Abstract
本发明公开了一种微藻凝胶的新应用,所述微藻凝胶中的微藻通过光合作用和呼吸作用可促进创面愈合。伤口修复用敷料,包括一敷料部件以及将敷料部件固定在患处的固定部件;所述敷料部件包括储料囊、凝胶球、抗菌透气内衬和膏药贴;本发明还提出一种伤口修复用敷料的使用方法,包括以下步骤:S1、将微藻‑高分子材料混合液注射至氯化钙水溶液中制得凝胶球,成形的凝胶球转移至储囊袋内密封制得伤口敷料。本发明藻凝胶伤口敷料贴附于创面后,通过隔绝创面和外环境,使伤口不接触环境中的病原体。
Description
技术领域
本发明涉及微藻凝胶与透气透水薄膜组合物用于伤口愈合技术领域,尤其涉及一种伤口修复用敷料及其使用方法。
背景技术
现有技术中,针对伤口部位,尤其是各种慢性创面往往缺乏氧供、易于感染,因此愈合困难,同时,现有的搭配伤口敷料使用的配件均为一次性的,使用过后就需要全部进行更换,造成了浪费,为解决上述技术难题,我们提出一种伤口修复用敷料及其使用方法。
发明内容
本发明的目的是为了解决现有技术中存在的缺点,而提出的一种伤口修复用敷料及其使用方法,其藻凝胶伤口敷料贴附于创面后,通过隔绝创面和外环境,使伤口不接触环境中的病原体;制得的凝胶球载体可缓慢释放气体,使得产品维持时间延长,减少更换频率,节约成本。
为了实现上述目的,本发明采用了如下技术方案:
一种微藻凝胶的新应用,所述微藻凝胶中的微藻通过光合作用和呼吸作用,提高伤口表面的氧气浓度、二氧化碳浓度和湿度,氧浓度提高直接增效创面愈合;二氧化碳浓度提高可通过玻尔效应增强创面的血氧供应;湿度提高有利于气体在创面的渗透扩散,以及通过湿性愈合的方式促进创面愈合。
优选的,所述微藻凝胶与抗菌透气内衬组合,用于制备含有微藻凝胶的伤口修复用敷料。
优选的,包括一敷料部件以及将敷料部件固定在患处的固定部件;
所述敷料部件包括储料囊、凝胶球、抗菌透气内衬和膏药贴;
所述凝胶球设置在储料囊内,所述抗菌透气内衬嵌设在储料囊的下端,所述膏药贴粘贴在抗菌透气内衬的下端;
所述储料囊的两端设置有一搭台。
优选的,所述储料囊的上端设置有一注液口,且注液口上配置有密封塞;所述抗菌透气内衬为亲水性的且厚度为0.1-0.22um孔径的PES或PTFE薄膜。
优选的,所述橡胶绑带上设置有可将绑带粘贴在皮肤表面上的粘贴部件,所述粘贴部件设置在橡胶绑带的端部上。
优选的,所述固定部件包括一橡胶绑带,所述橡胶绑带的面部开设有用于容纳储料囊的开口,所述开口的上端边缘处设置有一缺口。
一种含有微藻凝胶的伤口修复用敷料的使用方法,包括以下步骤:
S1、将微藻-高分子材料混合液注射至氯化钙水溶液中制得凝胶球,成形的凝胶球转移至独立包装的药袋中密封,备用;
S2、使用时,患者将独立包装的药袋中的凝胶球通过注射口注射到储料囊内,并通过密封塞对注射口密封;
S3、将储料囊两侧的搭台搭在缺口上,并通过粘贴部件固定;
S4、将膏药贴贴敷在患处,并通过绑带对患处进行固定。
优选的,所述S1中的高分子材料包括但不限于羟丙甲基纤维素、葡萄糖酸内酯、海藻酸钠、明胶、果胶、壳聚糖或微晶纤维素中的一种或几种。
优选的,所述微藻包括但不限于聚球藻、顶钝螺旋藻、极大螺旋藻、小球藻或念珠藻中的一种或几种。
优选的,所述微藻包括但不限于聚球藻、顶钝螺旋藻、极大螺旋藻、小球藻或念珠藻中的一种或几种。
本发明具有以下有益效果:
1、藻凝胶伤口敷料贴附于创面后,通过隔绝创面和外环境,使伤口不接触环境中的病原体;
2、微藻通过光合作用和呼吸作用,提高敷料和创面内部的氧气浓度,二氧化碳浓度和湿度,氧浓度提高直接增效创面愈合;二氧化碳浓度提高可通过玻尔效应增强创面的血氧供应;
3、湿度提高有利于气体在创面的渗透扩散,以及通过湿性愈合的方式促进创面愈合;
4、用于搭在敷料的部件可循环利用,降低了成本。

具体实施方式
为使本发明的上述目的、特征和优点能够更加明显易懂,下面结合附图对本发明的具体实施方式做详细的说明。在下面的描述中阐述了很多具体细节以便于充分理解本发明。但是本发明能够以很多不同于在此描述的其它方式来实施,本领域技术人员可以在不违背本发明内涵的情况下做类似改进,因此本发明不受下面公开的具体实施的限制。
需要说明的是,当元件被称为“固定于”另一个元件,它可以直接在另一个元件上或者也可以存在居中的元件。当一个元件被认为是“连接”另一个元件,它可以是直接连接到另一个元件或者可能同时存在居中元件。本文所使用的术语“垂直的”、“水平的”、“左”、“右”以及类似的表述只是为了说明的目的,并不表示是唯一的实施方式。
除非另有定义,本文所使用的所有的技术和科学术语与属于本发明的技术领域的技术人员通常理解的含义相同。本文中在本发明的说明书中所使用的术语只是为了描述具体的实施例的目的,不是旨在于限制本发明。本文所使用的术语“及/或”包括一个或多个相关的所列项目的任意的和所有的组合。
如图1所示,一种微藻凝胶的新应用,所述微藻凝胶中的微藻通过光合作用和呼吸作用,提高伤口表面的氧气浓度、二氧化碳浓度和湿度,氧浓度提高直接增效创面愈合;二氧化碳浓度提高可通过玻尔效应增强创面的血氧供应;湿度提高有利于气体在创面的渗透扩散,以及通过湿性愈合的方式促进创面愈合。所述微藻凝胶与抗菌透气内衬24组合,用于制备含有微藻凝胶的伤口修复用敷料。
参照图1-2,一种含有微藻凝胶的伤口修复用敷料,包括一敷料部件2以及将敷料部件2固定在患处的固定部件1;
敷料部件2包括储料囊21、凝胶23、抗菌透气内衬24和膏药贴25;
凝胶23设置在储料囊21内,抗菌透气内衬24嵌设在储料囊21的下端,膏药贴25粘贴在抗菌透气内衬24的下端,膏药贴25在使用后可更换掉,减少了浪费,抗菌透气内衬24为PES聚醚砜树脂或PTFE聚四氟乙烯薄膜,且PES聚醚砜树脂或PTFE聚四氟乙烯薄膜的孔径为0.1-0.22um,PES聚醚砜树脂或PTFE聚四氟乙烯薄膜允许气体和水分高效通透的同时,阻碍了微藻和细菌的跨膜流动,使伤口不易感染和滋生细菌;
凝胶23通过将海藻酸钠-纯种微藻混合液滴加入CaCl2溶液成胶制得;
储料囊21的两端设置有一搭台22;
固定部件1包括一橡胶绑带13,橡胶绑带13的面部开设有用于容纳储料囊21的开口,开口的上端边缘处设置有一缺口14。
储料囊21的上端设置有一注液口,且注液口上配置有密封塞26。
橡胶绑带13上设置有可将绑带粘贴在皮肤表面上的粘贴部件12,粘贴部件12设置在橡胶绑带13的端部11上。
凝胶23通过将海藻酸钠-纯种微藻混合液滴加入CaCl2溶液成胶制得,凝胶23中还添加有培养基、碳酸钠和葡萄糖,通过添加有培养基、碳酸钠和葡萄糖可以有效地提高微藻的活性,增强光合作用、呼吸作用的效率及产生的氧气、二氧化碳总量;此外,在凝胶球中加入山梨酸钾防腐剂,对微藻活性没有影响但可以选择性抑制其他细菌;微藻凝胶球主要作用是改变局部环境的气体组成,尤其是水、二氧化碳和氧气浓度。
敷料贴附于创面后,通过隔绝创面和外环境,使伤口不接触环境中的病原体。
此外,微藻通过光合作用和呼吸作用,提高敷料和创面内部的氧气浓度,二氧化碳浓度和湿度。
氧浓度提高直接增效创面愈合;二氧化碳浓度提高可通过玻尔效应增强创面的血氧供应;湿度提高有利于气体在创面的渗透扩散,以及通过湿性愈合的方式促进创面愈合。
本发明还提出一种伤口修复用敷料的使用方法,包括以下步骤:
S1、将微藻-高分子材料混合液注射至氯化钙水溶液中制得藻凝胶球,成形的凝胶球转移至独立包装的药袋中密封,备用;所述S1中的高分子材料包括但不限于羟丙甲基纤维素、葡萄糖酸内酯、海藻酸钠、明胶、果胶、壳聚糖或微晶纤维素中的一种或几种。
S2、使用时,患者将独立包装的药袋中的凝胶23通过注射口注射到储料囊21内,并通过密封塞26对注射口密封;
S3、将储料囊21两侧的搭台22搭在缺口14上,并通过粘贴部件固定;
S4、将膏药贴25贴敷在患处,并通过绑带对患处进行固定。
患者将独立包装的药袋中的凝胶23通过注射口注射到储料囊21内,并通过密封塞26对注射口密封;将储料囊21两侧的搭台22搭在缺口14上,并通过粘贴部件固定;将膏药贴25贴敷在患处,并通过绑带对患处进行固定。
敷料贴合于创面之后,微藻通过呼吸作用、光合作用,在密封环境中维持创面的高氧气浓度、高二氧化碳浓度、高湿度和无菌环境,从而起到促进各类伤口,尤其是慢性伤口,更尤其是糖尿病慢性伤口修复的作用。
微藻包括但不限于聚球藻、顶钝螺旋藻、极大螺旋藻、小球藻或念珠藻中的一种或几种。
使用完毕后,患者只需将绑带13拆下,然后对膏药贴25进行更换,将储料囊21内的凝胶23从注射口挤出,然后注入新的凝胶23即可,使用完毕后直接更换新的敷料。
实验1:为了证明微藻凝胶可以促进糖尿病慢性创面愈合,将患有糖尿病的Balb/C小鼠随机等数量分为三组:对照组、微藻凝胶组、局部通氧组。将背部表皮和真皮切除,制作创面。对照伤口暴露在空气中,微藻凝胶组将新鲜制备的微藻凝胶敷料每天局部覆盖在伤口上,局部通氧组每天定时定量进行高压氧疗。在手术当天及之后每2天同一时间测量伤口面积,直到伤口完全愈合。如图4,使用微藻凝胶能够缩短伤口愈合时间,加速创面愈合进程。
实验2:为了对比微藻凝胶球与微藻溶液的产氧差异,将微藻溶液与高分子凝胶均匀混合,使用注射器将混合溶液滴入氯化钙中,制成大小相同的藻凝胶球。在持续光照条件下,通过氧电极测量溶液中溶解氧浓度,比较等量的藻凝胶球与微藻溶液产氧量差异,对照组为培养基溶液。如图5,可以看出固定化的微藻凝胶球在相同时间内匀速产氧量与微藻溶液相近。
实验3:为了验证微藻凝胶能够促进糖尿病伤口周围细胞的生长,将人皮肤纤维上皮细胞置于高葡萄糖浓度培养基中,培养至一定密度后等量分为三组:对照组暴露于缺氧环境中,作为特定缺氧组;微藻凝胶组将藻凝胶球置于细胞上方小室中,近红外发光二极管照射;持续通氧组为医用级纯氧匀速通氧处理。光照培养相同时间后,通过染色鉴别细胞存活状态。如图6,相比之下微藻凝胶能够更好地促进细胞生长。
以上所述,仅为本发明较佳的具体实施方式,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,根据本发明的技术方案及其发明构思加以等同替换或改变,都应涵盖在本发明的保护范围之内。
Claims (10)
1.一种微藻凝胶的新应用,其特征在于,所述微藻凝胶中的微藻通过光合作用和呼吸作用,提高伤口表面的氧气浓度、二氧化碳浓度和湿度,氧浓度提高直接增效创面愈合;二氧化碳浓度提高可通过玻尔效应增强创面的血氧供应;湿度提高有利于气体在创面的渗透扩散,以及通过湿性愈合的方式促进创面愈合。
2.根据权利要求1所述一种微藻凝胶的新应用,其特征在于,所述微藻凝胶与抗菌透气内衬(24)组合,用于制备含有微藻凝胶的伤口修复用敷料。
3.一种含有微藻凝胶的伤口修复用敷料,其特征在于,包括一敷料部件(2)以及将敷料部件(2)固定在患处的固定部件(1);
所述敷料部件(2)包括储料囊(21)、凝胶球(23)、抗菌透气内衬(24)和膏药贴(25);
所述凝胶球(23)设置在储料囊(21)内,所述抗菌透气内衬(24)嵌设在储料囊(21)的下端,所述膏药贴(25)粘贴在抗菌透气内衬(24)的下端;
所述储料囊(21)的两端设置有一搭台(22)。
4.根据权利要求3所述的一种含有微藻凝胶的伤口修复用敷料,其特征在于,所述储料囊(21)的上端设置有一注液口,且注液口上配置有密封塞(26);所述抗菌透气内衬(24)为亲水性的且孔径为0.1-0.22um的PES或PTFE薄膜。
5.根据权利要求3所述的一种含有微藻凝胶的伤口修复用敷料,其特征在于,所述橡胶绑带(13)上设置有可将绑带粘贴在皮肤表面上的粘贴部件(12),所述粘贴部件(12)设置在橡胶绑带(13)的端部(11)上。
6.根据权利要求3所述的一种含有微藻凝胶的伤口修复用敷料,其特征在于,所述固定部件(1)包括一橡胶绑带(13),所述橡胶绑带(13)的面部开设有用于容纳储料囊(21)的开口,所述开口的上端边缘处设置有一缺口(14)。
7.一种含有微藻凝胶的伤口修复用敷料的使用方法,其特征在于,包括以下步骤:
S1、将微藻-高分子材料混合液注射至氯化钙水溶液中制得凝胶球,成形的凝胶球转移至独立包装的药袋中密封,备用;
S2、使用时,患者将独立包装的药袋中的凝胶球(23)通过注射口注射到储料囊(21)内,并通过密封塞(26)对注射口密封;
S3、将储料囊(21)两侧的搭台(22)搭在缺口(14)上,并通过粘贴部件固定;
S4、将膏药贴(25)贴敷在患处,并通过绑带对患处进行固定。
8.根据权利要求7所述一种含有微藻凝胶的伤口修复用敷料的使用方法,其特征在于,所述S1中的高分子材料包括但不限于羟丙甲基纤维素、葡萄糖酸内酯、海藻酸钠、明胶、果胶、壳聚糖或微晶纤维素中的一种或几种。
9.根据权利要求1所述一种微藻凝胶的新应用,其特征在于,所述微藻包括但不限于聚球藻、顶钝螺旋藻、极大螺旋藻、小球藻或念珠藻中的一种或几种。
10.根据权利要求7所述一种含有微藻凝胶的伤口修复用敷料的使用方法,其特征在于,所述微藻包括但不限于聚球藻、顶钝螺旋藻、极大螺旋藻、小球藻或念珠藻中的一种或几种。
