Description
Technical Field
The invention belongs to the technical field of additives, and particularly relates to a puppy colostrum substitute additive, a preparation method and application thereof.
Background
Currently, the closest prior art: at present, most of commercially available puppy colostrum substitutes are stacked of nutritional ingredients, and the characteristics of the nutritional ingredients required by the growth of the puppies and intestinal flora thereof are not completely considered, such as the goat milk powder, the ingredients show high nutritional content, but the ingredients contain nutritional ingredients which are not easy to absorb by the puppies, the situations of dyspepsia, abdominal distension, diarrhea and the like are easy to occur after eating, and some puppies are weak or even die due to unbalanced nutrition. Chitosan oligosaccharides are demonstrated to have a higher cell transduction potential and are completely absorbable through the intestinal epithelium due to their cationic globules exposed on shorter N-glucosamine units. Therefore, the chitosan oligosaccharide can be used as a carrier of macromolecular nutrient substances to promote the absorption of nutrient components, and the chitosan oligosaccharide can improve the disease resistance of the puppies, regulate intestinal microorganisms and help to quickly establish normal flora. The additive comprises various chemical additives such as feed additives, food additives, concrete additives, engine oil additives and the like. The food additive refers to a chemically synthesized or natural substance added to food for improving the quality and color, flavor, taste of food, and for the need of preservation and processing. The food additive is not food, and has no nutritive value, but must meet the concept defined above, i.e. has no influence on the nutritive value of food, and has the effects of preventing food spoilage, enhancing food organoleptic properties or improving food quality.
The colostrum substitute is needed to feed the puppies when the newborn puppies cannot feed due to insufficient milk production of the bitches, mastitis and other reasons. The normal intestinal flora of the newborn puppies is not established, the digestive ability is weak, the immunity is low, the requirement on nutrition is high, and the requirement on food intake is high. Most of the existing colostrum substitutes have nutrient components which are not easy to absorb. By adding the chitosan oligosaccharide, the digestive ability and the body immunity of the puppies can be enhanced, and nutrient substances can be absorbed more easily, so that the whole function of the colostrum substitute is exerted. The existing commercially available puppy colostrum substitute mostly has the defects of dyspepsia, abdominal distension, diarrhea and the like, and some puppies are weak or even die due to unbalanced nutrition.
In summary, the problems of the prior art are as follows: the existing colostrum substitute for puppies is easy to cause dyspepsia, abdominal distension and diarrhea.
The difficulty of solving the technical problems is as follows: the digestive system of animal body is very complex, and the working mechanism is difficult to be completely found. The digestive system of the dogs as omnivorous animals is more abnormal and complex, and newborn puppies are not completely adapted to the external environment, are more easily attacked by pathogens and have higher requirements on nutrient substances. The colostrum of the female dog contains rich nutrient substances, and can improve the disease resistance of the puppy. It is desirable that colostrum substitutes contain not only components that are easily absorbed and have a high nutritional value, but also certain substances that enhance immunity.
The significance of solving the technical problems is as follows: by adding the chitosan oligosaccharide, the nutrient substances of the colostrum substitute can be absorbed more easily, the body immunity of the puppies is enhanced, and the survival rate of the puppies is greatly improved. The chitosan oligosaccharide is a natural substance extracted from the shells of marine organisms, and the application of the chitosan oligosaccharide realizes the reutilization of resources.

The invention is realized by the following steps that the additive is a puppy colostrum substitute, and the additive is chitosan oligosaccharide; according to the mass ratio, the chitosan oligosaccharide: colostrum substitute | ═ 1: 200.
the invention also aims to provide a preparation method of the additive for the colostrum substitute of the puppy, which comprises the following steps:
firstly, preparing food-grade chitosan oligosaccharide powder;
secondly, sealing and then placing the mixture in steam high-pressure sterilization equipment for 15-20 minutes at the temperature of 121 ℃ and under the pressure of 0.12 MPa; sterilizing under high pressure, oven drying at 56 deg.C, moisture-proof packaging, and storing; the autoclaving is to prevent the pollution in the production, transportation and weighing processes, and to achieve the sterility; the drying is to prevent the chitosan oligosaccharide powder from being agglomerated into blocks by water vapor in the process of autoclaving, so that the effect is not influenced; the storage at 2-8 deg.C is for stabilizing chemical properties of chitosan oligosaccharide.
And thirdly, storing the mixture at the temperature of 2-8 ℃ for later use.
Furthermore, the food-grade chitosan oligosaccharide powder is light yellow or milk yellow powder, has the molecular weight of less than or equal to 1600Da, the content of more than or equal to 90 percent, the granularity of more than or equal to 140 meshes, the water content of less than or equal to 8 percent, the ash content of less than or equal to 0.85 percent, the pH value of 7.0-8.0 and is easy to dissolve in water.
Further, the mixture is dried for 2 to 3 hours at 56 ℃ after being sterilized by high pressure.
The invention also aims to provide an application of the additive in the colostrum substitute of the puppy.
In summary, the advantages and positive effects of the invention are: chitosan oligosaccharide has a low molecular weight, a high degree of deacetylation, a high degree of polymerisation, a low viscosity and complete water solubility, which gives it significant biological properties such as antibacterial, antioxidant, anti-inflammatory and anti-hypertensive, and drug/DNA delivery capabilities. In addition, it shows antidiabetic, antiobesity, anti HIV-1, anti Alzheimer's disease, hypocholesterolemic, calcium absorption and hemostatic effects. In addition, chitosan oligosaccharides proved to have higher cell transduction potential and were completely absorbable through the intestinal epithelium due to their cationic globules exposed to shorter N-glucosamine (N-GLC) units. It is reported in periodicals both at home and abroad. Due to the unique biological properties of the chitosan oligosaccharide, the intestinal digestion function of the puppies can be obviously improved, and the quality of the colostrums substitute of the puppies is improved. The invention uses the chitosan oligosaccharide as the additive of the colostrum substitute of the puppy, and solves the problems of most of the products sold in the market. The immunity of the newborn puppies is low, the newborn puppies are easy to be damaged by pathogens, intestinal flora is not completely established, and the chitosan oligosaccharide has the functions of improving the immunity, inhibiting the growth of cancer cells, promoting the formation of liver and spleen antibodies, promoting the absorption of calcium and mineral substances, proliferating beneficial flora of human bodies such as bifidobacteria, lactic acid bacteria and the like. After the chitosan oligosaccharide is added, the disease resistance of the newborn puppies can be improved, normal intestinal flora can be rapidly established, the absorption of nutrient substances is promoted, and the problems of dyspepsia, abdominal distension, diarrhea and even death caused by feeding most of commercially available puppy formula milk powder are solved.
The invention applies the chitosan oligosaccharide extracted from the marine organism carapace to the colostrum substitute additive of the puppy, fully exerts the advantages of the chitosan oligosaccharide, enhances the feeding effect of the colostrum substitute of the puppy, realizes the reutilization of resources, and meets the requirement of ecological development. The invention improves the survival rate of the puppies, reduces the diarrhea rate of the puppies and increases daily gain.

The chitin of the invention is an important natural high molecular polysaccharide, and the content of the chitin is second to that of cellulose in nature. Chitin exists in its natural state as ordered crystalline fibers and is widely found in arthropod hulls and in the cell walls of fungi, nematodes, and algae. Chitosan is a product of chitin after being partially deacetylated under the action of alkaline conditions or enzymes, and has various biological activities, but the low solubility of chitosan under neutral or alkaline conditions limits the functional development of chitosan in the fields of food and biomedicine. The chitosan oligosaccharide is an oligomer obtained by enzymatic reaction or chemical degradation of chitosan, has the polymerization degree of 2-20, and is the only alkaline amino oligosaccharide with positive charge in the natural world at present.
1. Acidolysis method
Free amino in chitosan molecules can be combined with hydrogen ions in an acid solution, so that hydrogen bonds among the molecules in the chitosan long chain are broken, and a plurality of molecules with different polymerization degrees are formed, namely the chitosan oligosaccharide.
2. Oxidation process
The oxidation method is a preparation method for forming chitosan oligosaccharide with different polymerization degrees by utilizing free radicals formed by an oxidant in an aqueous solution to break glycosidic bonds of chitosan.
3. Physical method
The physical method is a method for breaking the glycosidic bond of chitosan by radiation, and the main means are microwave, electromagnetic wave, ultrasonic wave and the like. Under certain conditions, the longer the irradiation time, the more chitosan is degraded, and the smaller the relative molecular mass of the oligosaccharide is.
4. Enzymolysis method
The method for degrading chitosan by using enzyme is mainly divided into degradation by using specific enzyme (chitinase and chitosanase) and non-specific enzyme (lipase, pectinase, cellulose and the like).
5. Combined method
The chitosan oligosaccharide is prepared by two or more methods, the obtained product has more products and good activity, the cost is lower than that of a single method, and the method is widely applied to industrial production.
The preparation and storage of the chitosan oligosaccharide additive of the invention are as follows:
(1) food-grade chitosan oligosaccharide powder: the appearance is light yellow or milk yellow powder, the molecular weight is less than or equal to 1600Da, the content is more than or equal to 90 percent, the granularity is more than or equal to 140 meshes, the water content is less than or equal to 8 percent, the ash content is less than or equal to 0.85 percent, the pH value (1 percent aqueous solution) is 7.0-8.0, and the powder is easy to dissolve in water.
(2) The high-pressure sterilization requirement is as follows: sealing, placing in steam autoclave for 15-20 min at 121 deg.C under 0.12 MPa. Autoclaving, oven drying at 56 deg.C (about 2-3 hr), moisture-proof packaging, and storing.
(3) And (3) storage: storing at 2-8 deg.C for use, and the effective period is 60 days.
The use of the chitosan oligosaccharide additive of the invention: the commercially available colostrum substitute for puppies within the valid period of legal production is prepared into directly edible emulsion according to the instruction, 150mg (150mg/kg) of chitosan oligosaccharide is added into each kilogram of emulsion, the mixture is uniformly mixed, the mixture is kept at 25 ℃ for artificial feeding of the puppies, the mixture is prepared immediately before use, the product is used up once within 2.5 hours, and the product is discarded after time out. Feeding with a baby bottle, the number of times, the quantity and the change with the age of the day are carried out according to the specifications of the products sold in the market.
The above description is only for the purpose of illustrating the preferred embodiments of the present invention and is not to be construed as limiting the invention, and any modifications, equivalents and improvements made within the spirit and principle of the present invention are intended to be included within the scope of the present invention.
