技术领域
本发明涉及一种萘环类苯硫酚荧光探针以及其在检测苯硫酚中的应用。
背景技术
苯硫酚(PhSH),又名苯硫醇,常温下是一种带有恶臭味的无色液体。对皮肤、粘膜呼吸道及眼睛有强烈的刺激作用,吸入后可引起支气管痉挛、喉、水肿及化学性肺水肿、肺炎。中毒表现有咳嗽、喘息、喉炎、气短、烧灼感、恶心、头痛、呕吐。苯硫酚对小鼠的半数致死量为502mg/kg,被美国环境保护局添加到优先级的污染物中。苯硫酚是重要的医药、农药、高分子材料合成中间体,在化学工业中被广泛应用。
有机小分子荧光探针检测法由于其具有检测快速、灵敏高、选择性强及操作简便等优点,是一种非常有效的检测方法,目前开发出众多的有机小分子荧光探针。因此,以荧光探针作为检测工具,测定苯硫酚的含量,对环境中苯硫酚的特异性检测具有重要的意义。
发明内容
本发明目的是提供一种检测苯硫酚的荧光探针,它适于药品、食品等产品中苯硫酚的检测。
更具体的是,本发明涉及一种检测苯硫酚的荧光探针,它通过下式表示:

图1是在磷酸缓冲盐溶液(pH=7.4)中,随着不同浓度苯硫酚的加入,本发明的荧光探针(10μM)的荧光强度变化。嵌入图:苯硫酚的浓度。

图3自然光下,本发明的荧光探针试纸浸染不同浓度苯硫酚(0,5,10,50,100μM)后试纸颜色变化情况。
具体实施方式
实施例1.检测苯硫酚的荧光探针的制备
合成路线如下式所示:
将化合物1(4.88g,20mmol)和化合物2(5.24g,20mmol)加入到100mL单口烧瓶中,加入THF(50mL),加热回流4h,得到化合物3;待反应瓶冷却至室温,向反应瓶中加入20mL 2MNaOH溶液,并加入24mL甲醇和28mLH2O,室温搅拌过夜,抽滤得黄色固体化合物4。
化合物5(1.0g,5.8mmol)溶于10mL DMF,并加入Et3N(0.607g,6mmol),在冰水浴条件下,逐滴滴加入化合物6(3.097g,11.6mmol,10mL DMF);滴加完毕后升温至40℃反应3h后,抽滤得白色固体,重结晶得化合物7。
化合物7(0.5g,1.2mmol)和化合物4(0.634g,1.5mmol)溶于THF,加入NaH(0.003g,0.12mmol)回流8h,抽滤得黄色固体,重结晶得化合物8,产率47.83%。
产物经过核磁氢谱及核磁碳谱表征。
1H NMR(300MHz,DMSO),δ(ppm):9.15(d,J=2.2Hz,1H),8.59(dd,J=8.7,2.3Hz,1H),8.43(d,J=14.0Hz,5H),8.33-7.79(m,8H),7.38(dd,J=8.9,2.4Hz,1H);13C NMR(75MHz,DMSO),δ(ppm):191.57,154.09,137.37,133.83,132.74,130.63,128.89,128.65,124.55,122.19,120.65,119.75,116.85.
实施例2.检测苯硫酚的荧光探针试纸的制备
将滤纸剪裁成1cm×1cm大小的正方形试纸,并将其浸泡于1mM本发明荧光探针溶液(DMSO溶剂)中。将滤纸放在表面皿中,50℃条件下真空干燥,得本发明的荧光探针试纸。
本发明的荧光探针的荧光强度随着苯硫酚的变化如图1所示,依据该变化可作出该荧光探针(10μM)荧光强度随着苯硫酚浓度变化的标准曲线,标准曲线如图2所示,通过该标准曲线的建立,本发明荧光探针可用于苯硫酚的定量检测。本发明的荧光探针试纸浸染不同浓度苯硫酚后溶液颜色变化情况如图3所示,随着苯硫酚浓度的增加,本发明的荧光探针试纸由无色逐渐变为粉红色,本发明的荧光探针可用于苯硫酚的定性及半定量检测。
