Experimental example 2 preparation and Effect test of Ligusticum chuanxiong Hort extract-Ligusticum chuanxiong Hort essential oil
1. Pretreatment
Cleaning rhizoma Ligustici Chuanxiong, crushing, and drying to obtain rhizoma Ligustici Chuanxiong decoction pieces;
2. supercritical carbon dioxide extraction of essential oil
(1) Adding carbon dioxide in supercritical state into rhizoma Ligustici Chuanxiong decoction pieces, and extracting with supercritical carbon dioxide extractor to obtain essential oil, wherein the specific extraction parameters are shown in Table 2:
TABLE 2 supercritical extraction Process parameters
Figure BDA0002501075720000051
(2) And (3) filtering: filtering with 200 mesh filter cloth for three times;
(3) concentration: concentrating with rotary evaporator, recovering ethanol, and controlling concentration temperature at 60 + -5 deg.C for 1-2 hr;
(4) and (3) filtering: filtering the concentrated essential oil with 200 mesh filter cloth for 2 times;
(5) standing: standing for 90 days to obtain;
3. mosquito repellent effect test of ligusticum wallichii essential oil
The prepared ligusticum wallichii essential oil is combined with permethrin and a high polymer slow release agent to prepare PD-1, PD-2 and PD-3 long-acting repellents, a repellent effect test is carried out on the termite albopictus in a laboratory, and compared with the repellent effects of Deet crude oil and 3 commercially available repellents, the results are as follows: human body presses 1.5μV cm2Or 1.5mg/cm2After treatment, the effective protection rate of the PD type repellent reaches over 90 percent, and the PD type repellent is obviously superior to Deet crude oil and commercial repellent and has obvious effect.
4. Analysis of effect of Ligusticum chuanxiong essential oil mosquito repellent prepared by supercritical carbon dioxide extraction
The effect of the supercritical carbon dioxide extraction of the essential oil for avoiding mosquitoes is better:
1) the distillation extraction temperature is too high, so that the top note components are difficult to recover; the heating temperature is kept at 100 ℃ or above, so that some components (such as wax and the like) with low volatility are also brought out and mixed into the essential oil, the quality of the essential oil is seriously influenced by excessive wax components, the essential oil extracted by supercritical carbon dioxide has no problems, and the mosquito repelling effect is better;
2) the distillation extraction oil-water separation is difficult: the ligusticum wallichii essential oil is a mixture consisting of a plurality of micromolecular compounds, has certain water solubility, and a large amount of water is added in distillation extraction, so that the oil-water ratio is increased, the essential oil is difficult to separate from water, and the critical carbon dioxide extraction uses an organic solvent, so that the essential oil can be volatilized at normal temperature, and the quality of the essential oil is improved.
Test example 3 comparative test
1. Test drugs
Comparative 1 group composition: 30% of extract components (10-20 parts of ethyl butyl levulinate, 1-2 parts of menthol, 1-6 parts of menthone glycerol ketal, 5-10 parts of citronella extract and 0.1-1 part of thymol), 20% of ethanol and 50% of deionized water
Comparative 2 group composition: 30% of ligusticum wallichii containing essential oil component (10-20 parts of ethyl butyl levulinate, 1-2 parts of menthol, 1-6 parts of menthone glycerol ketal, 5-20 parts of ligusticum wallichii essential oil, 5-10 parts of citronella extract and 0.1-1 part of thymol), 20% of ethanol and 50% of deionized water
Comparative 3 group composition: 30% of ligusticum wallichii essential oil, 20% of ethanol and 50% of deionized water

Volunteers (18-38 years old, 30 persons; 38-58 years old, 30 persons) in different age groups are respectively selected as trial test objects.
Test method 1: a sample of a fixed volume comparison group is taken and smeared on the arms of both hands of a volunteer, the hands are placed in a fixed cage, a certain number of female mosquitoes are in the cage, the number of times of biting is counted from the beginning of placing the female mosquitoes in the cage, and after 1 hour, the biting results and the using feeling scores of each group are observed and compared. The usage scoring items refer to the effects of relieving itching, stimulating feelings and overall experience comfort.
The test method 2 comprises the steps of taking samples of a fixed volume comparison group, smearing the samples on arms and legs of volunteers, enabling each group of volunteers to walk in a forest, and carrying out effective duration registration and use scoring every two hours. The usage scoring items refer to relieving and itching relieving effects, stimulating feelings and overall experience comfort level in the using process
The total using effect of each item in the score is 5 points: score 5 is the highest score, indicating very satisfactory; 4 points are better and relatively satisfactory; score 3 is acceptable; an unacceptable range is 3 or less. The number of bites is counted.
3. Test results
The results of the test of the effect of the 3 comparative drugs are shown in tables 3 and 4

Effects of use Comparative group 1 Comparative group 2 Comparative group 3
Number of bites 18 times of 6 times of 13 times (twice)
Relieving itching 4.5 4.96 4.80
Degree of irritation 4.84 4.96 4.67
Feeling of comfort 4.88 4.93 4.78
