后的生理作用可分为快速消化淀粉、慢消化淀粉和抗性淀粉。慢消化淀粉对控制血糖、减少餐后循环游离脂肪酸和降低氧化应激有着积极的影响。大口黑鲈作为天生的“糖尿病患者”,对血糖的调控能力较差,因此,消化速度慢的淀粉更有利于维持鱼类的血糖稳定,促进鱼体生长和保持鱼体健康。
本发明使用豌豆淀粉替代常规饲料中的马铃薯淀粉或小麦淀粉,豌豆淀粉中直链含量较高,是一种较好的慢消化淀粉,可以稳定鱼类餐后血糖水平;另一方面,普鲁兰酶是一种能专一地切开α-1,6-糖苷键的一种降支酶,通过普鲁兰酶处理淀粉能有效地产生更多的直链淀粉,普鲁兰酶水解后的豌豆淀粉,有较低的消化性,可以稳定鱼类餐后血糖水平。
本发明还进一步搭配中药复合剂,由葛根、熟地黄和玄参按特定重量份配比搭配组成,其中,葛根中含有葛根素,葛根素的将血糖作用明显,且降糖作用持久。熟地黄中的地黄素,具有降血糖作用,并能抑制实验性高血糖。玄参具有降低血压、降低血糖作用,动物试验表明玄参流浸膏可使正常家兔的血糖下降。一方面,三种组分搭配使用可以稳定餐后血糖,减缓血糖上升速度,而且没有副作用,另一方面,三种组分搭配上述普鲁兰酶酶解的豌豆淀粉,使得饲料的提高耐糖性效能大大提高。
第二方面,本发明还提供一种制备上述提高大口黑鲈糖耐受性的配合饲料的方法,包括:
(1)、用醋酸钠缓冲溶液制成豌豆淀粉溶液,加入普鲁兰酶,在35-65℃pH 3-5条件下酶解,灭酶,自然冷却后45-75℃烘干;
(2)、将除豆油、大豆磷脂油以外的组分原料和上述步骤(1)的烘干物,进行一次混合,得到一次混合物;
(3)、将一次混合物进行超微粉碎,再将超微粉碎后的一次混合物进行二次混合,得到二次混合物;
(4)、将二次混合物依次通过调质、膨化制粒、烘干,得到饲料半成品;
(5)、将豆油、大豆磷脂油的混合油喷涂于饲料半成品的颗粒表面上,得到所述提高大口黑鲈糖耐受性的配合饲料。
优选的,上述步骤(1)为:用弱酸缓冲液(醋酸钠缓冲溶液)制成20%的豌豆淀粉溶液,充分搅拌后灭菌,冷却至室温,加入普鲁兰酶6g/kg,在55℃PH4.5条件下酶解8h,然后灭酶10min,自然冷却后60℃烘干备用。
在一个优选的实施例中,一种制备上述提高大口黑鲈糖耐受性的配合饲料的方法,包括:
(1)、用醋酸钠缓冲溶液制成20%的豌豆淀粉溶液,充分搅拌后灭菌,冷却至室温,加入普鲁兰酶6g/kg,在55℃PH4.5条件下酶解8h,然后灭酶10min,自然冷却后再60℃烘干备用;
(2)、准确称量除豆油、大豆磷脂油以外的组分原料,将称量好的固体原料进行一次混合,得到一次混合物;
(3)、接着将一次混合物进行超微粉碎,再将超微粉碎后的一次混合物进行二次混合,得到二次混合物;
(4)、然后将二次混合物依次通过调质、膨化制粒、烘干,得到饲料半成品;
(5)、再将豆油、大豆磷脂油的混合油喷涂于饲料半成品的颗粒表面上,冷却后包装即得所述提高大口黑鲈糖耐受性的配合饲料。
优选的,在所述制备方法中,经过超微粉碎后的一次混合物,通过80目筛网的粉碎物颗粒的重量占比等于或大于95%。
第三方面,本发明还提供了上述制备方法制备得到的提高大口黑鲈糖耐受性的配合饲料。
第四方面,本发明还提供一种上述提高大口黑鲈糖耐受性的配合饲料的应用,用于大口黑鲈养殖过程中,所述饲料用于10g以上大口黑鲈的养殖,每日投喂2餐,日投喂量约为鱼体体重的2-6%。
上述配合饲料的应用中,用于提高大口黑鲈的生长性能,提高饲料利用率,稳定餐后血糖,降低糖原积累。
与现有技术相比,本发明的有益效果在于:
本发明在充分满足大口黑鲈生长的营养需要的基础上,能显著提高大口黑鲈的生长性能和饲料利用率,且血糖水平的波动较小,稳定餐后血糖,降低糖原积累,有利于鱼体健康,有利于水产养殖业的可持续发展。

以下结合具体实施例对本发明进行进一步说明。
在本发明的实施例中,采用的复合维生素为:每千克复合维生素包括以下组分与重量:维生素A,1.7×105IU;维生素D3,8.5×104IU;维生素K3,1.5克;维生素Bl,1.8克;维生素B2,4.9克;维生素B6,3克;维生素Bl2,0.03克;维生素E,16克;维生素C,80克;烟酸胺,8克;泛酸钙,7.5克;叶酸,0.7克;生物素,0.05克;肌醇,32克;氯化胆碱,150克;L-肉碱,10克,其余用脱脂米糠填充。
在本发明的实施例中,采用的复合矿物质为:每千克复合矿物质包括以下组分与重量:CuSO4 0.2克;ZnSO4 3.5克;MnS04 0.6克;FeSO4 2.2克;Ca(IO3)2 0.2克;Na2Se03 0.02克;CoC12 0.03克;MgS04·H20 5.3克,其余用沸石粉填充。

养殖试验例1
2019年5月13日在广东省广州市番禺区海大集团海鸥岛试验基地,开展对比实验8周。每组饲料设4个重复,网箱规格为2m×2m×2m,每个网箱放100尾鱼。其终末均重、成活率、特定增长率、饲料效率和肝糖原含量如表1所示。餐后血糖含量如表2所示。
其中,大口黑鲈的普通商品料由以下重量份数的组分组成:鱼粉45份、鸡肉粉5份、猪肉粉4份、大米蛋白粉3份、玉米蛋白粉7份、豆粕7份、高筋面粉15份、豆油4份、大豆磷脂油2.5份、啤酒酵母2份、谷朊粉3份、磷酸二氢钙1.5份、复合维生素0.5份、复合矿物质1.5份、抗氧化剂0.07份、防霉剂0.01份、米糠粕0.92份。
结果如表1和表2所示。

表1和表2的实验结果表明,与普通商品料、对比例1和对比6相比,经过实施例1至实施例3的饲料养殖后的大口黑鲈,其终末均重和饲料效率显著提高,而肝糖原含量显著降低。
经过血糖水平测试发现,饲料摄入后的血糖水平没有明显区别,但是随着时间的推移,特别是在3-12小时候,实施例1-3的饲料喂养的大口黑鲈的血糖升高明显减缓并且升高的水平也显著更低,证明本发明的饲料有显著的稳定餐后血糖及降低血糖积累的效果。
上述试验结果,结合发明人的进一步证实,配合饲料中的中药复合剂的葛根、熟地黄和玄参之间具有协同作用,并且其重量份配比对于效果也具有显著影响。
上述试验结果,结合发明人的进一步证实,配合饲料中的组分,特别是例如豌豆淀粉等组分,对于饲料的糖耐受性效果具有显著影响,而与其他组分结合其在饲料中的耐糖性效果,在本发明饲料范围内效果显著。
以上所述,仅为本发明的较佳的具体实施例,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,根据本发明的技术方案及其构思加以等同替换或改变,都应涵盖在本发明的保护范围内。

5.根据权利要求1至4中任一所述的配合饲料,其特征在于,所述配合饲料中将豌豆淀粉以采用普鲁兰酶酶解的方式使用。
6.一种制备权利要求1-5中任一所述的提高大口黑鲈糖耐受性的配合饲料的方法,其特征在于,包括:
(1)、用弱酸缓冲液制成豌豆淀粉溶液,加入普鲁兰酶,在35-65℃pH 3-5条件下酶解,灭酶,自然冷却后45-75℃烘干;
(2)、将除豆油、大豆磷脂油以外的组分原料和上述步骤(1)的烘干物,进行一次混合,得到一次混合物;
(3)、将一次混合物进行超微粉碎,再将超微粉碎后的一次混合物进行二次混合,得到二次混合物;
(4)、将二次混合物依次通过调质、膨化制粒、烘干,得到饲料半成品;
(5)、将豆油、大豆磷脂油的混合油喷涂于饲料半成品的颗粒表面上,得到所述提高大口黑鲈糖耐受性的配合饲料。
7.根据权利要求6所述的制备方法,其特征在于,上述步骤(1)为:用弱酸缓冲液,例如醋酸钠缓冲溶液制成20%的豌豆淀粉溶液,充分搅拌后灭菌,冷却至室温,加入普鲁兰酶6g/kg,在55℃PH4.5条件下酶解8h,然后灭酶10min,自然冷却后60℃烘干备用。
8.根据权利要求7所述的制备方法,其特征在于,包括:

10.一种权利要求1-5中任一所述或权利要求9所述的提高大口黑鲈糖耐受性的配合饲料的应用,其特征在于,用于大口黑鲈养殖过程中,所述饲料用于10g以上大口黑鲈的养殖,每日投喂2餐,日投喂量约为鱼体体重的2-6%。
