较早的实验表明,悬浮在营养液液滴中的微生物在最多一小时的相对短的静置时间(或“静止时间”)之后,以微生物的沉积物的形式沉积在平坦的基底上。例如,通过使吸收性布与液滴接触,可以将以一种“生物膜”沉积的微生物与残留的液体和残留的悬浮颗粒小心地分离。在该“脱水”之后,可以通过随后的质谱测定可靠地确定微生物的种类,请参见国际申请WO 2018/099500 A1。该发现之所以令人惊讶是因为与预期相反,发现目的微生物的细胞沉积物在被抽出时并未与液体一起被除去。这一发现可以培养(或培育)用于促进生长的微生物,也可以在一个相同的基底(例如用于插入到质谱仪的离子源中的样品支持板,或用于插入到红外光谱仪的测定槽中的样本载玻片)上进行分析测定的准备。
尚无法获得针对平坦基底上液滴中该微生物行为的完整科学解释,但可以认为样品支持物表面与微生物细胞之间的物理相互作用,以及来自微生物细胞表面的生物化学和生物物理特性的粘附过程决定了在基底上的优选粘附或沉降。
在从WO 2018/099500 A1已知的现有技术的进一步发展中,申请EP 3376202 A1提出了一种具有凹痕或孔的掩模,其内表面与液滴的外围区域接触并通过毛细管力侧向去除液滴。在此过程中,不接触液滴的中心(大部分微生物沉积物位于其下方),因为担心与沉积物的直接接触必定会导致微生物耗尽(“平板效应”),因此,在随后的分析测定中会导致相关的灵敏度损失。
如在EP 3 376 202 A1中所描述的,通过具有轮廓的掩模或通过将刚性板放置在围绕液滴的垂直间隔件上来局部、非接触地去除液体的缺点是液滴中的充足的残余液体可能仍会残留在基底上的沉积物上,从而干扰随后对微生物沉积物的分析,因此可能需要采取进一步的纯化措施。
因此,需要从覆盖的液体液滴中充分完全地分离出单独的微生物沉积物,同时确保在该分离过程中尽可能少的微生物沉积物材料与液体一起被除去。通过阅读以下公开内容,本发明要实现的其他目的对于本领域技术人员而言是立即清楚的。
发明内容
本发明涉及一种在光谱样品支持物上制备样品的方法,包括以下步骤:(i)提供样品支持物,该样品支持物包括单个液体液滴的排列,每个液体液滴中都包裹有微生物沉积物,(ii)提供由吸收性材料制成的板,(iii)将板垂直降低到样品支持物上,以使微生物沉积物与板接触,从而使液体液滴吸收到吸收性材料中,(iv)提起局部富含液滴液体的板使其离开样品支持物,从而暴露出耗尽液体的微生物沉积物,并且之后根据需要,使可能残留在微生物的细胞沉积物上的任何残留液体膜干燥,(v)准备暴露的微生物沉积物以用于光谱测定,例如随后的红外光谱测定或质谱测定。
在不同的实施方案中,板可以包含溶胀的吸收性材料并且可以降低直到其与样品支持物之间仅有小的间隙,使得液滴的上清液与板接触并被板吸收,由此吸收性材料在已经吸收液滴液体的位置处发生局部溶胀,从而与微生物沉积物接触。已经发现,由材料具有合适的溶胀特性的吸水纸板(blotting board)制成的板特别适用于该形式。吸水纸板特别出名的是对湿气敏感的艺术品的修复和存档,它基本上是一种中性、无酸、吸水且低绒毛的羊毛板,棉纤维比例高,可占超过50%、尤其是超过60%。
最初不允许接触的间隙尺寸的选择可以非常简单地通过将涂覆有容易擦掉的颜色的板降低到样品支持物上来测试。如果在再次提起板后在样品支持物上发现颜色痕迹,则间隙未大到足以防止直接接触,因此必须重新调整。因此,很容易通过实验设置毫米级的间隙,当发生局部液体吸收直到与微生物沉积物接触时,可以通过吸收性材料(例如吸水纸板)的溶胀反应来封闭这些间隙。
在各种可替代实施方案中,可将板直接放置在样品支持物上并轻轻向下压,以使排列和板全面接触。
本发明基于出乎意料且因此令人吃惊的发现,即可以将由吸收性材料制成的低绒毛平板(flat and low-lint plate)降低到平坦的样品支持物并使之接触,或者甚至将板部分地放置到样品支持物上,并将其略微向下压,以使毛细管力将液滴的几乎所有液体上清液吸入吸收性材料中,而隐藏在下面的微生物沉积物不会被消耗到这样的程度,即,随后对样品支持物进行光谱分析时由于生物材料太少而无法提供有用的结果。尽管将板子(垂直)提起后仍会留有液体膜,但该液体膜太稀薄,以至于在几秒钟内变干。
优选地,样品支持物和/或板相对于彼此以弹簧方式安装,从而当向下压板时施加的过大压力被弹簧吸收。弹簧的弹性变形可确保板在样品支持物上施加恒定的接触压力。压力可以通过设置合适的弹簧常数来适应要求,并进行调节,以使微生物沉积物不会因受到施加的压力而受到不利影响。
在不同实施方案中,面向样品支持物的板表面可以在降低之前被稍微润湿,例如用去离子水或甲醇/乙醇润湿,以加速从液滴吸收液体。位于板周边的纤维的轻微润湿和表面润湿特别有利于在沉积于样品支持物上的液滴和板的纤维基质之间形成液体接触或液桥,从而使位于周边的纤维织物从一开始就从表面进行润湿,并且当板与样品支持物上的液滴发生液体接触时,不必先进行耗时的润湿或浸泡。
优选地,板具有由纤维素和/或植物纤维,特别是棉花、硝化纤维素、尼龙、玻璃纤维、聚偏二氟乙烯或其组合制成的吸收性纤维。作为特别合适的材料的吸水纸板可以包含例如60%的棉纤维和40%的漂白的(pH中性且无酸)纤维素的混合物。
液滴的常规体积可以对应于大约1微升至12微升。这些均匀液滴形式的体积大致对应于整个样品支持物的表面的两或三毫米的直径。
在红外光谱或质谱分析中,特别是例如MALDI飞行时间分析中,可以将48、96、384或1536个单独的液滴以预定(和规则)模式排列在样品支持物上。当然,也可以用优选的液滴点,例如在疏液环境中的亲液圆形区域标记样品支持物。然后,液滴的直径将根据亲液圆形区域的尺寸进行自我调整,就像AnchorChipTM类型的标准靶板(anchor plate)那样。
液滴尤其可以主要包含液体,其用于红外光谱法或质谱法的样品制备方法和样品处理方法中。液滴可以包含例如营养液,而沉积的材料可以包含微生物,这些微生物已经在营养液的这些液滴中培养然后沉积。从沉积的微生物中分离营养液可特别用作根据物种/亚种或微生物的不同表征(例如快速确定微生物对抗菌物质(如抗生素和抗真菌剂)的耐药性/敏感性)进行红外光谱或质谱鉴定(例如,借助IR透射光谱或MALDI飞行时间质谱)的样品的制备步骤。
如有必要,可将抗微生物物质添加到营养液中,例如,通过在富含抗生素或抗真菌剂的营养液中进行培育以确定最小抑菌浓度。
附加地或可选地,液滴可以包含用于样品制备方法的洗涤液(例如水溶液或纯去离子水)或其他液体工作介质。例如,被预干燥的微生物沉积物或被干燥的基质物质和微生物沉积物覆盖的样品点的洗涤液可以被板吸收。优选选择洗涤液,以使基质物质或具有嵌入的样品晶体的基质晶格不被(例如)用于原位脱盐的α-氰基-4-羟基肉桂酸亲和制剂溶解(参加Gobom等,Anal.Chem.73,2001,434-438)。
在各种实施方案中,可以使用金属、玻璃或陶瓷板作为光谱样品支持物。可能的实施方案尤其是抛光的不锈钢板或其改进型式,例如所谓的靶板(AnchorChipTM,BrukerDaltonik公司),其包含在不锈钢基材上彼此分离的交替的亲液和疏液表面。也可以使用玻璃板作为用于透射测定的红外光谱样本载玻片。样品支持物可以重复使用或一次性使用。
用于通过基质辅助激光解吸(MALDI)进行电离的常规、标准化的样品支持物可以包含48、96、384或1536个样品点的规则排列。样品支持物和吸收性材料板的尺寸可以分别为127.76mm(长)×85.48mm(宽),相当于微量滴定板,其厚度大约为2至5毫米,只要它们是合适的即可。然而,样品点的排列以及因此样品支持物上的液滴的排列覆盖的面积小于样品支持物本身的面积。
还可以提供用于板的框架或用于样品支持物的导向装置,将板插入其中,在将板降低至液滴排列上时,框架有助于板的导向和对齐,如在EP 3 376 202 A1中所描述的,其全部内容一起通过引用并入本文。框架可以例如永久性地连接到板上,特别是通过折叠吸收性材料的突出的、按尺寸裁剪的边界,然后用聚合物浸渍(聚合物然后固化)而将框架永久地连接到板上。也可以使用合适的聚合物将框架注塑到板的外周上。
板优选地由坚硬且低绒的织物制成。植物纤维和/或纤维素对此是理想的。板可以(例如)具有纤维材料,例如棉纤维、硝化纤维、尼龙、玻璃纤维、聚偏二氟乙烯或它们的组合,例如包含大约60%棉纤维和40%漂白(pH中性且不含酸)纤维素的吸水纸板。刚性材料特别适合于吸收液体的自动化方法。可以通过专门改装过的处理设备(例如机械手)轻松地从商店中取出这些刚性板,然后再将其固定到改装过的支架中,转移到液体吸收步骤,然后放到所需的位置,并在必要时处置。
关于板的生产,可以提供连续长度的吸收性材料,然后根据所需的外部轮廓切下板件。为了在微生物实验室中使用,适宜的是选择板的外部尺寸,以使吸收的液体以及其仍可能包含的任何悬浮的微生物不能渗透到板的边缘或顶表面,使得在板被液体局部富集后,所述板可以在没有任何污染风险的情况下被抓紧和移动。

通过参考以下图示可以更好地理解本发明。图示中的元件不一定按比例显示,而是主要用于图示(大体上示意性地)本发明的原理。在图示中,相同的附图标记在不同的视图中指示相应的元件。
图1A至图1E示出了一种方法的示例性实施方案的示意图,其中将吸收性材料的板放置在带有液滴的样品支持物上。
图2A至图2D示意性地示出了该方法的一种变型,其中,弹簧吸收了将板压在样品支持物上时施加的过大压力,从而确保了压力的均匀施加。
图3是方法的示例实施方案的示意图,其中由溶胀的材料制成的板朝着样品支持物降低,直到板与样品支持物之间仅有小的间隙。
图4描绘了从肠细菌沉积物中去除培养上清液后,通过MALDI飞行时间质谱仪获得的光谱,当使用非接触式方法时,如顶部图A所示,例如,从E.A.Idelevich等人,Clin.Microbiol.Infect.2018(7):738-743所知的那样,其中将纤维素滤纸用作吸收性材料,当使用根据本发明原理的接触方法时,如底部图B所示;纵轴:强度,以任意单位表示;横轴:质荷比m/z。
具体实施方式
尽管已经参考多个实施方案说明和解释了本发明,但是本领域技术人员将认识到,可以在不背离所附专利权利要求书中定义的技术教导的范围内对其形式和细节进行各种改变。
图1A描绘了样品支持物(4)上一行8个的液滴(2)的排列,其可以对应于适于红外光谱的MALDI样品支持物板或样本载玻板。吸收性材料板(6)位于样品支持物(4)上方。板(6)在垂直方向上朝着样品支持物(4)缓慢移动,由此液滴(2)从前进的一定的下降点开始与板(6)的吸收性材料接触,从而液滴液体通过毛细管力被板材(6)吸收,如图1B所示。与先前的公开相反,如图1C所示,该下降移动可以以使板(6)搁置在样品支持物(4)上而结束。因此,可以在板(6)和液滴排列(2)之间产生全面接触,从而在微生物沉积物上留下较少的液滴(2)的残余液体,从而便于后续的沉积物的光谱测定。
令人惊讶地发现,先前假定的“平板效应”明显不如人们所担忧的那样明显。术语“平板效应”是指当许多微生物细胞与板(6)的吸收性材料直接接触时,它们会通过转移到板上而从沉积物中去除,这样使后续测定变得更加困难,因为减少了可用的生物材料的量,这就是为什么在较早的公开内容中非接触去除液体是有利的原因。申请人在室内进行了实验,实验中对于用于吸收液体的板进行了培养,结果表明,在板表面上已吸收液体的那些部位可能会形成微生物菌落。然而,“通过平板去除”的生物材料的数量如此之小,与预期相反,以至于随后对样品支持物(4)上残留的材料进行的光谱分析未受到不利影响。
对于这些有利的特性,尚无法找到全面发展的科学解释,但可以认为,板材料的纤维与微生物之间的相互作用非常小,以至于二者均无法很好地粘附在一起,从而抵消了它们之间的平板效应。此外,与先前公开中推荐的横向移动的情况不同,据认为,将板移动限制为仅垂直向下下降到样品支持物上,同时避免横向剪切移动,也可以减少并可能消除微生物生物膜被涂抹的风险,涂抹可能导致细胞从样品支持物转移到板材料上。
接触后,液滴液体会在很短的时间内被吸收,通常只有几分之一秒,必要时可通过稍微预湿板材料来帮助吸收。之后,可以再次提起局部富集液体的板(6),并将其移走,如图1D所示。吸收的液体牢固地保留在板(6)的毛细管基质中,因此没有在垂直提起板时会再次滴落从而污染样品支持物(4)的危险。相反,这是一种非常安全可靠的去除液体的方法。通常将部分浸泡的板(6)作为可消耗材料处置,这对于微生物学应用是有利的。但是,合适的话,它也可以洗后再使用。
如上文所述,封闭在液滴(2)中的微生物沉积物不会通过毛细管力从而被液体的温和吸收去除,而是大部分集中保留在样品支持物(4)的表面上液滴(2)沉积的位置。因此,从中已经去除了液体的沉积材料可用于进一步处理,例如用于红外光谱的样品制备,通过基质辅助激光解吸进行的电离或类似的处理步骤,以工作介质(例如MALDI的基质物质)的移液指示,如图1E中的(8)所示。
板边缘和最接近的液滴之间的距离优选至少对应于样品支持物的表面上的液滴半径。因为实验室人员可能会抓住板的边缘,所以为了在液体被吸收到吸收性材料中时避免将生物危害性成分拉到边缘附近,对应于液滴直径是更安全的。对于板的最小厚度,应遵守相似的安全距离,以防止吸收的液体渗透到背离液体吸收的板表面。
在上述方法的一种变型中,板(16)在朝着样品支持物(14)下降并与之接触后,也可以轻轻地压在样品支持物上,以确保板(16)和排列(12)或样品支持物表面之间完全接触,这使得可以最完全地吸收液体。为了防止施加过大的压力,尤其是用手(例如,经验不足的实验室工作人员)将其压下时,可能会对微生物沉积物产生不利影响,板(16)和样品支持物(14)可以是相对于彼此以弹簧形式安装,如图2A至图2D中的螺旋弹簧(18)示意性地所示。应该理解的是,样品支持物(14)、板(16)或这两个元件可以弹簧形式安装以实现所需的效果,但是仅样品支持物(14)的弹簧示出在了图2所示的顺序中。
如图2A所示,将由吸收性材料制成的板(16)朝样品支持物(14)放下,该样品支持物承载着液体液滴(12)。在该状态下,弹簧(18)不被压缩。一旦板(16)与液滴上清液接触,就将液滴的液体吸收到板(16)的吸收性材料中,板(16)与液滴上清液的接触甚至发生在板与样品支持物表面的接触之前,如图2B所示。在该状态下,弹簧(18)仍然没有被压缩。一旦板(16)接触到样品支持物(14)时,降低运动的阻力就变得明显。现在可以施加压力以确保板(16)与样品支持物表面或排列(12)之间的均匀接触,尤其是全面接触,并尽可能吸收残余液体,如图2C所示。现在,弹簧(18)根据所施加的压力N和弹簧常数而被压缩,从而防止过大的压力施加在微生物沉积物上。在几分之一秒的时间间隔后,当液体已被吸收到板材料中时,板(16)可以再次被垂直提起,从而暴露出微生物沉积物,这些微生物沉积物现在已耗尽液体并且可以用于在样品支持物(14)进行进一步处理,图2D所示。然后,弹簧(18)在此过程中再次松弛。
应当理解,来自前述示例的螺旋弹簧(18)仅用于说明缓冲原理。在本文提出的方法中,也可以使用其他缓冲机制,只要本领域的专业人员认为有利即可。
上面已经描述了吸收板(26)与样品支持物(24)直接接触的实施方案。但是,也可以在板(26)和样品支持物(24)之间保持微小的间隙,例如,当板由溶胀的吸收性材料制成时,通过将板放置在侧缘(28)上来实现,图3。当板表面与液滴上清液接触时,液体会局部吸收到板(26)中,这会导致板中的体积增加(“溶胀”),从而使溶胀的材料变得与微生物沉积物的温和的接触,请参见图3的放大部分。已经发现,这种温和的接触不会对沉积物产生不利影响,特别是只有极少数微生物细胞转移到板表面上(“平板效应”),但与以前发布的技术相比,它确实可以更完全地吸收液滴的液体,以前发布的技术由于使用了非溶胀性材料,因此在有间隙下严格地操作。因此,减少了局部饱和的板(26)被(垂直)提起之后残留在样品支持物(24)上的残余液体的量,从而有利于随后的光谱测定。
为了简化板的处理,可以将其插入或夹到框架中。例如,如EP 3 376 202 A1中所描述的那样,可以将框架的尺寸确定为使得其围绕样品支持物齐平地装配,液滴阵列位于样品支持物上。它既可以设计成可与饱和板一起丢弃的一次性物品,也可以设计成可清洗、可重复使用的物品。可能的实施方案包括具有阶梯状内部轮廓的框架,可将板放置在框架上以摩擦锁定。如果框架围绕样品支持物的外轮廓向下滑动,则从特定点开始与液体建立接触。框架还具有确保板相对于液滴的阵列可靠的对准并引导的优点。
在可选的实施方案中,原则上同样如在EP 3 376 202 A1中描述的,框架可以牢固地固定在板上。可以将吸收性材料的突出的、按尺寸裁剪的边界折叠起来,然后例如用聚合物浸渍,聚合物然后固化以确保稳定性和刚性(单件变体)。必要时,也可以使用合适的聚合物将框架在外周边注塑到板上。
从EP 3 376 202 A1中同样已知一种技术,通过该技术,将承载有液滴阵列的样品支持物插入到导向器中,该导向器将样品支持物的全部边都围住。然后,板的尺寸可以与样品支持物相似,并从顶部导向器开口缓慢向下滑动到样品支持物上。导向器壁上的抓握凹口可以使插入样品支持物和板并再次将它们抬起变得更加容易。
图4描绘了测量数据,这些数据说明了本文描述的方法的优点。根据本公开的原理,底部图B,(几乎)完全去除了直接在MALDI样品支持物上的液滴上清液,这里是细菌培育后的培养物或营养液上清液,因此也去除了会干扰后续测定的污染物(例如,营养介质或盐的成分),从而以更高的离子信号强度和改善的信噪比的形式带来更好的质量测量数据。
关于所分析的革兰氏阴性细菌的抗生素敏感性(头孢菌素抗生素:头孢他啶)的测定,与常规营养液微稀释法(培育20小时)相比,根据本发明的接触方法和根据现有技术的非接触方法在95%/100%/100%和93%/93%/92%分别培育4小时/4.5小时/5小时后,产生了一致的结果。这突出了这些工序的巨大优势。
除了示例性描述的实施方案之外,还可以想到本发明的其他实施方案。根据本公开的知识,本领域技术人员可以容易地设计用于使用解吸电离方法进行红外光谱或质谱测定的其他有利的样品制备方法,这些方法将被专利权利要求的保护范围覆盖,包括任何的等同形式。
Claims (13)
1.一种在光谱样品支持物上制备样品的方法,包括以下步骤:
-提供样品支持物,该样品支持物包括单个液体液滴的排列,每个液体液滴中都包裹有微生物沉积物,
-提供由吸收性材料制成的板,
-将所述板垂直降低到所述样品支持物上,以使所述微生物沉积物与所述板接触,从而使所述液体液滴吸收到所述吸收性材料中,
-提起局部富含液滴液体的所述板使其离开所述样品支持物,从而暴露出耗尽液体的微生物沉积物,以及
-准备暴露的微生物沉积物以用于光谱测定。
2.根据权利要求1所述的方法,其中,所述板包含溶胀的吸收性材料,并且将所述板降低直到其与所述样品支持物之间仅有小的间隙,使得所述液滴的上清液与所述板接触并被所述板吸收,由此所述吸收性材料在液滴液体被吸收的位置处发生局部溶胀,并与所述微生物沉积物接触。
3.根据权利要求1所述的方法,其中,将所述板直接放置在所述样品支持物上,并轻轻地压在所述样品支持物上,以使所述排列和所述板全面接触。
4.根据权利要求3所述的方法,其中,所述样品支持物和/或所述板相对于彼此以弹簧形式安装,使得当所述板被向下按压时,弹簧吸收施加的过大压力。
5.根据权利要求1所述的方法,其中,在降低所述板之前,将所述板的面向所述样品支持物的表面稍微润湿,以加速从所述液滴中吸收液体。
6.根据权利要求1所述的方法,其中,所述板具有纤维素的吸收性纤维。
7.根据权利要求1所述的方法,其中,液滴的体积等于一至十二微升。
8.根据权利要求1所述的方法,其中,所述微生物沉积物存在于营养液或洗涤液的液体液滴中。
9.根据权利要求8所述的方法,其中,将抗微生物物质添加到所述营养液中。
10.根据权利要求1所述的方法,其中,在准备所述暴露的微生物沉积物之后,对所述样品支持物进行红外测定或质谱测定。
11.根据权利要求1所述的方法,其中,在提起所述板与所述准备之间存在短暂的等待,以使微生物细胞沉积物上的任何残留的残留液膜干燥。
12.根据权利要求1所述的方法,其中,所述板具有纤维素的吸收性纤维和植物纤维。
13.根据权利要求1所述的方法,其中,所述板具有植物纤维。
