红花饮片的炮制方法主要包括烘干,微炒,炒炭,醋炙等,但无论哪种方法,都需要加热干燥,在加热过程中,多种有效成分不可避免的会遭受损失,导致红花的药效降低。红花饮片成品的保存过程中有效成分随时间的推移会发生氧化降解或挥发,也会导致红花的药效降低。
发明内容
本发明所要解决的技术问题:如何提高单位重量红花饮片成品中的有效成分含量,并在红花饮片制成后,长时间保持饮片中的有效成分不丢失。
为解决上述技术问题,本发明提供以下的技术方案:
高品质红花饮片炮制工艺,具体步骤如下:
(1)将新采收的红花去萼片,烘干得红花干品;
(2)将红花干品用增强液闷润12h,冻干得干品二,所述红花干品与闷润增强液的重量比为1:5~8。
(3)将干品二加酿造食用醋喷匀并完全吸收后,以微火炒至焦红色得干品三;
(4)将干品三进行蜡封后的红花饮片。
优选地,所述干品二与醋的重量比为10:1~3,所述微火的温度为110~130℃。
优选地,所述蜡封的具体步骤为:将蜂蜡加热融化为液体状停止加热,将干品三加入热蜡液中,搅拌均匀后静置凝固,切片即可,所述蜂蜡与干品三的重量比为3:2~8。
优选地,所述醋为米醋,果醋、白醋中的一种。
优选地,所述增强液的制备方法如下:
(1)将活血化瘀中药材按1:10~20重量比加水煮沸,采用蒸汽回流提取法提取油性挥发液;
(2)提取完成后将剩余的水溶液过滤,除渣,将滤液采用大孔树脂进行红花黄色素的提取,提取获得的红花黄色素原液与油性挥发液混合后即得增强液。
优选地,所述活血化瘀中药材为红花干品。
优选地,所述活血化瘀中药材为20重量份三七、10重量份益母草、10重量份丹参、20重量份川芎、15重量份当归、25重量份丹皮、5重量份生蒲黄、5重量份乳香、200重量份红花。
本发明获得的有益效果:
通过用增强液闷润红花干品,有效提高红花半成品中的活血有效成分含量,抵消部分加工过程中的损耗,使得红花饮片成品中的有效成分含量提高,同时利用蜂蜡对红花饮片进行包封,固定有效成分,隔绝空气,减少存贮过程中红花饮片的有效成分丧失,提高存贮的红花饮片质量。醋炙可降低红花饮片水溶液的pH值,有效提高红花黄色素的溶出率,提高药效。
具体实施方式
下面通过对实施例的描述,对本发明的具体实施方式作进一步详细的说明,以帮助本领域的技术人员对本发明的发明构思、技术方案有更完整、准确和深入的理解。
实施例1:按如下方法制备高品质红花饮片,具体步骤如下:
(1)将新采收的红花去萼片,烘干得红花干品;
将红花干品用增强液闷润12h,冻干得干品二,所述红花干品与闷润增强液的重量比为1:5~8。所述增强液的制备方法如下:
(A)将红花干品按料液比1:10重量比加水煮沸,采用蒸汽回流提取法提取油性挥发液;提取溶媒为纯水;温度60℃;提取1次,每次10min。
(B)提取完成后将剩余的水溶液过滤,除渣,将滤液采用大孔树脂H103进行红花黄色素的提取,上柱样品量与树脂量用量之比为1∶10(W:V),洗脱速度1.0mL/min·cm2。提取获得的红花黄色素原液与油性挥发液混合后即得增强液。
(2)将干品二加酿造食用醋喷匀并完全吸收后,以微火炒至焦红色得干品三;所述干品二与米醋的重量比为10:1,所述微火的温度为110℃。
(3)将干品三进行蜡封后的红花饮片,所述蜡封的具体步骤为:将蜂蜡加热融化为液体状停止加热,将干品三加入热蜡液中,搅拌均匀后静置凝固,切片即可,所述蜂蜡与干品三的重量比为3:2。
实施例2:按如下方法制备高品质红花饮片,具体步骤如下:
(1)将新采收的红花去萼片,烘干得红花干品;
(2)将红花干品用增强液闷润12h,冻干得干品二,所述红花干品与闷润增强液的重量比为1:8。所述增强液的制备方法如下:
(A)将20重量份三七、10重量份益母草、10重量份丹参、20重量份川芎、15重量份当归、25重量份丹皮、5重量份生蒲黄、5重量份乳香、200重量份红花称重配制后按料液比1:20重量比加水煮沸,采用蒸汽回流提取法提取油性挥发液;提取溶媒为pH=3的酸水;温度70℃;提取4次,每次4h。
(B)提取完成后将剩余的水溶液过滤,除渣,将滤液采用大孔树脂H103进行红花黄色素的提取,上柱样品量与树脂量用量之比为1∶15(W∶V),洗脱速度1.0mL/min·cm2。提取获得的红花黄色素原液与油性挥发液混合后即得增强液。
(3)将干品二加酿造食用醋喷匀并完全吸收后,以微火炒至焦红色得干品三;所述干品二与果醋的重量比为10:3,所述微火的温度为130℃。
(4)将干品三进行蜡封后的红花饮片,所述蜡封的具体步骤为:将蜂蜡加热融化为液体状停止加热,将干品三加入热蜡液中,搅拌均匀后静置凝固,切片即可,所述蜂蜡与干品三的重量比为3:8。
实施例3:按如下方法制备高品质红花饮片,具体步骤如下:
(1)将新采收的红花去萼片,烘干得红花干品;将红花干品用增强液闷润24h,冻干得干品二,所述红花干品与闷润增强液的重量比为1:6.5。所述增强液的制备方法如下:
(A)将红花干品按料液比1:15重量比加水煮沸,采用蒸汽回流提取法提取油性挥发液;提取溶媒为pH=3的酸水;温度65℃;提取3次,每次1h。
(B)提取完成后将剩余的水溶液过滤,除渣,将滤液采用大孔树脂H103进行红花黄色素的提取,上柱样品量与树脂量用量之比为1∶10(W∶V),洗脱速度1.0mL/min·cm2。提取获得的红花黄色素原液与油性挥发液混合后即得增强液。
(2)将干品二加酿造食用醋喷匀并完全吸收后,以微火炒至焦红色得干品三;所述干品二与白醋的重量比为10:2,所述微火的温度为120℃。
(3)将干品三进行蜡封后的红花饮片,所述蜡封的具体步骤为:将蜂蜡加热融化为液体状停止加热,将干品三加入热蜡液中,搅拌均匀后静置凝固,切片即可,所述蜂蜡与干品三的重量比为3:5。
对照实施例1:其余均与实施例1相同,不同之处在于未进行蜡封步骤。
对照实施例2:其余均与实施例2相同,不同之处在于未进行增强液闷润步骤。
将实施例1~3及对照实施例1、2新制的红花饮片分别采用纯水煮制30min,料液比1:30,过滤取下层水溶液得粗提液,将粗提液作为样品,采用HPLC测定其中的有效成分含量,采用GC测定挥发油含量,结果如下:

表1结果显示,采用增强液闷润后制备的红花饮片,其红花黄色素、聚炔、挥发油的含量或存留量显著提高。主要是由于增强液中含有大量的红花黄色素、聚炔、挥发油成分,抵消加工过程中的损耗,使得主要药效成分在热加工后仍有提高。蜡封步骤对新制红花饮片中有效成分含量几乎没有影响。
将实施例2及对照实施例1新制的红花饮片分别置于37℃,空气接触条件下,放置6、12、24个月后采样测定其中的有效成分含量,结果如下:

表1和表2结果显示没有进行蜡封时,挥发油的降低速度最快,其次为聚炔,挥发油主要是挥发至空气中而丧失,而聚炔则是由于在空气中聚变而丧失,红花黄色素在长时间的贮存后,也有较大的损失,因此,相同贮存条件下,蜡封可隔绝饮片与空气,使得饮片中的有效成分可长期存留与红花饮片中,显著提高了饮片的保藏时间,提高饮片的加工经济效益。
综上所述,通过用增强液闷润红花干品,有效提高红花半成品中的活血有效成分含量,抵消部分加工过程中的损耗,使得红花饮片成品中的有效成分含量提高,同时利用蜂蜡对红花饮片进行包封,固定有效成分,隔绝空气,减少存贮过程中红花饮片的有效成分丧失,提高存贮的红花饮片质量。醋炙可降低红花饮片水溶液的pH值,有效提高红花黄色素的溶出率,提高药效。

Claims (7)
1.高品质红花饮片炮制工艺,其特征在于,具体步骤如下:
(1)将新采收的红花去萼片,烘干得红花干品;
(2)将红花干品用增强液闷润12h,冻干得干品二,所述红花干品与闷润增强液的重量比为1:5~8。
(3)将干品二加酿造食用醋喷匀并完全吸收后,以微火炒至焦红色得干品三;
(4)将干品三进行蜡封后的红花饮片。
2.根据权利要求1中所述的高品质红花饮片炮制工艺,其特征在于:所述干品二与醋的重量比为10:1~3,所述微火的温度为110~130℃。
3.根据权利要求1中所述的高品质红花饮片炮制工艺,其特征在于:所述蜡封的具体步骤为:将蜂蜡加热融化为液体状停止加热,将干品三加入热蜡液中,搅拌均匀后静置凝固,切片即可,所述蜂蜡与干品三的重量比为3:2~8。
4.根据权利要求1中所述的高品质红花饮片炮制工艺,其特征在于:所述醋为米醋,果醋、白醋中的一种。
5.根据权利要求4中所述的高品质红花饮片炮制工艺,其特征在于:所述增强液的制备方法如下:

7.根据权利要求5中所述的高品质红花饮片炮制工艺,其特征在于:所述活血化瘀中药材为20重量份三七、10重量份益母草、10重量份丹参、20重量份川芎、15重量份当归、25重量份丹皮、5重量份生蒲黄、5重量份乳香、200重量份红花。
