본 발명은 AAV 전달을 위한 조성물 및 AAV 전달 효율 스크리닝 방법에 관한 것이다. 본 연구는 보건복지부 및 과학기술정보통신부의 재원으로 치매극복연구개발사업단 선정 한국보건산업진흥원의 보건의료기술연구개발사업 지원에 의하여 이루어진 것임 (과제고유번호 : HU20C0452).
AAV는 파보비리다에(Parvoviridae)과 및 데펜도바이러스(Dependovirus) 속에 속하는데, 이것의 구성원은 복제를 촉진하기 위해 아데노바이러스와 같은 헬퍼 바이러스와 공동-감염을 필요로 하며, AAV는 헬퍼 없이 잠재적 감염을 확립한다. 비리온은 2개의 오픈 리딩 프레임: rep 및 cap을 지니는 4.9 kb 단일 가닥 DNA 게놈을 포함하는 25㎚ 20면체 캡시드로 구성된다. 비구조적 rep 유전자는 바이러스 복제를 위해 필수적인 4개의 조절 단백질을 암호화하는 반면, cap은 60-량체 캡시드 껍질로 조립되는 3개의 구조적 단백질(VP1 내지 3)을 암호화할 수 있다. 이 바이러스 캡시드는 세포 표면 수용체 결합, 엔도사이토시스, 세포내 교환 및 핵 내 언패키징(unpackaging)을 포함하는 바이러스 형질도입의 다수의 생물학적 장벽을 극복하는 AAV 벡터의 능력을 매개한다.
이러한 AAV 벡터는 병원성이 없고 면역반응 또한 유도하지 않는 것으로 알려져 있어, 최근 이를 이용한 유전자 치료제 개발이 활발히 진행되고 있다. 다만, 뇌신경계 질환에 AAV 벡터를 적용하기 위해선 혈액뇌장벽(blood brain barrier)을 극복해야 한다.
유전자 치료제의 치료효과를 극대화하기 위해선 AAV 벡터를 정확하게 뇌내 전달해야 하며 뇌전체로 AAV가 분포되어야 뇌전체 광범위한 병변이 확인되는 퇴행성 뇌질환 같은 질환에 적용될 수 있다.
뇌실질 및 뇌실와 같은 뇌로 직접 투여할 수 있는 경로들은 정확하게 원하는 위치에 AAV를 전달할 수 있는 장점이 있지만 투여한 위치 외 뇌 전체에서 AAV 벡터의 효과를 관찰하기엔 부족하며, 뇌로 직접 투여하는 것은 임상적인 적용이 어려움이 있다는 문제점이 있다.
이러한 문제점을 극복하기 위해, 본 발명에서는 AAV 벡터를 이용한 뇌신경계 질환에 유전자 치료제를 적용하기 위한 최적의 투여 경로 및 투여 조건을 확립하고자 하였다.
일 양상은 캡시드 및 바이러스 게놈을 포함하는 아데노-부속 바이러스(AAV) 입자로서, 상기 캡시드가 AAV 입자의 전달 후 혈액 뇌 장벽을 투과하는, 아데노-관련 바이러스(AAV) 입자를 제공하는 것이다.
다른 양상은 AAV 캡시드 및 바이러스 게놈을 포함하는 AAV 입자를 포함하는 약제학적 조성물로서, 상기 AAV 캡시드가 상기 AAV 입자의 전달 후 혈액 뇌 장벽을 투과하는, 약제학적 조성물을 제공하는 것이다.
또 다른 양상은 상기 조성물을 세포에 투여하는 단계를 포함하는, 세포 내 관심 표적 유전자의 발현을 감소시키거나 억제하는 방법을 제공하는 것이다.
또 다른 양상은 상기 조성물을 대상체에게 투여하는 단계; 및 상기 대상체에서 조직면역염색 (immunohistochemistry; IHC)을 통해 AAV의 분포를 확인하는 단계를 포함하는, AAV 벡터 전달 효율 스크리닝 방법을 제공하는 것이다.
본 발명은 캡시드 및 바이러스 게놈을 포함하는 아데노-부속 바이러스(AAV) 입자로서, 상기 캡시드가 AAV 입자의 전달 후 혈액 뇌 장벽을 투과하는, 아데노-부속 바이러스(AAV) 입자를 제공한다.
본 명세서에서 "AAV"는 아데노-부속 바이러스를 의미하며, 바이러스 그 자체 또는 이의 유도체를 지칭하기 위해 사용될 수 있다. 예를 들어, AAV의 모든 하위유형 및 자연적으로 발생하는 형태와 재조합 형태를 포함할 수 있다. 상기 "AAV"는 AAV 1형(AAV1), AAV 2형(AAV2), AAV 3형(AAV3), AAV 4형(AAV4), AAV 5형(AAV5), AAV 6형(AAV6), AAV 7형(AAV7), AAV 8형(AAV8), AAV 9형 (AAV9) 조류 AAV, 소 AAV, 개 AAV, 말 AAV, 영장류 AAV, 비-영장류 AAV 및 양 AAV을 포함할 수 있다. 본 발명의 일 실시예에서, 상기 AAV 벡터는 AAV9일 수 있다.
AAV의 다양한 혈청형의 게놈 서열뿐만 아니라 천연 말단 반복체(terminal repeat: TR)의 서열, Rep 단백질 및 캡시드 서브유닛은 당업계에 공지되어 있다. 이러한 서열은 문헌에서 또는 젠뱅크(GenBank)와 같은 공용 데이터베이스에서 찾을 수 있다. 예를 들어, 젠뱅크(GenBank) 등록번호 NC_002077(AAV-1), AF063497(AAV-1), NC_001401(AAV-2), AF043303(AAV-2), NC_001729(AAV-3), NC_001829(AAV-4), U89790(AAV-4), NC_006152(AAV-5), AF513851(AAV-7), AF513852(AAV-8) 및 NC_006261(AAV-8)을 참조하며; 이것의 개시내용은 AAV 핵산 및 아미노산 서열을 교시하기 위해 본 명세서에 참조로서 포함된다. 또한 예를 들어 문헌[Srivistava et al. 본 발명은 혈액 뇌 장벽 (Blood Brain Barrier; BBB)를 통과하여 광범위한 뇌내 분포를 관찰할 수 있는 AAV 벡터 투여 방법에 관한 것이다. 구체적으로, 본 발명은 AAV 벡터의 광범위한 뇌내 분포를 관찰할 수 있는 최적의 조건 (AAV 벡터투여 용량 및 희생 시기 등등)을 확립하였다.
본 발명에서 "혈액-뇌 장벽((血液腦障壁, Blood-Brain Barrier, BBB) 또는 뇌혈관장벽)"은 뇌와 혈액을 보다 격리시키는 혈관 장벽으로 높은 선택적 투과성을 갖고 있어 뇌를 포함한 중추신경계의 조절기능을 세균 등과 같은 혈액으로 운반될 수 있는 병원체와 혈액 내의 잠재적인 위험 물질로부터 격리시키는 역할을 한다.
본 발명의 일 실시예에서, 상기 AAV의 캡시드 단백질은 서열번호 1로 기재된 아미노산 서열로 이루어진 것인, 아데노-부속 바이러스 입자일 수 있다.
본 발명의 일 실시예에서, 상기 전달은 경동맥내 투여에 의한 것인, 아데노-관련 바이러스 입자일 수 있다.
본 발명의 일 실시예에서, 상기 AAV 벡터는 GFP를 발현하는 것일 수 있다.
또한, 본 발명은 AAV 캡시드 및 바이러스 게놈을 포함하는 AAV 입자를 포함하는 약제학적 조성물로서, 상기 AAV 캡시드가 상기 AAV 입자의 전달 후 혈액 뇌 장벽을 투과하는, 약제학적 조성물을 제공한다.
또한, 본 발명은 상기 조성물을 세포에 투여하는 단계를 포함하는, 세포 내 관심 표적 유전자의 발현을 감소시키거나 억제하는 방법을 제공한다.
본 발명의 일 실시예에서, 상기 세포는 포유동물 세포인, 세포 내 관심 표적 유전자의 발현을 감소시키거나 억제하는 방법일 수 있으며, 구체적으로, 상기 포유동물 세포는 CNS의 세포일 수 있다.
또한, 본 발명은 상기 조성물을 대상체에게 투여하는 단계를 포함하는, 대상체의 CNS 내 관심 단백질의 수준을 감소시키는 방법을 제공한다.
본 발명의 일 실시예에서, 상기 투여 경로는 경동맥내 동맥 투여인, 방법일 수 있다.
본 발명의 일 실시예에서, 상기 AAV 벡터는 뇌실질 및 뇌실와 같은 부위를 통해 뇌로 직접 투여되거나, 경동맥 등의 혈관을 통해 투여될 수 있다. 일 구체예에서, 상기 AAV 벡터는 경동맥을 통해 투여될 수 있다.
내경동맥 (internal carotid artery; ICA) 투여의 장점은 약물을 내경동맥으로 투여하였을 시 대뇌동맥고리 (Circle of Willis)를 통해 뇌 전체로 약물 전달이 가능하다는 것이다.
본 발명에서 경동맥을 통해 AAV 벡터를 투여한 경우, 뇌 내에 전달 효과가 우수한 것을 확인하였으며, 형질도입된 신경세포가 뇌 전체에서 분포한 것을 확인하였다.
또한, 본 발명은 신경계 질환의 치료를 필요로 하는 대상체에서 상기 신경계 질환을 예방, 치료 및/또는 개선하는 방법으로서, 상기 방법이 청구항 4의 약제학적 조성물의 치료적 유효량을 상기 대상체에게 투여하는 단계를 포함하는, 방법을 제공한다.
본 발명의 일 실시예에서, 상기 방법은 상기 AAV 입자를 포함하는 조성물을 경동맥내 동맥 전달에 의해 상기 대상체에 투여하는 방법일 수 있다.
본 발명에서 신경계 질환은 신경변성 질환, 장애 또는 병태 중 하나이며, 예를 들어, 알츠하이머병, 근위축성 측삭경화증, 파킨슨병, 헌팅턴병, 일차 진행형 다발성 경화증; 이차 진행형 다발성 경화증, 피질기저 핵변성(corticobasal degeneration), 레트 증후군(Rett syndrome), 연령 관련 황반 변성 및 망막색소 변성증으로 이루어진 군으로부터 선택된 망막 변성 장애; 전방 허혈성 시신경 병증(anterior ischemic optic neuropathy); 녹내장; 포도막염; 우울증; 외상 연관 스트레스(trauma-associated stress) 또는 외상 후 스트레스 장애, 전두측두엽 치매(frontotemporal dementia), 루이소체 치매(Lewy body dementia), 경도 인지 장애, 후두 피질 위축증(posterior cortical atrophy), 일차 진행형 실어증 또는 진행성 핵상 안근마비(progressive supranuclear palsy)로부터 선택된 하나 이상일 수 있다.
본 발명에서 사용되는 용어, "예방"이란 본 발명에 따른 약학적 조성물의 투여에 의해 신경계 질환을 억제시키거나 발병을 지연시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어, "치료"란 본 발명에 따른 약학적 조성물의 투여에 의해 신경계 질환에 대한 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명에 따른 상기 조성물은 약학적으로 허용 가능한 담체를 더 포함할 수 있다. 상기 약학적으로 허용 가능한 담체는 제제 시에 통상적으로 이용되는 것으로서, 식염수, 멸균수, 링거액, 완충 식염수, 사이클로덱스트린, 덱스트로즈 용액, 말토덱스트린 용액, 글리세롤, 에탄올, 리포좀 등을 포함하지만 이에 한정되지 않으며, 필요에 따라 항산화제, 완충액 등 다른 통상의 첨가제를 더 포함할 수 있다. 또한 희석제, 분산제, 계면활성제, 결합제, 윤활제 등을 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 주입 백과 같은 주입제, 에어로졸 제제와 같은 분무제, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다. 적합한 약학적으로 허용되는 담체 및 제제화에 관해서는 레밍턴의 문헌에 개시되어 있는 방법을 이용하여 각 성분에 따라 바람직하게 제제화할 수 있다. 본 발명의 약학 제제는 제형에 특별한 제한은 없으나 주사제, 주입제, 분무제형, 액상제형 또는 피부 외용제 등으로 제제화할 수 있다.
본 발명의 상기 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여(예를 들어, 정맥 내, 피하, 복강 내 또는 안구를 포함한 국소에 적용)할 수 있으며, 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 시간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다.
본 발명의 상기 조성물은 약학적으로 유효한 양으로 투여할 수 있다. 본 발명에 있어서 "약학적으로 유효한 양"은 의학적 치료 또는 진단에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료 또는 진단하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 질환 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여시간, 투여 경로 및 배출비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명에 따른 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
구체적으로 본 발명의 조성물의 유효량은 환자의 연령, 성별, 상태, 체중, 체내에 활성 성분의 흡수도, 불활성률 및 배설속도, 질병종류, 병용되는 약물에 따라 달라질 수 있으며, 일반적으로는 체중 1 ㎏ 당 0.001 내지 150 ㎎, 바람직하게는 0.01 내지 100 ㎎을 매일 또는 격일 투여하거나, 1일 1 내지 3회로 나누어 투여할 수 있다. 그러나 투여 경로, 비만의 중증도, 성별, 체중, 연령 등에 따라서 증감 될 수 있으므로 상기 투여량이 어떠한 방법으로도 본 발명의 범위를 한정하는 것은 아니다.
본 명세서에서 상기 병용 투여는 병행 투여와 교차하여 사용할 수 있으며, 병용 투여 형태는 펩타이드 또는 펩타이드 유사체와 기타 화합물을 동시 투여하거나, 또는 별도 투여하는 형태를 모두 포함할 수 있다.
또한, 본 발명은 상기 조성물을 대상체에게 투여하는 단계; 및
상기 대상체에서 조직면역염색 (immunohistochemistry; IHC)을 통해 AAV의 분포를 확인하는 단계;를 포함하는, AAV 벡터 전달 효율 스크리닝 방법을 제공한다.
본 발명에서 "대상체"란 질병의 치료를 필요로 하는 대상을 의미하고, 보다 구체적으로는 인간 또는 비-인간인 영장류, 생쥐(mouse), 쥐(rat), 개, 고양이, 말 및 소 등의 포유동물을 의미한다.
본 발명의 일 실시예에서, 상기 조직면역염색은 GFP 항체를 이용하는 것일 수 있다.
본 발명의 일 실시예에서, 상기 스크리닝 방법은 상기 AAV 벡터를 뇌내 전달한 다음 AAV의 형질도입 효율 즉 뇌내 분포를 확인하는 단계를 포함할 수 있다.
본 발명의 일 구체예에서, 투여 4주 후에 sacrifice한 그룹에서, 투여 1주 후에 sacrifice한 그룹에 비해 AAV 벡터로 형질도입된 세포의 분포가 마우스 뇌내 광범위하게 관찰되었다.
일 실시예에서, 아데노부속바이러스 벡터(adeno-associated virus vector; AAV벡터)를 정상 마우스의 경동맥으로 주입하여 AAV벡터의 신경세포 형질도입 (transduction), 즉 뇌내 AAV 벡터의 분포를 확인하였다.
일 실시예에서, 뇌내 분포 되어있는 여러 세포들 중 어느 세포 종류 (신경세포, 미세아교세포, 별아교세포 등등)에 따른 형질도입 효율 차이를 확인하였다.
또한, 본 발명은 신경계 질환의 예방 또는 치료를 위한 약제의 제조에 사용하기 위한 AAV 캡시드 및 바이러스 게놈을 포함하는 AAV 입자를 포함하는 약제학적 조성물의 용도를 제공한다.
또한, 본 발명은 신경계 질환의 예방 또는 치료를 위한 약제의 제조에 사용하기 위한 AAV 캡시드 및 바이러스 게놈을 포함하는 AAV 입자의 용도를 제공한다.
본원에 기재된 상기 각 특징들은 조합되어 사용될 수 있으며, 상기 각 특징들이 특허청구범위의 서로 다른 종속항에 기재된다는 사실은 이들이 조합되어 사용될 수 없음을 나타내는 것은 아니다.
본 발명에 따라 최적화된 조건으로 AAV 벡터를 경동맥 투여하여 다양한 뇌질환에 적용될 수 있고 유전자 치료제의 치료 효과를 극대화시킬 수 있다.
본 발명의 AAV 벡터의 뇌내 전달 방법은 경동맥 투여는 임상에서 추가적인 수술 없이 대퇴동맥 (femoral artery)를 통해 경동맥으로 투여가 가능하여 수술의 대한 부담이 없고 실제 임상에서 널리 사용될 수 있어, 추후 임상 적용이 용이하다.

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예 및 실험예에 한정되는 것은 아니다.
실시예 및 실험예
실험예 1. AAV 벡터 제작
pCAG 프로모터 (promoter)의 조절을 받으며 포유류의 세포에서 녹색 형광 단백질 (green fluorescent protein; GFP)을 발현할 수 있는 재조합 아데노부속바이러스 (adeno associated virus; AAV)를 생산하였고 (도 1 참조), 캡시드 (capsid) 단백질의 serotype 은 AAV9을 이용하였다.
인간 배아 신장에서 유래한 HEK293 세포주에 AAV9의 capsid를 발현하는 플라스미드 (plasmid)와 helper plasmid, 그리고 pCAG-GFP plasmid를 모두 형질주입(transfection) 하였고, 72시간 후 세포와 상층액 (supernatant)을 모두 모았다.
상기 시료에서 순도 높은 바이러스를 정제하기 위해, 용해 완충액 (lysis buffer)과 benzonase를 2시간 동안 처리한 다음 세포를 분쇄하였다. 상기 분쇄물을 AAV에 특화된 컬럼 (column; Thermo Fisher Scientific, USA)이 장착된 fast protein liquid chromatography (FPLC)를 이용하여 순도 높은 AAV를 정제하였다 (도 2 참조).
실시간 중합효소연쇄반응 (quantitative polymerase chain reaction; qPCR)을 이용하여 정제된 virus의 역가 (titer)를 측정한 결과, 1L의 샘플로부터 2.231012 의 viral genome (VG)을 얻었고 농축 후 농도는 1 mL 당 1.121012 VG 인 것을 확인하였다 (도 3 참조).
실험예 2. AAV 벡터 경동맥 투여 (intra-arterial; IA injection)
누운 자세 (supine position)의 정상 마우스 (wild-type; WT)에 2% 이소플루란 (isoflurane)으로 호흡마취를 유지한 상태에서 수술을 진행하였다.
멸균된 수술도구로 피부를 절개한 후 오른쪽 대동맥 (common carotid artery; CCA), 오른쪽 외경동맥 (external carotid artery; ECA), 그리고 오른쪽 내경동맥 (ICA)을 분리하여 각각의 혈관을 확보하였다.
CCA의 아래 부분을 실크 실로 묶은 다음 ECA의 혈류 또한 실크실을 이용하여 일시적으로 중단시켰다. 이 후 ICA-ECA 분지 (bifurcation)에서 CCA를 묶은 부분의 중간 위치에서 CCA를 살짝 절개하였다. 상기 절개한 위치에 catheter를 주입하여 ICA에 안전하게 고정한 후 AAV 벡터를 투여하였다. 총 100 μL를 투여하였는데 (1.12×1011 viral genome; VG) 실험군은 AAV 벡터를 투여하였고, 대조군은 똑같은 용량의 인신완충생리식염수 (phosphate-buffered saline; PBS)를 투여하였다 (도 4 참조).
투여 후 catheter를 제거하고 CCA를 절개한 부분 바로 위 (ICA-ECA 분지 아래)를 실크실로 묶은 후 절개하였던 부위는 봉합하였다. AAV 벡터 혹은 PBS를 ICA로 투여한 후 1주일 (post 1 week), 그리고 4주뒤 (post 4 week) 안락사 (sacrifice)를 진행하여 뇌를 적출하였다 (도 4 참조).
실험예 3. 경동맥 투여로 인한 AAV 벡터의 뇌내 전달 확인
경동맥 투여 1주일 (post 1 week), 그리고 4주 뒤 (post 4 week) 안락사를 통해 채취한 뇌조직은 4% 포름알데하이드 (paraformaldehyde; PFA)에 고정시킨 후 파라핀 블록 (paraffin block)을 만들어 절편 제작 (두께: 4 μm)을 하였다.
상기 제작한 AAV 벡터는 GFP가 발현되므로 현광현미경으로 GFP의 signal (초록색 형광)를 관찰할 수 있다. GFP 항체 (abcam, USA)를 이용하여 조직면역염색 (immunohistochemistry; IHC)를 진행하였다. 현광현미경으로 통해 초록색 형광을 나타내는 세포/뇌 부위를 관찰하였다. 초록색 형광은 GFP의 signal를 의미하며 AAV가 성공적으로 신경세포에 형질도입 (transduction)되었다는 것을 의미한다.
AAV 벡터 경동맥 투여 1주일 뒤 시상부 (thalamus)쪽에서 GFP를 발현하는 세포 (GFP positive cell)를 (빨간색 화살표) 일부 발견하였다 (도 5 참조). 오른쪽 내경동맥으로 AAV 벡터 혹은 PBS를 투여하였는데 오른쪽 (right) 그리고 왼쪽 반구 (left hemisphere) 에서도 GFP signal를 확인할 수 있었다. PBS를 투여 받은 그룹에서는 GFP signal를 관찰하지 못하였다. Post 4 week에 sacrifice한 PBS그룹에서도 GFP signal를 관찰하지 못하였고 비특이적 (non-specific)인 signal 만 (하얀색 화살표) 관찰하였다 (도 6 참조).
AAV 벡터 그룹에서는 thalamus 뿐만 아니라 다양한 부위에서 GFP signal를 관찰할 수 있었다 (도 4 및 도 5 참조). 대뇌피질 (cerebral cortex), 시상하부 (hypothalamus), 해마 (hippocampus), 그리고 중간뇌 (midbrain)에서 GFP를 발현하는 신경세포를 일부 발견하였다. Right (도 7 참조) 외 left hemisphere에서도 GFP signal를 확인할 수 있었다 (도 8 참조). Post 1 week에 비해 post 4 week에 sacrifice한 AAV 벡터 그룹에서 GFP positive cell들을 더 많이 발견할 수 있었다. 또한 post 4 week에 sacrifice를 수행하였을 때 특정 부위가 아닌 뇌 전체 광범위하게 GFP positive cell들을 관찰할 수 있었다.
실험예 4. AAV 벡터 뇌실 투여 (intracerebroventricular; ICV injection)
실험예 1~3에서는 경동맥 투여로 인한 AAV 뇌내 전달을 확인하였다. 혈관으로 주입하였을 때 보다 직접적으로 뇌로 투여하였을 때 AAV 뇌내 전달의 차이를 비교해보고자 뇌실 투여 (intracerebroventricular; ICV injection)를 수행하였다 [도9 참조]. 뇌실(lateral ventricle; LV)을 채우고 있는 뇌척수액 (cerebrospinal fluid; CSF)의 순환(flow)으로 AAV가 뇌 전체 광범위하게 전달될 수 있다.
WT 마우스를 2% isoflurane으로 호흡마취를 유지한 상태에서 stereotaxic 수술을 진행하였다. 마우스의 머리를 stereotaxic frame의 ear bar로 고정한 다음 멸균된 수술도구로 피부를 절개하여 bregma 위치를 확인하였다. 투여 위치의 좌표는 다음과 같았다: anterior/posterior (A/P): -0.4 mm, medial/lateral (M/L): -1.0 mm, dorsal/ventral (D/V): -2.3 mm. 지정된 좌표에서 burr hole를 만든 다음 hamilton syringe 그리고 injection pump를 이용하여 투여를 진행하였다. WT 마우스를 두 그룹으로 나누어 PBS (5 μL)혹은 AAV (1.5Х1010 vg / 5 μL) 를 왼쪽 뇌실에 단회 투여 하였다 (injection rate: 1 μL / min). PBS 혹은 AAV의 loss를 줄이기 위해 투여를 마치고 5 분 delay 후 뇌내 삽입된 hamilton syringe를 천천히 빼 주었다. 절개하였던 피부 부위는 봉합하였다. AAV 벡터 혹은 PBS를 ICV로 투여한 후 4주 뒤 (post 4 week) 안락사 (sacrifice)를 진행하여 뇌를 적출하였다.
실험예 5. 뇌실 투여로 인한 AAV 벡터의 뇌내 전달 확인
뇌실 투여 4주 뒤 (post 4 week) 안락사를 통해 채취한 뇌조직은 4% PFA에 고정시킨 후 paraffin block을 만들어 절편 제작 (두께: 4 μm)을 하였다. GFP 항체 (abcam, USA)를 이용하여 조직면역염색 (immunohistochemistry; IHC)를 진행하였다. 현광현미경으로 통해 초록색 형광을 나타내는 세포/뇌 부위를 관찰하였다. 초록색 형광은 GFP의 signal를 의미하며 AAV가 성공적으로 신경세포에 형질도입 (transduction)되었다는 것을 의미한다.
PBS를 투여 한 그룹에서는 GFP signal이 발견되지 않았다. AAV를 투여한 그룹에서는 뇌실벽 (lateral ventricle wall) 그리고 맥락얼기 (choroid plexus)에서 GFP positive cell들을 발견하였다 [도10 참조]. 뇌실벽에 있는 뇌실막세포 (ependymal cell)에서 GFP signal이 관찰되었다. LV에 존재하는 choroid plexus에서도 GFP positive cell들을 확인할 수 있었다. 투여를 진행한 왼쪽 LV 외 반대편 오른쪽 LV에 존재하는 choroid plexus에서도 GFP signal를 발견하였다. 이 결과는 CSF flow로 왼쪽 LV에 투여한 AAV가 반대편 반구로 이동하여 transduction이 되었다는 것을 의미한다.
Lateral ventricle wall 그리고 choroid plexus 외 뇌실질내 GFP signal가 관찰되지 않았다. 이것은 ICV로 전달된 AAV가 혈액-뇌척수액 장벽 (blood-csf barrier)를 침투하지 못하여 뇌실질내 존재하는 세포에 transduction이 안 되었다는 것을 의미한다. ICV route를 이용한 뇌내 직접 투여 보다 IA route를 이용하였을 때 뇌실질내 GFP positive cell 즉 AAV transduction을 확인할 수 있었다.

- 캡시드 및 바이러스 게놈을 포함하는 아데노-부속 바이러스(AAV) 입자로서, 상기 캡시드가 AAV 입자의 전달 후 혈액 뇌 장벽을 투과하는, 아데노-부속 바이러스(AAV) 입자.
- 청구항 1에 있어서, 상기 캡시드는 서열번호 1로 기재된 아미노산 서열로 이루어진 것인, 아데노-부속 바이러스 입자.
- 청구항 1에 있어서, 상기 전달은 경동맥내 투여에 의한 것인, 아데노-부속 바이러스 입자.
- AAV 캡시드 및 바이러스 게놈을 포함하는 AAV 입자를 포함하는 약제학적 조성물로서, 상기 AAV 캡시드가 상기 AAV 입자의 전달 후 혈액 뇌 장벽을 투과하는, 약제학적 조성물.
- 청구항 4의 조성물을 세포에 투여하는 단계를 포함하는, 세포 내 관심 표적 유전자의 발현을 감소시키거나 억제하는 방법.
