Abstract

本发明公开了一种水稻增强子及鉴定方法。该水稻增强子为622bp的DNA分子,序列如SEQ ID NO.1。构建OsENHANCER1 reporter载体、正对照载体CaMV35S‑GFP reporter(SEQ ID NO.2)和负对照载体Enhancerless reporter(SEQ ID NO.3),通过水稻原生质体转化进行鉴定。本发明增强子在基因转录过程中发挥重要作用,鉴定方法快速,便捷。

一种水稻增强子及鉴定方法
技术领域
本发明涉及增强子及鉴定方法,特别涉及一种水稻增强子及鉴定方法。
背景技术
增强子作为顺式调控元件,主要促进远距离的目标基因的转录。增强子无论位于基因的上游、下游或是基因内部,都可以发挥增强转录的作用。研究表明增强子具有细胞或组织特异性,这取决于细胞或组织中反式作用因子。增强子可以与一些特定反式作用因子而起作用。且与其自身序列方向无关。真核生物基因的转录是一个复杂的过程,增强子与多种调控因子共同参与转录调控,在细胞分化及个体发育中发挥着重要作用。近年来关于增强子的研究取得了很大进展。据报道,增强子可以作为转录因子的平台,其中一研究认为增强子区域会与启动子区域相互作用,形成环状结构,使作用于该增强子区域的转录因子可以促进目的基因转录。因此增强子是基因工程中重要的研究对象。
发明内容
发明目的:本发明目的是提供一种水稻增强子。
本发明的另一目的是提供所述水稻增强子的鉴定方法。
技术方案:本发明提供一种水稻增强子,基因序列如SEQ ID NO.1。
进一步地,所述水稻增强子来源于禾本科稻属粳稻日本晴(Oryza sativaL.spp.Japonica var.Nipponbare),具体用引物以粳稻日本晴的基因组DNA为模板,扩增得到662bp的扩增产物。
进一步地,所述引物序列如下:
正向:5’-gtacaagtgactcgagctacggtttcgaggaaaaggggtt-3’
反向R:5’-atcttcatcttcatatatccgaacagcggatctgacgacg-3’。
所述的水稻增强子的鉴定方法,包括如下步骤:
(1)通过生物信息学分析,结合本实验室水稻DNaseI数据对日本晴基因组进行预测,获得增强子OsENHANCER1(SEQ ID NO.1);
(2)将OsENHANCER1(SEQ ID NO.1)扩增后插入mini35s-GFP reporter(SEQ IDNO.4),构建OsENHANCER1 reporter载体;
(3)构建正对照载体CaMV35S-GFP reporter(SEQ ID NO.2)和负对照载体Enhancerless reporter(SEQ ID NO.3);
(4)水稻原生质体的制备与转化;
(5)转化后对OsENHANCER1的活性进行验证。
进一步地,所述步骤(3)中正对照载体构建方法为:以P1300LV载体为模板,扩增CaMV35S启动子;使用限制性内切酶AscI和XbaI将mini35s-GFP reporter载体线性化,用无缝连接试剂盒将上述2个片段快速连接,转化至大肠杆菌DH5 α中,筛选阳性转化子,得到正对照载体CaMV35S-GFP reporter(SEQ ID NO.2)。
进一步地,所述步骤(3)中负对照载体构建方法为:使用限制性内切酶BsaI将mini35s-GFP reporter载体线性化,将一段无功能序列插入线性化载体,连接后转化至大肠杆菌DH5 α中,筛选阳性转化子,得到负对照载体Enhancerless reporter(SEQ IDNO.3)。
鉴定原理:负对照载体利用mini35s启动子不能启动的荧光蛋白GFP表达,由于增强子可远距离促进转录,所以负对照载体中GFP序列后插入增强子序列,当增强子有功能时荧光蛋白则可发光。若以相同策略插入非增强子序列时,荧光蛋白不能发光。
有益效果:本发明提供了一种水稻增强子序列,本质是DNA分子,622bp,对转录具有较好的增强效果,同时,提供了快速便捷的鉴定方法。

具体实施方式
本实施例的增强字获取及鉴定方法如下:
1.OsENHANCER1介绍
水稻基因组开放染色质数据(DNase-seq)在植物开放染色质数据库PlantDHS(http://plantdhs.org/)中获得,结合水稻组蛋白修饰数据(ChIP-seq),预测基因组中增强子位点。增强子通常位于无核小体保护的开放染色质区域,可以与调控蛋白共同作用于远端基因上游的启动子区域,共同促进下游基因的转录,与基因的H3K27ac组蛋白修饰高度相关。经分析获得OsENHANCER1(SEQ ID NO.1)。
设计序列特异性引物,制备如下引物:
正向:5’-gtacaagtgactcgagctacggtttcgaggaaaaggggtt-3’
反向R:5’-atcttcatcttcatatatccgaacagcggatctgacgacg-3’。
用以上引物以粳稻日本晴的基因组DNA为模板,扩增得到662bp的扩增产物。
经测序,扩增产物的核苷酸序列如SEQ ID NO.1的自5’端第一位-662位所示。
2.检测
将SEQ ID NO.1所示的片段插入载体mini35s-GFP reporter(由电子科技大学张勇课题组提供,SEQ ID NO.4)中的BsaI酶切位点中,构建OsENHANCER1 reporter载体(如图2所示)。
同时构建正对照载体:以P1300LV载体为模板,扩增CaMV35S启动子;使用限制性内切酶AscI和XbaI将mini35s-GFP reporter载体线性化,用无缝连接试剂盒将上述2个片段快速连接,转化至大肠杆菌DH5 α中,筛选阳性转化子,得到正对照载体CaMV35S-GFPreporter(如图1所示)。
负对照载体:使用限制性内切酶BsaI将mini35s-GFP reporter载体线性化,将一段无功能序列插入线性化载体,连接后转化至大肠杆菌DH5 α中,筛选阳性转化子,得到负对照载体Enhancerless reporter(如图1所示)。
5.水稻原生质体制备与转化
5.1水稻原生质体制备:
(1)叶片准备。取无菌培养14天左右生长良好的新鲜水稻植株40-60株,去掉根部,备用。
(2)酶解。将水稻叶片和茎段用锋利刀片切成宽度为0.5mm的条状,速度要快,防止叶片脱水,将切好的叶片放入盛有10mL酶解液的培养皿中,置于密闭容器中用真空泵抽真空30min,用封口膜封口,置于60rpm转速的摇床上25℃黑暗条件下过夜酶解。
(3)过滤。将过夜酶解的粗酶液通过预先用2mL W5 washing Buffer润洗过的孔径40μm的细胞筛进行过滤,去除酶解不充分的杂质。
(4)提纯。将过滤过的细胞酶解液转移到两个15mL离心管中,每管6mL细胞酶解液,用装有长针头的注射器吸取16mL0.55M蔗糖,每管细胞酶解液底部加入8mL 0.55M蔗糖,注意:加蔗糖时,针头伸入底部,推动注射器,随着液面升高,缓慢上移注射器,动作要轻柔,防止对原生质体细胞造成破坏。1000g离心5min。
(5)清洗。取50mL离心管一个,加入10mL Washing buffer,置于试管架上。用装有长针头的注射器吸取8mL Washing buffer,再将针头伸到上步离心后的中间细胞层,慢慢吸取该层的所有原生质体,然后小心转移至盛有10mL Washing buffer的50mL离心管中。轻弹混匀,100g室温离心5min。
(6)重复清洗一次。移除上清,沿壁慢慢加入10mL Washing buffer,轻弹混匀,100g离心2min。
(7)重悬。去掉上清,加入5mL Washing buffer,注意沿壁缓慢加入,轻弹混匀,使细胞保持悬浮。
(8)计数。吸取100μL细胞用于计数,取6-7pL细胞置于细胞计数板进行计数,如果细胞数目太多可采用MGG稀释后再进行计数。
(9)细胞稀释。根据计数结果取适量细胞悬浮液进行离心,然后用MGG Buffer重悬细胞,最终将细胞稀释至1×106个/mL,用于原生质体转化。
5.2水稻原生质体的转化:
(1)质粒的准备。采用质粒小提试剂盒(TIANGEN,Cat#DP-106)提取以下质粒:mini35s-GFP reporter,OsENHANCER1 reporter,Enhancerless reporter和CaMV35S-GFPreporter。确保其浓度尽量在500ng/μL以上,整个过程最好为无菌操作。质粒置于-20℃冰箱冻存。使用前以离心机最高转速进行离心10min处理,使杂质沉淀于离心管底部,避免因杂质影响原生质体转化的效率。处理好的质粒用MGG Buffer稀释到实验所需质粒浓度,稀释后充分混匀备用。
(2)PEG介导的转化。将上述准备好的质粒(mini35s-GFP reporter,OsENHANCER1reporter,Enhancerless reporter和CaMV35S-GFP reporter)中加入200μL按照2.2.6制备过程制备的原生质体细胞,轻弹混匀。然后,每管加入210-230μL 40%PEG(PEG加入量与质粒体积相关,确保等体积加入),轻弹混匀,直至细胞均匀悬浮于PEG当中,从第一管加入PEG开始计时,计时20min。从第一管加入到最后一管加入的时间尽量控制在5min以内。
(3)清洗。加入PEG反应20min后,每管加入1mL W5 Washing Buffer,终止反应,轻轻颠倒混匀,250g离心5min。
(4)重复清洗一次。用枪轻轻吸掉上清,重新加入800μL Washing Buffer,轻轻颠倒混匀,150g离心5min,轻轻吸掉上清。
(5)细胞重悬与培养。取40mm培养皿加入2mL W5 Buffer,再取培养皿中的W5Buffer重悬离心管中的细胞,并全部转移至培养皿中,25℃黑暗培养,两天后在显微镜下观察荧光。
6.激光共聚焦显微镜观察
使用Zeiss 700激光共聚集显微镜进行观察。GFP使用的激发波长为488nm,吸收波长为490-540nm。所有的荧光实验至少重复三次。mini35s-GFP reporter,OsENHANCER1reporter,Enhancerless reporter和CaMV35S-GFP reporter质粒分别转化原生质体后,在转化子中观察,结果如图1所示,正对照CaMV35S-GFP reporter有较强荧光;负对照mini35s-GFP reporter和Enhancerless reporter均没有荧光信号;图2所示,连有OsENHANCER1的OsENHANCER1 reporter质粒中能观察到明显的GFP荧光信号。

(2)将OsENHANCER1,其序列如SEQ ID NO.1所示,扩增后插入mini35s-GFP reporter,其序列如SEQ ID NO.4所示,构建OsENHANCER1 reporter载体;
(3)构建正对照载体CaMV35S-GFP reporter,其序列如SEQ ID NO.2所示,和负对照载体Enhancerless reporter,其序列如SEQ ID NO.3所示;
(4)水稻原生质体的制备与转化;
(5)转化后对OsENHANCER1的活性进行验证。
5.根据权利要求4所述的水稻增强子的鉴定方法,其特征在于:所述步骤(3)中正对照载体构建方法为:以P1300LV载体为模板,扩增CaMV35S启动子;使用限制性内切酶AscI和XbaI将mini35s-GFP reporter载体线性化,用无缝连接试剂盒将上述2个片段快速连接,转化至大肠杆菌DH5α中,筛选阳性转化子,得到正对照载体CaMV35S-GFP reporter,其序列如SEQ ID NO.2所示。
6.根据权利要求4所述的水稻增强子的鉴定方法,其特征在于:所述步骤(3)中负对照载体构建方法为:使用限制性内切酶BsaI将mini35s-GFP reporter载体线性化,将一段无功能序列插入线性化载体,连接后转化至大肠杆菌DH5α中,筛选阳性转化子,得到负对照载体Enhancerless reporter ,其序列如SEQ ID NO.3所示。
