本发明公开了一种高效制备山奈提取物的方法。方法步骤如下:取土木香粉碎50‑80目,加入超临界CO2萃取釜,设置温度50~60℃、压力15~30MPa,通入液态CO2,流量为2~3ml/min/g原料,同时通入甲醇做夹带剂,流量为0.1~0.4ml/min/g,萃取60~100min,解析得萃取物;上述萃取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙酸乙酯萃取,萃取液浓缩结晶,结晶物再用乙醇溶解,采用高速逆流色谱法分离,根据色谱收集流分,减压浓缩干燥即得。采用本方法制备山奈提取物,制备工艺简单,污染小,易于实现工业化生产。

技术领域
本发明属于生物技术领域,特别是涉及一种制备山奈提取物的方法。
背景技术
山柰,别名:沙姜、三柰、山辣, 多年生宿根草本,为姜科山柰属植物山柰(Kaempferia galanga Linn.) 的根茎,根茎块状,单生或数枚连接,淡绿色或绿白色,芳香;分布于广东、广西、云南、台湾等省区,栽培。山柰根茎为芳香健胃剂,有散寒、祛湿、温脾胃、辟恶气的功效,亦可作调味香料。在民间,山柰一直作为药食两用的植物使用,其根茎、叶常用于白切鸡、白斩鸡的食用佐料。据中国药典记载,其味辛,性温,有行气温中、消食、止痛的作用 ;用于胸膈胀满,脘腹冷痛,饮食不消。
发明内容
本发明的目的是克服现有技术的缺陷和不足,提供一种高效、环保的制备山奈提取物方法。
为了解决上述技术问题,本发明采用以下技术方案来实现的:
一种制备山奈提取物的方法,其特征在于以下步骤:
a.取土木香粉碎50-80目,加入超临界CO2萃取釜,设置温度50~60℃、压力15~30MPa,通入液态CO2,流量为2~3ml/min/g原料,同时通入甲醇做夹带剂,流量为0.1~0.4ml/min/g,萃取60~100min,解析得萃取物;
b.上述萃取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙酸乙酯萃取,萃取液浓缩结晶,结晶物再用乙醇溶解,采用高速逆流色谱法分离,根据色谱收集流分,减压浓缩干燥即得。
步骤b中所述的高速逆流色谱分离为以乙腈、甲醇、磷酸氢二钠溶液(PH6.5)为溶剂系统,按3-5:1-4:3-7混合,取上相做固定相、下相为流动相。
本方法的优点在于采用超临界CO2萃取,提取效率高,整个工艺操作简单,产品收率高,而且易实现工业化生产。
下面将结合具体实施方式对本发明作进一步阐述,但本发明要求保护的范围并不局限于下列实施方式。
具体实施方式:
实施例1:
取土木香粉碎50目,加入超临界CO2萃取釜,设置温度50℃、压力15MPa,通入液态CO2,流量为2ml/min/g原料,同时通入甲醇做夹带剂,流量为0.1ml/min/g,萃取60min,解析得萃取物;上述萃取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙酸乙酯萃取,萃取液浓缩结晶,结晶物再用乙醇溶解,采用高速逆流色谱法分离,以乙腈、甲醇、磷酸氢二钠溶液(PH6.5)为溶剂系统,按3:1:3混合,取上相做固定相、下相为流动相根据色谱收集流分,减压浓缩干燥即得。
实施例2:
取土木香粉碎80目,加入超临界CO2萃取釜,设置温度60℃、压力30MPa,通入液态CO2,流量为3ml/min/g原料,同时通入甲醇做夹带剂,流量为0.4ml/min/g,萃取100min,解析得萃取物;上述萃取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙酸乙酯萃取,萃取液浓缩结晶,结晶物再用乙醇溶解,采用高速逆流色谱法分离,以乙腈、甲醇、磷酸氢二钠溶液(PH6.5)为溶剂系统,按5: 4: 7混合,取上相做固定相、下相为流动相根据色谱收集流分,减压浓缩干燥即得。
实施例3:
取土木香粉碎70目,加入超临界CO2萃取釜,设置温度60℃、压力20MPa,通入液态CO2,流量为2ml/min/g原料,同时通入甲醇做夹带剂,流量为0.3ml/min/g,萃取80min,解析得萃取物;上述萃取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙酸乙酯萃取,萃取液浓缩结晶,结晶物再用乙醇溶解,采用高速逆流色谱法分离,以乙腈、甲醇、磷酸氢二钠溶液(PH6.5)为溶剂系统,按4:3:5混合,取上相做固定相、下相为流动相根据色谱收集流分,减压浓缩干燥即得。

1.一种高效制备山奈提取物的方法,其特征在于以下步骤:
a.取土木香粉碎50-80目,加入超临界CO2萃取釜,设置温度50~60℃、压力15~30MPa,通入液态CO2,流量为2~3ml/min/g原料,同时通入甲醇做夹带剂,流量为0.1~0.4ml/min/g,萃取60~100min,解析得萃取物;
b.上述萃取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙酸乙酯萃取,萃取液浓缩结晶,结晶物再用乙醇溶解,采用高速逆流色谱法分离,根据色谱收集流分,减压浓缩干燥即得。
2.根据权利要求1所述的制备山奈提取物的方法,其特征在于步骤b中所述的高速逆流色谱分离为以乙腈、甲醇、磷酸氢二钠溶液(PH6.5)为溶剂系统,按3-5:1-4:3-7混合,取上相做固定相、下相为流动相。
