Abstract
本发明公开了白介素18在抑制创面血管新生中的应用,属于生物医药技术领域,本发明首次公开了IL‑18在抑制血管新生方面的作用,经过实验证实,IL‑18作用于脐静脉内皮细胞可显著抑制90%的血管形成;作用于小鼠模型中,有效抑制新生血管可达90%。因而,有望将IL‑18开发为创面修复过程中抑制血管过度增生从而避免病理性瘢痕形成的药物。
Description
技术领域
本发明属于生物医学技术领域,涉及白介素18(以下简称IL-18)在抑制创面血管新生中的应用。
背景技术
血管生成是一个由多种促血管新生因子和抗血管新生因子共同调控的一个平衡过程。血管新生与组织发育、创伤修复以及肿瘤生长等生理病理过程密切相关。其中,组织的生长与发育需要新生的血管供给养分,创伤创面的修复也需要新生的血管供给营养和各种促细胞生长因子。
新生血管是影响创面愈合的最主要原因之一,在创面修复过程中必需的氧和营养物质只有经过血流才能输送到创面,而局部产生的大部分毒性物质及代谢物等也主要经血流才能输送出创面。在创面修复后期,过度的血管新生则易导致病理性瘢痕的产生,因而这个阶段需要采取相应的办法对血管新生进行抑制,这对于外科创面的治疗及病理性瘢痕的预防具有重要的意义。
在血管新生过程中,每个环节均受众多分子的严密调控,这些血管生成相关因子在血管新生各个环节发挥了重要的调控作用。目前,作用明确的抑制血管新生的分子有20多种。
但是,当前还未见IL-18在抑制创面修复过程中血管新生方面作用的相关报道。
发明内容
为了克服上述现有技术的缺点,本发明的目的在于提供白介素18在抑制创面血管新生中的应用。
为了达到上述目的,本发明采用以下技术方案予以实现:
本发明公开了白介素18在制备治疗抑制血管新生的药物中的应用。
优选地,所述的药物为抑制创面血管新生的药物。
进一步优选地,所述的药物为作用于创面新生血管内皮细胞的药物。
进一步优选地,所述的药物为抑制血管内皮细胞迁移的药物。
优选地,白介素18在体外的有效抑制浓度≥100ng/ml。
本发明还公开了一种抑制血管新生的药物,由白介素18和及药学上可接受的药用辅料制成。
优选地,该药物中白介素18的浓度≥10μg/ml。
与现有技术相比,本发明具有以下有益效果:
本发明首次公开了IL-18在抑制血管新生方面的作用,经过实验证实,IL-18作用于脐静脉内皮细胞可显著抑制90%的血管形成;作用于小鼠模型中,有效抑制新生血管可达90%。因而,有望将IL-18开发为抑制新生血管生成的药物。

具体实施方式
为了使本技术领域的人员更好地理解本发明方案,下面将结合本发明实施例中的附图,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分的实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都应当属于本发明保护的范围。
需要说明的是,术语“包括”和“具有”以及他们的任何变形,意图在于覆盖不排他的包含,例如,包含了一系列步骤或单元的过程、方法、系统、产品或设备不必限于清楚地列出的那些步骤或单元,而是可包括没有清楚地列出的或对于这些过程、方法、产品或设备固有的其它步骤或单元。
下面结合附图对本发明做进一步详细描述:
参见图1,为IL-18单体化学结构图,IL-18单体包括193个氨基酸,编码IL-18的基因ID为3606。
在96孔板中培养细胞,采用celltrack细胞示踪剂将细胞标记绿色荧光。待细胞长满后除去中间栓塞,并在实验组分别加入50ng/ml,100ng/ml,200ng/ml的IL-18,采用Cytation细胞成像仪对细胞进行观测并拍照,记录20小时内细胞迁移情况。结果参见图2,可以看出,IL-18浓度为100ng/ml组和200ng/ml组的细胞迁移均受到明显的抑制。经统计学分析,100ng/ml组IL-18对细胞迁移的抑制作用显著高于50ng/ml组,且差异具有统计学意义,200ng/ml组IL-18对细胞迁移的抑制作用与100ng/ml组比较,差异不显著,因而IL-18浓度≥100ng/ml为有效抑制浓度。
在96孔板中培养细胞,实验组加入100ng/ml的IL-18,分别在第1天,第3天和第5天分别加入等量体积的培养基和CellTiter-Glo试剂,用多孔扫描分光光度计进行发光检测,并对结果进行统计分析。参见图3,结果显示,实验组中IL-18分别在第3天和第5天对细胞活力产生了显著的抑制作用(p<0.05)。
8周龄无胸腺裸小鼠采用60ul/20g的剂量注射1%的戊巴比妥钠麻醉。用70%的酒精对小鼠背侧皮肤进行消毒,手术将填充了Matrigel基质胶的组织工程小室植入其背侧皮下。
将12只裸小鼠随机分为4组:
阳性对照组(n=3),组织工程小室内填充含有促血管生成因子的Matrigel凝胶(含肝素和VEGF);
阴性对照组(n=3),组织工程小室内填充无促血管生成因子的Matrigel凝胶;
IL-18组(n=6),组织工程小室内填充含促血管生成因子的Matrigel凝胶和IL-18(10μg/ml)。
移植后6天取出组织工程小室,使用dispase(conning,Cat.No.354235)消化凝胶,并用细胞回收液回收细胞,然后采用FITC-lectin标记细胞,用流式细胞仪分析各组细胞荧光强度,结果参见图5,结果显示,实验组(IL-18组)恢复细胞的荧光强度明显较低,与阳性对照组相比,差异显著,IL-18组新生血管显著受到抑制。本实验结果说明,IL-18在体内也具有明确的抑制血管新生的作用。
参见图4,为体内动物实验验证IL-18对血管新生的抑制作用效果照片;其中,A为组织工程小室;B为小室内加入基质胶,植入裸鼠背部皮下;C为植入体内后6天取出的小室。也说明,IL-18在体内也具有明确的抑制血管新生的作用。
综上所述,IL-18作用于脐静脉内皮细胞可显著抑制90%的血管形成;作用于小鼠模型中,有效抑制新生血管可达90%。因而,有望将IL-18开发为抑制新生血管生成的药物。

1.白介素18在制备治疗抑制血管新生的药物中的应用。
2.如权利要求1所述的应用,其特征在于,所述的药物为抑制创面血管新生的药物。
3.如权利要求2所述的应用,其特征在于,所述的药物为作用于创面新生血管内皮细胞的药物。
4.如权利要求3所述的应用,其特征在于,所述的药物为抑制血管内皮细胞迁移的药物。
5.如权利要求1~3所述的应用,其特征在于,白介素18在体外的有效抑制浓度≥100ng/ml。
6.一种抑制血管新生的药物,其特征在于,由白介素18和及药学上可接受的药用辅料制成。
7.如权利要求6所述的抑制血管新生的药物,其特征在于,该药物中白介素18的浓度≥10μg/ml。
