Abstract
本发明涉及一种抑制人结直肠癌细胞SW480增殖的玫瑰花提取物及其应用,属于食品加工技术领域。该玫瑰花提取物经如下步骤得到:新鲜食用玫瑰花在40‑80℃下烘干,将其超微粉碎,按粉碎后的玫瑰花:沸水=1g:1‑200mL的比例浸提30分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液经传统的浓缩、干燥后即得。细胞试验表明,此玫瑰花提取物能显著抑制人结直肠癌细胞SW480的增殖,能作为爱吃红肉人士的健康伴侣,应用于食品和保健品中。
Description
技术领域
本发明属于食品加工技术领域,具体的说,涉及一种抑制人结直肠癌细胞SW480增殖的玫瑰花提取物及其应用。
背景技术
结直肠癌是世界常见的恶性肿瘤之一,据国际癌症研究理事会最新统计,仅2013年就新增结直肠癌患者102480例,因结直肠癌而死亡的人数达50 830例。30年来,我国结直肠癌发病率年均上升3%~4%。我国结直肠癌发病率和死亡例数分别占全世界发病和死亡总例数的18.6%和20.1%,均居第1位。尽管当前结肠癌治疗方面涌出一些新的方法,如西妥昔单抗、贝伐单抗等分子靶向药物,但总体上来看结肠癌治疗效果仍然较差,尚无满意的非侵入性治疗方案。
现代研究证明,玫瑰花能够抗氧化、抗衰老及降血糖,能够平衡及强化胃肠道功能,调节消化液的分泌,促进消化和吸收;对消化不良及胃酸分泌过多等慢性疾患有一定的调节和改善作用;对情绪紧张、压抑等引起的胃痛有明显的改善功能。本发明以玫瑰花为原料,制备一种具有抑制人结直肠癌细胞SW480增殖的玫瑰花提取物。经文献检索,未见与本发明相同的公开报道。

本发明是目的在于提供一种抑制人结直肠癌细胞SW480增殖的玫瑰花提取物及其应用,
为实现上述目的,本发明是通过如下技术方案实现的:
所述的玫瑰花提取物,采用水浸提法制备而得,具体包括以下步骤:新鲜食用玫瑰花在40-80℃下烘干,将其超微粉碎,按粉碎后的玫瑰花:沸水=1g:1-200mL的比例浸提1-60分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液经浓缩、干燥后,即得玫瑰花提取物。
进一步优选,所述干燥采用真空冷冻干燥。
进一步优选,所述干燥采用40-80℃条件下烘干处理。
玫瑰花提取物在抑制人结直肠癌细胞SW480增殖产品中的应用,所述玫瑰花提取物为冻干粉或烘干粉。
玫瑰花提取物在抑制人结直肠癌细胞SW480增殖产品中的应用,所述玫瑰花提取物是利用上述制备方法获得。

本发明的有益效果:该玫瑰花提取物富含多酚类、黄酮类、多糖,能抑制人结直肠癌细胞SW480的增殖,可应用于食品和保健品中。
具体实施方式
为了使本发明的目的、技术方案和有益效果更加清楚,下面将对本发明的优选实施例进行详细的说明,以方便技术人员理解。
【实施例1】
玫瑰花提取物,采用水浸提法制备而得,具体步骤如下:新鲜食用玫瑰花在65℃下烘干后将其超微粉碎,按粉碎后的玫瑰花:沸水=1g:10mL的比例浸提30分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液减压浓缩后,经真空冷冻干燥,得到玫瑰花提取物(冻干)。
【实施例2】
玫瑰花提取物,采用水浸提法制备而得,具体步骤如下:新鲜食用玫瑰花在65℃下烘干后将其超微粉碎,按粉碎后的玫瑰花:沸水=1g:10mL的比例浸提30分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液减压浓缩后,浓缩液在65℃下烘干得到玫瑰花提取物(烘干)。
实施分析
将实施例1、2得到的玫瑰花提取物进行细胞试验,用MTT法检测玫瑰花提取物对人结直肠癌细胞SW480增殖的影响,其结果如下:采用不同浓度的玫瑰花提取物(冻干)培养SW480细胞,随着培养时间的延长,细胞相对存活率降低;培养时间达72h,玫瑰花提取物(冻干)浓度≥400mg/l时,SW480细胞存活率显著降低,表明其能显著抑制人结直肠癌细胞SW480增殖。采用不同浓度的玫瑰花提取物(烘干)培养SW480细胞,培养时间仅为24h时,细胞存活率即显著降低。培养时间为24h和48h时,不同浓度玫瑰花提取物(烘干)对SW480细胞的存活率无显著性差异;培养时间为72h时,玫瑰花提取物(烘干)浓度越高,则细胞存活率越低,显示出较好的剂量效应关系。表1表明,玫瑰花提取物(烘干)对人结直肠癌细胞SW480增殖的抑制优于玫瑰花提取物(冻干)。

研究结果表明:玫瑰花提取物能抑制人结直肠癌细胞SW480的增殖,玫瑰花提取物(烘干)对人结直肠癌细胞SW480增殖的抑制优于玫瑰花提取物(冻干)。玫瑰花提取物能直接加工成粉剂、胶囊、含片及饮料等,使用方便,便于携带;也能够作为添加剂用于食品中。

1.一种玫瑰花提取物,其特征在于:其制备方法包括以下步骤:新鲜食用玫瑰花在40-80℃下烘干,将其超微粉碎,按粉碎后的玫瑰花:沸水=1g:1-200mL的比例浸提1-60分钟,过滤得到滤液,重复上述浸提步骤2次,合并滤液,滤液经浓缩、干燥后,即得玫瑰花提取物。
2.根据权利要求1所述的一种玫瑰花提取物,其特征在于:所述干燥采用真空冷冻干燥。
