Description
1.技术领域
本公开内容总体上涉及腺相关病毒(AAV)载体(vector)及其用途。更具体地,本公开内容涉及载体,所述载体显示出对于某些靶组织,诸如中枢神经系统(CNS)和脂肪组织的特异性向性(specific tropism),并且可以用于转导细胞以用于将感兴趣的基因引入到靶组织中。
2.背景
腺相关病毒(AAV)是目前正被用于基因疗法应用的单链DNA病毒。AAV是细小病毒科(Parvoviridae),依赖病毒属(Dependovirus)的成员。约4.7kb长(1,2)的AAV基因组包含侧接反向末端重复元件(ITR)的两个开放阅读框(ORF)rep和cap(3)。存在11种已知的具有不同的细胞靶和抗原性质的AAV血清型(Wu等人,2006)。最近,已经发现了这些主要AAV血清型的约100种基因组变体(6)。

3.概述

5.详述
5.1重组AAV血清型

对于病毒颗粒的繁殖,细胞通常是允许AAV病毒复制的细胞。可以使用本领域已知的任何合适的细胞,诸如哺乳动物细胞。同样合适的是提供从复制缺陷型辅助病毒中缺失的功能的反式互补包装细胞系,例如293细胞或其他E1A反式互补细胞。
在实施方案中,生产重组病毒颗粒的方法包括在体外向细胞提供(a)载体基因组和(b)AAV rep序列和AAV cap序列,所述载体基因组包括(i)异源核酸和(ii)足以将载体基因组衣壳化为病毒颗粒的包装信号序列(诸如AAV末端重复序列),该AAV rep序列和AAVcap序列足以将载体基因组复制和衣壳化为病毒颗粒。载体基因组核酸和AAV rep序列和AAV cap序列在使得包括载体基因组的重组病毒颗粒被包装在细胞中产生的衣壳内的条件下被提供。

5.2.重组病毒载体的用途

实施例I

AAV滴度比较
每种血清型病毒在五个150mm的板中产生。来自每个板的每个载体储备液的病毒基因组滴度通过实时PCR来确定,并计算每个板中的病毒产率(每个细胞的病毒基因组颗粒,vg/细胞)。
小鼠
十四周龄的雄性C57BL/6小鼠(Charles River Laboratories,Wilmington,MA,USA)以四只为一组,在12h光照/黑暗周期(在1800点时熄灯)下安置,并自由提供食物和水。所有动物的使用通过俄亥俄州立大学动物保护和使用委员会(Ohio State UniversityAnimal Care and Use Committee)批准,并且符合NIH指南。
对纹状体的rAAV注射
小鼠用单剂量的氯胺酮/赛拉嗪(100mg/kg和20mg/kg;i.p.)麻醉,并且放置在Kopf立体定位框架上。纹状体的注射坐标是(从前囟):前-后,+1.0mm;中侧(medio-lateral),±1.7mm;背腹侧,–3.5mm(Franklin and Paxinos,1997)。将1μL AAV载体(1x1013vg/ml)以0.1μL/min的速率,使用附接至Micro4微型注射泵控制器(WorldPrecision Instruments Inc.,Sarasota,USA)的10μL Hamilton注射器双侧递送到背侧海马体和腹侧海马体。手术后监测动物,直到从麻醉中恢复。
免疫组织化学的组织制备
载体注射之后4周,通过戊巴比妥钠过量(20μL,i.p.)处死小鼠,并且用1xPBS,随后是4%PFA经心灌注。在30%蔗糖中冷冻保护之后,用低温恒温器切割40μm的冠状脑切片用于免疫组织化学。
免疫组织化学
将脑切片在含有0.25%Triton X-100的1×PBS(PBST)中冲洗,并且在含有1%血清的PBST中在室温封闭持续1小时。在去除封闭缓冲液之后,将切片与兔抗-NeuN抗体(Abcam,1:500)或山羊抗-GFAP抗体(Santa Cruz Biotechnology,Inc.,1:100)在4℃孵育过夜。第二天,将切片在PBST中彻底地洗涤,并与二级抗体,Cy3-缀合的山羊抗-兔IgG(Jackson ImmunoResearch Laboratories,Inc.,1:250)或驴抗-山羊IgG-TR(Santa CruzBiotechnology,Inc.,1:250)孵育持续3小时。然后冲洗切片,固定在载玻片上,并且在具有荧光封片介质(Vector Laboratories,Inc.,Burlingame,CA)的情况下,盖上盖玻片。
共聚焦显微术
脑切片在共聚焦显微镜(Olympus FluoViewTM FV1000,Tokyo,Japan)上可视化。GFP、Cy3和Texas的荧光使用488nm氩激光和543nm HeNe激光依次激发。使用×40油浸物镜依次采集图像。使用Olympus Viewer生成合并的图像。
立体测量学
脑组织的转导体积使用Stereo Investigator 7(MBF Bioscience,Willeston,VT)中的Cavalieri Estimator进行立体测量定量。显示了包含GFP阳性免疫反应性的每个切片的区域的轮廓,并且将标记放置在100μm的网格尺寸处,以估计每个切片内的转导面积。测量每第12个40μm切片中的面积(根据转基因表达,每个脑测量了10-12个切片),然后取平均值并乘以第一个切片和最后一个切片之间的纵轴距离(rostrocaudal distance),以给出转导体积的估计值。
GFP表达的亮度
每个脑组织中表达的GFP的强度使用Stereo Investigator 7(MBF Bioscience,Willeston,VT)中的收集亮度信息命令(Collect Luminance Information command)来测量。获取每个脑的具有最强荧光的切片的图像,并显示了GFP表达区域的轮廓。测量轮廓内每个像素的亮度,并且然后取平均值。对于每个像素,亮度具有从0至255的范围。黑色像素具有0的亮度,而白色像素具有255的亮度。对于彩色像素,亮度被定义为(0.299*红色)+(0.579*绿色)+(0.114*蓝色)。
统计学分析
来自不同的实验组的平均值使用单因素ANOVA,随后是通过学生t检验的成对比较进行比较。所有统计学分析使用JMP软件(SAS Institute Inc.,Cary,NC,USA)完成,显著性被设置为P<0.05,所有数据被表示为平均值±平均值的标准误差(S.E.M)。
结果
将四种新的灵长类动物来源的杂合重组AAV载体(AAVRec1-4)在小鼠脑中的转导效率与用野生型衣壳(AAV1、AAV8、AAV9)假型化(pseudotyped)的载体进行比较。纹状体内GFP表达组织的体积使用无偏倚的立体测量学方法进行定量。总体上,rAAVRec3和AAV9示出了最广泛的GFP表达,随后是rAAVRec4。(图2;单因素ANOVA,P<0.0001)。rAAVRec1和rAAVRec2产生与AAV1相当的转导体积,且AAV8表现出最小的转导体积。强烈的GFP荧光还在注射了AAV9、rAAVRec3和rAAVRec4的脑的苍白球、丘脑、皮质和丘脑中被观察到。对注射了rAAVRec3的脑的更详细的检查揭示了在对侧未注射的纹状体中、在苍白球中和在黑质中的GFP阳性纤维。此外,GFP阳性细胞在丘脑和皮质中被观察到。这样的皮质和丘脑细胞转导可以通过载体通过皮质纹状体和丘脑纹状体传入的逆向运输发生。转导的皮质和丘脑神经元在远至距离注射部位1mm处被检测到,这一距离被一些人认为太远,无法通过病毒溶液的简单扩散来解释(Aschauer等人,2013)。
有趣的是,最近已经显示在所测试的载体中以最高的效率转导棕色脂肪组织和白色脂肪组织两者的新的血清型rAAVRec2(Liu等人,2014)没有改善靶向脑的转基因递送。相比之下,rAAVRec3、rAAVRec4和AAV9以高效率转导脑,但脂肪组织转导不良。这些工程化的血清型的独特组织向性是扩展当前的AAV载体工具包用于基础研究和临床应用两者的有用特征。
为了比较多种血清型的转基因表达的强度,选择了来自每个脑的具有最强GFP荧光的切片,并且测量亮度并取平均值。rAAVRec3示出了最高的GFP荧光强度,该GFP荧光强度是通过AAV8介导的GFP荧光强度的2倍(图2A)。rAAVRec4介导的转基因表达与AAV9相当。结果表明,使用rAAVRec3载体在靶部位处实现的转基因蛋白表达的最大水平更高。这可能是由于转导细胞内增加的转基因表达或新的杂合重组血清型的更高的转导密度(每mm3转导的细胞)。
为了确定rAAVRec1-4的细胞向性,使用共聚焦显微术来进行使用针对神经元(NeuN)和星形细胞(GFAP)的细胞类型特异性表位的抗体进行的不同神经细胞类型的不同细胞标志物的GFP荧光和免疫荧光的共同定位可视化。对于所测试的所有血清型,大多数GFP阳性细胞与神经元标志物NeuN是免疫反应性的,每个切片仅有2-3个可检测的星形细胞特异性GFAP-阳性细胞(图3),表明rAAVRec 1-4主要转导神经元。如预期的,rAAVRec1-4不改变细胞的向性,这与转导细胞的表型明显取决于所使用的启动子的事实一致(Lawlor等人,2009)。通过AAV载体转导星形细胞可能需要掺入胶质细胞特异性启动子(glial-specific promoter)。此外,脑区域还可以影响不同AAV血清型的细胞向性。例如,Aschauer和同事(2013)最近示出,虽然皮质中的星形细胞在用AAV8转导之后表现出与用AAV6载体转导相比更高的GFP水平,但是在海马体中没有观察到星形细胞转导的这种差异(Aschauer等人,2013)。有趣的是,相同的研究示出AAV8能够转导星形细胞和少突胶质细胞,并且AAV1显示出对小胶质细胞的一些转导。这可能是所采用的不同方法的反映。尽管我们报道了转导模式的更定性的描述(图3),但Aschauer等人定量地评估了不同细胞类型的每个被转导的细胞内的GFP信号强度,因此即使少数细胞具有高信号强度也可能导致高的细胞类型特异性表达。尽管如此,结果清楚地证明了四种rAAVRec载体的向神经性质(neurotropicnature),其中rAAVRec3还证明了对星形细胞的中度向性(图3)。在这些杂合载体的生产期间,注意到尽管由相同的人进行相同的生产方法,不同的载体导致不同的产品产率(通过ANOVA分析的总体差异,P<0.0001)。结果在表1中呈现。

具体地,rAAVRec2和rAAVRec1表现出与其他载体相比的最大产率。尽管rAAVRec3滴度几乎是AAV8的2倍高,但差异没有达到统计学显著性。值得注意的是,尽管AAV9在脑中产生高度有效的转导,但所产生的滴度比rAAVRec3低超过8倍(P<0.001)。增加的产率具有实际关联,因为它转化为相同生产成本的更大的转导体积。
通过在不同AAV血清型之间互换病毒衣壳蛋白序列生成的本发明的rAAV载体可以提供增强的转导效率和更好的生产产率。本发明的杂合载体可以在规避免疫应答中作为用于再施用的第二载体使用。这些杂合载体进一步扩展了当前的AAV工具包,并且是用于神经学研究的有用的生物学工具。
