Abstract
The invention relates to the field of health-care food, in particular to a medicinal and edible compound health-care beverage with the functions of enriching blood and tonifying qi, which comprises the following components in parts by weight: 7-12 parts of medicinal and edible Chinese herbal medicine liquid extract, 15-20 parts of fruits and vegetables, 0.01-0.05 part of lactobacillus plantarum, 8-10 parts of white granulated sugar, 0.1-0.5 part of edible essence and 50-70 parts of purified water. The invention also provides a preparation method of the lactobacillus health-care beverage, which comprises the steps of preparing medicinal and edible Chinese herbal medicine liquid extract, uniformly mixing the medicinal and edible Chinese herbal medicine liquid extract, fruits and vegetables, white granulated sugar and water, adding lactobacillus plantarum for fermentation, and finally sequentially filtering, blending, filling and sterilizing to obtain the health-care beverage. The health-care beverage of the invention comprehensively conditions organs such as kidney, liver, spleen and the like of a human body, introduces high-efficiency lactobacillus plantarum for fermentation, regulates intestinal microecology by fermentation products, enhances the absorption effect of the human body on nutrient components and functional components, and has the effects of tonifying qi and blood, tonifying kidney and strengthening yang, regulating organism immunity, loosening bowel to relieve constipation and the like.

Background
With the rapid increase of economy in China, the life style and the life rhythm of people are continuously changed, the life quality and the body health consciousness are continuously enhanced, but simultaneously, the problems of body weakness, fatigue, weak immunity of the organism, constipation and the like caused by excessive overdraft of the body due to high pressure, high intensity, long-time unhealthy work and life style are troubled by more people all the time. Despite the fact that various medicines and health care products with various colors and various functions sold in the market at present have certain effects, the medicines and the health care products can only cure the exterior of the medicines in a short time but cannot consolidate the basis, and the medicines and the health care products can cause side effects on physical functions after being eaten for a long time, are expensive and are generally inconvenient to eat.
The nation carries out the theory of 'preventive treatment of disease' of traditional Chinese medicine, advocates a health preserving way that 'medicine tonifying is inferior to food tonifying', but generally, the general household basically uses one or a few traditional Chinese medicines or medicinal and edible raw materials to be matched with soup or used as ingredients for eating, the eating times and the processing way are simple and limited, and the food tonifying effect is difficult to achieve. The health beverage of the invention is composed of medicinal and edible Chinese herbal medicine liquid extract, fruits and vegetables, lactobacillus plantarum, purified water, white granulated sugar and edible essence, and the health beverage is produced by fermenting the components by combining the components. Kudzu vine root, cool in nature, sweet and pungent in flavor, has multiple functions of relieving exterior syndrome, allaying fever, promoting fluid production, promoting eruption, invigorating yang, relieving diarrhea and the like, and can be used for treating various diseases such as fever and headache caused by exogenous pathogens, neck pain caused by hypertension, thirst, diabetes, measles without adequate eruption, heat eruption, diarrhea and the like. The liquorice, which belongs to heart, lung, spleen and stomach channels, tonifies spleen and qi, clears heat and toxic materials, eliminates phlegm and stops cough, relieves spasm and pain, and harmonizes the drugs, is used for treating weakness of spleen and stomach, lassitude and hypodynamia, palpitation and shortness of breath, cough and profuse sputum, abdominal and limb spasm and pain, carbuncle and swelling and sore toxicity, and relieves drug toxicity and pungency. The sparrow's broom contains rich antioxidant flavonoid, and has remarkable curative effects of scavenging free radicals of human body, regulating functions of human body, and preventing and controlling various diseases. The peony flower has the functions of softening blood vessels, tranquilizing mind and calming the heart. Euglena, strong photosynthesis, contains rich nutrient components: the euglena polysaccharide can adsorb redundant substances in the human body such as cholesterol, neutral fat, heavy metal, alcohol and the like to be discharged out of the human body due to an internal porous structure, so that the euglena polysaccharide has strong effects of resisting oxidation and viruses and scavenging free radicals. In order to achieve better effect, the medicinal and edible Chinese herbal medicine also comprises cordyceps militaris and lindera aggregata leaves. The fruit and vegetable is one or more of fructus Myrtilli, fructus Vitis Viniferae, fructus Mali Pumilae, fructus Ananadis Comosi, fructus Lycopersici Esculenti, semen Sojae Atricolor, radix Dauci Sativae, folium Allii tuberosi, fructus Lycopersici Esculenti, fructus Colocasiae Esculentae, fructus Pyri, and fructus Citri Limoniae.
The dosage of each raw material of the medicinal and edible Chinese herbal medicine liquid extract is within the following weight portion range, and the extract has better curative effect:
3-6 parts of longan, 2-4 parts of rhizoma polygonati, 2-4 parts of red date, 2-4 parts of kudzu root, 2-4 parts of liquorice, 2-4 parts of gorse lingua, 2-4 parts of paeonia suffruticosa and 2-4 parts of euglena.
Preferably: 5 parts of longan, 2 parts of rhizoma polygonati, 4 parts of red date, 3 parts of kudzu root, 3 parts of liquorice, 3.5 parts of gorse senna, 3.5 parts of peony and 4 parts of euglena.
The dosage of the raw materials of the medicinal and edible Chinese herbal medicine liquid extract can also be within the following weight portion ranges: 3-6 parts of longan, 2-4 parts of rhizoma polygonati, 2-4 parts of red date, 2-4 parts of radix puerariae, 2-4 parts of liquorice, 2-4 parts of gorse lingua, 2-4 parts of paeonia suffruticosa, 2-4 parts of euglena, 2-4 parts of cordyceps militaris and 2-4 parts of lindera aggregata leaves.
Preferably: 5 parts of longan, 2 parts of rhizoma polygonati, 4 parts of red date, 3 parts of radix puerariae, 3 parts of liquorice, 3.5 parts of sparrow's online, 3.5 parts of peony, 4 parts of euglena, 2.5 parts of cordyceps militaris and 3 parts of lindera aggregata leaves.
The health-care beverage is prepared by adopting the following conventional method: firstly, preparing medicinal and edible Chinese herbal medicine liquid extract for later use; secondly, fermenting the fruit and vegetable and the medicinal and edible Chinese herbal medicine liquid extract mixture; finally, blending the fermentation stock solution, the edible essence and the purified water according to a proportion, and sterilizing and cooling to obtain the health-care beverage; in order to make the medicinal and edible herbs exert better medicinal effects, alcohol precipitation is preferably carried out on the far raw material water extract. These should not be used to limit the scope of the invention.
Preferably, the preparation method of the health beverage of the invention is as follows:
s1, preparing medicinal and edible Chinese herbal medicine liquid extract
a) Identifying and screening raw materials: randomly sampling each medicinal and edible Chinese herbal medicine, detecting the content of marker components in the extracted sample by referring to Chinese pharmacopoeia (2015) and a new food raw material detection method to determine whether the raw materials are true or false, and screening out raw materials which are mildewed, deteriorated and the like and do not meet the requirements; weighing longan, rhizoma polygonati, red date, radix puerariae, liquorice, caragana sericea, paeonia suffruticosa and euglena according to corresponding weight ratio;
b) crushing and cleaning raw materials: crushing the raw materials screened in the step a) by a universal crusher, quickly washing the crushed raw materials by purified water, and feeding the crushed raw materials;
c) and (3) cooking and concentrating: according to the amount of the raw materials, cooking for three times according to a certain material-liquid ratio, and combining the filtrates collected for three times;
d) alcohol precipitation: adding 5% ethanol into the filtrate obtained in the step c), repeatedly carrying out sedimentation for several times, and removing insoluble substances;
e) centrifugal separation: pouring the filtrate obtained in the step d) into a three-foot type centrifuge, adopting a 200-mesh screen, removing filter residue at the rotation speed of 4000r/min, and collecting the centrifugate to obtain the medicinal and edible Chinese herbal medicine liquid extract for later use.
S2, fermentation
f) Pretreatment of fermentation raw materials: cleaning fruits and vegetables with water, dispersing the fruits and vegetables with a coarse crusher, mixing the fruits and vegetables with the medicinal and edible Chinese herbal medicine liquid extract in the step e), adding white granulated sugar and water, uniformly mixing, and keeping the mixture at the temperature of 140 ℃ for 10s for ultrahigh-temperature instantaneous sterilization;
g) controlling the temperature of the slurry in the step f) to be between 40 and 43 ℃, adding lactobacillus plantarum, uniformly mixing, and fermenting for 2 to 15 days;
h) and (3) filtering: putting the pulp obtained in the step g) into a three-legged centrifuge, adopting a 200-mesh screen, centrifuging at the rotating speed of 4000r/min, removing filter residues, and taking supernatant to obtain fermentation stock solution of Chinese medicinal materials and fruits and vegetables for later use;
s3, blending
i) Weighing and mixing the traditional Chinese medicinal materials prepared in the step h), the fruit and vegetable fermentation stock solution, the edible essence and the purified water according to a specified proportion, and controlling the temperature of a mixed material system to be 70-80 ℃ until the materials are completely dissolved;
s4, filling
j) Filling the mixed material prepared in the step i) into bottled or aluminum-plastic composite membrane bagged beverage with the net content of 50ml by adopting a full-automatic beverage filling production line, wherein the temperature of the mixed material is controlled to be 60-70 ℃ in the filling process;

1. Experiment of blood enriching action of fermentation stock solution
Test materials: donkey-hide gelatin oral liquid for enriching blood and tonifying qi (Yuandao Huangshi flying cloud pharmaceutical Co., Ltd.); acetylphenylhydrazine (APH) (source leaf biotechnology bio ltd); physiological saline; the invention relates to three formulas of fermentation stock solutions;
the test method comprises the following steps: 90 Kunming mice, half of male and female, 6-8 weeks old are taken and randomly divided into a normal control group, a positive control group, a model group, a No. 1 formula high and low dose group, a No. 2 formula high and low dose group, a No. 3 formula high and low dose group, and 10 mice in each group. The dose groups are calculated according to the daily functional drink human body intake of 120ml/d, calculated according to 60kg body weight, equivalent to 2ml/kg, the equivalent dose conversion coefficient of the discount mouse is 9.1, equivalent to 18.2ml/kg, the fermentation stock solution group is designed into a low dose group and a high dose group according to 0.5 and 2 times of equivalent dose, and the positive control group, the blank group and the model group are all designed according to 1 time of equivalent dose. The grouping is as follows:
(1) blank control group: 18.2ml/kg (physiological saline)

Nine groups of mice are administrated according to the dose, the administration mode is that the stomach is drenched once a day for 27 continuous days, except for a blank control group, other groups are respectively injected with physiological saline solution of Acetylphenylhydrazine (APH) subcutaneously on the 1 st, 4 th and 7 th days of the experiment, the APH dose is respectively 0.2g/kg, 0.1g/kg and 0.1g/kg, the administration is carried out according to the body weight of 0.01mL/g, and the blank group is injected with the same amount of physiological saline. The food is restricted, and the food is administered at 5g per day. The blank control group and the model group are administered with gavage normal saline, and the gavage medicine is administered for 3 weeks 1 time per day at low dose and high dose according to the above dosage schedule. After administration for 30 minutes at 3 weeks, each mouse was removed from the eyeball and bled, placed in a centrifuge tube of anticoagulant, and immediately sent to a reagent company for biochemical index detection and analysis, and peripheral blood leukocytes (WBC), Red Blood Cells (RBC), Hemoglobin (HGB), and Platelets (PLT) were detected using a fully automatic blood analyzer.
The results show that the weight of the mice has no significant difference (p is more than 0.05) compared with the blank control group after the mice take the fermentation stock solution for 21 days, the weight of each group can not cause the excessive increase of the weight, and the normal development of the mice can not be inhibited, and the results are shown in the table below.

The results, combined with fig. 1 and fig. 2, show that, after the cyclophosphamide is used for intraperitoneal injection molding, the RBC and HGB of the model group mice are significantly lower than those of the other groups (P is less than 0.05 and P is less than 0.01), which indicates that the hemopoietic function of the cyclophosphamide generates obvious damage effect. However, after fermentation stock solutions with different proportions and different dosages are used, a typical dose effect relationship is not presented, namely within a certain range, the higher the dosage is, the more remarkable the improvement effect is not shown. After the high-dose fermentation stock solution is used, the content of red blood cells and hemoglobin are increased to different degrees compared with the blank group. When the dosage is smaller, no obvious growth effect is shown, which indicates that the high-dosage fermentation stock solution can effectively reduce the inhibition effect of cyclophosphamide on mouse platelet cells.
In conclusion, the fermentation stock solution can effectively relieve anemia damage caused by cyclophosphamide and improve the immune function of mice, and has a remarkable effect, but if the dosage is too low, the effect of enriching the blood is not obvious.
2. Qi tonifying effect experiment of the health care beverage
Test materials: liver glycogen ELISA kit (enzyme immunoassay Biotechnology Ltd.); physiological saline; three formulas of fermentation stock solutions; donkey-hide gelatin oral liquid for enriching blood and replenishing qi (Yuandao Huangshi Feiyun pharmaceutical Co., Ltd.)
The test method comprises the following steps: 80 Kunming mice, each half of male and female, are randomly divided into a normal control group, a positive control group, a No. 1 enzyme high and low dose group, a No. 2 enzyme high and low dose group and a No. 3 enzyme high and low dose group, each group comprises 10 enzymes, and the high and low dose groups are respectively infused with gastric lavage to give fermentation stock solution. The dose groups are calculated according to the human body intake of 120ml/d of daily functional drink and 60kg of body weight, which is equivalent to 2ml/kg, the equivalent dose conversion coefficient of the mouse is 9.1, which is equivalent to 18.2ml/kg, the fermentation stock solution is designed into two low and high dose groups according to 0.5 and 2 times of equivalent dose, and the positive control group and the blank group are designed according to 1 time of equivalent dose. The grouping is as follows:
(1) blank control group: 18.2ml/kg (physiological saline)
(2) Positive control group: 18.26ml/kg (donkey-hide gelatin oral liquid for tonifying qi and enriching blood)
(3) Formula 1 low dose group: 9.1ml/kg
(4) Formula 1 high dose group: 36.4ml/kg
(5) Formula 2 low dose group: 9.1ml/kg
(6) Formula 2 high dose group: 36.4ml/kg
(7) Formula No. 3 low dose group: 9.1ml/kg
(8) Formula No. 3 high dose group: 36.4ml/kg
Mice were gavaged once daily for 14 days in the groups and doses described above. After 30 minutes of the last administration, the eyeballs are picked and blood is taken, after the eyeballs are placed at room temperature for 30 minutes, the eyeballs are centrifuged at 3000r/min and 40 ℃ for 20-30 minutes, serum is separated, and the liver glycogen content of each group of mice is measured according to an Elisa kit.
As a result, the body weight of the mice has no significant difference (p is more than 0.05) compared with that of the blank control group 14 days after the mice take the fermentation stock solution. None of the groups caused an excessive increase in body weight nor inhibited normal development of the mice. The results are shown in the following table.

The experiment can research the qi tonifying effect of the medicine from five aspects of fatigue resistance, hypoxia tolerance, cold resistance, immune function and substance metabolism. The anti-fatigue and anti-anoxia test mainly shows the pushing action of qi, and qi deficiency can not carry oxygen and fine substances to nourish the whole body, so that the fatigue and anoxia can be shown; cold tolerance test is relative to warming effect; the immune index corresponds to the defense function; substance metabolism is an expression form of gasification, and because saccharides are main energy sources of substance metabolism, and glycogen of liver is a repository of blood sugar, glucose is continuously output to supply energy needs of organisms, and the improvement of glycogen storage has important significance for relieving exercise-induced fatigue, the experiment mainly observes the metabolism of glycogen.
The results show that the experiment observes the influence of the fermentation stock solution on the liver glycogen in the body of the mouse, and proves that the different dosages of the fermentation stock solution obviously improve the liver glycogen content of the mouse (P is less than 0.01), thereby prompting that the fermentation stock solution has the effect of tonifying qi. Compared with the positive medicine, the low dosage can not show better qi tonifying effect, but the high dosage of the fermentation stock solution shows excellent qi tonifying effect (P is less than 0.01). Although the experiment reveals that the fermentation stock solution has the effect of tonifying qi, the specific mechanism of tonifying qi has not been studied deeply, and further experiments are needed to be carried out if the mechanism of tonifying qi needs to be understood deeply.
The invention only tests the efficacy of enriching the blood and tonifying qi, so that no test is carried out on the formula of adding the cordyceps militaris and the lindera aggregata leaves, and other efficacies brought by the cordyceps militaris and the lindera aggregata leaves, such as the efficacies of tonifying lung yin, tonifying kidney yang, treating dyspepsia and the like, are not taken as the main efficacies of the health-care beverage.

A medicine-food homologous compound health-care beverage with the effects of enriching blood and tonifying qi is characterized by comprising the following components in parts by weight: 7-12 parts of medicinal and edible Chinese herbal medicine liquid extract, 15-20 parts of fruits and vegetables, 0.01-0.05 part of lactobacillus plantarum, 8-10 parts of white granulated sugar, 0.1-0.5 part of edible essence and 50-70 parts of purified water
The medicinal and edible Chinese herbal medicine liquid extract comprises the following raw materials in parts by weight: 3-6 parts of longan, 2-4 parts of rhizoma polygonati, 2-4 parts of red date, 2-4 parts of kudzu root, 2-4 parts of liquorice, 2-4 parts of gorse lingua, 2-4 parts of paeonia suffruticosa and 2-4 parts of euglena.
2. The compound health beverage with the effects of enriching blood and tonifying qi as claimed in claim 1, which is characterized in that: the fruit and vegetable is one or more of blueberry, grape, apple, pineapple, tomato, black bean, carrot, leek, tomato, taro, snow pear and lemon.

The compound health beverage with the effects of enriching blood and tonifying qi as claimed in claim 1, wherein the extract of the medicinal and edible Chinese herbal medicine liquid is prepared from the following raw materials in parts by weight: 5 parts of longan, 2 parts of rhizoma polygonati, 4 parts of red date, 3 parts of kudzu root, 3 parts of liquorice, 3.5 parts of gorse senna, 3.5 parts of peony and 4 parts of euglena.

The compound health beverage with the effects of enriching blood and tonifying qi as claimed in claim 1, wherein the extract of medicinal and edible Chinese herbal medicine liquid according to parts by weight of the raw materials can further comprise: 2-4 parts of cordyceps militaris and 2-4 parts of lindera root leaves.

The compound health beverage with the effects of enriching blood and tonifying qi as claimed in claim 4, wherein the extract of the medicinal and edible Chinese herbal medicine liquid is prepared from the following raw materials in parts by weight: 5 parts of longan, 2 parts of rhizoma polygonati, 4 parts of red date, 3 parts of radix puerariae, 3 parts of liquorice, 3.5 parts of sparrow's online, 3.5 parts of peony, 4 parts of euglena, 2.5 parts of cordyceps militaris and 3 parts of lindera aggregata leaves.
