Abstract

Technical Field

By adding the foaming agent and the thickening agent, the prepared sterilization and disinfection composition is emulsion-shaped, is more stable to store and has longer storage period; when the hand sanitizer is used for hand disinfection, the use is more convenient. Further preferably, the sterilization and disinfection composition comprises the following components in percentage by weight: 0.3-0.36% of polyhexamethylene guanidine, 1-2% of foaming agent, 25-30% of thickening agent and 67.64-73.7% of pure water.

s3, adding the mixture A into the mixture B, and continuing stirring for 5-15min at the rotating speed of 600-800rpm to obtain the emulsion-shaped sterilization and disinfection composition.
In summary, compared with the prior art, the application has the following beneficial effects:
(1) by adding the foaming agent and the thickening agent, the prepared sterilization and disinfection composition is emulsion-shaped, is more stable to store and has longer storage period; when the hand sanitizer is used for hand disinfection, the use is more convenient. (3) this application uses pearl powder as the thickener, except that the viscidity that improves disinfection product, after the use, pearl powder dehydration is separated out, can guarantee that the hand is dry smooth, and it is required that the skin is pressed close to the affinity, better moistening and protection skin.

The present application will be described in detail below with reference to the drawings and examples 1 to 5.
The pearl powder in the application is purchased from Wuhanxin Jiali biotechnology limited; the cassava starch and the corn starch are purchased from chemical products limited of jiana in Henan;
sapindus saponin was purchased from Sapindus Ridgeon Biotech Co., Ltd; sodium cocoyl amphoacetate was purchased from Lossen chemical Co., Linyi; n-alkyl aspartic acid- β -alkyl esters are available from guangzhou daily oil science co.
Example 1:
a sterilization and disinfection composition comprises, by weight, 0.3% of polyhexamethylene guanidine, 0.5% of soapberry saponin, 20% of pearl powder and 79.2% of pure water, and is prepared by the following steps as shown in figure 1:
s1, according to the proportion, taking 26.4kg of pure water, adding 0.3kg of polyhexamethylene guanidine and 0.5kg of soapnut saponin, and stirring for 10min at the rotating speed of 150rpm to obtain a mixture A;
s2, adding 20kg of pearl powder into 52.8kg of pure water, and stirring at 600rpm for 20min to obtain a mixture B;
s3, adding the mixture A into the mixture B, and continuing stirring for 15min at the rotating speed of 600rpm to obtain the emulsion-shaped sterilization and disinfection composition.
Example 2:
the sterilizing and disinfecting composition consists of polyhexamethylene guanidine 0.35 wt%, soapberry saponin 1.5 wt%, pearl powder 30 wt% and pure water 68.15 wt%, and is prepared through the following steps:
s1, taking 22.7kg of pure water according to the proportion, adding 0.35kg of polyhexamethylene guanidine and 1.5kg of soapnut saponin, and stirring for 10min at the rotating speed of 150rpm to obtain a mixture A;
s2, adding 45.45kg of pure water into 30kg of pearl powder, and stirring at 600rpm for 20min to obtain a mixture B;
s3, adding the mixture A into the mixture B, and continuing stirring for 15min at the rotating speed of 600rpm to obtain the emulsion-shaped sterilization and disinfection composition.
Example 3:
the sterilizing and disinfecting composition consists of polyhexamethylene guanidine 0.36 wt%, soapberry saponin 2.5 wt%, pearl powder 35 wt% and pure water 62.14 wt%, and is prepared through the following steps:
s1, taking 20.7kg of pure water according to the proportion, adding 0.36kg of polyhexamethylene guanidine and 2.5kg of soapberry saponin, and stirring for 10min at the rotating speed of 150rpm to obtain a mixture A;
s2, adding 41.44kg of pure water into 35kg of pearl powder, and stirring at 600rpm for 20min to obtain a mixture B;
s3, adding the mixture A into the mixture B, and continuing stirring for 15min at the rotating speed of 600rpm to obtain the emulsion-shaped sterilization and disinfection composition.
Example 4:
the sterilizing and disinfecting composition consists of, by weight, 0.3% of polyhexamethylene guanidine, 0.5% of sodium cocoyl amphoacetate, 20% of cassava starch and 79.2% of pure water, and is prepared by the following steps:
s1, according to the proportion, 26.4kg of pure water is taken, 0.3kg of polyhexamethylene guanidine and 0.5kg of sodium cocoyl amphoteric acetate are added, and the mixture is stirred for 12.5min at the rotating speed of 225rpm, so that a mixture A is obtained;
s2, taking 52.8kg of pure water, adding 20kg of cassava starch, and stirring at the rotating speed of 700rpm for 17.5min to obtain a mixture B;

the sterilizing and disinfecting composition consists of polyhexamethylene guanidine 0.3 wt%, N-alkyl aspartic acid-beta-alkyl ester 0.5 wt%, corn starch 20 wt% and pure water 79.2 wt%, and is prepared through the following steps:
s1, according to the proportion, taking 26.4kg of pure water, adding 0.3kg of polyhexamethylene guanidine and 0.5kg of N-alkyl aspartic acid-beta-alkyl ester, and stirring for 5min at the rotating speed of 300rpm to obtain a mixture A;
s2, taking 52.8kg of pure water, adding 20kg of corn starch, and stirring at the rotating speed of 800rpm for 15min to obtain a mixture B;
s3, adding the mixture A into the mixture B, and continuing stirring for 5min at the rotating speed of 800rpm to obtain the emulsion-shaped sterilization and disinfection composition.
Comparative example 1: a disinfectant solution having a disinfecting component of polyhexamethylene guanidine was used as comparative example 1.
Comparative example 2: the composition comprises the following components: a disinfectant solution of 0.3% polyhexamethylene guanidine and 99.7% pure water was used as comparative example 2.
Comparative example 3: the composition comprises the following components: a disinfectant solution of 0.36% polyhexamethylene guanidine and 99.64% pure water was used as comparative example 3.
Comparative example 4: the composition comprises the following components: 0.3% polyhexamethylene guanidine, 20% pearl powder and 79.7% pure water were used as comparative example 4.
Comparative example 5: the composition comprises the following components: 0.3% polyhexamethylene guanidine, 0.5% soapnut saponin and 99.2% pure water were used as comparative example 5.
Performance testing
1. Sensory evaluation
The test method comprises the following steps: the 50 subjects were equally divided into 10 groups and given samples at random according to the "double-blind" test, and were asked to wash their hands at random with the amount of disinfectant used and the time spent.
Evaluation criteria: the subjects were scored according to their feelings using a 5-point scale, wherein smooth feeling, moist feeling and irritation were all 1 point worst and 5 points best, and the test results are shown in table 1.

As can be seen from the test results in Table 1, the disinfectant of examples 1 to 5 had a smooth feeling, moist feeling, and irritation evaluation score of 4.5 to 4.8, 3.5 to 4.7, and 5, and the overall score evaluation was high. The disinfectant in comparative examples 1-3 has smooth feeling and moistening feeling of only 1 point, and irritation of 4.5-4.6 points, i.e. the disinfectant prepared has no smooth feeling and moistening feeling and poor use experience under the condition of not adding foaming agent and thickening agent. The disinfectant liquids of comparative examples 4 to 5 were found to have a smooth feeling and a moist feeling of 3.2 to 4.5 minutes and 4.2 to 4.8 minutes, which were slightly inferior to those of examples 1 to 5 and better than those of comparative examples 1 to 3.
2. Sterilization test strains: staphylococcus aureus (ATCC 6538); escherichia coli (8099); candida albicans (ATCC 10231).
Preparing a bacterial suspension: staphylococcus aureus, Escherichia coli and Candida albicans cultured for 24h were diluted with 5mL of sterile distilled water into bacterial suspensions in 0.03 mol/L phosphate buffer containing 1% peptone, respectively, at pH 7.2-7.4.
And (3) sterilization experiment: taking 1mL of the bacterial suspension into a sterile test tube, keeping the temperature in a water bath at 19-21 ℃, adding a certain amount of disinfectant, uniformly mixing, oscillating, acting for a specified time, taking 0.5mL of the bacterial-drug mixed solution, uniformly mixing in 4.5mL of a neutralizer, taking 1mL of the solution or diluent after the neutralization for 10min, inoculating a sterile plate, pouring a common nutrient agar culture medium, culturing for 48h in a 37 ℃ incubator, counting the number of bacterial colonies of the plate, averaging the experiments for three times, taking an average value, calculating the sterilization rate to be more than or equal to 99.96%, oscillating the water bath for the time and adding the disinfectant, and recording the test results into Table 2.

As can be seen from the test results in Table 2, the time required for the disinfection solution in examples 1-5 to reach a sterilization rate of 99.96% or more is 1.5-1.7min, and the minimum required amount is 2 mL; the time required for the sterilization rate of the disinfectant in comparative examples 1, 2 and 4 to reach more than 99.96 percent is 2.6-4.0min, and the minimum required dosage is 3-4 mL; in comparative examples 3 and 5, as the foaming agent and the thickening agent are not added, the sterilization time is 2.6min and 2.2min respectively, and the minimum consumption of the required disinfectant is 3mL and 2mL respectively. The polyhexamethylene guanidine can play a role in synergism after being added with the foaming agent and the thickening agent, can achieve a good sterilization effect in a short time only by using less disinfectant, and greatly reduces the cost.

3. The germicidal disinfectant composition as claimed in claim 2, wherein said germicidal disinfectant composition comprises the following components in percentage by weight: 0.35% of polyhexamethylene guanidine, 1.5% of foaming agent, 30% of thickening agent and 68.15% of pure water.
4. A germicidal disinfectant composition as claimed in any one of claims 1 to 3 wherein said foaming agent is selected from the group consisting of sapindoside, sodium cocoamphoacetate, and β -alkyl N-alkylaspartate.
5. The germicidal disinfectant composition as claimed in claim 4, wherein said foaming agent is sapindoside.
6. The bactericidal disinfectant composition as set forth in any one of claims 1 to 3, wherein said thickener is one selected from the group consisting of pearl powder, tapioca starch, and corn starch.
7. The sterilization and disinfection composition of claim 6, wherein the thickener is pearl powder.
8. A process for the preparation of a germicidal disinfectant composition as claimed in any one of claims 1 to 7 comprising the steps of:
s1, adding 1/3-1/2 pure water into polyhexamethylene guanidine and a foaming agent, and stirring at the rotating speed of 150-300rpm for 5-10min to obtain a mixture A;
s2, adding the thickener into the residual pure water, and stirring at the rotating speed of 600-800rpm for 15-20min to obtain a mixture B;
s3, adding the mixture A into the mixture B, and continuing stirring for 5-15min at the rotating speed of 600-800rpm to obtain the emulsion-shaped sterilization and disinfection composition.
